| 1 | Discovery of a series of dimethoxybenzene FGFR inhibitors with 5H-pyrrolo[2,3-b]pyrazine scaffold: structure–activity relationship, crystal structural characterization and in vivo study显示文摘Genomic alterations are commonly found in the signaling pathways of fibroblast growth factor receptors(FGFRs). Although there is no selective FGFR inhibitors in market, several promising inhibitors have been investigated in clinical trials, and showed encouraging efficacies in patients. By designing a hybrid between the FGFR-selectivity-enhancing motif dimethoxybenzene group and our previously identified novel scaffold, we discovered a new series of potent FGFR inhibitors, with the best one showing sub-nanomolar enzymatic activity. After several round of optimization and with the solved crystal structure, detailed structure–activity relationship was elaborated. | 3 | Genetic Diversity and Drug Susceptibility of Mycobacterium tuberculosis Isolates in a Remote Mountain Area of China显示文摘Objective We determined the genetic diversity of Mycobacterium tuberculosis(MTB) in a remote mountainous area of southwest China and evaluated the resolving ability of single nucleotide polymorphism(SNP) genotyping combined with variable number tandem repeat(VNTR) genotyping for Beijing family strains in association with drug resistance status.Methods Three hundred thirty-one MTB strains were isolated from patients living in mountainous regions of southwest China,and 8-loci SNP,VNTR-15 genotyping assays,and drug susceptibility testing of 9 drugs were performed.Results A total of 183 [55.29%(183/331)] strains were classified into the Beijing family.Of the 183 strains,111(60.66%) were defined as modern Beijing strains.The most predominant modern Beijing sub-lineage and ancient Beijing sub-lineage were Bmyc10 [39.34%(72/183)] and Bmyc25 [20.77%(38/183)],respectively.Of the isolates,19.64%(65/331) were resistant to at least 1 of the 9 anti-TB drugs and 17 [4.98%(17/331)] MTB isolates were multi-drug resistant tuberculosis(MDR-TB).Two hundred sixty-one isolates showed a clustering rate of 14.18%(37/261) and a discriminatory index of 0.9990.The Beijing lineage exhibited a significantly higher prevalence of MDR-TB,as well as resistance to isoniazid(INH),rifampin(RIF),and para-aminosalicylic acid(PAS) when analyzed independently(P = 0.005,P = 0.017,P = 0.014,and P = 0.006 respectively).The Beijing lineage was not associated with genetic clustering or resistance to any drug.In addition,genetic clustering was not associated with drug resistance.Conclusion MTB strains demonstrate high genetic diversity in remote mountainous areas of southwest China.Beijing strains,especially modern Beijing strains,are predominant in remote mountainous area of China.The combination of 8-loci SNPs and VNTR-15 genotyping is a useful tool to study the molecular epidemiology of MTB strains in this area. | ma ai jing wang sheng fen fan jia le zhao bing he guang xue zhao yan lin | 2018 | Biomedical and Environmental Sciences2018,31,5: | 2 |

| 5 | Lactate anions participate in T cell cytokine production and function显示文摘After antigen stimulation, T cells preferentially increase aerobic glycolysis to meet the bioenergetic and biosynthetic demands of T cell activation, proliferation, and effector functions. Lactate, a by-product of glycolysis, has been reported to function as an important energy source and signaling molecule. Here, we found that lactate anions are involved in cytokine production in Tcells after TCR activation. During ex vivo T cell activation, the addition of excess sodium lactate(Na L) increased the production of cytokines(such as IFNγ/IL-2/TNFα) more than the addition of sodium chloride(NaCl). This enhanced cytokine production was dependent on TCR/CD3 activation but not CD28 activation. In vivo, Na L treatment inhibited tumour growth in subcutaneously transplanted tumour models in a T cell-dependent manner, which was consistent with increased T cell cytokine production in the Na L treatment group compared to the Na Cl treatment group. Furthermore, a mechanistic experiment showed that this enhanced cytokine production was regulated by GAPDH-mediated post-transcriptional regulation. | 12 | A Novel Robust Watermarking Algorithm for Encrypted Medical Image Based on DTCWT-DCT and Chaotic Map显示文摘In order to solve the problem of patient information security protection in medical images,whilst also taking into consideration the unchangeable particularity of medical images to the lesion area and the need for medical images themselves to be protected,a novel robust watermarking algorithm for encrypted medical images based on dual-tree complex wavelet transform and discrete cosine transform(DTCWT-DCT)and chaotic map is proposed in this paper.First,DTCWT-DCT transformation was performed on medical images,and dot product was per-formed in relation to the transformation matrix and logistic map.Inverse transformation was undertaken to obtain encrypted medical images.Then,in the low-frequency part of the DTCWT-DCT transformation coefficient of the encrypted medical image,a set of 32 bits visual feature vectors that can effectively resist geometric attacks are found to be the feature vector of the encrypted medical image by using perceptual hashing.After that,different logistic initial values and growth parameters were set to encrypt the watermark,and zero-watermark technology was used to embed and extract the encrypted medical images by combining cryptography and third-party concepts.The proposed watermarking algorithm does not change the region of interest of medical images thus it does not affect the judgment of doctors.Additionally,the security of the algorithm is enhanced by using chaotic mapping,which is sensitive to the initial value in order to encrypt the medical image and the watermark.The simulation results show that the pro-posed algorithm has good homomorphism,which can not only protect the original medical image and the watermark information,but can also embed and extract the watermark directly in the encrypted image,eliminating the potential risk of decrypting the embedded watermark and extracting watermark.Compared with the recent related research,the proposed algorithm solves the contradiction between robustness and invisibility of the watermarking algorithm for encrypted medical images,and it has good results against both conventional attacks and geometric attacks.Under geometric attacks in particular,the proposed algorithm performs much better than existing algorithms. | Jing Liu Jingbing Li Jieren Cheng Jixin Ma Naveed Sadiq Baoru Han Qiang Geng Yang Ai | 2019 | Computers, Materials & Continua2019,,8: | 1 |

| 14 | Fertility outcome analysis after modified laparoscopic microsurgical tubal anastomosis显示文摘Modified laparoscopic microsurgical tubal anastomosis is an alternative for microsurgical anastomosis via laparotomy to reverse sterilization in women with renewed child wish.The current study aims to evaluate the fertility outcome after modified laparoscopic microsurgical tubal anastomosis.A retrospective study was performed.Fifty-eight women who underwent modified laparoscopic microsurgical tubal anastomosis were monitored to investigate the fertility outcome and characteristics of this new technology.Of the 58 patients,the cumulative pregnancy rate(PR)in the 42 patients with follow-up data was 23.8%(10/42),57.1%(24/42),66.7%(28/42),and 73.8%(31/42)within 6,12,24,and 36 months after surgery,respectively.The intrauterine PR was 69.0%(29/42).Two patients(4.8%)had ectopic pregnancies that occurred within 24 months of surgery;three cases ended in spontaneous abortion.The delivery rate was 83.9%(26/31).The length of operating time was 1.20.3 h,with a range of 1.0–2.5 h(60–145 min),and the mean time was approximately 75 min.The blood loss was relatively small,between 10 and 50 ml with an average amount of 22 ml.Thus,the modified laparoscopic tubal anastomosis is a highly successful procedure and a viable alternative to open abdominal microsurgical approaches.Compared with the traditional laparoscopic tubal sterilization reversal,this modified approach has three advantages:(1)less invasive approach via a trocar reduction;(2)remodeling of tube is better performing tied together after 3–4 sutures;and(3)faster operating time. | Jihui Ai Pei Zhang Lei Jin Yufeng Li Jing Yue Ding Ma Hanwang Zhang | 2011 | Frontiers of Medicine2011,5,3: | 0 |
| 15 | Monitoring rock desert formation caused by two different origins(ice-snow melting and drying)in the Qinghai-Tibet Plateau of China by considering topographic and meteorological elements显示文摘Monitoring rock desert formation caused by two different origins(ice-snow melting and drying)through remote sensing is crucial to our understanding of the interaction between the underlying surface of different rock desert and land-atmosphere types,as well as the relationship between bare land and soil erosion.A number of achievements have been made in remote sensing monitoring of desert areas,but there is a lack of accurate classification and remote sensing identification of rock desert types based on formation mechanism.In this study,the north and south sides of the eastern Kunlun Mountains in the northern part of the Qinghai-Tibet Plateau of China were taken as the study areas.Landsat operational landscape imager,digital elevation model,and precipitation and temperature grid data were used as data sources.By identifying the bare areas based on the normalized difference vegetation index(NDVI),we used the multi-element fusion method of contours,isotherms,and isohyets to identify the rock desert types in the ice-snow melting and dry areas.The results showed that:(1)the rock desert areas identified by remote sensing based on topographic and meteorological elements were highly accurate,with an overall accuracy of 88.45%and kappa coefficient of 0.77.The multi-element fusion method of contours,isotherms,and isohyets could effectively identify the rock desert types in the ice-snow melting and dry areas;(2)the optimal segmentation range of the ice-snow melting and dry areas was 3600 m contour,-2°C-2°C isotherms,and 100-130 mm isohyets.The areas with elevation less than 3600 m,annual average temperature higher than 2°C,and average annual precipitation less than 100 mm were rock desert in the dry areas.The range of-2°C-2°C isotherms and 100-130 mm isohyets was the transition area between the ice-snow melting and dry areas.The areas with elevation higher than 3600 m,annual average temperature less than-2°C,and average annual precipitation higher than 130 mm were rock desert in the ice-snow melting areas;and(3)the identification accuracy of the bare areas based on the NDVI method was better,specifically,the identification accuracy of plain bare areas was generally better than that of mountain bare areas.The remote sensing identification method considers not only the topographic factors that have great influence on the spatial distribution of the two types of rock desert areas,but also the meteorological factors,which can provide a scientific reference for the effective identification of the two types of rock desert areas. | JIA Wei SHI Peijun WANG Jing'ai MA Weidong XIA Xingsheng ZHOU Yuantao | 2022 | Journal of Arid Land2022,14,8: | 0 |
