| 2 | Correlations Between Single Nucleotide Polymorphisms,Cognitive Dysfunction,and Postmortem Brain Pathology in Alzheimer's Disease Among Han Chinese显示文摘In this study, the distribution of five Alzheimer's disease(AD)-related single nucleotide polymorphisms(SNPs) in the Han population was examined in combination with the evaluation of clinical cognition and brain pathological analysis. The associations among SNPs,clinical daily cognitive states, and postmortem neuropathological changes were analyzed in 110 human brains from the Chinese Academy of Medical Sciences/Peking Union Medical College(CAMS/PUMC) Human Brain Bank.APOE ?4(OR = 4.482, P = 0.004), the RS2305421 GG genotype(adjusted OR = 4.397, P = 0.015), and the RS10498633 GT genotype(adjusted OR = 2.375,P = 0.028) were associated with a higher score on the ABC(Ab plaque score, Braak NFT stage, and CERAD neuritic plaque score) dementia scale. | 5 | Distribution characteristics and source tracing of petroleum hydrocarbons in the northeastern South China Sea显示文摘In recent years,oil spills caused by human activities have occurred frequently,and the resultant oil pollution has received extensive attention worldwide.In this paper,a total of 50 water samples were collected from the northeastern part of the South China Sea,and total petroleum hydrocarbons(TPHs)and n-alkane content in the samples were analyzed by gas chromatography-flame ionization detector(GC-FID)technology.The petroleum hydrocarbon characteristic indices,such as carbon predominance index(CPI)and terrigenous/aquatic ratio(TAR),were calculated to trace the source of petroleum hydrocarbons.The measured value of TPHs ranged from 121.31-603.02μg/L.For surface waters,the TPHs in the no rthern coastal waters and the central waters were higher than that in the far shore.For ve rtical waters,the TPHs sharply decreased at first,and then increased slowly and finally reached a steady state.The n-alkanes in the water samples were concentrated in C10-C38,and they were mainly from terrestrial higher plant.The waters in the near shore,mid-layer and deep sea showed a strong reducing terrestrial characteristic,while the surface waters in the open sea showed an obvious oxidizing endogenous characteristic. | Shengying Gong Wenhu Liu Yazhuo Li Jibiao Zhang Changlin Chen Jie Fu | 2020 | Chinese Chemical Letters2020,31,10: | 1 |

| 8 | Allosteric conformational changes of G proteins upon its interaction with membrane and GPCR显示文摘Current resolved structures of GPCRs and G protein complexes provided important insights into G protein activation. However, the binding or dissociation of GPCRs with G protein is instantaneous and highly dynamic in the intracellular environment. The conformational dynamic of G protein still needs to be addressed. In this study, we applied ^(19)F solution NMR spectroscopy to monitor the conformational changes of G protein upon interact with detergent mimicking membrane and receptor. Our results show that there are two states equilibria in the G_(α)in apo states. The interaction of G_(α)with detergents will accelerate this conformational transformation and induce a state that tends to bind to GPCRs. | 9 | A direct spino-cortical circuit bypassing the thalamus modulates nociception显示文摘Nociceptive signals are usually transmitted to layer 4 neurons in somatosensory cortex via the spinothalamic-thalamocortical pathway.The layer 5 corticospinal neurons in sensorimotor cortex are reported to receive the output of neurons in superficial layers;and their descending axons innervate the spinal cord to regulate basic sensorimotor functions.Here,we show that a subset of layer 5 neurons receives spinal inputs through a direct spino-cortical circuit bypassing the thalamus,and thus define these neurons as spino-cortical recipient neurons(SCRNs).Morphological studies revealed that the branches from spinal ascending axons formed a kind of disciform structure with the descending axons from SCRNs in the basilar pontine nucleus(BPN).Electron microscopy and calcium imaging further confirmed that the axon terminals from spinal ascending neurons and SCRNs made functional synaptic contacts in the BPN,linking the ascending sensory pathway to the descending motor control pathway.Furthermore,behavioral tests indicated that the spino-cortical connection in the BPN was involved in nociceptive responses.In vivo calcium imaging showed that SCRNs responded to peripheral noxious stimuli faster than neighboring layer 4 cortical neurons in awake mice.Manipulating activities of SCRNs could modulate nociceptive behaviors.Therefore,this direct spino-cortical circuit represents a noncanonical pathway,allowing a fast sensory-motor transition of the brain in response to noxious stimuli. | Bing Cai Dan Wu Hong Xie Yan Chen Huadong Wang Sen Jin Yuran Song Anan Li Shiqi Huang Sashuang Wang Yingjin Lu Lan Bao Fuqiang Xu Hui Gong Changlin Li Xu Zhang | 2023 | Cell Research2023,33,10: | 0 |
