1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Seafloor hydrothermal activity and polymetallic sulfide exploration on the southwest Indian ridge显示文摘Polymetallic sulfides have attracted more attentions,which are regarded as one of the potential seabed mineral resources for their high grade of precious metal elements such as Cu,Zn,Pb,Au,and Ag.Since2007,there are four China dayang cruises(CDCs)with eight legs totally,which have been carried out to investigate polymetallic sulfides on the southwest Indian ridge(SWIR),and eight hydrothermal vents have been found.In2011,the China Ocean Mineral Resources Research and Development Association(COMRA)and International Seabed Authority(ISA)signed a 15-year contract for exploration for polymetallic sulfides located on the SWIR between 46°S and 53°S.The area allocated to the COMRA is approximately 10,000 km2.In this paper,we review the recent research advances about hydrothermal activity of the SWIR by the CDCs.As for the sulfides exploration in the future,some aspects should be emphasized,such as the controlling factors of sulfide mineralization on the SWIR,prospecting methods of inactive and buried sulfide deposits,sulfides resources assessment method,and the nearbottom sulfides exploration technology systems.Chunhui Tao Huaiming Li Xiaobing Jin Jianping Zhou Tao Wu Yonghua He Xianming Deng Chunhua Gu Guoyin Zhang Weiyong Liu 2014Chinese Science Bulletin2014,59,19:25
4Genetic analysis of 'all-fish' growth hormone gene transferred carp (Cyprinus carpio L.) and its F_1 generation显示文摘Recornbinant 'all-fish' growth hormone gene (GH) was microinjected into the fertilized eggs of carp. A comparison between the growth traits of transgenics and non-transgenics was carried out, and the transgenic individuals with significant 'fast-growing' effect were successfully gained. A comparison on the reproductivities was also given out between the transgenics and their non-transgenic siblings, and showed that the reproductive capacity of transgenics was substantially equivalent to those of the non-transgenics. On the other hand, the genetic separation and the characteristic distribution of the FI generation were genetically analyzed, which gave solid evidence for the hypothesis that 2-3 chromosomes are integrated with trans-gene. In addition, the distinct biological effects for multi-site-integrated transgenes were further discussed. 6Focal depths for moderate-sized aftershocks of the Wenchuan M_S8.0 earthquake and their implications显示文摘Sliding-window cross-correlation method is firstly adopted to identify sPn phase, and to constrain focal depth from regional seismograms, by measuring the time separation between sPn and Pn phases. We present the focal depths of the 17 moderate-sized aftershocks (MS≥5.0) of the Wenchuan MS8.0 earth-quake, using the data recorded by the regional seismic broadband networks of Shaanxi, Qinghai, Gansu, Yunnan and Sichuan. Our results show focal depths of aftershocks range from 8 to 20 km, and tend to cluster at two average depths, separate at 32.5°N, i.e., 11 km to the south and 17 km to the north, indicating that these aftershocks are origin of upper-to-middle crust. 9Biological soil crust distribution in Artemisia ordosica communities along a grazing pressure gradient in Mu Us Sandy Land, Northern China显示文摘This study investigated the distribution pattern of biological soil crust (BSC) in Artemisia ordosica communities in Mu Us Sandy Land. Three experimental sites were selected according to grazing pressure gradient. In each experimental site, the total vegetation cover, A. ordosica cover, BSC cover, litter-fall cover, BSC degree of fragmentation, BSC thickness and soil properties were investigated in both fixed and semi-fixed sand dunes and simultaneously analyzed in the laboratory. The results showed that at the same grazing pressure, BSC cover and composition were significantly affected by the fixation degree of sand dunes. In addition, BSC cover in the fixed sand dunes was 83.74% on average, whereas it is proportionally dominated by 28% mosses, 21% lichens, and 51% algae. Meanwhile, BSC cover in the semi-fixed sand dunes was 23.54% on average, which is proportionally dominated by 6.3% mosses, 2.5% lichens, and 91.2% algae. Fine sand, organic matter, and total nitrogen (N) contents in the fixed sand dunes were all significantly higher than those in the semi-fixed sand dunes. Litter-fall cover decreased along the grazing pressure gradient, whereas BSC fragmentation degree increased. Fine sand content decreased along with the increase of grazing pressure, whereas medium sand content increased in both fixed and semi-fixed dunes. The organic matter and total N contents in the no grazing site were significantly higher than those in light and normal grazing sites. However, there were no significant differences between the light and normal grazing sites. In addition, there were also no significant differences in BSC thickness between the light and normal grazing sites in the fixed sand dunes. However, a significant decrease was observed in both BSC cover and thickness in the normal grazing site. 10Chromosome-Level Reference Genome and Population Genomic Analysis Provide Insights into the Evolution and Improvement of Domesticated Mulberry (Morns alba)显示文摘Mulberry(Morns spp.)is the sole plant consumed by the domesticated silkworm.However,the genome of domesticated mulberry has not yet been sequenced,and the ploidy level of this species remains unclear.Here,we report a high-quality,chromosome-level domesticated mulberry(Morus alba)genome.Analysis of genomic data and karyotype analyses confirmed that M.alba is a diploid with 28 chromosomes(2/7=2x=28).Population genomic analysis based on resequencing of 134 mulberry accessions classified domesticated mulberry into three geographical groups,namely,Taihu Basin of southeastern China(Hu mulberry),northern and southwestern China,and Japan.Hu mulberry had the lowest nucleotide diversity among these accessions and demonstrated obvious signatures of selection associated with environmental adaptation.Further phylogenetic analysis supports a previous proposal that multiple domesticated mulberry accessions previously classified as different species actually belong to one species.This study expands our understanding of genome evolution of the genus Morus and population structure of domesticated mulberry,which would facilitate mulberry breeding and improvement.Feng Jiao Rongsong Luo Xuelei Dai Hui Liu Gang Yu Shuhua Han Xin Lu Chao Su Qi Chen Qinxia Song Caiting Meng Fanghong Li Hongmei Sun Rui Zhang Tian Hui Yonghua Qian Aichun Zhao Yu Jiang 2020Molecular Plant2020,13,7:10
11Migraine and stroke显示文摘Migraines are generally considered a relatively benign neurological condition.However,research has shown an association between migraines and stroke,and especially between migraine with aura and ischaemic stroke.Patients can also suffer from migrainous infarction,a subset of ischaemic stroke that often occurs in the posterior circulation of younger women.The exact pathogenesis of migrainous infarct is not known,but it is theorised that the duration and local neuronal energy level from cortical spreading depression may be a key factor.Other factors contributing to migrainous infarct may include vascular,inflammatory,endothelial structure,patent foramen ovale,gender,oral contraceptive pill use and smoking.Vasoconstrictors such as the triptan and ergot class are commonly used to treat migraines and may also play a role.Migraine is also shown to be correlated to haemorrhagic stroke,although studies do not demonstrate causation versus association,and further studies are warranted.There are also some rare genetic diseases such as cerebral autosomal-dominant arteriopathy with subcortical infarcts and leukoencephalopathy,retinal vasculopathy with cerebral leukodystrophy and others,which can cause both migraines and infarcts.On imaging,many migraineurs are found to have white matter changes similar to those seen in patients with stroke.These may be caused in part by alterations in resting cerebral blood flow and vasoconstrictor use.In treating patients with migraines,it is important to identify and modify any vascular risk factors such as hypertension,smoking,oral contraceptive pill use and lifestyle factors.Further studies will determine if more aggressive treatment of migraines can ultimately lead to fewer strokes in this population.Yonghua Zhang Aasheeta Parikh Shuo Qian 2017Stroke & Vascular Neurology2017,2,3:8

13Surrogate production of genome-edited sperm from a different subfamily by spermatogonial stem cell transplantation显示文摘The surrogate reproduction technique,such as inter-specific spermatogonial stem cells(SSCs)transplantation(SSCT),provides a powerful tool for production of gametes derived from endangered species or those with desirable traits.However,generation of genome-edited gametes from a different species or production of gametes from a phylogenetically distant species such as from a different subfamily,by SSCT,has not succeeded.Here,using two small cyprinid fishes from different subfamilies,Chinese rare minnow(gobiocypris rarus,for brief:Gr)and zebrafish(danio rerio),we successfully obtained Gr-derived genome-edited sperm in zebrafish by an optimized SSCT procedure.The transplanted Gr SSCs supported the host gonadal development and underwent normal spermatogenesis,resulting in a reconstructed fertile testis containing Gr spermatids and zebrafish testicular somatic cells.Interestingly,the surrogate spermatozoa resembled those of host zebrafish but not donor Gr in morphology and swimming behavior.When pou5f3 and chd knockout Gr SSCs were transplanted,Gr-derived genome-edited sperm was successfully produced in zebrafish.This is the first report demonstrating surrogate production of gametes from a different subfamily by SSCT,and surrogate production of genome-edited gametes from another species as well.This method is feasible to be applied to future breeding of commercial fish and livestock.Fenghua Zhang Yongkang Hao Xianmei Li Yi Li Ding Ye Ru Zhang Xiaosi Wang Mudan He Houpeng Wang Zuoyan Zhu Yonghua Sun 2022Science China(Life Sciences)2022,65,5:7

16Recent progress in Chinese polar upper-atmospheric physics research: review of research advances supported by the Chinese Arctic and Antarctic expeditions显示文摘It has been more than 30 years since the first Chinese Antarctic Expedition took place. Polar upper atmospheric observations started at this time. First began at Great Wall Station and then at Zhongshan Station in Antarctica, and later in the Arctic at Yellow River Station, Kjell Henriksen Observatory on Svalbard, and at the China-Iceland Joint Aurora Observatory in Iceland. In this paper, we reviewed the advances in polar upper atmosphere physics(UAP) based on the Chinese national Arctic and Antarctic research over the last five years. These included newly deployed observatories and research instruments in the Arctic and Antarctic; and new research findings, from ground-based observations, about polar ionosphere dynamics, aurora and particle precipitation, polar plasma convection, geomagnetic pulsations and space plasma waves, space weather in the polar regions, simulations of the polar ionosphere-magnetosphere. 17Source apportionment of ambient PM_(10) in urban areas of Wuxi,China显示文摘A total of 168 PM_(10)samples were collected during the year of 2005 at eight sites in the city of Wuxi in China.Fifteen chemical elements,three water-soluble ions,total carbon and organic carbon were analyzed.Six source categories were identified and their contributions to ambient PM_(10)in Wuxi were estimated using a nested chemical mass balance method that reduces the effects of colinearity on the chemical mass balance model.In addition,the concentrations of secondary aerosols,such as secondary organic carbon,sulfate and nitrate,were quantified.The spatially averaged PM_(10)was high in the spring and winter(123μg·m^(-3)and low in the summer-fall(90μg·m^(-3)).According to the result of source apportionment,resuspended dust was the largest contributor to ambient PM_(10),accounting for more than 50%of the PM_(10)mass.Coal combustion(14.6%)and vehicle exhaust(9.4%)were also significant source categories of ambient PM_(10).Construction and cement dust,sulfates,secondary organic carbon,and nitrates made contributions ranging between 4.1%and 4.9%.Other source categories such as steel manufacturing dust and soil dust made low contributions to ambient PM_(10).Bo HAN Xiaohui BI Yonghua XUE Jianhui WU Tan ZHU Baogui ZHANG Jianqing DING Yuanxin DU 2011Frontiers of Environmental Science & Engineering2011,5,4:5
18Preparation and Characterization of Waterborne Polyurethane/Cellulose Nanocrystal Composite Membrane from Recycling Waste Paper显示文摘Cellulose plays a key role in abundant organic natural materials meeting the increasing demand for green and biocompatible products.The highly crystalline nanoscale component of cellulose nanocrystals has recently attracted great attention due to the versatile performance as filler or matrix in producing functional materials.In this work,we prepared the waterborne polyurethane via a prepolymer process,and obtained cellulose and cellulose nanocrystals from waste paper via a facile acid hydrolysis process.After that,the cellulose nanocrystals were assembled into film and mixed with polyurethane to prepare flexible polyurethane/cellulose nanocrystals composite membrane with different soaking time.The correlation between the bulk structure and applied properties including thermal resistance and mechanical property was investigated by using Fourier transform infrared spectroscopy(FTIR),X-ray diffraction(XRD),X-ray photoelectron spectroscopy(XPS),scanning electron microscopy(SEM),thermogravimetric analysis(TGA),differential scanning calorimetry(DSC)and folding test.The structure analysis indicates that cellulose nanocrystals prepared from used paper have a quality similar to that of commercial cellulose.Meanwhile,the cellulose nanocrystals have been mixed with polyurethane uniformly.Polyurethane can significantly benefit to the thermal resistance and mechanical property of the cellulose nanocrystals film.The polyurethane/cellulose nanocrystals composite membrane present good flexibility and may hold a significantly potential application as visual and flexible material.Xing Zhou Xin Zhang Dong Wang Changqing Fang Wanqing Lei Zhigang Huang Yonghua Song Xinyu He Yingwei Huang 2020Journal of Renewable Materials2020,8,6:5
19Differential patterns of SH and P wave velocity structures in the transition zone beneath northwestern Tibet显示文摘Triplicate waveform modeling is used to resolve SH (Vs) and P (Vp ) wave velocity structures in the upper mantle transition zone (TZ) beneath northwestern (NW) Tibet. Focal depth move out stacking is proposed to enhance the identification of triplicate phases, and can be used to test consistency of our data. Our results show that the Vs and Vp structures are decorrelated, and that a large Vs jump occurred across the 660-km discontinuity, with a small Vs gradient above it. Conversely, the Vp model is characterized by a relatively small contrast across the discontinuity, accompanied by a high Vp gradient in the TZ. There seem no significant depth anomalies of the 660-km discontinuity in both models. The seismic structure in TZ beneath NW Tibet are similar to recent studies beneath the central Qiangtang and western Lhasa terrains. Taking the lower TZ structures under India as references, Vs is normal but Vp appears slightly high, and thus a high ratio of Vp/Vs was indicated beneath NW Tibet. Combined results with experiment information from mineral studies, we suggest that the differential anomalies of Vp and Vs can be attributed to a chemical heterogeneity, such as increased Al content in the lower TZ. 20Self-crosslinkable chitosan-hyaluronic acid dialdehyde nanoparticles for CD44-targeted siRNA delivery to treat bladder cancer显示文摘Bladder cancer is one of the concerning malignancies worldwide,which is lacking effective targeted therapy.Gene therapy is a potential approach for bladder cancer treatment.While,a safe and effective targeted gene delivery system is urgently needed for prompting the bladder cancer treatment in vivo.In this study,we confirmed that the bladder cancer had CD44 overexpression and small interfering RNAs(siRNA)with high interfere to Bcl2 oncogene were designed and screened.Then hyaluronic acid dialdehyde(HAD)was prepared in an ethanol-water mixture and covalently conjugated to the chitosan nanoparticles(CS-HAD NPs)to achieve CD44 targeted siRNA delivery.The in vitro and in vivo evaluations indicated that the siRNA-loaded CS-HAD NPs(siRNA@CS-HAD NPs)were approximately 100 nm in size,with improved stability,high siRNA encapsulation efficiency and low cytotoxicity.CS-HAD NPs could target to CD44 receptor and deliver the therapeutic siRNA into T24 bladder cancer cells through a ligand-receptor-mediated targeting mechanism and had a specific accumulation capacity in vivo to interfere the targeted oncogene Bcl2 in bladder cancer.Overall,a CD44 targeted gene delivery system based on natural macromolecules was developed for effective bladder cancer treatment,which could be more conducive to clinical application due to its simple preparation and high biological safety.Ye Liang Yonghua Wang Liping Wang Zhijuan Liang Dan Li Xiaoyu Xu Yuanbin Chen Xuecheng Yang Hongbo Zhang Haitao Niu 2021Bioactive Materials2021,6,2:5
