| 2 | Additive and Over-dominant Effects Resulting from Epistatic Loci Are the Primary Genetic Basis of Heterosis in Rice显示文摘A set of 148 F9 recombinant inbred lines(RILs) was developed from the cross of an indica cultivar 93-11 and japonica cultivar DT713,showing strong F1 heterosis.Subsequently,two backcross F1(BCF1) populations were constructed by backcrossing these 148 RILs to two parents,93-11 and DT713.These three related populations(281BCF1 lines,148 RILs) were phenotyped for six yield-related traits in two locations.Significant inbreeding depression was detected in the population of RILS and a high level of heterosis was observed in the two BCF1 populations.A total of 42 main-effect quantitative trait loci(M-QTLs) and 109 epistatic effect QTL pairs(E-QTLs) were detected in the three related populations using the mixed model approach.By comparing the genetic effects of these QTLs detected in the RILs,BCF1 performance and mid-parental heterosis(HMP),we found that,in both BCF1 populations,the QTLs detected could be classified into two predominant types:additive and over-dominant loci,which indicated that the additive and over-dominant effect were more important than complete or partially dominance for M-QTLs and E-QTLs.Further,we found that the E-QTLs detected collectively explained a larger portion of the total phenotypic variation than the M-QTLs in both RILs and BCF1 populations.All of these results suggest that additive and over-dominance resulting from epistatic loci might be the primary genetic basis of heterosis in rice. | Xiaojin Luo Yongcai Fu Peijiang Zhang Shuang Wu Feng Tian Jiayong Liu Zuofeng Zhu Jinshui Yang Chuanqing Sun | 2009 | Journal of Integrative Plant Biology2009,51,4: | 20 |
| 3 | Natural Variations at TIG1 Encoding a TCP Tran scription Factor Contribute to Plant Architecture Domestication in Rice显示文摘The modification of plant architecture is a crucial target in rice domestication and modern genetic improvement.Although several genes regulating rice plant architecture have been characterized,the molecular mechanisms underlying rice plant architecture domestication remain largely unclear.Here we show that the inclined tiller growth in wild rice is controlled by a single dominant gene,TILLER INCLINED GROWTH 1 (T/Gf),which is located on chromosome 8 and encodes a TCP transcriptional activator.TIG1 is primarily expressed in the adaxial side of the tiller base,promotes cell elongation,and enlarges the tiller angle in wild rice.Variations in the TIG1 promoter of indica cultivars {tig1 allele) resulted in decreased expression of TIG1 in the adaxial side of tiller base and reduced cell length and tiller angle,leading to the transition from inclined tiller growth in wild rice to erect tiller growth during rice domestication.TIG1 positively regulates the expression of EXP A3,EXPB5,and SAUR39 to promote cell elongation and increase the tiller angle.Selective sweep analysis revealed that the tig1 allele was selected in indica cultivars by human beings.The cloning and characterization of TIG1 supports a new scenario of plant architecture evolution in rice. | Weifeng Zhang Lubin Tan Hongying Sun Xinhui Zhao Fengxia Liu Hongwei Cai Yongcai Fu Xianyou Sun Ping Gu Zuofeng Zhu Chuanqing Sun | 2019 | Molecular Plant2019,12,8: | 13 |
| 4 | Association study of the single nucleotide polymorphisms in adiponectin-associated genes with type 2 diabetes in Han Chinese显示文摘Single-nucleotide polymorphisms (SNPs) of ADIPOQ, ADIPOR1, and ADIPOR2 have been associated with type 2 diabetes mellitus (T2DM), but there are many conflicting results especially in Chinese populations. To investigate the contribution of the adiponectin genes and their receptors to T2DM, a case-control study was performed and 11 SNPs of ADIPOQ, ADIPOR1, and ADIPOR2 were genotyped in 985 T2DM and 1,050 control subjects. rs16861194 (-11426 A>G) in the putative promoter of ADIPOQ was associated with T2DM (P = 0.007; OR = 1.29, 95% CI 1.08-1.55). None of the other 10 SNPs were associated with T2DM in this study, although rs2241766 and rs1501299 were reported to be associated with T2DM in previous Chinese studies. There was also no significant difference found from the ADIPOQ haplotype analysis, which contains rs16861194. In addition, we also assessed potential gene-gene interactions in three genes and no interactions were found. | 5 | Analysis of QTLs for yield-related traits in Yuanjiang common wild rice (Oryza rufipogon Griff.)显示文摘Using an accession of common wild rice (Oryza rufipogon Griff.) collected from Yuanjiang County, Yunnan Province, China, as the donor and an elite cultivar 93-11, widely used in two-line indica hybrid rice production in China, as the recurrent parent, an advanced backcross populations were developed. Through genotyping of 187 SSR markers and investigation of six yield-related traits of two gen- erations (BC4F2 and BC4F4), a total of 26 QTLs were detected by employing single point analysis and interval mapping in both genera-tions. Of the 26 QTLs, the alleles of 10 (38.5%) QTLs originating from O. rufipogon had shown a beneficial effect for yield-related traits in the 93-11 genetic background. In addition, five QTLs controlling yield and its components were newly identified, indicating that there are potentially novel alleles in Yuanjiang common wild rice. Three regions underling significant QTLs for several yield-related traits were detected on chromosome 1, 7 and 12. The QTL clusters were founded and corresponding agronomic traits of those QTLs showed highly significant correlation, suggesting the pleiotropism or tight linkage. | 6 | 乳腺导管内增生性病变及合并浸润性导管癌的临床病理分析(英文)显示文摘Objective:The purpose of this study was to study the clinical and pathological characteristics of breast intraductal proliferative lesions(IDPLs) and associated with invasive breast cancer.Methods:We reviewed 327 cases of breast intraductal proliferative lesions including 53 cases of usual ductal hyperplasia,57 cases of atypical ductal hyperplasia,89 cases of ductal carcinoma in situ,and 128 cases coexist with invasive ductal carcinomas.Cases of pure invasive cancer without intraductal proliferative lesions were excluded.The mult IDPLs biological parameters including the express of ER,PR,HER2,HIF-1α and Ki-67 detected by immunohistochemistry S-P method(n=327) and the levels of CA153,TSGF,CA125 and CEA both in nipple discharge and serum(n=179) measured with Electrochemiluminescence method and their relationship were studied,and 30 cases of normal pregnant women were compared with.Results:A single histologic subtype was present in 49.85%(163/327) of the cases,two subtypes in 33.03%(108/327),and three in 17.13%(56/327).The most common subtypes present were cribriform(43.12%,141/327) and solid(38.53%,126/327),while the comedo(16.35%,54/327),and micropapillary(12.84%,42/327) subtypes were less common.Comedo and solid were frequently found together for coexpression as were micropapillary and papillary subtypes.However,Comedo subtype was much less likely to be found with papillary,cribriform or micropapillary subtypes.Additionally,comedo subtypes tend to be hormone receptor negative,Her2 positive and high-grade while the cribriform and solid subtype tends to be hormone receptor positive,Her2 negative and low grade.Papillary subtype was least likely to be associated with an invasive cancer.Furthermore,the nipple discharge and serum levels of CA153,TSGF,CA125 and CEA in coexist with invasive ductal carcinomas patients were significantly higher than those in the benign breast disease(pure intraductal proliferative lesions) and normal pregnant women(P<0.01).Additionally,the levels of CA153,TSGF,CA125 and CEA in nipple discharge were significantly higher than in the serum(P<0.01),and had a positive correlation with the Ki-67,grade,clinical stage,lymph node metastasis and tumor recurrence(P<0.05),and negative correlation with the level of ER and PR(P<0.05).The sensitivity of the four serum tumor markers in combination was only 69.77%,in contrast,the combined detection both in discharge and serum was 97.67%,and the negative predictive value was 99.03%.The sensitivity of combined detection both in nipple discharge and serum were significantly higher than other detection(P<0.05).Conclusion:IDPLs often present more than one histologic subtype and the most common subtypes are cribriform and solid,while comedo and micropapillary subtypes are less common.Our results suggest that the levels of CA153,TSGF,CA125 and CEA in nipple discharge were significantly higher than those in the serum,and is associated with HIF-1α.The aberration of HIF-1α may play a key role during oncogenesis and promote breast cellular transformation into malignancy,a finding useful for further understanding of tumorigenesis.Nipple discharge can be the earliest presenting symptom of breast cancer.The dynamic combined detection of the four tumor markers both in nipple discharge and serum are helpful to the stratification of preoperative patients and benefit to better prewarning markers for monitoring their recurrence and metastasis and clinical staging of tumors in clinic,but cannot increase the sensitivity of judging the patients with early breast cancer. | Gangping Wang Hong Zhang Zuofeng Zhang Yun'ai Liang Ying Chen Lan Mei | 2013 | The Chinese-German Journal of Clinical Oncology2013,12,12: | 4 |

| 8 | Vascular endothelial growth factor expression and significance in different grades of the Breast Imaging Reporting and Data System显示文摘Objective Mammography is the only modality proven to reduce mortality in breast cancer,and ultrasonography is a well-known adjunct to mammography screening.The Breast Imaging Reporting and Data System(BI-RADS) classification is a practical tool and is correlated with histopathology and combined use with triple assessment(examination,imaging,and biopsy) of palpable diagnostic cases.This study aimed to investigate the relationship between vascular endothelial growth factor(VEGF) expression and different grades of BI-RADS in breast cancer.Methods Ninety-six patients with breast carcinoma were evaluated using BI-RADS by ultrasonography,mammography,and a combination of both modalities.In the combined imaging assessment,BI-RADS 1-4a grade was considered when the score of ultrasonography and mammography was lower than 4a,and BI-RADS 4b-5a grade was considered when the score of ultrasonography and mammography was higher than 4a.Immunohistochemical Ultra SensitiveTM S-P method was employed to evaluate the expression of VEGF in 96 patients.Fifty patients with benign breast disease were selected as the control group.The relationship between VEGF expression and different grades of BI-RADS and that between VEGF expression and other standard prognostic parameters associated with invasive breast cancer,such as size,grade,cancer stage,and metastasis were analyzed.Results The sensitivities of ultrasonography and mammography alone was 74.0% and 84.4%,respectively;However,the sensitivity of their combination increased to 90.6%.The positive rates of VEGF in invasive breast cancer BI-RADS 4b-5(59/87,67.8%) were higher than those in BI-RADS 4a(3/9,33.3%,P<0.05) and benign breast disease tissues(BI-RADS 1-4a,11/50,22.0%)(P<0.05).There was a positive correlation between VEGF overexpression and BI-RADS 4b-5,histological grade(Ⅲ),lymph node metastasis,and distant metastasis of invasive breast cancer.VEGF expression was not related to the age and size of the tumor in each group(P>0.05).Conclusion There was a positive correlation between VEGF overexpression and BI-RADS 4b-5 grade.The overexpression of VEGF might be an important biological marker for the invasion and metastasis of breast cancer. | Gangping Wang Jingjing Li Shuguang Yang Zuofeng Zhang Songjing Lei | 2019 | Oncology and Translational Medicine2019,5,6: | 1 |

| 10 | E-cadherin、β-catenin和E-cadherin-catenins复合体在乳腺癌及癌前病变中的表达及意义(英文)显示文摘Objective: The aim of our study was to observe the expressions and clinical Significance of E-cadherin, β-catenin and E-cadherin-catenins complex in breast cancer and precancerous lesions, and analyze the relationship between the expressions and clinicopathological features in breast cancer. Methods: Immunhistochemical UltraSensitiveTM S-P method was employed to detect the expression of E-cadherin, β-catenin and E-cadherin-catenins complex in 128 cases of invasive ductal carcinomas, 89 cases of ductal carcinoma in situ and 57 cases of atypical ductal hyperplasia, 53 cases of usual ductal hyperplasia breast tissues were selected as a control group. The express of E-cadherin, β-catenin and their relationship with mult biological parameters including histological grade, region lymph node metastasis, distant metastasis and recurrence on files were also assessed. Results:(1) The staining patterns character of E-cadherin, β-catenin and E-cadherin-catenins complex: In UDH breast tissues, E-cadherin and a-catenin were expressed on cell membrane of ductal and acinic cells, showing cellular contour and border among cells. The abnormal expression of the three proteins occurred in breast invasive ductal carcinomas, ductal carcinoma in situ and atypical ductal hyperplasia tissues, showing cytoplasmic or nuclear staining, decrease and loss of cytomembrane staining.(2) The abnormal expression rates of E-cadherin, β-catenin and E-cadherin-catenins complex in invasive ductal carcinomas were 53.91%, 65.63% and 81.25%, which were significantly higher than that in ductal carcinoma in situ, atypical ductal hyperplasia, usual ductal hyperplasia tissues(P < 0.01). Compared with usual ductal hyperplasia breast tissues group, the abnormal expression rates of E-cadherin, β-catenin and E-cadherin-catenins complex were significantly decreased(P < 0.01) in the breast cancer group. However, there was no significance of the abnormal expression rate between ductal carcinoma in situ and atypical ductal hyperplasia tissues groups(χ2 = 0.76, P = 0.38; χ2 = 0.14, P = 0.70; χ2 = 0.81, P = 0.37; χ2 = 2. 19, P = 0.14)(P > 0.05).(3) There was a significantly difference in the mean E-cadherin, β-catenin and Ecadherin-catenins complex frequency between estrogen receptor & progesterone receptor positive IDC group and negative group, epidermal growth factor receptor type 2(HER2/neu) positive and negative groups, Ki-67 proliferation index ≤ 14% and > 14% groups, histological grade(I + II) and grade III invasive ductal carcinomas groups, with lymph node metastasis, distant metastasis and recurrence groups(P < 0.05) and without groups(P < 0.05). However, there was no difference in the mean E-cadherin, β-catenin and E-cadherin-catenins complex frequency between age(≤ 50 years vs > 50 years), tumor diameter(≤ 2 cm vs > 2 cm)(P > 0. 05). Conclusion: In breast cancer, the expressions of E-cadherin, β-catenin and E-cadherin-catenins complex are abnormally decreased and are correlated with pathology grade, differentiation disturbance and metastasis. Ecadherin and β-catenin may be as the predictors for prognosis. | 13 | Comparison of time trends in the incidence of primary liver cancer between China and the United States: an age-period-cohort analysis of the Global Burden of Disease 2019显示文摘Background::China and the United States(US)ranked first and third in terms of new liver cancer cases and deaths globally in 2020.Therefore,a comprehensive assessment of trends in the incidence of primary liver cancer with four major etiological factors between China and the US during the past 30 years with age-period-cohort(APC)analyses is warranted.Methods::Data were obtained from the Global Burden of Disease 2019,and period/cohort relative risks were estimated by APC modeling from 1990 to 2019.Results::In 2019,there were 211,000 new liver cancer cases in China and 28,000 in the US,accounting for 39.4%and 5.2%of global liver cancer cases,respectively.For China,the age-standardized incidence rate(ASIR)consecutively decreased before 2005 but increased slightly since then,whereas the ASIR continuously increased in the US.Among the four etiological factors of liver cancer,the fastest reduction in incidence was observed in hepatitis B virus-related liver cancer among Chinese women,and the fastest increase was in nonalcoholic steatosis hepatitis(NASH)-related liver cancer among American men.The greatest reduction in the incidence of liver cancer was observed at the age of 53 years in Chinese men(-5.2%/year)and 33 years in Chinese women(-6.6%/year),while it peaked at 58 years old in both American men and women(4.5%/year vs.2.8%/year).Furthermore,the period risks of alcohol-and NASH-related liver cancer among Chinese men have been elevated since 2013.Simultaneously,leveled-off period risks were observed in hepatitis C viral-related liver cancer in both American men and women.Conclusions::Currently,both viral and lifestyle factors have been and will continue to play an important role in the time trends of liver cancer in both countries.More tailored and efficient preventive strategies should be designed to target both viral and lifestyle factors to prevent and control liver cancer. | Zhiyong Zou Zuofeng Zhang Ce Lu Hui Wang | 2022 | Chinese Medical Journal2022,135,17: | 1 |
