1Mechanical properties of electron beam welded dissimilar joints of TC17 and Ti60 alloys显示文摘Microstructure, hardness, tensile and high cycle fatigue(HCF) properties of the welded dissimilar joints of Ti60 and TC17 titanium alloys had been investigated in this study. A significant microstructural change was observed to occur after welding, with rod-like α and β phases in the fusion zone(FZ), equiaxed αphases, fine α laths and β phases in the heat-affected zone(HAZ) of TC17 side and acicular martensiteα' phases+ 'ghost' α phases in the HAZ of Ti60 side. The microhardness across the joints exhibited an inhomogeneous distribution with the highest hardness of ~404 HV in FZ and the lowest hardness of^304 HV in base material(BM) of Ti60. All the joints tested in tension fractured at BM of Ti60 side.Fatigue limits of the joints at 107 cycles were 425 MPa at room temperature and 380 MPa at 400℃,respectively. 2Genomic insights into the fast growth of paulownias and the formation of Paulownia witches' broom显示文摘Paulownias are among the fastest growing trees in the world,but they often suffer tremendous loss of wood production due to infection by Paulownia witches'broom(PaWB)phytoplasmas.In this study,we have sequenced and assembled a high-quality nuclear genome of Paulownia fortunei,a commonly cultivated paulownia species.The assembled genome of P.fortunei is 511.6 Mb in size,with 93.2%of its sequences anchored to 20 pseudo-chromosomes,and it contains 31985 protein-coding genes.Phylogenomic analyses show that the family Paulowniaceae is sister to a clade composed of Phrymaceae and Orobanchaceae.Higher photosynthetic efficiency is achieved by integrating C3 photosynthesis and the crassulacean acid metabolism pathway,which may contribute to the extremely fast growth habit of paulownia trees.Comparative transcriptome analyses reveal modules related to cambial growth and development,photosynthesis,and defense responses.Additional genome sequencing of PaWB phytoplasma,combined with functional analyses,indicates that the effector PaWB-SAP54 interacts directly with Paulownia PfSPLa,which in turn causes the degradation of PfSPLa by the ubiquitin-mediated pathway and leads to the formation of witches'broom.Taken together,these results provide significant insights into the biology of paulownias and the regulatory mechanism for the formation of PaWB.Yabing Cao Guiling Sun Xiaoqiao Zhai Pingluo Xu Liming Ma Minjie Deng Zhenli Zhao Haibo Yang Yanpeng Dong Zhonghai Shang Yujie Lv Lijun Yan Haifang Liu Xibing Cao Bingbing Li Zhe Wang Xiaogai Zhao Haiyan Yu Fan Wang Wen Ma Jinling Huang Guoqiang Fan 2021Molecular Plant2021,14,10:7
3Functional Metabolomics Reveals that Astragalus Polysaccharides Improve Lipids Metabolism through Microbial Metabolite 2-Hydroxybutyric Acid in Obese Mice显示文摘Polysaccharides are widely present in herbs with multiple activities,especially immunity regulation and metabolic benefits for metabolic disorders.However,the underlying mechanisms are not well under-stood.Functional metabolomics is increasingly used to investigate systemic effects on the host by iden-tifying metabolites with particular functions.This study explores the mechanisms underlying the metabolic benefits of Astragalus polysaccharides(APS)by adopting a functional metabolomics strategy.The effects of APS were determined in eight-week high-fat diet(HFD)-fed obese mice.Then,gas chromatography–time-of-flight mass spectrometry(GC–TOFMS)-based untargeted metabolomics was performed for an analysis of serum and liver tissues,and liquid chromatography–tandem mass spectrom-etry(LC–MS/MS)-based targeted metabolomics was performed.The potential functions of the metabo-lites were tested with in vitro and in vivo models of metabolic disorders.Our results first confirmed the metabolic benefits of APS in obese mice.Then,metabolomics analysis revealed that APS supplemen-tation reversed the HFD-induced metabolic changes,and identified 2-hydroxybutyric acid(2-HB)as a potential functional metabolite for APS activity that was significantly decreased by a HFD and reversed by APS.Further study indicated that 2-HB inhibited oleic acid(OA)-induced triglyceride(TG)accumula-tion.It was also found to stimulate the expression of proteins in lipid degradation in hepatocytes and TG lipolysis in 3T3-L1 cells.Moreover,it was found to reduce serum TG and regulate the proteins involved in lipid degradation in high-fat and high-sucrose(HFHS)-fed mice.In conclusion,our study demonstrates that the metabolic benefits of APS are at least partially due to 2-HB generation,which modulated lipid metabolism both in vitro and in vivo.Our results also highlight that functional metabolomics is practical for investigating the mechanism underlying the systemic benefits of plant polysaccharides.Bingbing Li Ying Hong Yu Gu Shengjie Ye Kaili Hu Jian Yao Kan Ding Aihua Zhao Wei Jia Houkai Li 2022Engineering2022,8,2:6

7Impact of the Rainfall Intensity and Seepage on Slope Stability in Loess Plateau显示文摘In recent years,with the increase of traffic construction in mountainous areas in China,road slope traffic accidents have become more and more common.In addition,natural disasters such as landslides,collapses and subgrade settlements caused by rainfall,driving load,seasonal variation and groundwater distribution are frequent.In particular,rainfall is one of the most common factors leading to slope instability(landslide).Therefore,this paper proposes the seep module based on the application software Geo-studio,and analyzes the slope soil parameters and slope stability under five types of rainfall conditions:light rain(10 mm/d),moderate rain(25 mm/d),heavy rain(50 mm/d),rainstorm(100 mm/d),and torrential rain(250 mm/d).The critical safety factor under rainfall intensity is fitted with nonlinear curve by sine function.The results show that the fitting curve of rainfall intensity and safety factor on the upper slope is excellent.The residual points are evenly distributed in the belt area of±0.1,and the data basically conform to the nonlinear sine model,indicating that the curve plays an essential role in slope health diagnosis.ZHANG Yu YANG Lixia HAN Bingbing WU Yajie 2020Earthquake Research in China2020,34,4:3
8Effects of Dielectric Barrier Discharge Plasma on the Combustion Performances of Reverse-Flow Combustor in an Aero-Engine显示文摘In order to apply plasma assisted combustion(PAC) into a reverse-flow aero-engine and verify the improvement of combustion performance, a feasible approach was proposed in this work. In this approach, based on the structure characteristics of the reverse-flow combustor, a parallel plate double dielectric barrier discharge(DBD) PAC actuator was designed to generate plasma. It was installed at the front of combustor. When the actuator is driven, the original air flow is not disturbed for the device’s structure and installation. Using aviation kerosene as fuel, the effects of plasma on ignition boundary and outlet temperature of the combustor were experimentally investigated at atmosphere pressures. Through the dual high voltage differential power supply, the large gap, large area and uniform plasma discharge was achieved. The results of PAC actuator discharge indicate that inlet air temperature has a small increase of 4–9 K. After PAC is applied, the combustion performances of reverse-flow combustor in an aero-engine are remarkably improved. Experimental results indicate that ignition boundary is widened by 3.7%–12.5% because of the impact of plasma. Outlet highest temperature of combustor is raised by 19–75 K;outlet temperature distribution coefficient is reduced by 11.1%–26.6%. 10High-temperature superconductivity in sulfur hydride evidenced by alternating-current magnetic susceptibility显示文摘The search for high-temperature superconductivity is one of the research frontiers in physics. In the sulfur hydride system, an extremely high T_c(~200 K) has been recently developed at pressure. However, the Meissner effect measurement above megabar pressures is still a great challenge. Here, we report the superconductivity identification of sulfur hydride at pressure, employing an in situ alternating-current magnetic susceptibility technique. We determine the superconducting phase diagram, finding that superconductivity suddenly appears at 117 GPa and Tcreaches 183 K at 149 GPa before decreasing monotonically with increasing pressure. By means of theoretical calculations, we elucidate the variation of T_c in the low-pressure region in terms of the changing stoichiometry of sulfur hydride and the further decrease in Tcowing to a drop in the electron–phonon interaction parameter λ. 11A modified pCas/pTargetF system for CRISPR-Cas9-assisted genome editing in Escherichia coli显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated nuclease 9(Cas9)-based genome editing tool pCas/pTargetF system that we established previously has been widely used in Escherichia coli MG1655.However,this system failed to manipulate the genome of E.coli BL21(DE3),owing to the potential higher leaky transcription of the gRNA-pMB1 specific to pTargetF in this strain.In this study,we modified the pCas/pTargetF system by replacing the promoter of gRNA-pMB1 with a tightly regulated promoter PrhaB,changing the replicon of pCas to a nontemperature-sensitive replicon,adding the sacB gene into pCas,and replacing the original N20-specific sequence of pTargetF with ccdB gene.We call this updated system as pEcCas/pEcgRNA.We found that gRNA-pMB1 indeed showed a slightly higher leaky expression in the pCas/pTargetF system compared with pEcCas/pEcgRNA.We also confirmed that genome editing can successfully be performed in BL21(DE3)by pEcCas/pEcgRNA with high efficiency.The application of pEcCas/pEcgRNA was then expanded to the E.coli B strain BL21 Star^(TM)(DE3),K-12 strains MG1655,DH5α,CGMCC3705,Nissle1917,W strain ATCC9637,and also another species of Enterobacteriaceae,Tatumella citrea DSM13699,without any specific modifications.Finally,the plasmid curing process was optimized to shorten the time from|$\sim$|60 h to|$\sim$|32 h.The entire protocol(including plasmid construction,editing,electroporation and mutant verification,and plasmid elimination)took only|$\sim$|5.5 days per round in the pEcCas/pEcgRNA system,whereas it took|$\sim$|7.5 days in the pCas/pTargetF system.This study established a faster-acting genome editing tool that can be used in a wider range of E.coli strains and will also be useful for other Enterobacteriaceae species.Qi Li Bingbing Sun Jun Chen Yiwen Zhang Yu Jiang Sheng Yang 2021Acta Biochimica et Biophysica Sinica2021,53,5:2

14Alleviating experimental pulmonary hypertension via co-delivering FoxO1 stimulus and apoptosis activator to hyperproliferating pulmonary arteries显示文摘Pulmonary hypertension(PH)is an insidious pulmonary vasculopathy with high mortality and morbidity and its underlying pathogenesis is still poorly delineated.The hyperproliferation and apoptosis resistance of pulmonary artery smooth muscle cells(PASMCs)contributes to pulmonary vascular remodeling in pulmonary hypertension,which is closely linked to the downregulation of forkhead box transcriptional factor O1(FoxO1)and apoptotic protein caspase 3(Cas-3).Here,PA-targeted co-delivery of a FoxO1 stimulus(paclitaxel,PTX)and Cas-3 was exploited to alleviate monocrotaline-induced pulmonary hypertension.The co-delivery system is prepared by loading the active protein on paclitaxel-crystal nanoparticles,followed by a glucuronic acid coating to target the glucose transporter-1 on the PASMCs.The co-loaded system(170 nm)circulates in the blood over time,accumulates in the lung,effectively targets the PAs,and profoundly regresses the remodeling of pulmonary arteries and improves hemodynamics,leading to a decrease in pulmonary arterial pressure and Fulton's index.Our mechanistic studies suggest that the targeted co-delivery system alleviates experimental pulmonary hypertension primarily via the regression of PASMC proliferation by inhibiting cell cycle progression and promoting apoptosis.Taken together,this targeted co-delivery approach offers a promising avenue to target PAs and cure the intractable vasculopathy in pulmonary hypertension.Bingbing Li Chao Teng Huiling Yu Xiaohong Jiang Xuyang Xing Qi Jiang Chenshi Lin Zongmin Zhao Ruifeng Zhang Wei He 2023Acta Pharmaceutica Sinica B2023,13,6:1

16Expression of phenylalanine ammonia lyase as an intracellularly free and extracellularly cell surface-immobilized enzyme on a gut microbe as a live biotherapeutic for phenylketonuria显示文摘Phenylketonuria(PKU),a disease resulting in the disability to degrade phenylalanine(Phe)is an inborn error with a 1 in 10,000 morbidity rate on average around the world which leads to neurotoxicity.As an potential alternative to a protein-restricted diet,oral intake of engineered probiotics degrading Phe inside the body is a promising treatment,currently at clinical stage II(Isabella,et al.,2018).However,limited transmembrane transport of Phe is a bottleneck to further improvement of the probiotic’s activity.Here,we achieved simultaneous degradation of Phe both intracellularly and extracellularly by expressing genes encoding the Phe-metabolizing enzyme phenylalanine ammonia lyase(PAL)as an intracellularly free and a cell surface-immobilized enzyme in Escherichia coli Nissle 1917(EcN)which overcomes the transportation problem.The metabolic engineering strategy was also combined with strengthening of Phe transportation,transportation of PAL-catalyzed trans-cinnamic acid and fixation of released ammonia.Administration of our final synthetic strain TYS8500 with PAL both displayed on the cell surface and expressed inside the cell to the Pah^(F263S)PKU mouse model reduced blood Phe concentration by 44.4%compared to the control Ec N,independent of dietary protein intake.TYS8500 shows great potential in future applications for PKU therapy.Yu Jiang Bingbing Sun Fenghui Qian Feng Dong Chongmao Xu Wuling Zhong Rui Huang Qiwei Zhai Yu Jiang Sheng Yang 2023Science China(Life Sciences)2023,66,1:1

18Recent advances in RNA structurome显示文摘RNA structures are essential to support RNA functions and regulation in various biological processes. Recently, a range of novel technologies have been developed to decode genome-wide RNA structures and novel modes of functionality across a wide range of species. In this review, we summarize key strategies for probing the RNA structurome and discuss the pros and cons of representative technologies. In particular, these new technologies have been applied to dissect the structural landscape of the SARS-CoV-2 RNA genome. We also summarize the functionalities of RNA structures discovered in different regulatory layers-including RNA processing, transport, localization, and mRNA translation-across viruses, bacteria, animals, and plants. We review many versatile RNA structural elements in the context of different physiological and pathological processes(e.g., cell differentiation, stress response, and viral replication). 19Electrochemically prepared coniferous leaf-like nickel black membrane for desalination by solar-thermal energy conversion显示文摘The structures of the solar-thermal membranes always influence the performance of light absorption and salt resistance in desalination.Inspired by the hierarchical structure of the coniferous leaves with excellent sunlight absorption in frigid regions,a coniferous leaf-like nickel black(L-Ni)membrane for desalination by solar-thermal energy conversion was prepared through electroplating method under a constant voltage.The light trapping effect of coniferous leaf-like structure led to the light absorption enhanced to 92%,the evaporation rate improved to 1.38 kg·m^(-2)·h^(-1),and the solar-vapor conversion efficiency of L-Ni membrane reaching up to 89.75%under 1 sun irradiation.The stability of the membrane was still excellent after 20 cycles desalination because the coniferous leaf-like structure could enhance the hydrophobicity(water contact angle:152°)of the L-Ni membrane,and it was beneficial to salt resistance.The promising performance of L-Ni membrane with coniferous leaf-like structure provides a possibility to replace the noble metal solar-thermal conversion materials.Dongmin Yue Bingbing Li De Sun Hao Zhang Meiling Liu Jingtong Yu 2023Nano Research2023,16,7:1
20Li-richening strategy in Li_(2)ZrCl_(6) lattice towards enhanced ionic conductivity显示文摘All-solid-state Li batteries(ASSLBs) with solid-state electrolytes(SSEs) are exciting candidates for nextgeneration energy storage and receive considerable attention owing to their reliability. Halide SSEs are promising candidates due to their excellent stability against 4 V-class layered cathodes. Compared with Li3InCl6or Li_(3)ScCl_(6), the low ionic conductivity of Li_(2)ZrCl_(6)(LZC) is a challenge despite its low raw-material cost. Herein, we report a family of Li-Richened chloride, Li_(2+2x)Zr_(1–x)MxCl_(6), which can be used in highperformance ASSLBs owing to its high ionic conductivity(up to 0.62 mS cm^(-1)). The theoretical(ab initio molecular dynamics simulations) and experimental results prove that the strategy of aliovalent substitution with divalent metals to obtain Li-Richened LZC is effective in improving Li^(+)conductivity in SSEs. By combining Li_(2.1)Zr_(0.95)Mg_(0.05)Cl_(6)(Mg5-LZC) with a Li–In anode and a LiCoO_(2)cathode, a room-temperature ASSLBs with excellent long-term cycling stability(88% capacity retention at 0.3C for 100 cycles) and highrate capability(121 m A h g^(-1)at 1C) is reported.
