| 2 | Involvement of aquaporins in a mouse model of rotavirus diarrhea显示文摘Rotavirus diarrhea is a major worldwide cause of infantile gastroenteritis; however, the mechanism responsible for intestinal fluid loss remains unclear. Water transfer across the intestinal epithelial membrane seems to occur because of aquaporins(AQPs). Accumulating evidence indicates that alterations in AQPs may play an important role in pathogenesis. Here, we focus on changes in AQPs in a mouse model of rotavirus diarrhea. In the present study, 32 of 35 mice developed diarrhea and mild dehydration within 24 hours after infection with rotavirus strain SA11. Intestinal epithelial cells demonstrated cytoplasmic vacuolation, malaligned villi, and atrophy. AQP1 expression was significantly attenuated in the ileum and colon in comparison with controls; likewise, AQP4 and-8 protein expression were significantly decreased in the colon of rotavirus diarrhea-infected mice. In contrast, AQP3 protein expression was significantly increased in the colon of rotavirus-infected mice in comparison with controls. These results indicate that rotavirus diarrhea is associated with the downregulation of AQP1,-4, and-8 expression. | 4 | IL-25 stimulates M2 macrophage polarization and thereby promotes mitochondrial respiratory capacity and lipolysis in adipose tissues against obesity显示文摘Obesity and associated metabolic diseases are characterized by a chronic low-grade inflammatory state with the infiltration of many inflammatory cells,especially macrophages.Immune molecules,including some cytokines,have a close relationship with metabolism.Interleukin(IL)-25 is a member of the IL-17 cytokine family that can regulate macrophages and alleviate some metabolic dysfunction;however,its role and mechanisms in lipid metabolism remain to be extensively clarified.Human serum and liver biopsy specimens,high-fat diet-induced obesity mice and DB/DB(Lepr−/−)animal models were used to examine IL-25 expression in obesity and nonalcoholic fatty liver diseases(NAFLD).To observe the role of IL-25 in lipid metabolism,model mice were administered with IL-25 or adoptively transferred with IL-25-educated macrophages in vivo,whereas bone marrow-derived macrophages,the macrophage cell line RAW264.7 and adipocytes differentiated from 3T3-L1 were used in vitro.IL-25 was decreased in NAFLD patients and obese mice.In addition,IL-25 reduced body weight gain and lipid accumulation,enhanced lipid uptake by macrophages and increased the expression of lipolysis andβ-oxidation enzymes via alternatively activating macrophages.IL-25 also promoted lipolysis and suppressed lipogenesis in adipocytes co-cultured with the IL-25-educated macrophages.Furthermore,IL-25 improved the mitochondrial respiratory capacity and oxygen consumption rate of macrophages and produced more NAD+/NADH and ATP.In conclusion,IL-25 can stimulate M2 macrophage polarization and thereby promote lipolysis and mitochondrial respiratory capacity,highlighting the potential for IL-25 to be used as a therapeutic agent against obesity and associated metabolic syndromes. | Juan Feng Lingyi Li Zhiying Ou Qiao Li Baoyong Gong Zhenxian Zhao Weiwei Qi Ti Zhou Jun Zhong Weibin Cai Xia Yang Aiping Zhao Guoquan Gao Zhonghan Yang | 2018 | Cellular & Molecular Immunology2018,15,5: | 10 |
| 5 | Enhanced methane production in an anaerobic digestion and microbial electrolysis cell coupled system with co-cultivation of Geobacter and Methanosarcina显示文摘The anaerobic digestion(AD)and microbial electrolysis cell(MEC)coupled system has been proved to be a promising process for biomethane production.In this paper,it was found that by co-cultivating Geobacter with Methanosarcina in an AD–MEC coupled system,methane yield was further increased by 24.1%,achieving to 360.2 m L/g-COD,which was comparable to the theoretical methane yield of an anaerobic digester.With the presence of Geobacter,the maximum chemical oxygen demand(COD)removal rate(216.8 mg COD/(L·hr))and current density(304.3 A/m_3)were both increased by 1.3 and 1.8 fold compared to the previous study without Geobacter,resulting in overall energy efficiency reaching up to 74.6%.Community analysis demonstrated that Geobacter and Methanosarcina could coexist together in the biofilm,and the electrochemical activities of both were confirmed by cyclic voltammetry.Our study observed that the carbon dioxide content in total gas generated from the AD reactor with Geobacter was only half of that generated from the same reactor without Geobacter,suggesting that Methanosarcina may obtain the electron transferred from Geobacter for the reduction of carbon dioxide to methane.Taken together,Geobacter not only can improve the performance of the MEC system,but also can enhance methane production. | Qi Yin Xiaoyu Zhu Guoqiang Zhan Tao Bo Yanfei Yang Yong Tao Xiaohong He Daping Li Zhiying Yan | 2016 | Journal of Environmental Sciences2016,28,4: | 9 |
| 6 | Comparison of the short-term and long-term outcomes of laparoscopic hysterectomies and of abdominal hysterectomies: a case study of 4,895 patients in the Guangxi Zhuang Autonomous Region, China显示文摘Background: To evaluate the short-term and long-term outcomes after laparoscopic hysterectomy(LH) compared with abdominal hysterectomy(AH) in case of benign gynecological disease. Methods: A multi-center cohort retrospective comparative study of population among 4,895 hysterectomies(3,539 LH vs.1,356 AH) between 2007 and 2013 was involved. Operative time(OT), estimated blood loss(EBL), intra-operative and post-operative complications, passing flatus; days with indwelling catheter, questionnaires covering pelvic floor functions and sexual functions were assessed.Results: The EBL(174.1±157.4 vs. 263.1±183.2 cc, LH and AH groups, respectively), passing flatus(38.7±14.1 vs. 48.1±13.2 hours), days with indwelling catheter(1.5±0.6 vs. 2.2±0.8 days), use of analgesics(6.5% vs. 73.1%), intra-operative complication rate(2.4% vs. 4.1%), post-operative complication rate(2.3% vs. 5.7%), post-operative constipation(12.1% vs. 24.6%), mild and serious stress urinary incontinence(SUI) post-operative(P<0.001; P=0.014), and proportion of Female Sexual Functioning Index(FSFI) total score <26.55 post-operative(P<0.001) of the LH group were significantly less than those of AH group. There wereno significant differences in OT(106.5±34.5 vs. 106.2±40.3 min) between the two groups.Conclusions: LH is a safe and efficient operation for improving patients' long-term quality of life(Qo L), and LH is a cost-effectiveness procedure for treating benign gynecological disease. | 8 | Clinical diagnosis and treatment of alpha-fetoprotein-negative small hepatic lesions显示文摘Objective: We examined 103 cases over the last five years and discussed diagnosis and treatment of alpha- fetoprotein (AFP)-negative small hepatic lesions. Background: Small hepatic lesions (less than 2 cm in diameter) usually have no typical imaging characteristics and therefore are difficult to diagnose, especially when AFP tests provide a negative result. Methods: A total of 103 patients with AFP-negative small hepatic lesions from January 2003 to December 2008 were retrospectively reviewed. Differential diagnosis was performed by digital subtraction angiography (DSA), dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI), contrast-enhanced ultrasound (CEUS), or positron emission tomography-computed tomography (PET-CT) based on the multiplicity of lesions. Ninety-four patients with suspected cancers underwent partial hepatectomy. Clinical data were collected from hospital records and follow-up questionnaires. Results: Hepatocellular carcinoma (HCC) diagnostic sensitivity of DSA, DCE-MRI, CEUS and PET-CT was 88.2%, 93.9%, 88.9% and 88.9%, respectively. The surgery-related complication rate was 6.4%. Prognosis was good, with 1- and 3-year survival rates of 98.8% and 76.1%, respectively. Conclusions: DSA, DCE-MRI, CEUS and PET-CT are valuable for diagnosis of small hepatic lesions. Partial hepatectomy is a preferred surgical procedure. | 10 | Preparation of rabbit anti-rat LRRN3 polyclonal antibody and study of its expression显示文摘BACKGROUND:Studies have shown that the LRRN3,a member of the Neuron Leucine-Rich Repeat superfamily,could be related to neural development,differentiation,information transmission,and other functions,but most studies have focused on nucleic acid levels and few have reported on LRRN3 protein levels.OBJECTIVE:To prepare rabbit anti-rat LRRN3 polyclonal antibody and to observe protein tissue expression profiles.DESIGN,TIME AND SETTING:In vitro,molecular,biological experiments were performed from October 2007 to April 2009 in Laboratory of Neurobiology at Xiangya School of Medicine,Central South University.MATERIALS:Immunization antigen,namely rat Mal-LRRN3C-His recombinant protein,was provided by the Laboratory of Neurobiology at Xiangya School of Medicine,Central South University.METHODS:Rat Mal-LRRN3C-His recombinant protein was used to immunize male,New Zealand rabbits,and rabbit anti-rat LRRN3 polyclonal antibody was prepared.MAIN OUTCOME MEASURES:Antibody purification was conducted using Protein A affinity chromatography,and the LRRN3 anti-serum titer was identified using enzyme-linked immunosorbent assay.Immunohistochemical techniques and Western blot preliminary tests were used to determine LRRN3 protein expression profiles in adult rats.RESULTS:A highly purified rabbit anti-rat LRRN3 polyclonal antibody was obtained.Western Blot results from rat brain total protein revealed a band at 79 kD,which was consistent with the size of LRRN3.Immunohistochemistry results showed that protein was mainly expressed in the central nervous system,and no significant positive signals were observed in other tissues.Positive cells included neurons of cerebral cortex and hippocampal dentate gyrus granule cell layer,and cerebellar Purkinje cells.There was no positive expression in glial cells.CONCLUSION:Rabbit anti-rat LRRN3 polyclonal antibody was successfully prepared at a high purity from the prokaryotic-expressed Mal-LRRN3C-His recombinant protein,which served as an antigen.Rat LRRN3 protein was primarily expressed in cerebral cortex neurons,hippocampal dentate gyrus granule cell layer neurons,and cerebellar Purkinje cells. | Jing Yang Yanjin Wang Yan Cai Zhiying Yang Ganqiu Wu Jianguo Wen Xuegang Luo He Huang | 2010 | Neural Regeneration Research2010,5,13: | 3 |

| 15 | The G-protein α subunit GhGPA positively regulates Gossypium hirsutum resistance to Verticillium dahliae via induction of SA and JA signaling pathways and ROS accumulation显示文摘Verticillium wilt, a devastating disease in cotton caused by Verticillium dahliae, reduces cotton quality and yield. Heterotrimeric GTP-binding proteins, consisting of Ga, Gb, and Gc subunits, transducers of receptor signaling, function in a wide range of biological events. However, the function of Ga proteins in the regulation of defense responses in plants is largely unexplored, except for a few reports on model species. In the present study, a cotton G-protein a-subunit-encoding gene(GhGPA) was isolated from Verticillium wilt-resistant Gossypium hirsutum(upland cotton) cv. ND601. GhGPA transcription was up-regulated under V. dahliae stress, with higher expression in tolerant than in susceptible cotton cultivars.Subcellular localization revealed GhGPA to be located in the plasma membrane. GhGPA shows high(85.0%) identity with Arabidopsis AT2 G26300(AtGPA1), and AtGPA1 gpa1-4 mutants displayed susceptibility to V. dahliae. Ectopic expression of GhGPA successfully restored the resistance of Arabidopsis gpa1-4 mutants to Verticillium wilt and made them more resistant than the wild type. Overexpression of GhGPA in Arabidopsis markedly increased the resistance and resulted in dramatic up-regulation of pathogenesis-related(PR) genes and increased in H2 O2 accumulation and salicylic acid(SA) and jasmonic acid(JA) contents. However, suppressing GhGPA expression via virus-induced gene silencing(VIGS)increased susceptibility to Verticillium wilt, down-regulated the expression of PR and marker genes in SA and JA signaling pathways, and reduced H2 O2 content. The contents of SA and JA in Arabidopsis gpa1-4 and VIGS cotton were lower than those in the wild type and empty-vector control. However,GhGPA-overexpressing Arabidopsis contained more SA and JA than the wild type when inoculated with V. dahliae. Thus, GhGPA plays a vital role in Verticillium wilt resistance by inducing SA and JA signaling pathways and regulating the production of reactive oxygen species. | 17 | Development of powdery mildew resistant derivatives of wheat variety Fielder for use in genetic transformation显示文摘Genetic transformation is widely used to improve target traits and to study gene function in wheat.However,transformation efficiency depends on the physiological status of the recipient genotype and that is affected by several factors including powdery mildew(PM)infection.The widely used recipient variety Fielder is very susceptible to PM.Therefore,it would be beneficial to develop PM resistant derivatives with high regeneration ability for use in genetic transformation.In the present study PM resistant lines CB037 and Pm97033 carrying genes Pm21 and PmV,respectively,were backcrossed to Fielder with selection for PM resistance.Five lines,NT89,NT90,NT154,and WT48 with Pm21 and line FL347 with PmV were developed,identified by molecular markers and genomic in situ hybridization(GISH)or fluorescent in situ hybridization(FISH),and further subjected to detailed assessment of agronomic traits and regeneration ability following genetic transformation capacity.Lines FL347,WT48,NT89 and NT154 assessed as being equal to,or superior,to Fielder in regeneration and transformation ability are recommended as suitable materials for the replacement of Fielder for wheat gene transfer and genome editing study. | Xiaona Liang Xiaomin Bie Yuliang Qiu Ke Wang Zujun Yang Yuqi Jia Zhiying Xu Mei Yu Lipu Du Zhishan Lin Xingguo Ye | 2023 | The Crop Journal2023,11,2: | 2 |
