| 1 | Mosquito-Associated Viruses in China显示文摘Mosquitoes are classified into approximately 3500 species and further grouped into 41 genera.Epidemiologically,they are considered to be among the most important disease vectors in the world and they can harbor a wide variety of viruses.Several mosquito viruses are considered to be of significant medical importance and can cause serious public health issues throughout the world.Such viruses are Japanese encephalitis virus(JEV),dengue virus(DENV),chikungunya virus(CHIKV),and Zika virus(ZIKV).Others are the newly recognized mosquito viruses such as Banna virus(BAV)and Yunnan orbivirus(YNOV)with unclear medical significance.The remaining mosquito viruses are those that naturally infect mosquitoes but do not appear to infect humans or other vertebrates.With the continuous development and improvement of mosquito and mosquito-associated virus surveillance systems in China,many novel mosquito-associated viruses have been discovered in recent years.This review aims to systematically outline the history,characteristics,distribution,and/or current epidemic status of mosquito-associated viruses in China. | Han Xia Yujuan Wang Evans Atoni Bo Zhang Zhiming Yuan | 2018 | Virologica Sinica2018,33,1: | 13 |
| 2 | Great progress in study on aerosol and its impact on the global environment显示文摘The transport from atmosphere is a more important pathway than that from rivers for certain elements and compounds in the remote ocean. The desert and those arid/semi-arid areas in the northwest of China and the Loess Plateau are the sources of those aerosols over the Pacific. Atmospheric iron is the limiting factor of the productivity in certain ocean regions. The dust storms must now be seen as repeated sources of pollution elements as well as soil elements to the East China Sea and, farther out, to the North Pacific Ocean and even to the USA. The long-range transport of aerosols, especially the dust storm, from China and the positive feedbacks of iron coupled with sulfur in atmosphere might be one of the important mechanisms that would affect the primary productivity in the Pacific and/or the global climate change and deserve to be further studied. | 3 | Rapid Detection of Filoviruses by Real-time TaqMan Polymerase Chain Reaction Assays显示文摘Ebola virus (EBOV) and Marburg virus (MARV) are causative agents of severe hemorrhagic fever with high mortality rates in humans and non-human primates and there is currently no licensed vaccine or therapeutics. To date, there is no specific laboratory diagnostic test in China, while there is a national need to provide differential diagnosis during outbreaks and for instituting acceptable quarantine procedures. In this study, the TaqMan RT-PCR assays targeting the nucleoprotein genes of the Zaire Ebolavirus (ZEBOV) and MARV were developed and their sensitivities and specificities were investigated. Our results indicated that the assays were able to make reliable diagnosis over a wide range of virus copies from 103 to 109, corresponding to the threshold of a standard RNA transcript. The results showed that there were about 1010 RNA copies per milliliter of virus culture supernatant, equivalent to 10,000 RNA molecules per infectious virion, suggesting the presence of many non-infectious particles. | 4 | Analysis of threshed rice mixture separation through vibration screen using discrete element method显示文摘The discrete element method was utilized to construct three-dimensional discrete element models for the rice mixture,and their motions were analyzed numerically on a planar vibration screening device.The results showed that,after falling onto the vibrating screen surface,the mixture undergoes a reciprocating motion within the same cycle.During the screening process,the mixture undergoes segregation,slides along the screen surface,passes through the screen and then falls.In comparing the movement of grains and shriveled grains,it can be seen that the velocity of shriveled grains experiences cyclical changes,which is consistent with the grains’motion cycle.The impact on grains is shown to be greater than that on shriveled grains,and the average speeds of shriveled grains and grains converge.The curve shows that the screening of repetitive movements has a significant effect on the average velocity of shriveled grains,but also the velocity of shriveled grains and the vibration parameters can be well represented by a fitting equation.It is beneficial for the separation of grains from shriveled grains to choose a greater vibration frequency and screen surface inclination in the range of commonly used. | Li Hua Wang Jinshuang Yuan Jianbo Yin Wenqing Wang Zhiming Qian Youzhang | 2017 | International Journal of Agricultural and Biological Engineering2017,10,6: | 10 |
| 5 | Hierarchical assembly of highly efficient visible-light-driven Ag/g-C_(3)N_(4)/kaolinite composite photocatalyst for the degradation of ibuprofen显示文摘A novel Ag/g-C_(3)N_(4)/kaolinite composite photocatalyst was fabricated for the first time through a two-step assembly strategy by employing in situ calcination and a photodeposition process.The synthesized Ag/g-C_(3)N_(4)/kaolinite composite reached a higher degradation rate of ibuprofen(IBP)with a reaction rate constant of 0.0113 min^(-1) at an Ag content of 7% under visible-light irradiation,which was approximately 1.87 times that of the Ag/g-C_(3)N_(4) composite.Based on the physicochemical properties,the enhanced photocatalytic activity was attributed to the stronger adsorption property,wider photoresponse range and more efficient separation and transfer of electron-hole pairs.Furthermore,the incorporation of monodispersed Ag nanoparticles onto the g-C_(3)N_(4)/kaolinite sheets provided more reactive sites for the IBP degradation.In addition,according to the EPR study and trapping experiments,it was demonstrated that holes(h^(+))should be the key reactive species.A possible pathway of IBP degradation was also proposed based on the detected intermediates.Overall,the results of this work may facilitate the design of a novel visible-light-driven photocatalyst with a high efficiency that is derived from a natural mineral for environmental remediation. | Zhiming Sun Xiangwei Zhang Xiongbo Dong Xiaorui Liu Ye Tan Fang Yuan Shuilin Zheng Chunquan Li | 2020 | Journal of Materiomics2020,6,3: | 8 |
| 6 | Micro-droplet Digital Polymerase Chain Reaction and Real-Time Quantitative Polymerase Chain Reaction Technologies Provide Highly Sensitive and Accurate Detection of Zika Virus显示文摘The establishment of highly sensitive diagnostic methods is critical in the early diagnosis and control of Zika virus(ZIKV)and in preventing serious neurological complications of ZIKV infection. In this study, we established micro-droplet digital polymerase chain reaction(ddPCR) and real-time quantitative PCR(RT-qPCR) protocols for the detection of ZIKV based on the amplification of the NS5 gene. For the ZIKV standard plasmid, the RT-qPCR results showed that the cycle threshold(Ct) value was linear from 10~1 to 10~8 copy/l L, with a standard curve R^2 of 0.999 and amplification efficiency of 92.203%;however, a concentration as low as 1 copy/l L could not be detected. In comparison with RT-qPCR, the dd PCR method resulted in a linear range of 10~1–10~4 copy/l L and was able to detect concentrations as low as 1 copy/l L. Thus, for detecting ZIKV from clinical samples, RT-qPCR is a better choice for high-concentration samples(above 10~1 copy/l L),while ddPCR has excellent accuracy and sensitivity for low-concentration samples. | 8 | Nucleoprotein-based indirect enzyme-linked immunosorbent assay(indirect ELISA) for detecting antibodies specific to Ebola virus and Marbug virus显示文摘Full-length nucleoproteins from Ebola and Marburg viruses were expressed as His-tagged recombinant proteins in Escherichia coli and nucleoprotein-based enzyme-linked immunosorbent assays(ELISAs) were established for the detection of antibodies specific to Ebola and Marburg viruses. The ELISAs were evaluated by testing antisera collected from rabbit immunized with Ebola and Marburg virus nucleoproteins. Although little cross-reactivity of antibodies was observed in antiEbola virus nucleoprotein rabbit antisera, the highest reactions to immunoglobulin G(Ig G) were uniformly detected against the nucleoprotein antigens of homologous viruses. We further evaluated the ELISA's ability to detect antibodies to Ebola and Marburg viruses using human sera samples collected from individuals passing through the Guangdong port of entry. With a threshold set at the mean plus three standard deviations of average optical densities of sera tested, the ELISA systems using these two recombinant nucleoproteins have good sensitivity and specificity. | 11 | Critical role of Dengue Virus NS1 protein in viral replication显示文摘Dengue virus(DENV) nonstructural protein 1(NS1) is a highly conserved 46-kDa protein that contains 2 glycosylation sites(Asn-130 and Asn-207) and 12 conserved cysteine(Cys) residues. Here, we performed site-directed mutagenesis to generate systematic mutants of viral strain TSV01. The results of the subsequent analysis showed that an alanine substitution at the second N-linked glycan Asn-207 in NS1 delayed viral RNA synthesis, reduced virus plaque size, and weakened the cytopathic effect. Three mutants at Cys sites(Cys-4, Cys-55, Cys-291) and a C-terminal deletion(ΔC) mutant significantly impaired RNA synthesis, and consequently abolished viral growth, whereas alanine mutations at Asn-130 and Glu-173 resulted in phenotypes that were similar to the wild-type(WT) virus. Further analysis showed that the Asn-207 mutation slightly delayed viral replication. A series of 2-D physical model experiments were conducted to measure the wave transmission coefficient, tension acting on the mooring line, and motion response of FB under a regular wave. | 15 | Inhibition of Japanese Encephalitis Virus Infection by Flavivirus Recombinant E Protein Domain Ⅲ显示文摘Japanese encephalitis virus (JEV) is a mosquito-borne flavivirus closely related to the human pathogens including yellow fever virus, dengue virus and West Nile virus. There are currently no effective antiviral therapies for all of the flavivirus and only a few highly effective vaccines are licensed for human use. In this paper, the E protein domain III (DIII) of six heterologous flaviviruses (DENV1-4, WNV and JEV) was expressed in Escherichia coli successfully. The proteins were purified after a solubilization and refolding procedure, characterized by SDS-PAGE and Western blotting. Competitive inhibition showed that all recombinant flavivirus DIII proteins blocked the entry of JEV into BHK-21 cells. Further studies indicated that antibodies induced by the soluble recombinant flavivirus DIII partially protected mice against lethal JEV challenge. | 20 | Mutagenesis of D80-82 and G83 Residues in West Nile Virus NS2B: Effects on NS2B-NS3 Activity and Viral Replication显示文摘Flaviviral NS2B is a required cofactor for NS3 serine protease activity and plays an important role in promoting functional NS2B-NS3 protease configuration and maintaining critical interactions with protease catalysis substrates. The residues D80DDG in West Nile virus (WNV) NS2B are important for protease activity. To investigate the effects of D80DDG in NS2B on protease activity and viral replication, the negatively charged region D80DD and the conserved residue G83 of NS2B were mutated (D80DD/E80EE, D80DD/K80KK, D80DD/A80AA, G83F, G83S, G83D, G83K, and G83A), and NS3 D75A was designated as the negative control. The effects of the mutations on NS2B-NS3 activity, viral translation, and viral RNA replication were analyzed using kinetic analysis of site-directed enzymes and a transient replicon assay. All substitutions resulted in significantly decreased enzyme activity and blocked RNA replication. The negative charge of D80DD is not important for maintaining NS2B function, but side chain changes in G83 have dramatic effects on protease activity and RNA replication.
