| 2 | Optimization of Protein Extraction and Decoloration Conditions for Tea Residues显示文摘To optimize alkaline method for extracting proteins from tea residue(TR), the effect of extraction conditions on tea protein extraction rate(TPER) was investigated. Single factor experiment showed the extraction temperature 80 °C, extraction time 100 min, p H value 13 and liquid–solid ratio 40:1 as the optimal extraction conditions. The orthogonal test revealed that the maximum TPER reached 29.71% under the following optimal combination of conditions: extraction temperature 70 °C, extraction time 60 min, p H 12 and liquid–solid ratio 50:1. For optimizing the purification of tea residue proteins, isoelectric point precipitation(p I), ammonium sulfate precipitation(a S) and isoelectric point plus ammonium sulfate precipitation(i PAS) were compared. The result showed that the highest protein precipitation rate(PPR) was 89.70% which was generated by using i PAS. | 3 | The Electric Field Detector (EFD) onboard the ZH-1 satellite and firstobservational results显示文摘Previous studies have reported that, before or after occurrences of strong earthquakes, some low earth orbit satellites recorded ionospheric disturbances, including electromagnetic emissions and plasma fluctuations over the epicenter region or its conjugate point.Theoretically speaking, due to some electromagnetic coupling effect, electromagnetic emissions from the earthquake preparation zone could propagate from the lithosphere to the atmosphere, and could reach the ionosphere, even up to the inner magnetosphere. This paper introduces the electric field detector(EFD) onboard the ZhangHeng-1 satellite(ZH-1). The EFD is designed to measure electric field fluctuations within the broad frequency range of DC to 3.5 MHz, divided into 4 channels: ULF(DC–16 Hz), ELF(6 Hz–2.2 kHz), VLF(1.8 kHz–20 kHz) and HF(18 kHz–3.5 MHz). The sampling rates of the channels are 125 Hz, 5 kHz, 50 kHz and 10 MHz, respectively. The EFD includes4 spherical probes mounted on a over 4.5 m boom and an electronic box inside the satellite module. The resolution of the EFD is 1μV·m-1·Hz-1/2 at frequencies from DC to 16 Hz, and the sensitivity is 0.1 μV·m-1·Hz-1/2 at frequencies from 6 Hz to 2.2 kHz, 0.05 μV·m-1·Hz-1/2 in the band 1.8 kHz to 20 kHz, and 0.1μV·m-1·Hz-1/2 from 20 kHz to 3.5 MHz. The dynamic range from DC to 20 kHz is over 120 dB, and over96 dB from 20 kHz to 3.5 MHz. The EFD has two observation modes: survey mode and burst mode. The survey mode concentrates primarily on electric field power density values; the burst mode provides high sampling rate waveform data. The detailed configuration of the EFD onboard the ZH-1 is also introduced in this paper. During the six months' orbit test phase, the EFD recorded a number of natural electromagnetic emissions. | 4 | Neonatal Maternal Deprivation Followed by Adult Stress Enhances Adrenergic Signaling to Advance Visceral Hypersensitivity显示文摘The pathophysiology of visceral pain in patients with irritable bowel syndrome remains largely unknown.Our previous study showed that neonatal maternal deprivation(NMD) does not induce visceral hypersensitivity at the age of 6 weeks in rats. The aim of this study was to determine whether NMD followed by adult stress at the age of 6 weeks induces visceral pain in rats and to investigate the roles of adrenergic signaling in visceral pain. Here we showed that NMD rats exhibited visceral hypersensitivity 6 h and 24 h after the termination of adult multiple stressors(AMSs). The plasma level of norepinephrine was significantly increased in NMD rats after AMSs. Whole-cell patch-clamp recording showed that the excitability of dorsal root ganglion(DRG) neurons from NMD rats with AMSs was remarkably increased. The expression of β_2 adrenergic receptors at the protein and mRNA levels was markedly higher in NMD rats with AMSs than in rats with NMD alone. Inhibition of β_2 adrenergic receptors with propranolol or butoxamine enhanced the colorectal distention threshold and application of butoxamine also reversed the enhanced hypersensitivity of DRG neurons. | 6 | Accumulation of polybrominated diphenyl ethers in breast milk of women from an e-waste recycling center in China显示文摘Polybrominated diphenyl ethers(PBDEs) can be transferred to infants through the ingestion of breast milk, resulting in potential health risk. In this study, PBDEs, hydroxylated polybrominated diphenyl ethers(OH-PBDEs) and 2,2′,4,4′,5,6′-hexachlorobiphenyl(CB-153)in human milk from women living adjacent to e-waste recycling sites of Wenling,China, were investigated. The median level of PBDEs in samples from residents living in the e-waste recycling environment > 20 years(R_20group, 19.5 ng/g lipid weight(lw))was significantly higher than that for residents living in Wenling < 3 years(R_3group,3.88 ng/g lw)(p < 0.05), likely ascribable to specific exposure to PBDEs from e-waste recycling activities. In the R_20 group, most congeners(except for BDE-209) were correlated with each other(p < 0.05). Moreover, CB-153 showed significant association with most PBDE congeners, rather than BDE-209. The relationship indicated that most BDE congeners other than BDE-209 shared common sources and/or pathways with CB-153, e.g., dietary ingestion. The correlations between BDE-209 and other congeners were different in the two groups, likely suggesting their different exposure sources and/or pathways for PBDEs.Although estimated dietary intake of PBDEs for infants via breast milk was lower than the minimum value affecting human health, the PBDE exposure of infants should be of great concern because of their potential effect on the development of neonates over long-term exposure. | 7 | Phyllosticta species associated with citrus diseases in China显示文摘Phyllosticta species associated with diseases of four commercial Citrus species grown in China are reported.Totally,496 Phyllosticta strains were isolated from mandarins(Citrus reticulata),pomeloes(C.maxima),oranges(C.sinensis)and lemons(C.limon)in the main citrus producing regions across China,and 74 strains were selected for phylogenetic analysis.Analyses inferred from the sequences of internal transcribed spacer region(ITS1,5.8S nrDNA and ITS2),partial translation elongation factor 1-alpha(TEF1)and partial actin gene(ACT),showed these representative Phyllosticta isolates clustered in four distinct clades corresponding to three known,and one undescribed species.The newly resolved taxon,Phyllosticta citrichinaensis was isolated from leaves and fruits of all four Citrus species and is introduced in this paper.This taxon caused minor damage,showing irregular spots or freckles.Phyllosticta citriasiana,associated with tan spot of pomeloes,was isolated only from pomeloes,and never from lemons,mandarins and oranges.Phyllosticta citricarpa,the citrus black spot pathogen,which is presently subjected to phytosanitary legislation in the EU and United States,was isolated from lemons,mandarins and oranges,but never from pomeloes.The isolates of P.citricarpa clustered in two subclades,one from mandarins,the other from oranges and lemons.P.capitalensis was isolated from all four Citrus species as an endophyte,causing false melanose,or together with P.citricarpa or P.citriasiana.Morphological,cultural and biochemical characters were consistent with the results of phylogenetic analysis.In addition,a specific primer pair Pca8/ITS4 was designed and selected,and its corresponding PCR procedure was developed for the detection of P.citriasiana in this study. | Xinghong Wang Guoqing Chen Feng Huang Jingze Zhang Kevin D.Hyde Hongye Li | 2012 | Fungal Diversity2012,,1: | 5 |
| 8 | PCL2 regulates p53 stability and functions as a tumor suppressor in breast cancer显示文摘Polycomblike2(PCL2) is a well-known component of polycomb repressive complex 2(PRC2) and plays important roles in H3 K27 methylation and homeotic gene silencing.However,the involvement of PCL2 in breast cancer development remains unclear.Here,we established PCL2 as a tumor suppressor gene in breast cancer.Expression level of PCL2 was significantly downregulated in breast cancer tissue samples observed at different TNM stages.Ectopic expression of PCL2 could significantly inhibit cell proliferation and promoted apoptosis.PCL2 also remarkably elevated levels of p53 and its targets by increasing p53 stability.Mechanistically,PCL2 protected p53 proteins from MDM2-mediated ubiquitination and degradation by sequestering MDM2 into the nucleolus.Overexpression of PCL2 also suppressed migration and invasion by inhibiting epithelial-mesenchymal transition.PCL2 expression was correlated with Ecadherin expression and was inversely correlated with vimentin expression.Furthermore,PCL2 knockdown could attenuate anti-tumor effect of MLN4924.Taken together,our findings indicated that PCL2 played a tumor suppressor role in development and progression of breast cancer and may be a prognostic and predictive marker for breast cancer. | Yiran Liang Yang Yang Renbo Guo Shuang Gao Xinghong Guo Danyang Li Meng Wang Haruhiko Koseki Xiangzhi Li | 2018 | Science Bulletin2018,63,10: | 4 |
| 9 | Materials Experiment on Tiangong-2 Space Laboratory显示文摘During the China's Tiangong-2(TG-2) flight mission, the experiments of 18 kinds of material samples were conducted in space by using a Multiple Materials Processing Furnace(MMPF) mounted in the orbital module of the TG-2 space laboratory. After the experiments of 12 kinds of samples of the first and second batches were completed successfully, astronauts packed and brought them back to the ground by ShenzhouII spacecraft. By studying processing and formation on semiconductor and optoelectronics materials, metal alloys and metastable materials, functional single-crystal, micro-and nano-composite materials encapsulated in sample ampoules both in space and on Earth, we expect to explore some physical and chemical processes and mechanism of the materials formation that are normally obscured and therefore are difficult to study quantitatively on the ground due to the gravity-induced convection, to obtain the processing and synthesis technology for preparing high quality materials, and lead to the improvement and development of materials processing techniques on Earth, and also develop the experiment device and comprehensive ability for materials experiment in microgravity environment. | 12 | The maize late embryogenesis abundant protein Zm DHN13 positively regulates copper tolerance in transgenic yeast and tobacco显示文摘Late embryogenesis abundant (LEA) proteins accumulate in the late stage of plant seed development, and are upregulated in most plants during drought, cold, heat, or salinity stress. LEA proteins can be classified by amino-acid sequence into seven groups. Dehydrins belong to LEA protein group Ⅱ. In previous studies, the maize KS type dehydrin ZmDHN13 increased the tolerance of transgenic tobacco to oxidative stress. In the present study, ZmDHN13 was identified under copper stress conditions, and the protein was then characterized using transgenic yeast and tobacco plants to investigate its functions. ZmDHN13 bound Cu2+. Its overexpression in transgenic tobacco conferred tolerance to copper stress by binding metals and reducing the accumulation of reactive oxygen species (ROS). | 13 | Bioavailability and tissue distribution of Dechloranes in wild frogs(Rana limnocharis) from an e-waste recycling area in Southeast China显示文摘Dechlorane Plus(DP), a flame retardant used as an alternative to decabromodiphenylether, has been frequently detected in organisms, indicating its bioaccumulation and biomagnification potential in aquatic and terrestrial species. However, little data is available on the bioaccumulation of DP in amphibians. Dechlorane Plus and its analogs(DPs) were detected in the liver, muscle and brain tissues of wild frogs(Rana limnocharis), which were collected from an e-waste recycling site, Southeast China. DP, Mirex, Dec 602 and a dechlorinated compound of DP(anti-Cl11-DP) varied in the range of 2.01–291, 0.650–179, 0.260–12.4, and not detected(nd)–8.67 ng/g lipid weight, respectively. No difference of tissue distribution was found for syn-DP, Mirex and Dec 602 between the liver and muscle tissue(liver/muscle concentration ratio close to 1, p > 0.05). However, higher retention was observed for anti-DP and anti-Cl11-DP in the frog muscle relative to the liver tissue(liver/muscle concentration ratio < 1, p < 0.05). Additionally, the blood-brain barrier was found to work efficiently to suppress these compounds entering brain tissues in this species(liver/brain concentration ratio > 1, p < 0.05), and the molecular weight was a key factor impacting the extent of the blood-brain barrier. | 17 | Valence electron structure and properties of the ZrO_2显示文摘To reveal the properties of ZrO2 at the atom and electron levels, the valence elec- tron structures of three ZrO2 phases were analyzed on the basis of the empirical electron theory of solids and molecules. The results showed that the hybridization levels of Zr and O atoms in the m-ZrO2 were the same as those in the t-ZrO2, while those in the c-ZrO2 rose markedly. The electron numbers and bond energies on the strongest covalent bonds in the m-ZrO2 phase were the greatest, the values were 0.901106 and 157.5933 kJ/mol, respectively. Those in the t-ZrO2 phase took second place, which were 0.722182 and 123.9304 kJ/mol, and those in the c-ZrO2 phase were the smallest, which were 0.469323 and 79.0289 kJ/mol. According to the product of the bond energy on the strongest covalent bond and equivalent bond number (this value reflected the crystal cohesive energy), the order from the greatness to smallness was the c-ZrO2> t-ZrO2 > m-ZrO2. This showed that the m-phase bonds were the tightest, their energy was the smallest, the crystal cohe- sive energy of the m-phase was the largest, and the m-phase existed most stably at room temperature.
