2A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 3Iron-doped Mn-Ce/TiO_2 catalyst for low temperature selective catalytic reduction of NO with NH_3显示文摘The catalysts of iron-doped Mn-Ce/TiO 2(Fe-Mn-Ce/TiO 2) prepared by sol-gel method were investigated for low temperature selective catalytic reduction(SCR) of NO with NH 3.It was found that the NO conversion over Fe-Mn-Ce/TiO 2 was obviously improved after iron doping compared with that over Mn-Ce/TiO 2.Fe-Mn-Ce/TiO 2 with the molar ratio of Fe/Ti = 0.1 exhibited the highest activity.The results showed that 96.8% NO conversion was obtained over Fe(0.1)-Mn-Ce/TiO 2 at 180°C at a space velocity of 50,000 hr 1.Fe-Mn-Ce/TiO 2 exhibited much higher resistance to H 2 O and SO 2 than that of Mn-Ce/TiO 2.The properties of the catalysts were characterized using X-ray diffraction(XRD),N 2 adsorption,temperature programmed desorption(NH 3-TPD and NOx-TPD),and Xray photoelectron spectroscopy(XPS) techniques.BET,NH3-TPD and NOx-TPD results showed that the specific surface area and NH3 and NOx adsorption capacity of the catalysts increased with iron doping.It was known from XPS analysis that iron valence state on the surface of the catalysts were in Fe3+ state.The doping of iron enhanced the dispersion and oxidation state of Mn and Ce on the surface of the catalysts.The oxygen concentrations on the surface of the catalysts were found to increase after iron doping.Fe-Mn-Ce/TiO2 represented a promising catalyst for low temperature SCR of NO with NH3 in the presence of H2 O and SO2.Boxiong Shen ,Ting Liu,Ning Zhao,Xiaoyan Yang,Lidan Deng College of Environmental Science and Engineering,Nankai University,Tianjin 300071,China. 2010Journal of Environmental Sciences2010,22,9:63
4Re-Os isotopes and PGE geochemistry of black shales and intercalated Ni-Mo polymetallic sulfide bed from the Lower Cambrian Niutitang Formation, South China显示文摘The Lower Cambrian Niutitang Formation consists of a thick black shale sequence with a regionally distributed conformable Ni Mo polymetallic sulfide horizon and a chert bed at its basal strata on theYangtze Platform, South China. In this paper, we discuss all available data on Re Os isotopes and Platinum Group Element (PGE) distribution pattern of the Ni Mo polymetallic sulfide ore and its host rocks (black shales, cherts, and phosphorites) from Guizhou and Hunan provinces. Our results show that the black shales and the Ni Mo sulfide ore have a high initial 187 Os/ 188 Os ratio of 0.78~0.86, indicating that the Early Cambrian ocean across the Yangtze Platform had a highly radiogenic Os value. This ratio is slightly lower than but still similar to present day seawater, possibly as a result of high continental weathering at that time. The Ni Mo sulfide ore yields a Re Os isochron of 537±10 Ma (MSWD=11.9), possibly representing the depositional age of the Niutitang Formation. The chondrite normalized PGE pattern, Pt anomaly (Pt/Pt *), Pt/Pd, Ir/Pd, Au/Ir and Re/Mo ratios of the Ni Mo sulfide ore and its host rocks from South China indicate a varying source contribution of the PGE and other metals for different rocks. 5Oncofetal antigen glypican-3 as a promising early diagnostic marker for hepatocellular carcinoma显示文摘BACKGROUND:Hepatocellular carcinoma (HCC) is characterized by a multi-cause,multi-stage and multi-focus process of tumor progression.Its prognosis is poor and early diagnosis is of utmost importance.This study was undertaken to investigate the dynamic expression of oncofetal antigen glypican-3 (GPC-3) and GPC-3 mRNA in hepatocarcinogenesis and to explore their early diagnostic value for HCC.METHODS:A hepatoma model was induced in male Sprague-Dawley rats with 0.05% 2-fluorenylacetamide and confirmed by hematoxylin and eosin staining and gamma-glutamyltransferase (GGT) expression.Total RNA was purified and transcribed into cDNA by reverse transcription.Fragments of the GPC-3 gene were amplified by nested RT-PCR,and confirmed by sequencing.GPC-3 was analyzed by immunohistochemistry,Western blotting or ELISA.RESULTS:Positive GPC-3 expression showed as brown granule-like staining localized in the cytoplasm.Histological examination of hepatocytes revealed three morphological stages of granule-like degeneration,atypical hyperplasia (precancerous),and cancer formation,with a progressive increase of liver total RNA and GGT expression.The incidence of liver GPC-3 mRNA and GPC-3,and serum GPC-3 was 100%,100% and 77.8% in the HCC group,100%,100%,and 66.7% in the precancerous group,83.3%,83.3%,and 38.9% in the degeneration group,and no expression in the liver or blood of the control group,respectively.There was a positive correlation between liver GPC-3 mRNA and total RNA level (r=0.475,P<0.05) or liver GPC-3 (r=1.0,P<0.001) or serum GPC-3 (r= 0.994,P<0.001).CONCLUSION:Abnormal oncofetal antigen GPC-3 and GPC-3 mRNA expression in hepatocarcinogenesis may be promising molecular markers for early diagnosis of HCC.Research Center of Clinical Medicine,Affiliated Hospital of Nantong University,Nantong 226001,China (Yao M,Yao DF,Qiu LW,Wu W,Sai WL,Yang JL and Zhang HJ) Department of Oncology,Yancheng First People’s Hospital,Yancheng 224001,China (Bian YZ) Department of Oncology,Second Affiliated Hospital,Nanjing Medical University,Nanjing 210011,China (Zhang CG) 2011Hepatobiliary & Pancreatic Diseases International2011,10,3:51
6Get effective polyclonal antisera in one month显示文摘According to the traditional immunization procedure, after the first injection of the sample A (emulsion of aimed antigen and Freund's complete adjuvant) to immunize rabbit, successive injections of the sample B (emulsion of aimed antigen and Freund's incomplete adjuvant) were followed every 2-4 weeks. In general,high titer of the corresponding polyclonal antisera will be observed after 4-5 injections of sample B in 3-4months. This report presents a simply modified procedure that was able to stimulate the antisera formation in one month and achieve enough avidity to satisfy either Western blot or immunohistochemistry analysis.It just applied an additional injection of the sample A to the rabbit at the 3rd day after the primary immunization injection. You could gain the high titer of the antisera right after the first sample B injection in one month. 9The deformation pattern and fault rate in the Tianshan Mountains inferred from GPS observations显示文摘Based on GPS measurements conducted from 1992 to 2006, we present the current crustal movement velocity field for approximately 400 sites in the Tianshan Mountains and their adjacent areas, and estimate slip rates on the major faults using a 2-D elastic dislocation model. Our studies show slip rates within the range of 1―4 mm/a on the NW-SE trending strike-slip faults (such as Talas-Fergana fault) in the Tianshan Mountains. We also found the slip rates on the approximately WE-SN trending gently-dipping detachment fault vary from 10―13 mm/a for the southwest Tianshan Mountains to 2―5 mm/a for the eastern Tianshan Mountains, and to 6―12 mm/a for the Kyrgrz Tianshan. The GPS velocity field reveals that the total convergence is not uniformly distributed across the Tianshan Mountains, with 80%―90% of the N-S shortening absorbed along the southern and northern edges, and relatively little deformation accommodated within the interior. This first-order feature of strain pattern is explained best by underthrusting of adjacent blocks beneath the Tianshan Mountains along a basal detachment fault. We found the occurrence of historical M7―8 earthquakes somewhere in the locked ramp that connects the creeping and locking segments of the detachment, thereby resulting in elastic strain concentration and accumulation around it. The elastic strain confined in the upper crustal layer above the detachment ultimately releases through infrequent great earthquakes in the Tianshan Mountains, resulting in considerable folding and faulting at their margins. 11Ultrahigh pressure (>7 GPa) gneissic K-feldspar (-bearing) garnet clinopyroxenite in the Altyn Tagh, NW China: Evi- dence from clinopyroxene exsolution in garnet显示文摘The exsolution of clinopyroxene and rutile in coarse-grain garnet is found in the gneissic K-feldspar(-bearing) garnet clinopyroxenite from Yinggelisayi in the Altyn Tagh, NW China. The maximum content of the exsolved clinopyroxene in the garnet is up to >5% by volume. The reconstructed precursor garnet (Grt1) before exsolution has a maximum Si content of 3.061 per formula uint, being of supersilicic or majoritic garnet. The peak-stage metamorphic pressure of >7 GPa is estimated using the geobarometer for volume percentage of exsolved pyroxene in garnet and the Si-(Al+Cr) geobarometer for majoritic garnet, and the temperature of about 1000℃ using the ternary alkali-feldspar geothermometer and the experimental data of ilmen- ite-magnetite solid solution. The protoliths of the rocks are intra-plate basic and intermediate ig- neous rocks, of which the geochemical features indicate that they are probably the products of the evolution of basic magma deriving from the continental lithosphere mantle. The rocks are in outcrops associated with ultrahigh pressure garnet-bearing lherzolite and ultrahigh pressure garnet granitoid gneiss. All of these data suggest that the ultrahigh pressure metamorphic rocks in the Altyn Tagh are the products of deep-subduction of the continental crust, and such deep- subduction probably reaches to >200 km in depth. 12Ages and Compositions of the Precambrian High-grade Basement of the Qilian Terrane and Its Adjacent Areas显示文摘Based on geological, chronological, geochemical and Nd isotopicstudies of the high-grade basement of the Qilian terrane, the authors have drawn the following main conclusions: (1) the high-grade basement of the Qilian terrane consists mainly of meta-argillo-arenaceous rocks and granites and its bulk part was formed in the period of 0.8-1.0 Ga (the Jinningian period); (2) most of the meta-argillo-arenaceous rocks and granitic rocks have strong negative Eu and Ba anomalies (Eu/Eu*= 0.47-0.71 and Ba/Ba*= 0.16-0.64), with tDM and εNd (1.0 Ga) ranging from 1.87 to 2.26 Ga and from -8.54 to -4.06 respectively, showing relatively high maturity; and (3) the Jinningian granitic rocks are a typical product of continent-continent collision, being probably related to the formation of the supercontinent Rodinia. These studies, com bined with the study of high-grade basement rocks near the Qilian terrane, suggest that before the Jinningian period, the Qilian-Qaidam northern-margin terrane and Dunhuang-Alxa terrane were separated from each other, belonging to differ ent plate systems of the North China craton and Yangtze platform respectively. 13Seismic Sequence in Carbonate Rocks By Vibrational Liquefaction显示文摘On the basis of the records of strong seismic events taking place in soft carbonate sediments, a new seismic sequence system of vibrational liquefaction is established, which consists of a series of units, such as escaped structure of micrite veins and liquefied deformation formed in the period of seismic liquefaction, land subsidence structure after liquefaction, tsunamic hummocky and turbidite produced by seismic events, This sequence is a generalization and summation of field observation in vast areas, which shows the whole process of a strong seismic event and provides an unified theoretical explanation. The pattern of the seismic sequence by vibrational liquefaction provides one of correlation standards for geologists in the field to discriminate events in carbonate sequences. 14The Sequence of Magmatic-Tectonic Events and Orogenic Processes of the Yanshan Belt, North China显示文摘This paper emphasizes that the interactive constraints of geology and isotopic dating is the best approach to construct the geological event sequence, and has compiled 106 data of reasonable isotopic ages for the igneous rocks of the Yanshan belt. We propose a sequence of mgmatic-tectonic events in the Jurassic-Cretaceous Yanshan orogen of North China. Five orogenic episodes are divided, (1) pre-and initial orogenic episode (Early Jurassic); (2) early orogenic episode (Middle Jurassic); (3) peak orogenic episode (Late Jurassic); (4) late orogenic episode (early Early Cretaceous),and (5) post-orogenic episode. Each episode is a short cycle, all of the orogenic processes construct a longer cycle, and they, in general, followed a counter-clockwise (ccw) PTt path. 15Effect of carbon source on the denitrification in constructed wetlands显示文摘The ability of constructed wetlands with different plants in nitrate removal were investigated. The factors promoting the rates of denitrification including organic carbon, nitrate load, plants in wetlands, pH and water temperature in field were systematically investigated. The results showed that the additional carbon source (glucose) can remarkably improve the nitrate removal ability of the constructed wetland. It demonstrated that the nitrate removal rate can increase from 20% to more than 50% in summer and from 10% to 30% in winter, when the nitrate concentration was 30–40 mg/L, the retention time was 24 h and 25 mg/L dissolved organic carbon (DOC) was ploughed into the constructed wetland. However, the nitrite in the constructed wetland accumulated a little with the supply of the additional carbon source in summer and winter, and it increased from 0.15 to 2 mg/L in the effuent. It was also found that the abilities of plant in adjusting pH and temperature can result in an increase of denitrification in wetlands. 18Decoupling of surface and subsurface sutures in the Dabie orogen and a continent-collisional lithospheric-wedging model: Sr-Nd-Pb isotopic evidences of Mesozoic igneous rocks in eastern China显示文摘There are significant differences of Nd and Pbisotopic compositions between Mesozoic mafic igneous rocks from the North China Block (NCB) and the South ChinaBlock (SCB). Mesozoic mantle-derived igneous rocks fromthe North China Block have very low e Nd values (-15 to -21), and 206Pb/204Pb ratios (< 17.9), while those in the SCB are characterized by e Nd > -10 and 206Pb/204Pb > 18.3. The verylow e Nd values (-16 to -20) and 206Pb/204Pb ratios (< 17.3) of the early Cretaceous mafic-ultramafic intrusions developedin the north part of the Dabie orogen (NDZ) suggest that the deep lithosphere underneath the NDZ belongs to the NCBbut not the SCB. Therefore, although the surface suture be-tween the NCB and SCB is located on the north side of theNDZ, the subsurface suture between the NCB and SCBshould be located to the south side of the NDZ. This is con-sistent with the previous suggestion that the subsurface su-ture in the Sulu terrane east of the Tanlu fault was the south displacement, but contradictory to northward continentalsubduction of the SCB. A continent-collisional lithospheric-wedging model can interpret the decoupling of the surface and subsurface sutures in the Dabie-Sulu orogen. After slabbreak-off, the continuing convergence of two continental blocks must increase the compression force acting on the suture zone, which might induce the lithosphere splitting of SCB. Thus, the lower crust and lithospheric mantle on thesouth margin of the NCB can wedge into the north margin of the lithosphere of the SCB along the Dabie-Sulu collisionzone. This process caused the overthrust of the mid-upper continental crust with exhumed ultrahigh pressure meta-morphic (UHPM) rocks and underthrust of the deep litho-sphere of the SCB. 20Metastatic human hepatocellular carcinoma models in nude mice and cell line with metastatic potential显示文摘Metastatic human HCC model is needed for the studies on mechanism and intervention of metastatic recurrence. By using orthotopic implantation of histologically intact tissues of 30 surgical specimens, a patient like metastatic model of human HCC in nude mice (LCI-D20)and a Iow metastatic model of human HCC in nude mice LCI-D35 ) have been established. All mice with transplanted LCI-D20 tumors exhibited extremely high metastatic ability including spontaneous metastasis to liver, lungs, lymph nodes and peritoneal seeding.Remarkable difference was also found in expression of some of the invasiveness related genes and growth factors between the LCI-D20 and LCI-D35 tumors. PAI-Iincreased gradually following tumor progression in LCID20 model, and correlated with tumor size and AFP level,Phasic expression of tissue intercellular adhesion molecule-I in this model was also observed. Using corneal micropocket model, it was demonstrated that the vascular response induced by LCI-D20 tumor was stronger than that induced by LCI-D35 tumor. Similar report on metastatic human HCC model in nude mice and human HCC cell line with metastatic potential was rarely found in the literature.
