| 3 | m^6A Regulates Neurogenesis and Neuronal Development by Modulating Histone Methyltransferase Ezh2显示文摘N6-methyladenosine (m6A),catalyzed by the methyltransferase complex consisting of Mettl3 and Mettl14,is the most abundant RNA modification in mRNAs and participates in diverse biological processes. However,the roles and precise mechanisms of m6A modification in regulating neuronal development and adult neurogenesis remain unclear. Here,we examined the function of Mettl3,the key component of the complex,in neuronal development and adult neurogenesis of mice. We found that the depletion of Mettl3 significantly reduced m6A levels in adult neural stem cells (aNSCs) and inhibited the proliferation of aNSCs. Mettl3 depletion not only inhibited neu-ronal development and skewed the differentiation of aNSCs more toward glial lineage,but also affected the morphological maturation of newborn neurons in the adult brain. m6A immunoprecip-itation combined with deep sequencing (MeRIP-seq) revealed that m6A was predominantly enriched in transcripts related to neurogenesis and neuronal development. Mechanistically,m6A was present on the transcripts of histone methyltransferase Ezh2,and its reduction upon Mettl3 knockdown decreased both Ezh2 protein expression and consequent H3K27me3 levels. The defects of neurogenesis and neuronal development induced by Mettl3 depletion could be rescued by Ezh2 overexpression. | 12 | A novel in vitro prognostic model of bladder cancer based on urine-derived living tumor cells显示文摘Bladder cancer(BLCA)remains a difficult malignancy to manage because of its high recurrence,intense follow-up,and invasive diagnostic and treatment techniques.Immune checkpoint inhibitors(ICIs)have forged a new direction for the treatment of BLCA,but it is currently challenging to predict whether an individual patient will be sensitive to ICIs.We collected 43 urine/tumor samples from BLCA patients for primary bladder cancer cells(BCCs)culturing using our previously reported BCC culture platform.We used flow cytometry(FCM)to measure the expression levels of Programmed Death-Ligand 1(PD-L1)on BCCs before and after interferon-gamma(IFN-γ)treatment and found that PD-L1 expression and the sensitivities to IFN-γvaried among patients.RNA-sequencing,western blotting,and programmed death-1(PD-1)binding assays confirmed that the BCC FCM-based PD-L1 detection platform(BC-PD-L1)was reliable and was not hindered by the glycosylation of PD-L1.In the subsequent retrospective study,we found that IFN-γ-stimulated PD-L1(sPD-L1)expression on BCCs detected by BC-PD-L1 could predict the prognosis of BLCA patients.Importantly,the prognostic value was similar or even better in urine-derived BC-PD-L1(UBC-PD-L1).Transcriptome analysis showed that BCCs with high sPD-L1 tended to enrich genes associated with the collagen-containing extracellular matrix,cell–cell adhesion,and positive regulation of the immune system.In addition,the UBC-PD-L1 also exhibited predictive value for ICI response in BLCA patients.In conclusion,as a novel personalized urine-detection method,UBC-PD-L1 may provide a rapid,accurate,and non-invasive tool for monitoring tumor progression,predicting therapeutic responses,and helping improve BLCA clinical treatment in future. | Jiaqi Wang Jiying Zhu Junchi Hu Ziruoyu Wang Xiaobo Wang Jianbo Pan Yiwei Chu Zengxia Li Wei Jiang Chunmin Liang Jun Hou Jianming Guo Yongjun Dang Shuai Jiang | 2023 | Genes & Diseases2023,10,6: | 0 |

| 15 | XCO_2 satellite retrieval experiments in short-wave and infrared spectra with SCIATRAN model for Sahara Desert显示文摘The spectra of O_2 A-band(0.76 μm) and CO_2 near-infrared emissions(1.6 μm) are simulated by the SCIATRAN radiative transfer model(V3.1.23), and compared with those observed by GOSAT-FTS(Greenhouse gases Observing SATellite-Fourier Transform Spectrometer). Systematic deviations between the observed and simulated spectra are found to exist,especially for the O_2 A-band. The discrepancies are characterized by their mean differences averaged over the observed spectral ranges. A correction is applied to the observed GOSAT-FTS L1B(V141.141) spectra by scaling the spectral intensity measured by TANSO-FTS(Thermal and Near infrared Sensor for carbon Observation Fourier Transform Spectrometer) onboard GOSAT.The average columnar CO_2 concentrations(XCO_2) are retrieved from the observed and the corrected GOSAT-FTS spectra by using the SCIATRAN inversion algorithm. Compared with the GOSAT-FTS L2 XCO_2 data products retrieved from the observed spectra of GOSAT-FTS, the SCIATRAN retrievals from the corrected spectra show a much better agreement, with the relative error less than 1%. But the results of GOSAT TANSO-FTS(V161.160) show smaller residuals than GOSAT TANSO-FTS(V141.141) without mean residual correction.
