1The impacts of conservation agriculture on crop yield in China depend on specific practices, crops and cropping regions显示文摘For smooth and wide application of conservation agriculture(CA), remaining uncertainties about its impacts on crop yield need to be reduced. Based on previous field experiments in China, a meta-analysis was performed to quantify the actual impacts of CA practices(NT: no/reduced-tillage only, CTSR: conventional tillage with straw retention, NTSR: NT with straw retention) on crop yields as compared to conventional tillage without straw retention(CT).Although CA practices increased crop yield by 4.6% on average, there were large variations in their impacts. For each CA practice, CTSR and NTSR significantly increased crop yield by 4.9%and 6.3%, respectively, compared to CT. However, no significant effect was found for NT. Among ecological areas, significant positive effects of CA practices were found in areas with an annual precipitation below 600 mm. Similar effects were found in areas with annual mean air temperature above 5 °C. For cropping regions, CA increased crop yield by 6.4% and 5.5%compared to CT in Northwest and South China, respectively, whereas no significant effects were found in the North China and Northeast China regions. Among crops, the positive effects of CA practices were significantly higher in maize(7.5%) and rice(4.1%) than in wheat(2.9%). NT likely decreased wheat yield. Our results indicate that there are great differences in the impacts of CA practices on crop yield, owing to regional variation in climate and crop types. CA will most likely increase maize yield but reduce wheat yield. 4Effect of emodin on Aquaporin 5 expression in rats with sepsis-induced acute lung injury显示文摘OBJECTIVE: To investigate the effects of emodin on aquaporin 5(AQP5) expression in rats with sepsis-induced acute lung injury.METHODS: We divided 60 adult male Sprague-Dawley rats, weighing 200-230 g, into four groups: control, sham surgery, model and emodin groups(n = 15 for each). We created a sepsis model with cecal ligation and puncture; the sham surgery group had their cecums replaced after exposure outside the abdominal cavity. Each group was further divided into three subgroups(n = 5 for each)and expressions of AQP5 m RNA and proteins in lung tissue were measured by real-time fluorescence polymerase chain reaction and western blot at 6, 12 and 24 h after surgery.RESULTS: AQP5 expression did not change over time in the control group and sham surgery group,but decreased over time in the model group. The lowest expression was found in 12-h subgroup,which significantly differed from the 6-h subgroup(P < 0.01). Compared with the model group, AQP5 expression in the emodin group was significantly higher in all the subgroups(all P <0.01). Expressions in the 12-h subgroup were the highest, and significantly differed from the other subgroups. We found that lung tissue damage,such as pulmonary edema, alveolar damage and the exudation of red blood cells in pulmonary interstitium and alveolar, was significantly milder in the emodin group under light microscope than the model group.CONCLUSION: AQP5 expression was significantly down-regulated in rats with sepsis-induced acute lung injury induced by cecal ligation and puncture.Early prophylactic use of emodin can significantly enhance the AQP5 expression, thus effectively reducing the degree of pulmonary edema in septic rats.Sun Yanni Sun Lijing Liu Shuqing Song Juan Cheng Jiejing Liu Jun 2015Journal of Traditional Chinese Medicine2015,35,6:5

7POST1/C12ORF49 regulates the SREBP pathway by promoting site-1 protease maturation显示文摘Sterol-regulatory element binding proteins(SREBPs)are the key transcriptional regulators of lipid metabolism.The activation of SREBP requires translocation of the SREBP precursor from the endoplasmic reticulum to the Golgi,where it is sequentially cleaved by site-1 protease(S1P)and site-2 protease and releases a nuclear form to modulate gene expression.To search for new genes regulating cholesterol metabolism,we perform a genome-wide CRISPR/Cas9 knockout screen and find that partner of site-1 protease(POSH),encoded by C120RF49,is critically involved in the SREBP signaling.Ablation of POSH decreases the generation of nuclear SREBP and reduces the expression of SREBP target genes.POSH binds S1P,which is synthesized as an inactive protease(form A)and becomes fully mature via a two-step autocatalytic process involving forms B/B and C/C.POSH promotes the generation of the functional S1P-C/C from S1P-B/B(canonical cleavage)and,notably,from S1P-A directly(non-canonical cleavage)as well.This POSH-mediated S1P activation is also essential for the cleavages of other S1P substrates including ATF6,CREB3 family members and the a/p-subunit precursor of N-acetylglucosamine-1-phospho-transferase.Together,we demonstrate that POSH is a cofactor controlling S1P maturation and plays important roles in lipid homeostasis,unfolded protein response,lipoprotein metabolism and lysosome biogenesis.Jian Xiao Yanni Xiong Liu-Ting Yang Ju-Qiong Wang Zi-Mu Zhou Le-Wei Dong Xiong-Jie Shi Xiaolu Zhao Jie Luo Bao-Liang Song 2021Protein & Cell2021,12,4:3

10Amelioration of imiquimod-induced psoriasis-like dermatitis in mice by DSW therapy inspired hydrogel显示文摘Psoriasis is a long-lasting and recurrent autoimmune disease which is incurable so far.Dead Sea water(DSW)therapy is an effective approach to help control the symptoms of psoriasis due to the abundant mineral ions in DSW,which inspired the material formulation in this study.Rubidium–Sodium alginate/Polyacrylamide hydrogels(Rb-SA/PAAm gels)composed of sodium alginate and polyacrylamide interpenetrating network structure with different concentrations of rubidium and certain magnesium and zinc content were prepared for the treatment of psoriasis.The obtained results suggest the good mechanical properties of the Rb-SA/PAAm gels including toughness and swelling performance.In terms of in vitro tests,the Rb-SA/PAAm gels not only show nontoxicity to human keratinocyte cell line(Hacats)but also inhibits the activity against inflammatory NF-κβ signaling pathway.Meanwhile,they can release Rb+which enable the Rb-SA/PAAm gels have better antibacterial ability to Streptococcus and Escherichia coli.The results obtained from in vivo tests indicate that these hydrogels could alleviate the symptoms of psoriasis caused by Imiquimod(IMQ)in mice by reducing the inflammatory factor in STAT3 pathway and therefore reduce the immune stimulation of the spleen.In conclusion,the 100Rb-SA/PAAm gel has demonstrated great potential to be a topical wettable dressing for psoriasis treatment.Xiang He Bing Zhu WeiJia Xie Yu He Jian Song Yi Zhang Chi Sun Hao Li QiYu Tang XinXin Sun Yanni Tan Yong Liu 2021Bioactive Materials2021,6,2:2
11Risk stratification in symptomatic intracranial atherosclerotic disease with conventional vascular risk factors and cerebral haemodynamics显示文摘Background and purpose Symptomatic intracranial atherosclerotic stenosis(sICAS)is associated with a considerable risk of recurrent stroke despite contemporarily optimal medical treatment.Severity of luminal stenosis in sICAS and its haemodynamic significance quantified with computational fluid dynamics(CFD)models were associated with the risk of stroke recurrence.We aimed to develop and compare stroke risk prediction nomograms in sICAS,based on vascular risk factors and these metrics.Methods Patients with 50%-99%sICAS confirmed in CT angiography(CTA)were enrolled.Conventional vascular risk factors were collected.Severity of luminal stenosis in sICAS was dichotomised as moderate(50%-69%)and severe(70%-99%).Translesional pressure ratio(PR)and wall shear stress ratio(WSSR)were quantified via CTA-based CFD modelling;the haemodynamic status of sICAS was classified as normal(normal PR&WSSR),intermediate(otherwise)and abnormal(abnormal PR&WSSR).All patients received guideline-recommended medical treatment.We developed and compared performance of nomograms composed of these variables and independent predictors identified in multivariate logistic regression,in predicting the primary outcome,recurrent ischaemic stroke in the same territory(SIT)within 1 year.Results Among 245 sICAS patients,20(8.2%)had SIT.The D2H2A nomogram,incorporating diabetes,dyslipidaemia,haemodynamic status of sICAS,hypertension and age≥50 years,showed good calibration(P for Hosmer-Lemeshow test=0.560)and discrimination(C-statistic 0.73,95%CI 0.60 to 0.85).It also had better performance in risk reclassification and provided larger net benefits in decision curve analysis,compared with nomograms composed of conventional vascular risk factors only,and plus the severity of luminal stenosis in sICAS.Sensitivity analysis in patients with anterior-circulation sICAS showed similar results.Conclusions The D2H2A nomogram,incorporating conventional vascular risk factors and the haemodynamic significance of sICAS as assessed in CFD models,could be a useful tool to stratify sICAS patients for the risk of recurrent stroke under contemporarily optimal medical treatment.Xuan Tian Hui Fang Linfang Lan Hing Lung Ip Jill Abrigo Haipeng Liu Lina Zheng Florence S Y Fan Sze Ho Ma Bonaventure Ip Bo Song Yuming Xu Jingwei Li Bing Zhang Yun Xu Yannie O Y Soo Vincent Mok Ka Sing Wong Thomas W Leung Xinyi Leng 2023Stroke & Vascular Neurology2023,8,1:2

17Hybridoma-derived neutralizing monoclonal antibodies against Beta and Delta variants of SARS-CoV-2 in vivo显示文摘Neutralizing monoclonal antibodies(mAb)are a major therapeutic strategy for the treatment of severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)infection.The continuous emergence of new SARS-CoV-2 variants worldwide has increased the urgency for the development of new mAbs.In this study,we immunized mice with the receptor-binding domain(RBD)of the SARS-CoV-2 prototypic strain(WIV04)and screened 35 RBDspecific mAbs using hybridoma technology.Results of the plaque reduction neutralization test showed that 25 of the mAbs neutralized authentic WIV04 strain infection.The 25 mAbs were divided into three categories based on the competitive enzyme-linked immunosorbent assay results.A representative mAb was selected from each category(RD4,RD10,and RD14)to determine the binding kinetics and median inhibitory concentration(IC_(50))of WIV04 and two variants of concern(VOC):B.1.351(Beta)and B.1.617.2(Delta).RD4 neutralized the B.1.617.2 variant with an IC50 of 2.67 ng/mL;however,it completely lost neutralizing activity against the B.1.351 variant.RD10 neutralized both variants with an IC50 exceeding 100 ng/mL;whereas RD14 neutralized two variants with a higher IC50(>1 mg/mL).Animal experiments were performed to evaluate the protective effects of RD4 and RD10 against various VOC infections.RD4 could protect Adv-hACE2 transduced mice from B.1.617.2 infection at an antibody concentration of 25 mg/kg,while RD10 could protect mice from B.1.351 infection at an antibody concentration of 75 mg/kg.These results highlight the potential for future modifications of the mAbs for practical use.Qianran Wang Lu Peng Yanqiu Nie Yanni Shu Huajun Zhang Zidan Song Yufeng Li Hengrui Hu Liushuai Li Xi Wang Jia Liu Jiang Li Zhengli Shi Fei Deng Yu Guo Yiwu Zhou Bing Yan Zhihong Hu Manli Wang 2023Virologica Sinica2023,38,2:0
18Quorum sensing-based interactions among drugs,microbes,and diseases显示文摘Many diseases and health conditions are closely related to various microbes,which participate in complex interactions with diverse drugs;nonetheless,the detailed targets of such drugs remain to be elucidated.Many existing studies have reported causal associations among drugs,gut microbes,or diseases,calling for a workflow to reveal their intricate interactions.In this study,we developed a systematic workflow comprising three modules to construct a Quorum Sensing-based Drug-Microbe-Disease(QSDMD)database(http://www.qsdmd.lbci.net/),which includes diverse interactions for more than 8,000 drugs,163 microbes,and 42 common diseases.Potential interactions between microbes and more than 8,000 drugs have been systematically studied by targeting microbial QS receptors combined with a docking-based virtual screening technique and in vitro experimental validations.Furthermore,we have constructed a QS-based drug-receptor interaction network,proposed a systematic framework including various drug-receptor-microbe-disease connections,and mapped a paradigmatic circular interaction network based on the QS-DMD,which can provide the underlying QS-based mechanisms for the reported causal associations.The QS-DMD will promote an understanding of personalized medicine and the development of potential therapies for diverse diseases.This work contributes to a paradigm for the construction of a molecule-receptor-microbe-disease interaction network for human health that may form one of the key knowledge maps of precision medicine in the future.Shengbo Wu Shujuan Yang Manman Wang Nan Song Jie Feng Hao Wu Aidong Yang Chunjiang Liu Yanni Li Fei Guo Jianjun Qiao 2023Science China(Life Sciences)2023,66,1:0

20Analysis of a Strong Sandstorm Process in Ulanqab City显示文摘Based on NCEP reanalysis data,EDAS data and ground observation data,the causes of a strong sandstorm process in Ulanqab City from March 14 to 15,2021 were analyzed.The analysis shows that'3.15'sandstorm,a sandstorm process accompanied by northwest gale behind the front,was caused by strong cold air in the north.In the early stage,the Mongolian cyclone resulted in the increase of temperature and decrease of pressure on the ground.When a strong surface cold high pressure at the rear of the cyclone invaded the city,a strong pressure gradient between high and low pressure provided dynamic conditions for the sandstorm outbreak.The transit of the surface cold front was the sign of the sandstorm outbreak,and the downward transmission of momentum at high altitudes increased the wind speed near the surface.Biyun LI Yu XUE Yanni SONG 2024Meteorological and Environmental Research2024,15,1:0
