2Herbal formula of Bushen Jianpi(补肾健脾方)combined with sorafenib inhibits hepatocellular carcinoma growth by promoting cell apoptosis and blocking the cell cycle显示文摘OBJECTIVE:To investigate the efficacy of an herbal formula of Bushen Jianpi(补肾健脾方,BSJP)combined with sorafenib on hepatocellular carcinoma(HCC)in vitro and in vivo,and to study the underlying mechanisms of action.METHODS:BSJP,a mixture of 12 raw herbs,was extracted in 70%alcohol/30%water and freeze-dried into a powder.The in vitro effects of BSJP alone,sorafenib alone,and their combination on cell survival,apoptosis,and cell cycle distribution were evaluated in HCC cell lines HCCLM3,HepG2,and SMMC-7721.The expression of B-cell lymphoma-2(Bcl-2),caspase-3,and caspase-9 in HCCLM3 cells was measured using Western blots after drug administration.The in vivo effects of BSJP and sorafenib were evaluated in a tumor surgical resection model using 4-week old male athymic BALB/c nude mice injected with HCCLM3 cells.Immunohistochemical analysis of tumor tissues was performed to evaluate the effects of BSJP alone,sorafenib alone,and their combination on the expression of caspase-3,caspase-9,and Bcl-2.RESULTS:BSJP decreased the survival rate of HCC cell lines,and the combination of BSJP and sorafenib further decreased the survival rate.BSJP significantly promoted cell apoptosis and blocked cell-cycle progression in HCCLM3,HepG2,and SMMC-7721 cells in a dose-dependent manner.Furthermore,the administration of BSJP and sorafenib inhibited the growth of HCCLM3 cell xenografts in nude mice,with no reduction in body weight.In vivo and in vitro experiments showed that BSJP combined with sorafenib could significantly decrease the expression of Bcl-2.CONCLUSION:Our findings suggest that the herbal formula of BSJP is a potential HCC antitumor agent.ZHOU Zhangjie LIU Xinhua WU Tingting QUE Zujue WU Zhonghua WU Weizhong FU Shujuan ZHANG Shiqiang YANG Yun JIANG Haiyan XIA Xiaoting LV Junqiang DU Boqian LI Yun LU Tao ZHANG Zhihui ZHONG Yi 2021Journal of Traditional Chinese Medicine2021,41,2:3
3Ground demonstration system based on in-orbit assembly oriented manipulator flexible force control显示文摘To eliminate the load weight limit of carrier rockets and reduce the burden on support structures,in-orbit assembly is a key technology to make design of scattering a large diameter telescope into submirror modules,which requires smooth operation of assembly robots,and flexible force control technology is necessary. A ground demonstration system is presented for in-orbit assembly focusing on flexible force control. A six-dimensional force/torque sensor and its data acquisition system are used to compensate for gravity. For translation and rotation,an algorithm for flexible control is proposed. A ground transportation demonstration verifies accuracy and smoothness of flexible force control,and the transportation and assembly task is completed automatically. 4Study on the Activation Effect of Fermented Organic Fertilizers on Selenium in the Soil of Southern Dragon Fruit Orchards显示文摘[Objectives]This study was conducted to explore the effects of using fermented organic fertilizers such as cow manure on the activity of selenium in the soil.[Methods]The project team applied the organic fertilizer prepared by fermenting cow manure and other raw materials to activate selenium in the soil at two dragon fruit orchard bases in Shilong Town,Guiping City and Dingdang Town,Long an County,and carried out many years of experiments.[Results]The contents of available selenium in the two experimental orchards in Shilong Town and Dingdang Town increased by 10%and 5%,respectively,and the selenium contents in dragon fruit increased by 228.6%and 200.0%,respectively.[Conclusions]The application of fermented organic fertilizers such as cow manure can increase available selenium content in the soil,thereby increasing the selenium content of dragon fruit.Boqian CHEN Liping PAN Ying XING Jinping CHEN Kun DENG Yongxian LIU 2021Agricultural Biotechnology2021,10,5:1

6Construction of Oryza sativa genome contigs by fingerprint strategy显示文摘We described the construction of BAC contigs of the genome of a indica variety of Oryza sativa, Guang Lu Ai 4.An entire representative (sixfold coverage of rice chromosomes) and genetically stable BAC library of rice genome constructed in this lab has been systematically analysed by restriction enzyme fragmentation and polyacrylamide gel electrophoresis. And all the images thus obtained were subject to image-processing, which consisted of preliminary location of bands, cooperative tracking of lanesby correlation of adjacent bands, a precise densitometric pass, alignment at the marker bands with the standard,optional interactive editing, and normalization of the accepted bands. The contigs were generated based on the Computer Software specially designed for genome map ping. The number of contigs with 600 kb in length on average was 464; of contigs with 1000 kb in length on average was 107; of contigs with 1500 kb in length on average was 23. Therefore, all the contigs we have obtained amounted up to 420 megabases in length. Considering the size of rice genome (430 megabased), the contigs generated in this lab have covered nearly 98% of the rice genome. 7Tree-ring response of Larix chinensis on regional climate and sea-surface temperature variations in alpine timberline in the Qinling Mountains显示文摘Tree-ring width chronologies of Larix chinensis were developed from the northern and southern slopes of the Qinling Mountains in Shaanxi Province,and climatic factors affecting the tree-ring widths of L.chinensis were examined.Correlation analysis showed that similar correlations between tree-ring width chronologies and climatic factors demonstrated that radial growth responded to climate change on both slopes.The radial growth of L.chinensis was mainly limited by temperature,especially the growing season.In contrast,both chronologies were negatively correlated with precipitation in May of the previous year and April of the current year.Spatial climate-correlation analyses with gridded land-surface climate data revealed that our tree-ring width chronologies contained a strong regional temperature signal over much of northcentral and eastern China.Spatial correlation with seasurface temperature fields highlights the influence of the Pacific Ocean,Indian Ocean,and North Atlantic Ocean.Wavelet coherence analysis indicated the existence of some decadal and interannual cycles in the two tree-ring width chronologies.This may suggest the influences of El Nin˜o-Southern Oscillation and solar activity on tree growth in the Qinling Mountains.These findings will help us understand the growth response of L.chinensis to climate change in the Qinling region,and they provide critical information for future climate reconstructions based on this species in semi-humid regions.Boqian Yan Jian Yu Qijing Liu Lihua Wang Lile Hu 2020Journal of Forestry Research2020,31,1:1
8Forest recovery after clear-cutting in Chinese pine(Pinus tabuliformis) plantations of North China显示文摘In Hebei Province of North China, forest was recovered with natural recruitment in plantations with large area of clear-cutting Chinese pine(Pinus tabuliformis). This study was aimed to demonstrate the dynamic characteristics of recruits during the natural recruitment. Both plot survey and the spatial point-pattern analysis were performed. Five developmental stages of natural recruitment were selected and studied, including 1 year before and 2, 5, 8, and 11 years after clear-cutting. Different slope aspects were also included. Natural recruitment was always dominated by Chinese pine with a proportion of higher than 90%. For plots of 1 year before clear-cutting on east-and north-facing slopes, recruit densities were 7886 and 5036 stems/hm2, the average heights were 0.78(±0.85) and 1.06(±1.15) m, and the average diameters at breast height(DBH) were 3.21(±1.38) and 2.91(±1.38) cm, respectively. After clear-cutting, recruit density was initially increased, then it was gradually declined with time; however, the variation of average DBH was contrary to that of recruit density. Both of them were no longer varied between 8 and 11 years after clear-cutting. The average height of recruits continued to increase after clear-cutting. For the plots of 11 years after clear-cutting on east-and north-facing slopes, average heights of recruits reached 2.00(±1.14) and 2.24(±1.20) m, respectively. The statuses of recruits on north-facing slopes were better than those on east-facing slopes after clear-cutting. Meanwhile, recruits on east-facing slopes were always aggregated at small scales, while spatial pattern of recruits varied with time on north-facing slopes. Moreover, forest was recovered more quickly by natural recruitment than by artificial afforestation after clear-cutting. The structural diversity was higher in naturally regenerated forests than in plantations of the same age. 14The Tianlin Mission:A 6m UV/Opt/IR Space Telescope to Explore Habitable Worlds and the Universe显示文摘It is expected that ongoing and future space-borne planet survey missions including Transiting Exoplanet Survey Satellite(TESS),PLATO and Earth 2.0 will detect thousands of small to medium-sized planets via the transit technique,including over a hundred habitable terrestrial rocky planets.To conduct a detailed study of these terrestrial planets,particularly the cool ones with wide orbits,the exoplanet community has proposed various follow-up missions.The currently proposed European Space Agency mission Ariel is the first step for this purpose,and it is capable of characterization of planets down to warm super-Earths mainly using transmission spectroscopy.The NASA Large Ultraviolet/Optical/Infrared Surveyor mission proposed in the Astro2020 Decadal Survey white paper will endeavor to further identify habitable rocky planets,and is expected to launch around 2045.In the meanwhile,China is funding a concept study of a 6 m class space telescope named Tianlin(a UV/Opt/NIR large aperture space telescope)that aims to start its operation within the next 10–15 yr and last for 5+yr.Tianlin will be primarily aimed at the discovery and characterization of rocky planets in the habitable zones around nearby stars and to search for potential biosignatures mainly using the direct imaging method.Transmission and emission spectroscopy at moderate to high resolution will be carried out as well on a population of exoplanets to strengthen the understanding of the formation and evolution of exoplanets.It will also be utilized to perform in-depth studies of the cosmic web and early galaxies,and constrain the nature of dark matter and dark energy.We describe briefly the primary scientific motivations and main technical considerations based on our preliminary simulation results.We find that a monolithic off-axis space telescope with primary mirror diameter larger than 6 m equipped with a high contrast coronagraph can identify water in the atmosphere of a habitable-zone Earth-like planet around a Sunlike star.More simulations for the detectability of other key biosignatures including O_(3),O_(2),CH_(4)and chlorophyll are coming.Wei Wang Meng Zhai Gang Zhao Shen Wang Jifeng Liu Jin Chang Xuejun Zhang Jihong Dong Boqian Xu Frank Grupp 2023Research in Astronomy and Astrophysics2023,23,9:0
15Determination of Polynomial Degree in the Regression of Drug Combinations显示文摘Studies on drug combinations are becoming more and more popular in the past few decades, with the development of computer and algorithms. One of the most common methods in optimizing drug combinations is regression of a polynomial model based on certain number of experimental observations. In this paper, we study how to determine the degree of polynomials in different circumstances of drug combination optimization. Using cross-validation, we have found that in most cases, a high degree results in failures of accurate prediction, named overfitting. 16Revisiting the space-time gradient method:A time-clocking perspective, high order difference time discretization and comparison with the harmonic balance method显示文摘This paper revisits the Space-Time Gradient(STG) method which was developed for efficient analysis of unsteady flows due to rotor–stator interaction and presents the method from an alternative time-clocking perspective. The STG method requires reordering of blade passages according to their relative clocking positions with respect to blades of an adjacent blade row. As the space-clocking is linked to an equivalent time-clocking, the passage reordering can be performed according to the alternative time-clocking. With the time-clocking perspective, unsteady flow solutions from different passages of the same blade row are mapped to flow solutions of the same passage at different time instants or phase angles. Accordingly, the time derivative of the unsteady flow equation is discretized in time directly, which is more natural than transforming the time derivative to a spatial one as with the original STG method. To improve the solution accuracy, a ninth order difference scheme has been investigated for discretizing the time derivative. To achieve a stable solution for the high order scheme, the implicit solution method of Lower-Upper Symmetric GaussSeidel/Gauss-Seidel(LU-SGS/GS) has been employed. The NASA Stage 35 and its blade-countreduced variant are used to demonstrate the validity of the time-clocking based passage reordering and the advantages of the high order difference scheme for the STG method. 20Antigenic Drift of the Hemagglutinin from an Influenza A(H1N1) pdm09 Clinical Isolate Increases its Pathogenicity In Vitro显示文摘The influenza A(H1 N1)pdm09 virus emerged in 2009 and has been continuously circulating in humans for over ten years.Here,we analyzed a clinical influenza A(H1 N1)pdm09-infected patient case hospitalized for two months in Guangdong(from December 14,2019 to February 15,2020).This isolate,named A/Guangdong/LCF/2019(LCF/19),was genetically sequenced,rescued by reverse genetics,and phylogenetically analyzed in the context of other relevant pdm09 isolates.Compared with earlier isolates,this pdm09 virus’s genetic sequence contains four substitutions,S186 P,T188 I,D190 A,and Q192 E,of the hemagglutinin(HA)segment at position 186–192(H3 numbering)in the epitope Sb,and two of which are located at the 190-helix.Phylogenetic analysis indicated that the epitope Sb started undergoing a rapid antigenic change in2018.To characterize the pathogenicity of this novel substitution motif,a panel of reassortant viruses containing the LCF/2019 HA segment or the chimeric HA segment with the four substitutions were rescued.Kinetic growth data revealed that the reassortant viruses,including the LCF/2019 with the PTIAAQE substitution,propagated faster than those rescued ones having the STTADQQ motif in the epitope Sb in Madin-Darby Canine Kidney(MDCK)cells.The HI test showed that the binding activity of escape mutant to 2018 pdm09 sera was weaker than GLW/2018,suggesting that old vaccines might not effectively protect people from infection.Due to the difference in the selection of vaccine strains,people vaccinated in the southern hemisphere could still suffer a severe infection if infected with this antigenic drift pdm09 virus.Lei Xing Yunbo Chen Boqian Chen Ling Bu Ying Liu Zhiqi Zeng Wenda Guan Qigao Chen Yongping Lin Kun Qin Honglin Chen Xilong Deng Xinhua Wang Wenjun Song 2021Virologica Sinica2021,36,5:0
