2Establishment of a dmp based maternal haploid induction system for polyploid Brassica napus and Nicotiana tabacum显示文摘Doubled haploid(DH) technology is used to obtain homozygous lines in a single generation, a technique that significantly accelerates the crop breeding trajectory. Traditionally, in vitro culture is used to generate DHs, but this technique is limited by species and genotype recalcitrance. In vivo haploid induction(HI) through seed is widely and efficiently used in maize and was recently extended to several other crops. Here we show that in vivo HI can be triggered by mutation of DMP maternal haploid inducer genes in allopolyploid(allotetraploid) Brassica napus and Nicotiana tabacum. We developed a pipeline for selection of DMP orthologs for clustered regularly interspaced palindromic repeats mutagenesis and demonstrated average amphihaploid induction rates of2.4% and 1.2% in multiple B. napus and N. tabacum genotypes, respectively. These results further confirmed the HI ability of DMP gene in polyploid dicot crops. The DMP-HI system offers a novel DH technology to facilitate breeding in these crops. 3Gas Occurrence and Accumulation Characteristics of Cambrian–Ordovician Shales in the Tarim Basin, Northwest China显示文摘The Tarim Basin is located in northwestern China and is the biggest basin in China with huge oil and gas resources. Especially the Lower to Middle Cambrian and Middle to Upper Ordovician possess the major marine source rocks in the Tarim Basin and have large shale gas resource potential. The Cambrian–Ordovician shales were mainly deposited in basin–slope facies with thicknesses between 30–180 m. For shales buried shallower than 4500 m, there is high organic matter abundance with TOC(total organic carbon) mainly between 1.0% and 6.0%, favorable organic matter of Type I and Type II, and high thermal maturity with RoE as 1.3%–2.75%. The mineral composition of these Cambrian–Ordovician shale samples is mainly quartz and carbonate minerals while the clay minerals content is mostly lower than 30%, because these samples include siliceous and calcareous shale and marlstone. The Cambrian and Ordovician shales are compacted with mean porosity of 4% and 3%, permeability of 0.0003×10^(-3)–0.09×10^(-3) μm^2 and 0.0002×10^(-3)–0.11×10^(-3) μm^2, and density of 2.30 g/m^3 and 2.55 g/m^3, respectively. The pores in the shale samples show good connectivity and are mainly mesopore in size. Different genetic types of pores can be observed such as intercrystal, intergranular, dissolved, organic matter and shrinkage joint. The reservoir bed properties are controlled by mineral composition and diagenesis. The maximum adsorption amount to methane of these shales is 1.15–7.36 cm^3/g, with main affecting factors being organic matter abundance, porosity and thermal maturity. The accumulation characteristics of natural gas within these shales are jointly controlled by sedimentation, diagenesis, hydrocarbon generation conditions, reservoir bed properties and the occurrence process of natural gas. The natural gas underwent short-distance migration and accumulation, in-place accumulation in the early stage, and adjustment and modification in the later stage. 4Comparisons of AGRI/FY-4A Cloud Fraction and Cloud Top Pressure with MODIS/Terra Measurements over East Asia显示文摘Fengyun-4 A(FY-4 A),the second generation of China’s geostationary meteorological satellite,provides high spatiotemporal resolution cloud products over East Asia.In this study,cloud fraction(CFR)and cloud top pressure(CTP)products in August 2017 derived from the Advanced Geosynchronous Radiation Imager(AGRI)aboard FY-4 A(AGRI/FY-4 A)are retrospectively compared with those from the Moderate Resolution Imaging Spectroradiometer(MODIS)aboard Terra(MODIS/Terra)over East Asia.To avoid possible errors in the comparison caused by the lower temporal coverage of MODIS/Terra products compared to that of AGRI/FY-4 A over the same region and to account for time lags between observations of the two instruments,we construct datasets of AGRI/FY-4 A CFR and CTP to match those of MODIS/Terra in each scan over East Asia in August 2017.Results show that the CFR and CTP datasets of the two instruments generally agree well,with the linear correlation coefficients(R)between CFR(CTP)data of 0.83(0.80)regardless of time lags.Though longer time lags contribute to the worse consistency between CFR(CTP)data derived from observations of the two instruments in most cases,large CFR/CTP discrepancies do not always match with long time lags.Large CFR discrepancies appear in the vicinity of the Tibetan Plateau(TP;28°–45°N,75°–105°E).These differences in the cloud detection by the two instruments largely occur when MODIS/Terra detects clear-sky while AGRI/FY-4 A detects higher values of CFR,and this accounts for 61%of the CFR discrepancy greater than 50%near the TP.In the case of CTP,the largest discrepancies appear in the eastern Iranian Plateau(IP;25°–45°N,60°–80°E),where there are some samples with long time lags(20–35 min)and fewer daily data samples are available for computing monthly means compared to other regions since there are many clearsky data samples there during the study period.Tao WANG Jiali LUO Jinglin LIANG Baojian WANG Wenshou TIAN Xiaoyan CHEN 2019Journal of Meteorological Research2019,33,4:2

8Geochemistry and origin of gas pools in the Gaoqing-Pingnan fault zone, Jiyang Depression显示文摘In the surroundings of the Gaoqing-Pingnan fault zone are developed quite a number of gas reservoirs. Based on gas compositions, they can be divided into two groups, i.e., CO2 and CH4. Their composition and isotope geochemistry were dealt with in this study. The CO2 contents range from 60.72%–99.99%, the δ13CCO2 values from -3.41‰– -9.8‰, and the 3He/4He ratios from 4.35×10-6–6.35×10-6 (i.e. R/Ra=4.45–4.35). Based on the data on composition and isotope geochemistry, deep geological background, deep faults and volcanic rocks, it is shown that CO2 ,distributed in the Gaoqing area, mostly originated from mantle-source inorganic matter which is associated with magmatic rocks. The favorable tectonic environment for the formation of CO2 reservoirs is the rift, which is related to great fault-magmatic activity, the formation of CO2 gas pools and their space-time correlation to the most recent magmatic activities. Hydrocarbon gas pools occur in the Huagou area. The CH4 contents are within the range of 88.83%–99.12%, and the δ13CCH4 values, -44.7‰– -54.39‰. This indicates that the hydrocarbon gas resulted from the decomposition of oil-type gas at high temperatures. Volcanic rocks in the CO2 gas pool-and CH4 gas pool-distributed areas show significant differences in Fe2O3 and FeO contents. This has proven that the hydrocarbon gas may have resulted from various chemical reactions. 16Pseudo-dynamic test and numerical simulation of high-strength concrete frame structure reinforced with high-strength rebars显示文摘This paper describes an investigation of a high-strength concrete frame reinforced with high-strength rebars that was tested in the structure engineering laboratory at Shenyang Jianzhu University. The frame specimen was pseudo- dynamically loaded to indicate three earthquake ground motions of different hazard levels, after which the test specimen was subjected to a pseudo-static loading. This paper focuses on the design, construction and experiment of the test frame and validation of the simulation models. Research shows that a high-strength concrete frame reinforced with high-strength rebars is more eff icient and economical than a traditional reinforced concrete frame structure. In addition to the economies achieved by effective use of materials, research shows that the frame can provide enough strength to exceed conventional reinforced concrete frames and provide acceptable ductility. The test study provides evidence to validate the performance of a high- strength concrete frame designed according to current seismic code provisions. Based on previous test research, a nonlinear FEM analysis is completed by using OpenSees software. The dynamic responses of the frame structure are numerically analyzed. The results of the numerical simulation show that the model can calculate the seismic responses of the frame by OpenSees. 17High-throughput haploid induction in species with bisexual flowers显示文摘Dear Editor,Doubled haploid(DH)technology can significantly accelerate the development of homozygous lines.DH breeding has achieved great success inmaize because of the discovery of the first haploid inducer,Stock6,and the development of a series of high-efficiency haploid inducers(Hu et al.,2016).Pioneering studies on the genetic basis of haploid induction(HI)revealed that loss-offunction mutation of the phospholipase gene ZmPLA1/MATL/NLD triggers HI and that the HI rate(HIR)can be dramatically enhanced by a single nucleotide substitution from T to C in ZmDMP(Jacquier et al.,2020).Remarkably,knockout of ZmPLA1/MATL/NLD homologs in rice,wheat,and foxtail millet results in HIRs of 2%–6%,5%–15%,and 2%–3%,respectively(Jacquier et al.,2020;Cheng et al.,2021).In addition,loss of function of ZmDMP-like genes enables HI in species including Arabidopsis,tomato,rapeseed,tobacco,etc.,with an average HIR of around 2%(Zhong et al.,2020,2022a,2022b).These successes have laid solid foundations for the construction of a universal DH breeding system in different crop species.More importantly,HI-Edit/IMGE systems that enable gene editing in elite germplasms have been established on the basis of HI,making HI even more important(Kelliher et al.,2019;Wang et al.,2019).Xiaolong Qi Jinchu Liu Zongkai Liu Chen Chen Baojian Chen Shuwei Guo Zhongfu Ni Yu Zhong Shaojiang Chen Chenxu Liu 2023Plant Communications2023,4,1:0
18Establishment of an efficient haploid identification system by engineering anthocyanin accumulation in the wheat embryo显示文摘Dear Editor,Bread wheat(Triticum aestivum)is one of the most important food crops and provides approximately 20%of the food calories for human consumption.A 70%increase in wheat production is needed by 2050 to keep pace with the growing global population(International Wheat Genome Sequencing,2014).Developing superior cultivars is an efficient way to improve yield.Nevertheless,conventional breeding is time consuming,as more than eight generations are needed to develop new plant varieties.Using doubled haploid(DH)technology,homozygous lines can be produced in only two generations,dramatically accelerating the breeding process.In wheat,haploids can be obtained by cross pollination with corn pollen followed by embryo rescue(Laurie and Bennett,1988).In maize,haploids can be induced by haploid inducer lines derived from Stock6(Liu et al.,2022).The cloning of two genes that control haploid induction(HI)in maize,MATL/ZmPLA1/NLD and ZmDMP,paved the way for DH breeding in more crop species(Jacquier et al.,2020).Further studies have shown that loss of function of TaPLAs triggers wheat HI with an efficiency of 5.88%to 31.6%(Liu et al.,2020a,2020b);this would be a promising approach for establishing a new,simple,and more efficient DH breeding method in wheat.Xiaolong Qi Shuwei Guo Yu Zhong Baojian Chen Zongkai Liu Tongzheng Yan Bin Feng Zhongfu Ni Shaojiang Chen Chenxu Liu 2023Plant Communications2023,4,3:0
19Recent progress in microbial cell surface display systems and their application on biosensors显示文摘Microbial cell surface display technology is a recombinant technology to express target proteins on the cell membrane,which can be used to redesign the cell surface with functional proteins and peptides.Bacterial and yeast surface display systems are the most common cell surface display systems of prokaryotic and eukaryotic proteins,that are widely applied as the core elements in the field of biosensors due to their advantages,including enhanced stability,high yield,good safety,expression of larger and more complex proteins.To further promote the performance of biosensors,the biomineralized microbial surface display technology was proposed.This review summarized the different microbial surface display systems and the biomineralized surface display systems,where the mechanisms of surface display and biomineralization were introduced.Then we described the recent progress of their applications on biosensors for different types of detection targets.Finally,the outlooks and tendencies were discussed and forecasted with the expectation to provide some general functions and enlightenments to this aspect of research.HAIYING CHEN YUQING WU BAOJIAN HUANG LEI HAN 2023BIOCELL2023,47,6:0
20Single-Cell RNA Sequencing Reveals Potential for Endothelial-to-Mesenchymal Transition in Tetralogy of Fallot显示文摘Background:Tetralogy of Fallot(TOF)is a very common cyanotic congenital heart disease.Endothelial-to-mesenchymal transition(EndoMT)is recognized as a physiological mechanism involved in embryonic heart development and endothelial formation.However,there is still a gap in the reports related to the mechanism of EndoMT development in TOF.Methods:First,transcriptomic data of single cell nuclei of TOF and Donor were obtained based on the Gene Expression Omnibus(GEO)database,and the data were normalized and clus-tered by dimensionality reduction using the Seurat package.Subsequently,differentially expressed genes(DEGs)between TOF and Donor were screened using the“FindMarkers”function,and the gene sets of interest were enriched.Finally,to characterize the dynamics of EndoMT occurrence in TOF,we performed pseudotime cell tra-jectory inference as well as utilized SCENIC analysis to probe the gene regulatory networks(GRNs)dominated by transcription factors(TFs)in endothelial cells.Results:We identified a total of six cell clusters based on single-cell nuclear transcriptome data from TOF and Donor.We found that 611 genes with up-regulated expression within TOF showed conversion to mesenchyme.By subdividing endothelial cell subtypes,endothelial cells 2 were shown to be involved in cell adhesion,migration and extracellular matrix processes.Pseudo-time and SCENIC analyses showed that endothelial cell 2 has EndoMT potential.In addition,ERG and TEAD1 are TFs that play key reg-ulatory roles in this subtype,and both of their target genes are also highly expressed in TOF.This demonstrates that ERG and TEAD1 effectively promote the EndoMT process.Conclusion:Our study reveals the molecular mechanisms underlying the development of EndoMT in TOF,which demonstrates that manipulating the endothelial-to-mesenchymal transition may offer unprecedented therapeutic potential for the treatment of TOF.Aisa Zulibiya Jing Wen Huiqing Yu Xiaoming Chen Lei Xu Xiao Ma Baojian Zhang 2023Congenital Heart Disease2023,18,6:0
