1A multi-locus backbone tree for Pestalotiopsis, with a polyphasic characterization of 14 new species显示文摘Pestalotiopsis is a taxonomically confused,pathogenic and chemically creative genus requiring a critical reexamination using a multi-gene phylogeny based on ex-type and ex-epitype cultures.In this study 40 isolates of Pestalotiopsis,comprised of 28 strains collected from living and dead plant material of various host plants from China were studied by means of morphology and analysis of ITS,β–tubulin and tef1 gene sequence data.Based on molecular and morphological data we describe 14 new species(Pestalotiopsis asiatica,P.chinensis,P.chrysea,P.clavata,P.diversiseta,P.ellipsospora,P.inflexa,P.intermedia,P.linearis,P.rosea,P.saprophyta,P.umberspora,P.unicolor and P.verruculosa)and three species are epitypified(P.adusta,P.clavispora and P.foedans).Of the 10 gene regions(ACT,β-tubulin,CAL,GPDH,GS,ITS,LSU,RPB 1,SSU and tef1)utilized to resolve cryptic Pestalotiopsis species,ITS,β–tubulin and tef1 proved to be the better markers.The other gene regions were less useful due to poor success in PCR amplification and/or in their ability to resolve species boundaries.As a single gene tef1 met the requirements for an ideal candidate and functions well for species delimitation due to its better species resolution and PCR success.Althoughβ-tubulin showed fairly good differences among species,a combination of ITS,β-tubulin and tef1 gene data gave the best resolution as compared to single gene analysis.This work provides a backbone tree for 22 ex-type/epitypified species of Pestalotiopsis and can be used in future studies of the genus.Sajeewa S.N.Maharachchikumbura Liang Dong Guo Lei Cai Ekachai Chukeatirote Wen Ping Wu Xiang Sun Pedro W.Crous D.Jayarama Bhat Eric H.C.McKenzie Ali H.Bahkali Kevin D.Hyde 2012Fungal Diversity2012,,5:9
2Effects of uniconazole with or without micronutrient on the lignin biosynthesis,lodging resistance,and winter wheat production in semiarid regions显示文摘Lodging stress results in grain yield and quality reduction in wheat. Uniconazole, a potential plant growth regulator significantly enhances lignin biosynthesis and thus provides mechanical strength to plants in order to cope with lodging stress. A field study was conducted during the 2015–2016 and 2016–2017 growing seasons, to investigate the effects of uniconazole sole application or with micronutrient on the lignin biosynthesis, lodging resistance, and production of winter wheat. In the first experiment, uniconazole at concentrations of 0(CK), 15(US1), 30(US2), and 45(US3) mg L^-1 was applied as sole seed soaking, while in the second experiment with manganese(Mn) at concentration of 0.06 g L^-1 Mn, 0.06 g L^-1 Mn+ 15 mg L^-1 uniconazole(UMS1), 0.06 g L^-1 Mn+30 mg L^-1 uniconazole(UMS2), and 0.06 g L^-1 Mn+45 mg L^-1 uniconazole(UMS3), respectively. Uniconazole sole application or with micronutrient significantly increased the lignin content by improving the lignin-related enzyme activities of phenylalanine ammonia-lyase, cinnamyl alcohol dehydrogenase, tyrosine ammonialyase, and peroxidase, ameliorating basal internode characteristics, and breaking strength. The spike length, spike diameter, spikes/plant, weight/spike, yield/spike, and grain yield increased and then decreased with uniconazole application at a higher concentration, where their maximum values were recorded with UMS2 and US2 treatments. The lignin accumulation was positively correlated with lignin-related enzyme activities and breaking strength while, negatively correlated with lodging rate. 3Use of genotype-environment interactions to elucidate the pattern of maize root plasticity to nitrogen deficiency显示文摘Maize(Zea mays L.) root morphology exhibits a high degree of phenotypic plasticity to nitrogen(N) de ficiency,but the underlying genetic architecture remains to be investigated Using an advanced BC_4F_3 population,we investigated the root growth plasticity under two contrasted N levels and identi fied the quantitative trait loci(QTLs) with QTL-environment(Q×E)interaction effects. Principal components analysis(PCA) on changes of root traits to N de ficiency(D LN-HN) showed that root length and biomass contributed for 45.8% in the same magnitude and direction on the first PC,while root traits scattered highly on PC_2 and PC_3. Hierarchical cluster analysis on traits for D LN-HN further assigned the BC_4F_3 lines into six groups,in which the special phenotypic responses to N de ficiency was presented These results revealed the complicated root plasticity of maize in response to N de ficiency that can be caused by genotype environment(G×E) interactions. Furthermore,QTL mapping using a multi-environment analysis identi fied 35 QTLs for root traits. Nine of these QTLs exhibited signi ficant Q×E interaction effects. 6Exosomes Released from Rabies Virus-Infected Cells May be Involved in the Infection Process显示文摘Exosomes are cell-derived vesicles that are secreted by many eukaryotic cells. It has recently attracted attention as vehicles of intercellular communication. Virus-infected cells release exosomes, which contain viral proteins, RNA, and pathogenic molecules. However, the role of exosomes in virus infection process remains unclear and needs to be further investigated.In this study, we aimed to evaluate the effects of exosomes on rabies virus infection. OptiPrep^(TM) density gradient centrifugation was used to isolate exosomes from rabies virus-infected cell culture supernatants. A rabies virus G protein enzyme-linked immunosorbent assay and acetylcholinesterase activity assays were performed to verify the centrifugation fractions. Exosomes were then characterized using transmission electron microscopy and Western blotting. Our results showed that rabies virus infection increased the release of exosomes. Treatment with GW4869 and si-Rab27 a, two exosomal secretion inhibitors, inhibited exosome release. Furthermore, the inhibitors reduced the levels of extracellular and intracellular viral RNA. These data indicated that exosomes may participate in the viral infection process. 8Serological Investigation of Laboratory-Confirmed and Suspected Ebola Virus Disease Patients During the Late Phase of the Ebola Outbreak in Sierra Leone显示文摘This study aimed to investigate the serological characteristics of Ebola virus(EBOV) infection during the late phase of the Ebola outbreak in Sierra Leone. In total, 877 blood samples from 694 suspected Ebola virus disease(EVD) cases assessed from March to December 2015, were analyzed via real-time reverse transcription polymerase chain reaction(RT-PCR) for viral RNA and enzyme-linked immunosorbent assay(ELISA) and Luminex to detect antibodies against EBOV. Viral load and EBOV-specific IgM/IgG titers displayed a declining trend during March to December 2015. Viral RNA load decreased rapidly at earlier stages after disease onset, while EBOV-specific IgM and IgG still persisted in 58.1%(18/31) and 93.5%(29/31) of the confirmed EVD patients and in 3.8%(25/663) and 17.8%(118/663) of the RNA-negative suspected patients in the later phase, respectively. 9Synthesis and characterization of caprolactone based polyurethane with degradable and antifouling performance显示文摘In this work,a degradable polyurethane composed of caprolactone(CL)and L-Lactide(LLA)as soft segments,and 4,40-methylenebis(cyclohexyl isocyanate)(H12 MDI)and polytetramethylene ether glycol(PTMEG)as hard segments,was prepared.Hydrolytic degradation experiment revealed that the degradable polyurethane(PU)could be degraded in artificial seawater.It also showed that caprolactone-copolyurethane(CL-PU)copolymer with higher crystallinity degraded much slower in artificial seawater.However,the introduction of LLA resulted in an increase in the hydrophilicity and reduction in the crystallinity of degradable PU,as demonstrated by the contact angle analysis.The result of the scanning electron microscope showed that the surface of degradable PU renewed under static condition.Moreover,degradable PU was able to be used as a carrier,and it controlled the release rate of 4,5-dichloro-2-octyl-isothiazolone(DCOIT).The anti-diatom(Navicula incerta)test demonstrated that the(caprolactone-co-L-lactide)-co-polyurethane 4(CL/LAx-PU4)with DCOIT contents prevented the adhesion of diatom Navicula incerta(88.37%reduction)due to their self-polishing and the release of antifoulants.Therefore,the degradable PU consisted of CL,LLA,and DCOIT could be a durable resin with good antifouling activity for the application in the marine anti-biofouling field.Abid Ali Lina Song Jiankun Hu Jingxian Jiang Qingqing Rao Muhammad Shoaib Shah Fahad Yongjie Cai Xiaoli Zhan Fengqiu Chen Qinghua Zhang 2021Chinese Journal of Chemical Engineering2021,34,6:2
10Genetic variation in LBL1 contributes to depth of leaf blades lobes between cotton subspecies, Gossypium barbadense and Gossypium hirsutum显示文摘Leaf is a essential part of the plants for photosynthetic activities which mainly economize the resources for boll heath. Significant variations of leaf shapes across the Gossypium sp. considerably influence the infiltration of sunlight for photosynthesis. To understand the genetic variants and molecular processes underlying for cotton leaf shape, we used F_2 population derived from upland cotton genotype P30 A(shallow-lobed leaf) and sea-island cotton genotype ISR(deeplobed leaf) to map leaf deep lobed phenotype controlling genes LBL1 and LBL2. Genetic analysis and localization results have unmasked the position and interaction between both loci of LBL1 and LBL2, and revealed the co-dominance impact of the genes in regulating depth of leaf blades lobes in cotton. LBL1 had been described as a main gene and member of transcription factor family leucine zipper(HD-ZIPI) from a class I homologous domain factor Gorai.002 G244000. The qRT-PCR results elaborated the continuous change in expression level of LBL1 at different growth stages and leaf parts of cotton. Higher expression level was observed in mature large leaves followed by medium and young leaves respectively. For further confirmation, plants were tested from hormonal induction treatments, which explained that LBL1 expression was influenced by hormonal signaling. 11Revealing the linguistic and geographical disparities of public awareness to Covid-19 outbreak through social media显示文摘The Covid-19 has presented an unprecedented challenge to public health worldwide.However,residents in different countries showed diverse levels of Covid-19 awareness during the outbreak and suffered from uneven health impacts.This study analyzed the global Twitter data from January 1st to June 30th,2020,to answer two research questions.What are the linguistic and geographical disparities of public awareness in the Covid-19 outbreak period reflected on social media?Does significant association exist between the changing Covid-19 awareness and the pandemic outbreak?We established a Twitter data mining framework calculating the Ratio index to quantify and track awareness.The lag correlations between awareness and health impacts were examined at global and country levels.Results show that users presenting the highest Covid-19 awareness were mainly those tweeting in the official languages of India and Bangladesh.Asian countries showed more disparities in awareness than European countries,and awareness in Eastern Europe was higher than in central Europe.Finally,the Ratio index had high correlations with global mortality rate,global case fatality ratio,and country-level mortality rate,with 21-31,35-42,and 13–18 leading days,respectively.This study yields timely insights into social media use in understanding human behaviors for public health research.Binbin Lin Lei Zou Nick Duffield Ali Mostafavi Heng Cai Bing Zhou Jian Tao Mingzheng Yang Debayan Mandal Joynal Abedin 2022International Journal of Digital Earth2022,15,1:2

14Rational design of eco-friendly Mn-doped nonstoichiometric CuInSe/ZnSe core/shell quantum dots for boosted photoelectrochemical efficiency显示文摘Colloidal core/shell quantum dots(QDs)with environment-friendly feature and controllable optoelectronic properties are promising building blocks in emerging solar technologies.In this work,we rationally design and tailor the eco-friendly CuInSe(CISe)/ZnSe core/shell QDs by Mn doping and stoichiometric optimization(i.e.,molar ratios of Cu/In).It is demonstrated that Mn doping in In-rich CISe/ZnSe core/shell QDs can effectively engineer the charge kinetics inside the QDs,enabling efficient photogenerated electrons transfer into the shell for retarded charge recombination.As a result,a solar-driven photoelectrochemical(PEC)device fabricated using the optimized Mn-doped In-rich CISe/ZnSe core/shell QDs(Cu/In ratio of 1/2)exhibits improved charge extraction and injection,showing a~3.5-fold higher photocurrent density than that of the pristine CISe/ZnSe core/shell QDs under 1 sun AM 1.5G illumination.The findings indicate that transition metal doping in“green”nonstoichiometric core/shell QDs may offer a new strategy for achieving high-efficiency solar energy conversion applications.Rui Wang Xin Tong Zhihang Long Ali Imran Channa Hongyang Zhao Xin Li Mengke Cai Yimin You Xuping Sun Zhiming Wang 2022Nano Research2022,15,8:1

20Local nitrogen application increases maize post-silking nitrogen uptake of responsive genotypes via enhanced deep root growth显示文摘Nitrogen(N)is unevenly distributed throughout the soil and plant roots proliferate in N-rich soil patches.However,the relationship between the root response to localized N supply and maize N uptake efficiency among different genotypes is unclear.In this study,four maize varieties were evaluated to explore genotypic differences in the root response to local N application in relation to N uptake.A split-root system was established for hydroponically-grown plants and two methods of local N application(local banding and local dotting)were examined in the field.Genotypic differences in the root length response to N were highly correlated between the hydroponic and field conditions(r>0.99).Genotypes showing high response to N,ZD958,XY335 and XF32D22,showed 50‒63%longer lateral root length and 36‒53%greater root biomass in N-rich regions under hydroponic conditions,while the LY13 genotype did not respond to N.Under field conditions,the root length of the high-response genotypes was found to increase by 66‒75%at 40‒60 cm soil depth,while LY13 showed smaller changes in root length.In addition,local N application increased N uptake at the post-silking stage by 16‒88%in the high-response genotypes and increased the grain yield of ZD958 by 10‒12%.Moreover,yield was positively correlated with root length at 40‒60 cm soil depth(r=0.39).We conclude that local fertilization should be used for high-response genotypes,which can be rapidly identified at the seedling stage,and selection for“local-N responsive roots”can be a promising trait in maize breeding for high nitrogen uptake efficiency.CHEN Zhe REN Wei YI Xia LI Qiang CAI Hong-guang Farhan ALI YUAN Li-xing MI Guo-hua PAN Qing-chun CHEN Fan-jun 2023Journal of Integrative Agriculture2023,22,1:0
