1Expression of ipt gene driven by tomato fruit specific promoter and its effects on fruit development of tomato显示文摘Fruit specific promoter (2A12) from Lycopersi-com esculentum and cDNA of isopentenyl-transferase (ipt) from Ti plasmid of Agrobacterium tumerfaciens C58 were cloned by PCR procedure respectively. Two plant expression vectors with 2A12/gus or 2A12/ipt were respectively constructed. These two chimeric genes were transferred into tomato by Agrobacterium mediated procedure. The results of Southern hybridization showed that the fusion genes had been integrated into tomatoes. The result of gus histochemi-cal staining showed that 2A12 had high fruit specific expressive capability in transgenic tomato. The ipt expression resulted in accumulation of high level of cytokinins (CTKs) in fruit lead to developmental changes in fruits and seeds. The fruit of ipt transformed tomato had the hyperplastic placenta with very few seeds or even seedless. The shelf life of transgenic fruits elongated for 1-2 weeks. 2Boosting the photodynamic therapy efficiency with a mitochondria-targeted nanophotosensitizer显示文摘Photodynamic therapy (PDT) is an attractive clinical technique for cancer treatment. However, the poor solubility and weak cellular internalization of the molecule-photosensitizers, as well as the exceedingly short lifetime and limited diffusion distance of the generated reactive oxygen species (ROS) in cytoplasm directly restricted the therapeutic efficiency of conventional PDT. In this study, we proposed a facile strategy for improving PDT of cancer based on a mitochondria-targeted nanophotosensitizer. The molecule-photosensitizer chlorin e6 was covalently attached on the internal and external surfaces of the mesoporous silica nanoparticles. Then, the triphenylphosphonium was anchored on the nanoparticles for selectively targeting mitochondria. When irradiated with laser, the nanophotosensitizer can generate a large amount of ROS in mitochondria, thus causing the mitochondrial dysfunction and irreversible cell apoptosis. 3Cloning and expression pattern of a dehydrin-like BDNI gene from drought-tolerant Boea crassifolia Hemsl.显示文摘A 500-bp cDNA fragment was amplified via RT-PCR from drought-induced total RNA of the drought-tolerant B. crassifolia Hemsl. using primers based on the sequence of published dehydrin conserved region. By using 5’ RACE, full-length coding region (1 148 bp) of BDN1 gene was produced. It is a new member of the dehydrin gene family. Southern analysis indicated that BDN1 is present in the 6. crassifolia genome as a single-copy gene. 4Enhanced late blight resistance of transgenic potato expressing glucose oxidase under the control of pathogen-inducible promoter显示文摘To engineer crop disease resistance by utilizing natural defense mechanism that was expressed in the incompatible host-pathogen interactions is expected to result in a durable and broad-spectrum resistance. In order to prove this viewpoint, we amplified the coding region of the glucose oxidase (GO) gene from Aspergillus nigervia PCR and fused it to the pathogen-inducible promoter, Prp1-1. The chimeric gene was cloned into a plant expression vector and conjugated into Agrobacterium. Twenty-three transgenic potato plants were obtained by Agrobacterium-mediated transformation. The integration of GO gene was confirmed by Southern hybridization and the GO gene expression was identified with Kl-starch color reaction. Phytophthora infestans inoculation revealed that the expression of the chimeric transgene was induced by pathogen infection. Most of the transgenic plants exhibited various degrees of enhanced disease resistance. 15A big-data approach for investigating destination image gap in Sanya City: When will the online and the offline goes parted?显示文摘Tourism destination images in terms of the gaps between the projected and perceived images are of great significance in the development of destinations.Additionally,the use of big-data in tourism studies remains under-utilized despite the boom in big-data applications and the increasing number of electronic User Generated Contents(UGC).Aiming to take advantage of tourism UGC to fully understand the destination image gap between official promotion materials and tourist perception of Sanya City in China,this study innovatively employed a big-data analysis technique,Tourism Sentiment Evaluation(TSE)model and proposed a new analysis framework integrating the“cognitive-affective”model with the gpp analysis of projected and perceived destination image to explore the destination image gap of Sanya It is found that Sanya's perceptive destination image is overall consistent with its official positioning;however,there also exist image gaps between the two groups in terms of the impact of festival events and tourists'attitude towards core scenic spots amongst others.This study's findings are discussed in light of their methodological,theoretical,and practical implications for destination positioning,marketing,and management.Lingkun Meng Yi Liu Yuanlei Wang Xiaojuan Li 2021Regional Sustainability2021,2,1:0

17Co-transformation to tobacco of Cre/lox site-specific recombination system and its precise recombination显示文摘For the temporally and spatially regulated expression of the barnase gene in plant, two kinds of plasmids with ere gene and its directly repeat recognition sites lox from bacteriophage P1 were constructed and co-transformed into tobacco by agrobacterium mediated procedure. The transgenic plants were conformed by PCR analysis. The blocking fragment between the two lox directly repeat sites was excised by Cre protein in the transgenic plant genome.
