1Abrogation of USP7 is an alternative strategy to downregulate PD-L1 and sensitize gastric cancer cells to T cells killing显示文摘Targeting immune checkpoints such as programmed cell death protein 1(PD-1)and programmed death ligand-1(PD-L1)have been approved for treating melanoma,gastric cancer(GC)and bladder cancer with clinical benefit.Nevertheless,many patients failed to respond to anti-PD-1/PD-L1 treatment,so it is necessary to seek an alternative strategy for traditional PD-1/PD-L1 targeting immunotherapy.Here with the data from The Cancer Genome Atlas(TCGA)and our in-house tissue library,PD-L1 expression was found to be positively correlated with the expression of ubiquitin-specific processing protease 7(USP7)in GC.Furthermore,USP7 directly interacted with PD-L1 in order to stabilize it,Gastric cancer;Immunosuppression;Cancer biologywhile abrogation of USP7 attenuated PD-L1/PD-1 interaction and sensitized cancer cells to T cell killing in vitro and in vivo.Besides,USP7 inhibitor suppressed GC cells proliferation by stabilizing P53 in vitro and in vivo.Collectively,our findings indicate that in addition to inhibiting cancer cells proliferation,USP7 inhibitor can also downregulate PD-L1 expression to enhance anti-tumor immune response simultaneously.Hence,these data posit USP7 inhibitor as an anti-proliferation agent as well as a novel therapeutic agent in PD-L1/PD-1 blockade strategy that can promote the immune response of the tumor.Zhiru Wang Wenting Kang Ouwen Li Fengyu Qi Junwei Wang Yinghua You Pengxing He Zhenhe Suo Yichao Zheng Hong-Min Liu 2021Acta Pharmaceutica Sinica B2021,11,3:5
2Ripple-associated high-firing interneurons in the hippocampal CA1 region显示文摘By simultaneously recording the activity of individual neurons and field potentials in freely behaving mice, we found two types of interneurons firing at high frequency in the hippocampal CA1 region, which had high correlations with characteristic sharp wave-associated ripple oscillations (100―250 Hz) during slow-wave sleep. The firing of these two types of interneurons highly synchronized with ripple oscillations during slow-wave sleep, with strongly increased firing rates corresponding to individual ripple episodes. Interneuron type I had at most one spike in each sub-ripple cycle of ripple episodes and the peak firing rate was 310±33.17 Hz. Interneuron type II had one or two spikes in each sub-ripple cycle and the peak firing rate was 410±47.61 Hz. During active exploration, their firing was phase locked to theta oscillations with the highest probability at the trough of theta wave. Both two types of interneurons increased transiently their firing rates responding to the startling shake stimuli. 3Effects of adding sodium dichloroacetate to low-protein diets on nitrogen balance and amino acid metabolism in the portaldrained viscera and liver of pigs显示文摘Background:Identifying regulatory measures to promote glucose oxidative metabolism while simultaneously reducing amino acid oxidative metabolism is one of the foremost challenges in formulating low-protein(LP)diets designed to reduce the excretion of nitrogen-containing substances known to be potential pollutants.In this study,we investigated the effects of adding sodium dichloroacetate(DCA)to a LP diet on nitrogen balance and amino acid metabolism in the portal-drained viscera(PDV)and liver of pigs.To measure nitrogen balance,18 barrows(40±1.0 kg)were fed one of three diets(n=6 per group):18%crude protein(CP,control),13.5%CP(LP),and 13.5%CP+100 mg DCA/kg dry matter(LP-DCA).To measure amino acid metabolism in the PDV and liver,15 barrows(40±1.0 kg)were randomly assigned to one of the three diets(n=5 per group).Four essential amino acids(Lys,Met,Thr,and Trp)were added to the LP diets such that these had amino acid levels comparable to those of the control diet.Results:The LP-DCA diet reduced nitrogen excretion in pigs relative to that of pigs fed the control diet(P<0.05),without any negative effects on nitrogen retention(P>0.05).There were no differences between the control and LP-DCA groups with respect to amino acid supply to the liver and extra-hepatic tissues in pigs(P>0.05).The net release of ammonia into the portal vein and production rate of urea in the liver of pigs fed the LP-DCA diet was reduced relative to that of pigs fed the control and LP diets(P<0.05).Conclusion:The results indicated that addition of DCA to a LP diet can efficiently reduce nitrogen excretion in pigs and maximize the supply of amino acids to the liver and extra-hepatic tissues.Weizhong Sun Yunxia Li Zhiru Tang Huiyuan Chen Ke Wan Rui An Liuting Wu Zhihong Sun 2020Journal of Animal Science and Biotechnology2020,11,3:2

9A covalent attraction between two molecular cation TTF·^+显示文摘The optimized structure of the tetrathiafulvalence radical-cation dimer(TTF·+-TTF·+) with all-real frequencies is obtained at MP2/6-311G level,which exhibits the attraction between two molecular cation TTF·+.The new attraction interaction is a 20-center-2-electron intermolecular covalent π /π bonding with a telescope shape.The covalent π /π bonding has the bonding energy of about -21 kcal·mol-1 and is concealed by the Coulombic repulsion between two TTF·+ cations.This intermolecular covalent attraction also influences the structure of the TTF·+ subunit,i.e.,its molecular plane is bent by an angle θ=5.6°.This work provides new knowledge on intermolecular interaction.WANG FangFang WANG Yi WANG BingQiang WANG YinFeng MA Fang Li ZhiRu 2009Science China Chemistry2009,52,11:1

11Imitating trumpet shells: Mbius container molecules显示文摘Mbius container molecules C64H8,C60N4H4,and C58N6H2 with topological one-sided characteristics were constructed at the first time by imitating natural trumpet shells.The structure is an open cage with an inner hexagonal bridge.The bridge joints the outer and inner surfaces of the cage to form a new one-sided Mbius structure.The optimized structures of the three molecules in the singlet(the ground state),triplet and quintet states are obtained using the density functional theory(B3LYP).For the ground state structures of the three Mbius molecules,their oxidizabilities are weaker than that of the C60 and reducibilities are close to that of the stable C80 cage and slightly stronger than that of the C60.These may show that the unusual Mbius structures have some stability.Their potential properties were predicted,for example,the special aromaticity of the bridge ring due to the unique interaction between the bridge and the cage wall.These findings enlarge the knowledge of Mbius molecules. 12Effects of different nitrogen forms and concentrations on seedling growth traits and physiological characteristics of Populus simonii × P. nigra显示文摘Numerous growth and physiological variables of 3-week-old Populus simonii × P.nigra seedlings were assessed after treatment with either nitrate nitrogen(NO_(3)^(−)--N)(0.1,0.5,1,5,or 10 mmol·L^(−1)) or ammonium nitrogen(NH_(4)^(+)+-N)(0.1,0.5,1,5,or 10 mmol·L^(−1)) to determine the best nitrogen form and concentration to optimize growth,biomass allocation,pigment content,and photosynthetic capacity.The results of combining membership function and an evaluation index suggested that,5 mmol·L^(−1) nitrogen,regardless of the form,yielded the highest comprehensive evaluation index and good growth.In addition,a Pearson correlation analysis and network visualization revealed that the total mass,shoot mass,root mass,leaf dry mass,plant height,leaf area,chlorophyll a and total chlorophyll had a physiological index connectivity degree≥15 for both nitrogen forms.Net photosynthetic rate,stomatal conductance,transpiration rate,maximum photochemical efficiency of PSII,total nitrogen content,ground diameter,chlorophyll b,and carotenoid were unique indices for evaluating NH_(4)^(+)+-N-based nutrition,which could provide a theoretical basis for evaluating the effects of nitrogen fertilizer on seedlings,cultivation periods,and stress tolerance in P.simonii× P.nigra.Zhongye Liu Wei Li Zhiru Xu Huihui Zhang Guangyu Sun Heng Zhang Chuanping Yang Guanjun Liu 2022Journal of Forestry Research2022,33,5:0
13Population Structure and Diversity of Plankton in the Wetland of Qilihai in Tianjin显示文摘The population structure and diversity of plankton in the wetland of Qilihai in Tianjin was investigated in May, July and September, 2012 by fi eld investigation and sampling examination. The results showed that there were 44 species of phytoplankton belonged to 6 phyla, including 11 species of Cyanophyta, 18 species of Chlorophyta, 8 species of Bacillariophyta, 4 species of Euglenophyta, 1 species of Pyrrophyta and 2 species of Cryptophyta. There were 13 genus and 22 species of zooplankton, among which 4 genus and 4 species belonged to Cladocera; 2 genus and 2 species belonged to Copepoda; 7 genus and 16 species belonged to Rotifera. The investigation results of density and biomass of plankton in May, July and September showed that the density and biomass of phytoplankton was shown as September>July>May, and that of zooplanktonic was shown as May>September>July. In addition, the diversity of phytoplankton and zooplanktonic were evaluated using Shannon-Wiener index, and the results demonstrated that the phytoplankton species were rich in May, July and September, while the zooplanktonic species were poor in May and general in July and September. 14Genome-Wide Analysis of the KANADI Gene Family and Its Expression Patterns under Different Nitrogen Concentrations Treatments in Populus trichocarpa显示文摘KANADI(KAN)is a plant-specific gene that controlled the polarity development of lateral organs.It mainly acted on the abaxial characteristics of plants to make the lateral organs asymmetrical.However,it had been less identified in woody plants.In this study,the members of the KAN gene family in Populus trichocarpa were identified and analyzed using the bioinformatics method.The results showed that a total of 8 KAN family members were screened out,and each member contained the unique GARP domain and conserved region of the family proteins.Phylogenetic analysis and their gene structures revealed that all KAN genes from P.trichocarpa,Arabidopsis thaliana,and Nicotiana benthamiana could be divided into four subgroups,while the eight genes in P.trichocarpa were classified into three subgroups,respectively.The analysis of tissue-specific expression indicated that PtKAN1 was highly expressed in young leaves,PtKAN6 was highly expressed in young leaves and mature leaves,PtKAN2,PtKAN5,and PtKAN7 were highly expressed in nodes and internodes,PtKAN8 was highly expressed in roots,and PtKAN3 and PtKAN4 showed low expression levels in all tissues.Among them,PtKAN2 and PtKAN6,and PtKAN4 and PtKAN5 might have functional redundancy.Under high nitrogen concentrations,PtKAN2 and PtKAN8 were highly expressed in mature stems and leaves,respectively,while PtKAN4,PtKAN5,and PtKAN7 were highly expressed in roots.This study laid a theoretical foundation for further study of the KAN genemediated nitrogen effect on root development.Minghui Niu Heng Zhang Xiangyang Li Zhibao Hu Hongjiao Zhang Zhiru Xu Chunpu Qu Guanjun Liu 2023Phyton-International Journal of Experimental Botany2023,92,7:0

16Mechanized Grafting Technology for Apple Seedlings显示文摘The current cultivation of apple trees mainly relies on grafted seedlings to achieve seedling regeneration of apple trees. The commonly used splice grafting,V-shaped grafting and tongue grafting mostly use plastic film for fixation and sealing. After one year,the wrapped plastic film needs to be removed to avoid affecting the growth of the seedlings. This paper introduces a brand new grafting technology which uses tongue grafting iron nails for fixation and growth fixation wax for sealing( SDRG). The process of mechanized grafting for apple nursery stock is determined,and the fixation and sealing problems with the grafting parts of seedlings are solved. 17Bioinformatics analysis of genes differentially expressed in autism and screening of hub genes in the occurrence and development of autism显示文摘Objective:to screen the genes differentially expressed in autism using bioinformatics methods,and to explore their functional enrichment,related signaling pathways and the tissue-specific expression of hub genes.Methods:the autism expression profile chip numbered GSE77103 in the Gene Expression Omnibus(GEO)database was selected for examination.R language and related R packages were used for the screening and visualization of the diflerentially expressed genes.Gene Ontology(GO)function enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)signaling pathway analysis of the differential genes were carried out using the relevant R package of R language.The database STRING was used to construct the interaction network of the proteins encoded by the differentially expressed genes,and the software Cytoscape was used to screen the hub genes in the network.The selected hub genes were imported into the BioGPS database to analyze the tissue-specific expression of the hub genes.Results:six hundred and sixty diflerentially expressed genes were screened out.Three hundred and seventy-three up-regulated genes and 287 down-regulated genes in the peripheral blood mononuclear cells of autistic children were compared with the peripheral blood mononuclear cells of healthy children.GO functional enrichment results showed that biological processes(BP)were mainly involved in viral response,negative regulation of viral genome replication,negative regulation of multiple biological processes,negative reg ulation reg ulation of viral life cycle,and defense responses to viruses.Cell components(CC)were involved in vesicles,lysosomal membranes,lysosomal lumen,etc.;molecular functions(MF)were involved in regulating glutathione transferase activity peroxidase activity,oxidoreductase activity,Glutathione peroxidase and transferase activity,etc.The results of KEGG signaling pathway analysis showed that the differentially expressed genes were related to the lysosomal pathway,the glutathione metabolism pathway and the arachidonic acid metabolism pathway.The hub genes screened by cytoHubba were:interferon regulatory factor 7(IRF7),interferon stimulated exonuclease gene 15(ISG15),XIAP associated factor 1(XAF1),MX dynamin like GTPase 1(MX1),interferon induced protein with tetratricopeptide repeats 1(IFIT1),interferon induced protein with tetratricopeptide repeats 5(IFIT5),29-55-oligoadenylate synthetase 3(OAS3),interferon induced protein 44(IFI44),HECT and the RLD domain containing E3 ubiquitin protein ligase 5(HERC5),interferon stimulated exonuclease gene 20(ISG20).Conclusion:there are genes that are diflerentially expressed in the peripheral blood mononuclear cells of autistic toddlers and healthy toddlers.IRF7,ISG15,XAF1,MX],IFIT1,IFIT5,OAS3,IFI44,HERC5,ISG20 are the hub regulatory genes of autism.Manli Li Xiaoli Ma Chendi Mai Zhiru Fan Yankai Ren 2022Journal of Translational Neuroscience2022,7,1:0
