| 1 | Active Fault Exploration and Seismic Hazard Assessment in Fuzhou City显示文摘It has been proven by a number of earthquake case studies that an active fault-induced earthquake beneath a city can be devastating. It is an urgent issue for seismic hazard reduction to explore the distribution of active faults beneath the urban area and identify the seismic source and the risks underneath. As a pilot project of active fault exploration in China, the project, entitled “Active fault exploration and seismic hazard assessment in Fuzhou City', started in early 2001 and passed the check before acceptance of China Earthquake Administration in August 2004. The project was aimed to solve a series of scientific issues such as fault location, dating, movement nature, deep settings, seismic risk and hazard, preparedness of earthquake prevention and disaster reduction, and etc. by means of exploration and assessment of active faults by stages, i.e., the preliminary survey and identification of active faults in target area, the exploration of deep seismotectonic settings, the risk evaluation of active seismogenic faults, the construction of geographic information system of active faults, and so on. A lot of exploration methods were employed in the project such as the detection of absorbed mercury, free mercury and radon in soil, the geological radar, multi-channel DC electrical method, tsansient electromagnetic method, shallow seismic refraction and reflection, effect contrast of explored sources, and various sounding experiments, to establish the buried Quaternary standard section of the Fuzhou basin. By summing up, the above explorations and experiments have achieved the following results and conclusions: (1)The results of the synthetic pilot project of active fault exploration in Fuzhou City demonstrate that, on the basis of sufficient collection, sorting out and analysis of geological, geophysical and borehole data, the best method for active fault exploration (location) and seismic risk assessment (dating and characterizing) in urban area is the combination of various approaches, that is, the possible location of active fault determined by using geochemical exploration as a guide “scout', the shallow seismic sounding as the main tool, the electromagnetic method as a supplement, establishing the standard Quaternary profile or stratigraphic sequence from drilling and various geophysical parameters from borehole logs as methods to correct and verify the data above. And in addition, the method also includes the field surveys on fault exposures, trenching, paleoearthquake investigation, dating and comparison of lithology, strata sequence, absolute or relative ages of the cores on the two sides of buried faults. (2)The Fuzhou basin locates under the regional seismotectonic settings which have the potential of moderate earthquake. Comparatively, the region is less affected by the “Taiwan dynamic Antenna'; (3)The activity of the major faults in Fuzhou basin is weak in general. All the six identified target faults are not Holocene faults, among which the Bayi Reservoir-Shanggan fault and the Minhou-Nanyu fault are dormant at least since the mid Epipleistocene time, and the rest are dormant since the Epipleistocene time; (4)In terms of deep-seated structures beneath the basin, there is no evidence indicating the possible occurrence of the underneath strong destructive earthquakes. | 2 | High expression of FABP4 in colorectal cancer and its clinical significance显示文摘Objective:To investigate the relationship between the fatty acid-binding protein 4(FABP4)and colorectal cancer(CRC).Methods:Using an enzyme-linked immunosorbent assay(ELISA),we measured the expression of FABP4 in plasma of50 patients who underwent surgery for CRC from October 2017 to May 2018 and 50 healthy controls.The content of the visceral fat area(VFA)as seen with abdominal computed tomography(CT)scanning was measured by ImageJ software.The expression levels of FABP4,E-cadherin,and Snail proteins in CRC and adjacent tissues were determined by immunohistochemistry.Results:The mean concentration of plasma FABP4 of CRC patients was higher than that of the control group(22.46 vs.9.82 ng/m L;P<0.05).The concentration of plasma FABP4 was related to the tumor,node,metastatis(TNM)stage and lymph node metastasis and was independent of age,body mass index(BMI),tumor size and location,and the degree of differentiation of CRC.The concentration of plasma FABP4 was positively correlated with high VFA and lipoprotein-a(LPA)(P<0.05);but it was not correlated with total cholesterol(TG),total triglyceride(TC),low-density lipoprotein(LDL),high-density lipoprotein(HDL),or apolipoprotein AI(Apo-AI).The expression of FABP4 protein in CRC tissues was positively correlated with the degree of CRC differentiation,tumor stage,and lymph node metastasis.The level of FABP4 protein was negatively correlated with E-cadherin protein(r=-0.3292,P=0.0196)and positively correlated with Snail protein(r=0.5856,P<0.0001).Conclusions:High LPA and VFA were risk factors for increased plasma FABP4 in CRC patients.FABP4 protein was highly expressed in CRC tissues and associated with TNM stage,differentiation,and lymph node metastasis of CRC.The level of FABP4 in CRC tissue was correlated with E-cadherin and Snail expression,suggesting that FABP4 may promote CRC progression related to epithelial-mesenchymal transition(EMT). | Yan ZHANG Wenjia ZHANG Min XIA Zhujun XIE Fangmei AN Qiang ZHAN Wenying TIAN Tianyue ZHU | 2021 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,2: | 5 |
| 3 | Expression Analysis of Genes Related to Auxin Metabolism at Different Growth Stages of Pak Choi显示文摘Pak choi is a low-temperature vernalized plant that readily undergoes premature bolting during spring, but little is known about the governing molecular regulation of vernalization. In order to enhance our understanding of mechanism about premature bolting, we discussed the relationship between auxin(indole-3-acetic acid, IAA) and flowering of pak choi. During vernalization, hormone metabolism is an important regulatory pathway, and IAA plays a specific role. IAA metabolism has been studied in Arabidopsis thaliana and other plants, but not in pak choi. In this paper, the IAA content in pak choi shoot apices during vernalization and different growth stages was compared. The IAA content decreased significantly after low-temperature treatment(4 °C) and then increased rapidly during vegetative growth. During floral bud initiation,the IAA content decreased rapidly and was the lowest. Expressions of genes encoding key IAA metabolic enzymes were analyzed, and a major synthetase-encoding gene was downregulated, while a key degrading enzyme-encoding gene was upregulated during each comparison period,resulting in decreased IAA content. Expressions of four genes(Bra034975, Bra030246, Bra012239 and Bra040296) were consistent with changes in the IAA content. | 4 | A Numerical Convex Lens for the State-Discretized Modeling and Simulation of Megawatt Power Electronics Systems as Generalized Hybrid Systems显示文摘Modeling and simulation have emerged as an indispensable approach to create numerical experiment platforms and study engineering systems.However,the increasingly complicated systems that engineers face today dramatically challenge state-of-the-art modeling and simulation approaches.Such complicated systems,which are composed of not only continuous states but also discrete events,and which contain complex dynamics across multiple timescales,are defined as generalized hybrid systems(GHSs)in this paper.As a representative GHS,megawatt power electronics(MPE)systems have been largely integrated into the modern power grid,but MPE simulation remains a bottleneck due to its unacceptable time cost and poor convergence.To address this challenge,this paper proposes the numerical convex lens approach to achieve state-discretized modeling and simulation of GHSs.This approach transforms conventional time-discretized passive simulations designed for pure-continuous systems into state-discretized selective simulations designed for GHSs.When this approach was applied to a largescale MPE-based renewable energy system,a 1000-fold increase in simulation speed was achieved,in comparison with existing software.Furthermore,the proposed approach uniquely enables the switching transient simulation of a largescale megawatt system with high accuracy,compared with experimental results,and with no convergence concerns.The numerical convex lens approach leads to the highly efficient simulation of intricate GHSs across multiple timescales,and thus significantly extends engineers’capability to study systems with numerical experiments. | Bochen Shi Zhengming Zhao Yicheng Zhu Zhujun Yu Jiahe Ju | 2021 | Engineering2021,7,12: | 3 |

| 14 | The effect of glutathione on glucosinolate biosynthesis through the sulfur assimilation pathway in pakchoi associated with the growth conditions显示文摘Glucosinolates(GSLs) are a group of nitrogen-and sulfur-containing secondary metabolites, synthesized primarily in members of the Brassicaceae family, that play an important role in food flavor, plant antimicrobial activity, resistance to insect attack, stress tolerance, and human anti-cancer effects. As a sulfur-containing compound, glutathione has a strong connection with GSLs biosynthesis as a sulfur donor or redox system, and exists in reduced(glutathione;GSH) and oxidized(glutathione disulfide;GSSG) forms. However, the mechanism of GSH regulating GSLs biosynthesis remainds unclear. Hence, the exogenous therapy to pakchoi under normal growth condition and sulfur deficiency condition were conducted in this work to explore the relevant mechanism. The results showed that exogenous application of buthionine sulfoximine, an inhibitor of GSH synthesis, decreased the transcript levels of GSLs synthesis-related genes and transcription factors, as well as sulfur assimilation-related genes under the normal growth condition. Application of exogenous GSH inhibited the expression of GSLs synthesis-and sulfur assimilation-related genes under the normal condition, while the GSLs biosynthesis and the sulfur assimilation pathway were activated by exogenous application of GSH when the content of GSH in vivo of plants decreased owing to sulfur deficiency. Moreover,exogenous application of GSSG increased the transcript levels of GSLs synthesis-and sulfur assimilation-related genes under the normal growth condition and under sulfur deficiency. | 15 | Impairment of rigidity sensing caused by mutant TP53 gain of function in osteosarcoma显示文摘Osteosarcoma(OS)is the most common primary malignant pediatric bone tumor and is characterized by high heterogeneity.Studies have revealed a wide range of phenotypic differences among OS cell lines in terms of their in vivo tumorigenicity and in vitro colony-forming abilities.However,the underlying molecular mechanism of these discrepancies remains unclear.The potential role of mechanotransduction in tumorigenicity is of particular interest.To this end,we tested the tumorigenicity and anoikis resistance of OS cell lines both in vitro and in vivo.We utilized a sphere culture model,a soft agar assay,and soft and rigid hydrogel surface culture models to investigate the function of rigidity sensing in the tumorigenicity of OS cells.Additionally,we quantified the expression of sensor proteins,including four kinases and seven cytoskeletal proteins,in OS cell lines.The upstream core transcription factors of rigidity-sensing proteins were further investigated.We detected anoikis resistance in transformed OS cells.The mechanosensing function of transformed OS cells was also impaired,with general downregulation of rigidity-sensing components.We identified toggling between normal and transformed growth based on the expression pattern of rigidity-sensing proteins in OS cells.We further uncovered a novel TP53 mutation(R156P)in transformed OS cells,which acquired gain of function to inhibit rigidity sensing,thus sustaining transformed growth.Our findings suggest a fundamental role of rigidity-sensing components in OS tumorigenicity as mechanotransduction elements through which cells can sense their physical microenvironment.In addition,the gain of function of mutant TP53 appears to serve as an executor for such malignant programs. | Ming Luo Mingyang Huang Ningning Yang Yufan Zhu Peng Huang Zhujun Xu Wengang Wang Lin Cai | 2023 | Bone Research2023,11,2: | 0 |

| 17 | Diversity of glucosinolates among common Brassicaceae vegetables in China显示文摘Brassicaceae vegetables are an important traditional daily food in China and around the world, which provide nutrients and phytochemicals that are beneficial for human health. Among them, Brassica and Raphanus are widely cultivated and eaten, have been evolved and/or bred for special characteristics during the long history of cultivation. Epidemiological studies suggest that the health benefits of Brassicaceae vegetables are mainly associated with glucosinolates(GSLs) and their hydrolytic products. In this review, we discuss the diversity of common consumed Brassicaceae vegetables and their GSL composition in edible parts. We also discuss the diversity factors affecting GSL content, and the diversity roles and functions of GSL.
