| 2 | Mutational analysis of the SDD sequence motif of a PRRSV RNA-dependent RNA polymerase显示文摘The subgenomic mRNA transcription and genomic replication of the porcine reproductive and respiratory syndrome virus (PRRSV) are directed by the viral replicase. The replicase is expressed in the form of two polyproteins and is subsequently processed into smaller nonstructural proteins (nsps). nsp9, containing the viral replicase, has characteristic sequence motifs conserved among the RNA-dependent RNA polymerases (RdRp) of positive-strand (PS) RNA viruses. To test whether the conserved SDD motif can tolerate other conserved motifs of RNA viruses and the influence of every residue on RdRp catalytic activity, many amino acids substitutions were introduced into it. Only one nsp9 substitution, of serine by glycine (S3050G), could rescue mutant viruses. The rescued virus was genetically stable. Alteration of either aspartate residue was not tolerated, destroyed the polymerase activity, and abolished virus transcription, but did not eliminate virus replication. We also found that the SDD motif was essentially invariant for the signature sequence of PRRSV RdRp. It could not accommodate other conserved motifs found in other RNA viral polymerases, except the GDD motif, which is conserved in all the other PS RNA viruses. These findings indicated that nidoviruses are evolutionarily related to other PS RNA viruses. | 4 | Effect of Dopamine Injection on the Hemocyte Count and Prophenoloxidase System of the White Shrimp Litopenaeus vannamei显示文摘Effects of dopamine injection on the hemocyte count, phenoloxidase activity, serine proteinase activity, proteinase in-hibitor activity and α2-macroglobulin-like activity in L. vannamei were studied. Results showed that dopamine injection resulted in a significant effect on the parameters measured (P < 0.05), while no significant difference was observed in the control group (0.85% NaCl). In the experimental groups,the hemocyte count reached the minimum in 3 h;granular and semi-granular cells became stable after 12 h and hyaline cells and the total hemocyte count became stable after 18 h. Phenoloxidase activity reached the minimum in 6 h, and then became stable after 9 h. Serine protease activity and proteinase inhibitor activity reached the minimum in 3 h, and α2-macro-globulin-like activity reached the maximum in 3 h,and all the three parameters became stable after 12 h. | 5 | Tandem Molecular Self-Assembly Selectively Inhibits Lung Cancer Cells by Inducing Endoplasmic Reticulum Stress显示文摘The selective formation of nanomaterials in cancer cells and tumors holds great promise for cancer diagnostics and therapy.Until now,most strategies rely on a single trigger to control the formation of nanomaterials in situ.The combination of two or more triggers may provide for more sophisticated means of manipulation.In this study,we rationally designed a molecule(Comp.1)capable of responding to two enzymes,alkaline phosphatase(ALP),and reductase.Since the A549 lung cancer cell line showed elevated levels of extracellular ALP and intracellular reductase,we demonstrated that Comp.1 responded in a stepwise fashion to those two enzymes and displayed a tandem molecular self-assembly behavior.The selective formation of nanofibers in the mitochondria of the lung cancer cells led to the disruption of the mitochondrial membrane,resulting in an increased level of reactive oxygen species(ROS)and the release of cytochrome C(Cyt C).ROS can react with proteins,resulting in endoplasmic reticulum(ER)stress and the unfolded protein response(UPR).This severe ER stress led to disruption of the ER,formation of vacuoles,and ultimately,apoptosis of the A549 cells.Therefore,Comp.1 could selectively inhibit lung cancer cells in vitro and A549 xenograft tumors in vivo.Our study provides a novel strategy for the selective formation of nanomaterials in lung cancer cells,which is powerful and promising for the diagnosis and treatment of lung cancer. | Debin Zheng Yumiao Chen Sifan Ai Renshu Zhang Zhengfeng Gao Chunhui Liang Li Cao Yaoxia Chen Zhangyong Hong Yang Shi Ling Wang Xingyi Li Zhimou Yang | 2019 | Research2019,,1: | 1 |

| 7 | Effects of Different Dietary Lipid Contents on Growth and Lipase Activity of Eriocheir sinensis Larvae显示文摘The effects of different dietary lipid content on the growth and lipase activity of Eriocheir sinensis larvae were studied in the paper.The results showed that the survival,metamorphic rate and weight gain of E.sinensis larvae at different stages of growth all varied significantly with lipid content (P<0.05).Further,the survival and metamorphosis rates were the highest during the larval phases Z3 to Z4,and the weight gain was the highest during the larval phases Z5 to M.During the first 20 h after metamorphosis of every larval stage,the lipase activity increased over time at Z1,Z2,Z3 and M and declined at Z4 and Z5,and was influenced significantly by lipid content (P<0.05).In addition,lipase activity at each larval stage began to respond to dietary lipid contents 4 h after the larvae were fed,and tended to be stable after 12 h.The diets with higher lipase activity and lower lipid content were selected to give the suitable recipe of lipid requirements at each larval stage.It was concluded that the suitable lipid requirements at Z1,Z2,Z3,Z4,Z5 and M were 6%,4%,6%,8%,8% and 10%,respectively. | ZHENG Debin PAN Luqing FANG Bo | 2011 | Journal of Ocean University of China2011,10,1: | 1 |

| 10 | Initiation of two ovarian cell lines from Fugu rubripes(Temminck et.Schlegel)显示文摘The ovary is an excellent system for studying stem cell renewal and differentiation,which is under the control of ovarian somatic cells.In order to understand oogenesis in Fugu rubripes(Temminck et.Schlegel) as a marine fish model of aquaculture importance,we established cell lines called TSOC1 and TSOC2 from a juvenile ovary of this organism.TSOC1 is composed of spindle epithelial-like cells,while the other is cobblestone-like cells.Therefore,TSOC1 and TSOC2 appear to consist of ovarian somatic cells.Growth requirement condition was investigated including temperature,concentration of FBS and p H.Significant fluorescent signals were observed after TSOC1 and TSOC2 cells were transfected with p EGFP-N3 vector,indicating its potential utility for genetic manipulation such as gene function studies.It is shown that these cell lines are effective for infection by the turbot reddish body iridovirus and flounder lymphosystis disease virus as evidenced by the appearance of cytopathic effect and virus propagation in the virus-infected cells,and most convincingly,the observation of viral particles by electron microscopy,demonstrating that TSOC1 and TSOC2 are suitable to study interactions between virus and host cells.It is believed that TSOC1 and TSOC2 will be useful tools to study sex-related events and interactions between primordial germ cells and oogonia cells during oogenesis.Therefore,establishment of ovary cell lines from Fugu rubripes seems to be significant for those research areas. | ZHENG Debin ZHANG Bo SONG Wenping PAN Luqing MA Chao XIAO Guangxia | 2015 | Acta Oceanologica Sinica2015,34,10: | 0 |
| 11 | 在PACS与CT工作站测量恶性孤立肺结节大小及密度的对比研究(英文)显示文摘Objective: The aim of our study was to determine the efficiency and effectiveness of picture archiving and communication system(PACS) workstation in detecting the sizes and attenuation of malignant solitary pulmonary nodules(SPNs). Methods: Forty patients with malignant SPNs(diameter ≤ 3 cm) underwent multidetector-row computed tomography(CT) of the chest in a single-breath-hold technique. The raw data were acquired with a collimation of 0.625 mm. The diameters and attenuation of malignant SPNs were measured on PACS and CT workstation respectively. The diameter was defined as the average value of the anteroposterior, lateral and superoinferior diameters on CT scans obtained with a mediastinal window setting. The superoinferior diameters were measured on MPR image. The diameters and attenuation of malignant SPNs and spending time in measuring were recorded. Results: The diameters of malignant SPNs measured on a PACS and CT workstation were 2.09 cm ± 0.87 cm, 2.07 cm ± 0.79 cm, respectively. There was not statistically significant difference in the diameters of malignant SPNs between that measured on a PACS workstation and that on a CT workstation(t = 1.580, P = 0.210 > 0.05). The attenuation of malignant SPNs measured on a PACS and CT workstation were 40.15 HU ± 7.53 HU, 39.99 HU ± 8.13 HU, respectively. There was not statistically significant difference in the attenuation of malignant SPNs between that measured on a PACS workstation and that on a CT workstation(t = 1.008, P = 0.298 > 0.05). The spending time in measuring on a PACS and CT workstation were 55 s ± 4.03 s, 56 s ± 3.95 s, respectively. No statistically significant difference was found in spending time in measuring between that on a PACS workstation and that on a CT workstation(t = 0.958, P = 0.315 > 0.05). Conclusion: The efficiency and effectiveness of PACS workstation is as same as those of CT workstation in detecting the sizes and attenuation of malignant SPNs. | 12 | 消化系统肿瘤CT随访中发现的肝脏高灌注异常的转归(英文)显示文摘Objective: The aim of this study was to investigate progression of hepatic hyperperfusion disorders revealed during follow-up contrast material-enhanced multi-slice spiral computed tomography(MSCT) scan of digestive system neoplasm. Methods: Three-phase contrast material-enhanced MSCT were performed during the follow-up in patients with digestive system malignant tumor confirmed histologically. The progression of hepatic hyperperfusion disorders revealed on contrast material-enhanced CT image were investigated at the 2 years follow-up with approximately 6 months interval. Results: The hepatic hyperperfusion disorders were showed in 39 patients on follow-up contrast material-enhanced MSCT scans. Among the 39 patients, initial hyperperfusion disorders were revealed in 6(15.38%), 26(66.67%), and 7(17.95%) patients in 6, 12, and 18 months during follow-up respectively. The initial hyperperfusion disorders revealed in 12 months were more frequent than those revealed in 6 months(χ2 = 14.82, P < 0.05) and 18 months(χ2 = 15.02, P < 0.05). Among the 39 patients, the hyperperfusion disorders progressed into liver metastasis based on typical CT findings in 37(94.87%) patients, and were not obvious changes in 2(5.13%) patients. Among the 37 patients, the hyperperfusion disorders progressed into metastasis in 10(25.64%) patients in 6 months after the hyperperfusion disorders were revealed, and in 27(69.23%) patients in 12 months. The hyperperfusion disorders developing into metastasis were more in 12 months than those in 6 months(χ2 = 14.98, P < 0.05). Conclusion: Most hepatic hyperperfusion disorders revealed at the follow-up of digestive system neoplasm may be early manifestations of liver metastasis. | 13 | Cloning,characterization,and expression of a novel member of proteasomal subunits gene in turbot,Scophthalmus maximus显示文摘The proteasome is a large, polymeric protease complex responsible for the degradation of intracellular proteins and generation of peptides that bind to class I major histocompatibility complex(MHC) molecules. This study identified a new member of proteasomal subunits in turbots(Scophthalmus maximus). The fulllength c DNA sequence of turbot proteasomal subunit was obtained. Sequence analysis indicated that its primary structure is highly similar to that of LMP7 from other vertebrates. The relationship between the turbot LMP7 expression and immune responses to pathogen infection was reported. Quantitative reverse transcriptase polymerase chain reaction showed that LMP7 was expressed differently in various tissues, with higher expression in the spleen, liver, muscle, and skin. The LMP7 expression was the highest at 96 h after challenge with lymphocyctis disease virus(LCDV) and at 12 h after challenge with Vibrio anguillarum in the turbot liver, kidney, and spleen. Furthermore, the LMP7 expression distinctly increased in turbot kidney cells at 24 h after challenge with V. anguillarum and at 96 h after challenge with LCDV.
