1Characterization of the receptor-binding domain(RBD)of 2019 novel coronavirus:implication for development of RBD protein as a viral attachment inhibitor and vaccine显示文摘The outbreak of Coronavirus Disease 2019(COVID-19)has posed a serious threat to global public health,calling for the development of safe and effective prophylactics and therapeutics against infection of its causative agent,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),also known as 2019 novel coronavirus(2019-nCoV).The CoV spike(S)protein plays the most important roles in viral attachment,fusion and entry,and serves as a target for development of antibodies,entry inhibitors and vaccines.Here,we identified the receptor-binding domain(RBD)in SARS-CoV-2 S protein and found that the RBD protein bound strongly to human and bat angiotensin-converting enzyme 2(ACE2)receptors.SARS-CoV-2 RBD exhibited significantly higher binding affinity to ACE2 receptor than SARS-CoV RBD and could block the binding and,hence,attachment of SARS-CoV-2 RBD and SARS-CoV RBD to ACE2-expressing cells,thus inhibiting their infection to host cells.SARS-CoV RBD-specific antibodies could crossreact with SARS-CoV-2 RBD protein,and SARS-CoV RBD-induced antisera could cross-neutralize SARS-CoV-2,suggesting the potential to develop SARS-CoV RBD-based vaccines for prevention of SARS-CoV-2 and SARS-CoV infection.Wanbo Tai Lei He Xiujuan Zhang Jing Pu Denis Voronin Shibo Jiang Yusen Zhou Lanying Du 2020Cellular & Molecular Immunology2020,17,6:35
2Regulation of activin receptor-interacting protein 2 expression in mouse hepatoma Hepa1-6 cells and its relationship with collagen type Ⅳ显示文摘AIM: To investigate the regulation of activin receptor-interacting protein 2 (ARIP2) expression and its possible relationships with collagen type Ⅳ (collagen Ⅳ) in mouse hepatoma cell line Hepal-6 cells. METHODS: The ARIP2 mRNA expression kinetics in Hepal-6 cells was detected by RT-PCR,and its regulation factors were analyzed by treatment with signal transduction activators such as phorbol 12-myristate 13-acetate (PMA),forskolin and A23187. After pcDNA3-ARIP2 was transfected into Hepal-6 cells,the effects of ARIP2 overexpression on activin type Ⅱ receptor (ActRⅡ) and collagen Ⅳ expression were evaluated. RESULTS: The expression levels of ARIP2 mRNA in Hapel-6 cells were elevated in time-dependent manner 12 h after treatment with activin A and endotoxin LPS,but not changed evidently in the early stage of stimulation (2 or 4 h). The ARIP2 mRNA expression was increased after stimulated with signal transduction activators such as PMA and forskolin in Hepal-6 cells,whereas decreased after treatment with A23187 (25.3% ± 5.7% vs 48.1% ± 3.6%,P < 0.01). ARIP2 overexpression could remarkably suppress the expression of ActRⅡA mRNA in dose-dependent manner,but has no effect on ActRⅡB in Hepal-6 cells induced by activin A. Furthermore,we have found that overexpression of ARIP2 could inhibit collagen Ⅳ mRNA and protein expressions induced by activin A in Hapel-6 cells. CONCLUSION: These findings suggest that ARIP2 expression can be influenced by various factors. 3Quantification of Panax notoginseng saponins metabolites in rat plasma with in vivo gut microbiota-mediated biotransformation by HPLC-MS/MS显示文摘Panax notoginseng saponins(PNS) are the major components of Panax notoginseng, with multiple pharmacological activities but poor oral bioavailability. PNS could be metabolized by gut microbiota in vitro, while the exact role of gut microbiota of PNS metabolism in vivo remains poorly understood. In this study, pseudo germ-free rat models were constructed by using broad-spectrum antibiotics to validate the gut microbiota-mediated transformation of PNS in vivo. Moreover, a high performance liquid chromatography-electrospray ionization tandem mass spectrometry(HPLC-ESI-MS/MS) was developed for quantitative analysis of four metabolites of PNS, including ginsenoside F1(GF1), ginsenoside Rh2(GRh2), ginsenoside compound K(GCK) and protopanaxatriol(PPT). The results showed that the four metabolites could be detected in the control rat plasma, while they could not be determined in pseudo germ-free rat plasma. The results implied that PNS could not be biotransformed effectively when gut microbiota was disrupted. 4Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 5Pyogenic liver abscesses associated with nonmetastatic colorectal cancers: An increasing problem in Eastern Asia显示文摘AIM: To elaborate the clinicopathologic features of colorectal cancer-related pyogenic liver abscess (PLA). METHODS: Reported cases of colorectal cancer-related PLAs were collected from the literature published up to October 2011 and evaluated for their clinicopathologic features. Data of collected cases included demographics, clinical presentation, microbial findings and treatment. Categorical variables were compared by 2 analysis and continuous variables were evaluated using Student's t test. RESULTS: A total 96 cases of colorectal cancer-related PLA were collected from the previous literature. Most patients (60%) were male and 40% cases occurred in the age group of 61-70 years. Apart from some special types of PLA, there were significant differences in the microbiological spectrum between Eastern Asia and non-Eastern Asian countries, which implied different risk factors and courses of the disease. Gram negative bacteria especially Klebsiella pneumoniae (K. pneu- moniae ) PLA was predominant in Eastern Asia (80.0%) in contrast to non-Eastern Asian countries (P < 0.01). Meanwhile, most of the Eastern Asian patients exhibited smaller size of liver abscess and atypical presentation. Sigmoid colon and rectum (72.73%) were the main sites of tumor in Eastern Asian patients, whereas tumor sites were uneven among most of the non-Easter Asian PLA patients. 7Panax notoginseng saponins prevent colitis-associated colorectal cancer development: the role of gut microbiota显示文摘Gut microbiota dysbiosis is a risk factor for colorectal cancer(CRC) in inflammatory bowel disease(IBD).In this study, the effects of Panax notoginseng saponins(PNS) on colitis-associated CRC progression were evaluated on an azoxymethane(AOM)/dextran sulfate sodium(DSS) mouse model.In vivo, PNS significantly relieved AOM/DSS-induced colon tumorigenesis and development by reducing the disease activity index(DAI) scores and colon tumor load.The 16S rRNA data of fecal samples showed that the microbiome community was obviously destructed, while PNS could recover the richness and diversity of gut microbiota.Especially, PNS could increase the abundance of Akkermansia spp.which was significantly decreased in model group and negatively correlated with the progression of CRC.Moreover, ginsenoside compound K(GC-K) was evaluated on the effects of human CRC cells,which was the main bio-transformed metabolite of PNS by gut microbiota.Our data showed that PNS played important role in the prevention of the progression of CRC, due to their regulation on the microbiome balance and microbial bio-converted product with antiCRC activity.CHEN Ling CHEN Man-Yun SHAO Li ZHANG Wei RAO Tai ZHOU Hong-Hao HUANG Wei-Hua 2020Chinese Journal of Natural Medicines2020,18,7:12

9Corrosion behavior of aluminum alloys in Na_2SO_4 solution using the scanning electrochemical microscopy technique显示文摘The corrosion behavior of aluminum alloys 1060 and 2A12 in a 10 mM Na2SO4+5 mM KI solution was investigated by scanning electrochemical microscopy(SECM) and scanning electron microscopy(SEM) . The potential topography and corrosion morphology results show that the potential of the sample surface over the same area changes with the increase of immersion time. The corrosion area becomes large,and the potential becomes more negative. The corrosion potential of the 2A12 alloy surface is lower than that of 1060 aluminum,and 2A12 alloy becomes easily corrosive. 13Rapid decolorization of water soluble azo-dyes by nanosized zero-valent iron immobilized on the exchange resin显示文摘Nanosized zero-valent iron (NZVI) supported on the cation exchange resin was synthesized and applied to decompose some water soluble azo dyes. The decomposition efficiency for azo dyes was evaluated by using the aqueous suspensions and parked column of this material. Batch experiments indicated that this novel material exhibited excellent degradation ability for 0.05 g·L1 of Acid Orange 7, Acid Orange 8, Acid Orange 10, Sunset Yellow, and Methyl Orange, with decolorization ratio up to 95% in 4 min; pH value was the key factor for degradation and H+ was one of the reactants; adsorption of azo dyes onto the material existed at the beginning but reduced gradually until disappearing completely. For the packed column system, 58%~90% of azo dyes were decomposed in the 1st circle of solution passing through the column, and the adsorption onto the materials could accelerate the degradation azo dyes with the increasing reaction time. During the degradation process, Fe2+, the product of NZVI, was exchanged to the resin again and could be reduced to Fe0 by KBH4 for reusing. The 10th refreshed NZVI possessed reductive activity up to 90% of the newly systhesized NZVI. 14Application of Contact Graph Routing in Satellite Delay Tolerant Networks显示文摘Satellite networks have many inherent advantages over terrestrial networks and have become an important part of the global network infrastructure.Routing aimed at satellite networks has become a hot and challenging research topic.Satellite networks,which are special kind of Delay Tolerant Networks(DTN),can also adopt the routing solutions of DTN.Among the many routing proposals,Contact Graph Routing(CGR) is an excellent candidate,since it is designed particularly for use in highly deterministic space networks.The applicability of CGR in satellite networks is evaluated by utilizing the space oriented DTN gateway model based on OPNET(Optimized Network Engineering Tool).Link failures are solved with neighbor discovery mechanism and route recomputation.Earth observation scenario is used in the simulations to investigate CGR's performance.The results show that the CGR performances are better in terms of effectively utilizing satellite networks resources to calculate continuous route path and alternative route can be successfully calculated under link failures by utilizing fault tolerance scheme.ZHU Laitao LI Yong ZHANG Junxiang WU Jing TAI Xiao ZHOU Jianguo 2015空间科学学报2015,35,1:5

19Effects of 1-MCP on Ethylene Synthesis and Metabolismof Apple During Storage显示文摘Red Fuji apple (Malus domestica Borkh var. Red Fuji) fruits were used to study the effect of 1-MCP on ethylene biosynthesis metabolism during storage. The results showed that 1-MCP maintained the firmness and inhibited the respiration rate, LOX activity and ethylene production rate of fruits. Further study indicated that 1-MCP inhibited ACS (ACC synthase) activity from the 15th day, increased ACC accumulation,and delayed the appearance of ACO (ACC oxidase) activity peak.
