| 1 | Novel insights into stress-induced susceptibility to influenza:corticosterone impacts interferon-βresponses by Mfn2-mediated ubiquitin degradation of MAVS显示文摘Although stress has been known to increase the susceptibility of pathogen infection,the underlying mechanism remains elusive.In this study,we reported that restraint stress dramatically enhanced the morbidity and mortality of mice infected with the influenza virus(H1N1)and obviously aggravated lung inflammation.Corticosterone(CORT),a main type of glucocorticoids in rodents,was secreted in the plasma of stressed mice.We further found that this stress hormone significantly boosted virus replication by restricting mitochondrial antiviral signaling(MAVS)protein-transduced IFN-βproduction without affecting its mRNA level,while the deficiency of MAVS abrogated stress/CORT-induced viral susceptibility in mice.Mechanistically,the effect of CORT was mediated by proteasome-dependent degradation of MAVS,thereby resulting in the impediment of MAVS-transduced IFN-βgeneration in vivo and in vitro.Furthermore,RNA-seq assay results indicated the involvement of Mitofusin 2(Mfn2)in this process.Gain-and loss-offunction experiments indicated that Mfn2 interacted with MAVS and recruited E3 ligase SYVN1 to promote the polyubiquitination of MAVS.Co-immunoprecipitation experiments clarified an interaction between any two regions of Mfn2(HR1),MAVS(C-terminal/TM)and SYVN1(TM).Collectively,our findings define the Mfn2-SYVN1 axis as a new signaling cascade for proteasome-dependent degradation of MAVS and a‘fine tuning’of antiviral innate immunity in response to influenza infection under stress. | Zhuo Luo Li-Fang Liu Ying-Nan Jiang Lu-Ping Tang Wen Li Shu-Hua Ouyang Long-Fang Tu Yan-Ping Wu Hai-Biao Gong Chang-Yu Yan Shan Jiang Yu-Hui Lu Tongzheng Liu Zhenyou Jiang Hiroshi Kurihara Yang Yu Xin-Sheng Yao Yi-Fang Li Rong-Rong He | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 5 |

| 4 | Formation of uniform reduced graphene oxide films on modified PET substrates using drop-casting method显示文摘In this work,uniform reduced graphene oxide(RGO) films were formed on poly-(ethylene terephthalate)(PET) substrates using a simple drop-casting method.We investigated four types of substrates:unmodified PET,polydopamine-coated PET.carboxyl-group-modified PET,and alkyl-group-modified PET.Upon water evaporation,the surface of the polydopamine-modified PET substrates can interact with the reduced graphene oxide sheets to form flattened and continuous RGO films,which exhibit a sheet resistance of 21.75 kΩ/sq at 82%transmittance.The result indicates that the properties of the surface groups determined whether uniform and flattened RGO films could be formed on the substrates.Hence,we proposed a simple and effective way to produce transparent and conductive films in which the catechol unit exhibits a great effect on the deposition of uniform RGO films on PET substrates. | Chunlin Zhao Li Xing Junhui Xiang Lijie Cui JianbinJiao Huazheng Sai Zhenyou Li Fei Li | 2014 | Particuology2014,12,6: | 4 |
| 5 | Pathological and physiological functional cross-talks ofα-synuclein and tau in the central nervous system显示文摘α-Synuclein and tau are abundant multifunctional brain proteins that are mainly expressed in the presynaptic and axonal compartments of neurons,respectively.Previous works have revealed that intracellular deposition ofα-synuclein and/or tau causes many neurodegenerative disorders,including Alzheimer’s disease and Parkinson’s disease.Despite intense investigation,the normal physiological functions and roles ofα-synuclein and tau are still unclear,owing to the fact that mice with knockout of either of these proteins do not present apparent phenotypes.Interestingly,the co-occurrence ofα-synuclein and tau aggregates was found in post-mortem brains with synucleinopathies and tauopathies,some of which share similarities in clinical manifestations.Furthermore,the direct interaction ofα-synuclein with tau is considered to promote the fibrillization of each of the proteins in vitro and in vivo.On the other hand,our recent findings have revealed thatα-synuclein and tau are cooperatively involved in brain development in a stage-dependent manner.These findings indicate strong cross-talk between the two proteins in physiology and pathology.In this review,we provide a summary of the recent findings on the functional roles ofα-synuclein and tau in the physiological conditions and pathogenesis of neurodegenerative diseases.A deep understanding of the interplay betweenα-synuclein and tau in physiological and pathological conditions might provide novel targets for clinical diagnosis and therapeutic strategies to treat neurodegenerative diseases. | Mingyue Jin Shengming Wang Xiaodie Gao Zhenyou Zou Shinji Hirotsune Liyuan Sun | 2024 | Neural Regeneration Research2024,19,4: | 1 |
