| 2 | Diversity of methanotrophs in Zoige wetland soils under both anaerobic and aerobic conditions显示文摘Zoige wetland is one of the most important methane emission centers in China. The oxidation of methane in the wetland affects global warming, soil ecology and atmospheric chemistry. Despite their global significance, microorganisms that consume methane in Zoige wetland remain poorly characterized. In this study, we investigated methanotrophs diversity in soil samples from both anaerobic site and aerobic site in Zoige wetland using pmoA gene as a molecular marker. The cloning library was constructed according to the pmoA sequences detected. Four clusters of methanotrophs were detected. The phylogenetic tree showed that all four clusters detected were affiliated to type I methanotrophs. Two novel clusters (cluster 1, cluster 2) were found to relate to none of the recognized genera of methanotrophs. These clusters have no cultured representatives and reveal an ecological adaptation of particular uncultured methanotrophs in Zoige wetland. Two clusters were belonging to Methylobacter and Methylococcus separately. Denaturing gradient gel electrophoresis gel bands pattern retrieved from these two samples revealed that the community compositions of anaerobic soil and aerobic soil were different from each other while anaerobic soil showed a higher metanotrophs diversity. Real-time PCR assays of the two samples demonstrated that aerobic soil sample in Zoige wetland was 1.5 times as much copy numbers as anaerobic soil. | 3 | Overexpression of SlOFP20 affects floral organ and pollen development显示文摘The OVATE gene was initially identified in tomato and serves as a key regulator of fruit shape.There are 31 OFP members in the tomato genome.However,their roles in tomato growth and reproductive development are largely unknown.Here,we cloned the OFP transcription factor SlOFP20.Tomato plants overexpressing SlOFP20 displayed several phenotypic defects,including an altered floral architecture and fruit shape and reduced male fertility.SlOFP20 overexpression altered the expression levels of some brassinosteroid(BR)-associated genes,implying that SlOFP20 may play a negative role in the BR response,similar to its ortholog OsOFP19 in rice.Moreover,the transcript accumulation of gibberellin(GA)-related genes was significantly affected in the transgenic lines.SlOFP20 may play an important role in the crosstalk between BR and GA.The pollen germination assay suggested that the pollen germination rate of SlOFP20-OE plants was distinctly lower than that of WT plants.In addition,the tomato pollen-associated genes SlCRK1,SlPMEI,LePRK3,SlPRALF,and LAT52 were all suppressed in the transgenic lines.Our data imply that SlOFP20 may affect floral organ and pollen development by modulating BR and GA signaling in tomato. | Shengen Zhou Zongli Hu Fenfen Li Shibing Tian Zhiguo Zhu Anzhou Li Guoping Chen | 2019 | Horticulture Research2019,6,1: | 4 |
| 4 | The characterization of auditory brainstem response(ABR) waveforms: A study in tree shrews(Tupaia belangeri)显示文摘To characterize the patterns of ABR waves in tree shrews, we must understand the hearing sensitivity and auditory function of healthy adult tree shrews. Fifteen tree shrews(30 ears) were stimulated with clicks and tone-pips at 11 different frequencies from 1 to 60 kHz. The ABR waves were recorded and analyzed. The ABR consisted of five to seven positive waves in the first 10 ms after a click stimulus, and the average hearing threshold of component III was 27.86 ± 3.78 dB SPL. Wave III was the largest and most clear. The ABR threshold was related to the tone-pip sitmulus by a 'U' shaped curve. The sensitive frequency was approximately 8 kHz in tree shrews. The latencies systematically decreased with increasing stimulus frequencies. The ABR amplitudes of wave III increased as the sound pressure level increased. | 6 | Three-dimensional numerical simulation of M_2 internal tides in the Luzon Strait显示文摘A three-dimensional isopycnic-coordinate internal tidal model is employed to investigate the generation,propagation, vertical structure and energy conversion of M2 internal tides in the Luzon Strait(LS) with mooring observations. Simulated results, especially the tidal current amplitudes, agree well with observations,demonstrating the reasonability and accuracy of the model. Results indicate that M2 internal tides mainly propagate into three directions horizontally, i.e., eastward towards the western Pacific Ocean, westward towards the Dongsha Island and southwestward towards the South China Sea Basin. In the horizontal direction, tidal current amplitudes decrease as distance increases away from the LS; in the vertical direction, they show an obvious decreasing tendency with depth. Between the double ridges of the LS, a clockwise gyre of M2 baroclinic energy flux appears, which is caused by reflections of M2 internal tides at supercritical topographies, and resonance of M2 internal tides happens along 19.5° and 21.5°N due to the heights and separation distance of the double ridges. | 9 | Estimation of the Reflection of Internal Tides on a Slope显示文摘Reflection occurs when internal tides impact on a steep continental slope. Separating reflected internal tide signals from incident ones is crucial to develop the parameterization of internal tide-driven turbulent mixing on the continental slopes. In this study, the performances of three different methods for estimating internal tide reflections are examined by using two different cases. The Hilbert transform-based method is found to be more suitable than two other methods for both cases considered in this study. The two other methods are effective for westward-propagating mode-1 internal tides impacting a slope, but inappropriate in the case where internal tides radiate from a Gaussian ridge impact the slope because of their inaccurate estimation of incident internal tides in the latter case. Such inaccurate estimation further influences the extraction of reflected signals and calculation of the reflected and cross term of energy fluxes.
