| 1 | TALEN or Cas9-Rapid,Efficient and Specific Choices for Genome Modifications显示文摘Precise modifications of complex genomes at the single nucleotide level have been one of the big goals for scientists working in basic and applied genetics,including biotechnology,drug development,gene therapy and synthetic biology.However,the relevant techniques for making these manipulations in model organisms and human cells have been lagging behind the rapid high throughput studies in the post-genomic era with a bottleneck of low efficiency,time consuming and laborious manipulation,and off-targeting problems.Recent discoveries of TALEs(transcription activator-like effectors) coding system and CRISPR(clusters of regularly interspaced short palindromic repeats) immune system in bacteria have enabled the development of customized TALENs(transcription activator-like effector nucleases) and CRISPR/Cas9 to rapidly edit genomic DNA in a variety of cell types,including human cells,and different model organisms at a very high efficiency and specificity.In this review,we first briefly summarize the development and applications of TALENs and CRISPR/Cas9-mediated genome editing technologies;compare the advantages and constraints of each method;particularly,discuss the expected applications of both techniques in the field of site-specific genome modification and stem cell based gene therapy;finally, propose the future directions and perspectives for readers to make the choices. | Chuanxian Wei Jiyong Liu Zhongsheng Yu Bo Zhang Guanjun Gao Renjie Jiao | 2013 | Journal of Genetics and Genomics2013,40,6: | 52 |

| 3 | Laparoscopic low anterior resection for rectal carcinoma:Complications and management in 132 consecutive patients显示文摘AIM:To analyze the clinical manifestations and risk fac-tors of complications in laparoscopic low anterior resection(LAR)for rectal cancer patients.METHODS:A series of 132 consecutive patients who received laparoscopic LAR for rectal cancer in our center were included.The etiology,diagnosis,treatment and prevention of rectal cancer were studied among the patients with surgery-related complications using both univariate and multivariate regression analysis.RESULTS:No conversion to open surgery was observed and 5 cases converted to hand-assisted laparoscopic operation.The overall morbidity rate was 20.5%.Complications occurred during the operation in 7 patients(5.3%),within 30 postoperative days in 24 patients(18.2%),and within 3 mo in 2 patients(1.5%).The most significant complications were anastomotic leak-age(9.1%)and anastomotic hemorrhage(5.3%).Sizeand location of tumor,pathological staging and preoperative nutrition were significant factors associated with LAR complications,while gender,age and pathological type showed no relevance.Binary logistics regression showed that the size and location of tumor,and pathological staging were independent factors of laparoscopic LAR.All the complications were treated during their onset of clinical manifestations by interventional or conservative therapy.CONCLUSION:Anastomotic leakage is a major com-plication in laparoscopic LAR.The complications may be associated with tumor size and site,and pathological stage.Interventional therapies are of value in the management of laparoscopic LAR complications. | Qian-Lin Zhu,Bo Feng,Ai-Guo Lu,Ming-Liang Wang,Wei-Guo Hu,Jian-Wen Li,Zhi-Hai Mao,Min-Hua Zheng,Department of General Surgery,Shanghai Ruijin Hospital,Shanghai Jiao Tong University School of Medicine,Shanghai 200025,China Shanghai Minimally Invasive Surgery Center,Shanghai 200025,China | 2010 | World Journal of Gastroenterology2010,16,36: | 23 |

| 5 | Lung transplantation as therapeutic option in acute respiratory distress syndrome for coronavirus disease 2019-related pulmonary fibrosis显示文摘Background:Critical patients with the coronavirus disease 2019(COVID-19),even those whose nucleic acid test results had turned negative and those receiving maximal medical support,have been noted to progress to irreversible fatal respiratory failure.Lung transplantation(LT)as the sole therapy for end-stage pulmonary fibrosis related to acute respiratory distress syndrome has been considered as the ultimate rescue therapy for these patients.Methods:From February 10 to March 10,2020,three male patients were urgently assessed and listed for transplantation.After conducting a full ethical review and after obtaining assent from the family of the patients,we performed three LT procedures for COVID-19 patients with illness durations of more than one month and extremely high sequential organ failure assessment scores.Results:Two of the three recipients survived post-LT and started participating in a rehabilitation program.Pearls of the LT team collaboration and perioperative logistics were summarized and continually improved.The pathological results of the explanted lungs were concordant with the critical clinical manifestation,and provided insight towards better understanding of the disease.Government health affair systems,virology detection tools,and modern communication technology all play key roles towards the survival of the patients and their rehabilitation.Conclusions:LT can be performed in end-stage patients with respiratory failure due to COVID-19-related pulmonary fibrosis.If confirmed positive-turned-negative virology status without organ dysfunction that could contraindicate LT,LT provided the final option for these patients to avoid certain death,with proper protection of transplant surgeons and medical staffs.By ensuring instant seamless care for both patients and medical teams,the goal of reducing the mortality rate and salvaging the lives of patients with COVID-19 can be attained. | Jing-Yu Chen Kun Qiao Feng Liu Bo Wu Xin Xu Guo-Qing Jiao Rong-Guo Lu Hui-Xing Li Jin Zhao Jian Huang Yi Yang Xiao-Jie Lu Jia-Shu Li Shu-Yun Jiang Da-Peng Wang Chun-Xiao Hu Gui-Long Wang Dong-Xiao Huang Guo-Hui Jiao Dong Wei Shu-Gao Ye Jian-An Huang Li Zhou Xiao-Qin Zhang Jian-Xing He | 2020 | Chinese Medical Journal2020,,12: | 19 |
| 6 | A minimally invasive alternative for the treatment of nutcracker syndrome using individualized three-dimensional printed extravascular titanium stents显示文摘Background: This study was designed to evaluate the clinical and radiographic outcomes of patients with nutcracker syndrome (NCS) who were treated with three-dimensional printing (3DP) extravascular titanium stents (EVTSs). The 3DP EVTS was expected to release the hypertension of the left renal vein (LRV) produced by its compression between the superior mesenteric artery (SMA) and the aorta without causing any complications. Method: The pre-operative kidney model of each patient was printed out to enable surgical planning. After that, the EVTS was designed based on the LRV’s primitive physiologic structure using computer-aided design software, and each stent was printed out with a precision setting of 20 μm. Seventeen patients who had been suffering from NCS underwent laparoscopic 3DP EVTS placement. The surgical procedure was designed for the placement of EVTS, taking great care in positioning and fixing the stent. Surgical data, which included patient demographic characteristics as well as pre- and post-operative test results, were collected and analyzed. Results: The mean duration of surgery was 75 ± 9 min, and the mean blood loss was 20 ± 5 mL. Computed tomography examinations revealed that the pre- and post-operative angle between the SMA and the aorta ranged from 18.7°± 4.3° to 48.0°± 8.8°(P < 0.05);in patients with left varicocele, the mean diameter of the left spermatic vein ranged from 3.7 ± 0.5 to 1.3 ± 0.2 mm (P < 0.05). Moreover, Doppler ultrasound examinations showed that the peak velocity of blood flow at the hilar area ranged from 12.4 ± 3.3 to 18.5 ± 3.4 cm/s (P < 0.05). No side effects were observed in the 24 to 42 months following surgery. | 10 | Damage characteristics and constitutive modeling of the 2D C/Si C composite: Part I – Experiment and analysis显示文摘This paper reports an experimental investigation on the macroscopic mechanical behaviors and damage mechanisms of the plain-woven(2D) C/Si C composite under in-plane on- and offaxis loading conditions. Specimens with 15, 30, and 45 off-axis angles were prepared and tested under monotonic and incremental cyclic tension and compression loads. The obtained results were compared with those of uniaxial tension, compression, and shear specimens. The relationships between the damage modes and the stress state were analyzed based on scanning electronic microscopy(SEM) observations and acoustic emission(AE) data. The test results reveal the remarkable axial anisotropy and unilateral behavior of the material. The off-axis tension test results show that the material is fiber-dominant and the evolution rate of damage and inelastic strain is accelerated under the corresponding combined biaxial tension and shear loads. Due to the damage impediment effect of compression stress, compression specimens show higher mechanical properties and lower damage evolution rates than tension specimens with the same off-axis angle. | 11 | Damage characteristics and constitutive modeling of the 2D C/SiC composite: Part Ⅱ–Material model and numerical implementation显示文摘In this work, a macroscopic non-linear constitutive model accounting for damage, inelastic strain and unilateral behavior is proposed for the 2D plain-woven C/Si C composite. A set of scalar damage variables and a new thermodynamic potential expression are introduced in the framework of continuum damage mechanics. In the deduced constitutive equations, the material's progressive damage deactivation behavior during the compression loading is described by a continuous function, and different deactivation rates under uniaxial and biaxial compression loadings are also considered. In damage evolution laws, the coupling effect among the damage modes and impediment effect of compression stress on the development of shear damage in different plane stress states are taken into account. Besides, the general plasticity theory is applied to describing the evolution of inelastic strain in tension and/or shear stress state. The Tsai–Wu failure criterion is adopted for strength analysis. | 12 | Research on the Influence of Cutting Condition on the Surface Microstruct ure of Ultra-thin Wall Parts in Ultrasonic Vibration Cutting显示文摘In many fields of high-tech industry the ultra-t hi n wall parts are employed. In this paper the experiments were carried out to dis cuss the surface microstructure of the camera’s guided drawtube by applying ult rasonic vibration cutting device to the traditional lathe. The influence rule of the cutting condition on the surface roughness was put forward, which was drawn by comparing the ultrasonic cutting with the common cutting by use of the cemen ted carbide tool and the polycrystalline diamond (PCD) tool. The test results sh owed that the ultrasonic cutting performs better than the common cutting in the same condition. According to the test results analyzing, the surface characteriz ation is influenced clearly by the rigidity of the acoustic system and the machi ne tool, as well the setting height of the tool tip. Otherwise, the dense regula r low frequency vibration ripples will be scraped on the machined surface. When the tool tip is set higher than the rotating center of the work piece by three t imes of the amplitude of ultrasonic vibration, the vibration ripples behave alig ht; they turn light and shade alternatively when the tool tip is lower than the rotating center of the work piece by three times of the amplitude of ultrasonic vibration. According to the test result analyzing, the following conclusions are put forward: 1) The surface roughness in ultrasonic cutting is better than that in common cutting. Under a one third critical cutting velocity, the value of th e surface roughness in ultrasonic cutting rise slightly along with the cutting v elocity, while in common cutting it decreases contrast to the cutting velocity; the curves of the surface roughness in ultrasonic cutting and common cutting see m to be alike, both increase along with the feed rate and the cutting depth, but the value in ultrasonic cutting is smaller in the same condition.2) The influen ce of the coolant on the surface roughness cannot be ignored. The kerosene can b e employed to improve the surface roughness in ultrasonic machining.3) In ultras onic cutting process of aluminum alloy ultra-thin wall work piece, the PCD tool performs better than the cemented carbide tools.4) The vibration ripples result from the not enough rigidity of the acoustic system and the improper setting he ight of the tool tip. | 13 | Electroactive barium titanate coated titanium scaffold improves osteogenesis and osseointegration with low-intensity pulsed ultrasound for large segmental bone defects显示文摘For large segmental bone defects,porous titanium scaffolds have some advantages,however,they lack electrical activity which hinders their further use.In this study,a barium titanate(BaTiO3)piezoelectric ceramic was used to modify the surface of a porous Ti6Al4V scaffold(pTi),which was characterized by scanning electron microscopy,energy dispersive spectroscopy,X-ray photoelectron spectroscopy,and roughness and water contact angle analyses.Low intensity pulsed ultrasound(LIPUS)was applied in vitro and in vivo study.The activity of bone marrow mesenchymal stem cells,including adhesion,proliferation,and gene expression,was significantly superior in the BaTiO3/pTi,pTi+LIPUS,and BaTiO3/pTi+LIPUS groups than in the pTi group.The activity was also higher in the BaTiO3/pTi+LIPUS group than in the BaTiO3/pTi and pTi+LIPUS groups.Additionally,micro-computed tomography,the mineral apposition rate,histomorphology,and the peak pull-out load showed that these scaffold conditions significantly enhanced osteogenesis and osseointegration 6 and 12 weeks after implantation in large segmental bone defects in the radius of rabbits compared with those resulting from the pTi condition.Consequently,the improved osteogenesis and osseointegration make the BaTiO3/pTi+LIPUS a promising method to promote bone regeneration in large segmental bone defects for clinical application. | Bo Fan Zheng Guo Xiaokang Li Songkai Li Peng Gao Xin Xiao Jie Wu Chao Shen Yilai Jiao Wentao Hou | 2020 | Bioactive Materials2020,5,4: | 9 |

| 15 | Dispersion polymerization of anionic polyacrylamide in an aqueous salt medium显示文摘Anionic polyacrylamide dispersions were prepared by dispersion polymerization in an aqueous salt medium, using acrylamide(AM) and acrylic acid(AA) as monomers and anionic polyelectrolytes as stabilizer. Effects of salt concentration, and molecular weight and concentration of stabilizers on the stability of the dispersions were investigated using a HAAKE rheometer and optical microscopy. The results showed that stable anionic polyacrylamide dispersions, consisting of smooth, spherical, polydisperse particles, could be obtained under the conditions of salt concentration ranging from 26 wt% to 30 wt%, concentration of stabilizers from 1.2 wt% to 1.8 wt%, and intrinsic viscosity of stabilizers from 2.98 dL·g-1 to 3.74 dL·g-1 . | 19 | Remaining useful life prediction based on variation coefficient consistency test of a Wiener process显示文摘High-cost equipment is often reused after maintenance, and whether the information before the maintenance can be used for the Remaining Useful Life(RUL) prediction after the maintenance is directly determined by the consistency of the degradation pattern before and after the maintenance.Aiming at this problem, an RUL prediction method based on the consistency test of a Wiener process is proposed.Firstly, the parameters of the Wiener process estimated by Maximum Likelihood Estimation(MLE) are proved to be biased, and a modified unbiased estimation method is proposed and verified by derivation and simulations.Then, the h statistic is constructed according to the reciprocal of the variation coefficient of the Wiener process, and the sampling distribution is derived.Meanwhile, a universal method for the consistency test is proposed based on the sampling distribution theorem, which is verified by simulation data and classical crack degradation data.Finally, based on the consistency test of the degradation model, a weighted fusion RUL prediction method is presented for the fuel pump of an airplane, and the validity of the presented method is verified by accurate computation results of real data, which provides a theoretical and practical guidance for engineers to predict the RUL of equipment after maintenance. | Juan LI Bo JING Hongde DAI Xiaoxuan JIAO Xiaodong LIU | 2018 | Chinese Journal of Aeronautics2018,31,1: | 5 |
| 20 | Supramolecular self-assembly and controllable drug release of thermosensitive hyperbranched multiarm copolymers显示文摘A novel temperature-responsive hyperbranched multiarm copolymer with a hydrophobic hyperbranched poly(3-ethyl-3-(hydroxymethyl)oxetane)(HBPO) core and thermosensitive poly(N-isopropylacrylamide)(PNIPAM) arms was synthesized via the atom transfer radical polymerization(ATRP) of NIPAM monomers from a hyperbranched HBPO macroinitiator.It was found that HBPO-star-PNIPAM self-assembled into multimolecular micelles(around 60 nm) in water at room temperature according to pyrene probe fluorescence spectrometry,1H NMR,TEM,and DLS measurements.The micelle solution showed a reversible thermosensitive phase transition at a lower critical solution temperature(LCST)(around 32°C) observed by variable temperature optical absorbance measurements.Variable temperature NMR and DLS analyses demonstrated that the LCST transition originated from the secondary aggregation of the micelles driven by increasing hydrophobic interaction due to the dehydration of PNIPAM shells upon heating.The drug loading and release properties of HBPO-star-PNIPAM micelles were also investigated using prednisone acetate as a model drug.The micelles showed a much improved drug encapsulation efficiency and temperature-dependent sustainable release behavior due to the special micellar structure.The micelles exhibited no apparent cytotoxicity against human HeLa cells. | GUO Bo,SUN XiaoYi,ZHOU YongFeng & YAN DeYue College of Chemistry and Chemical Engineering,Shanghai Jiao Tong University,Shanghai 200240,China | 2010 | Science China Chemistry2010,53,3: | 5 |
