| 1 | Molecular epidemiology demonstrates that imported and local strains circulated during the 2014 dengue outbreak in Guangzhou, China显示文摘The dengue virus(DENV) is a vital global public health issue. The 2014 dengue epidemic in Guangzhou, China, caused approximately 40,000 cases of infection and five deaths. We carried out a comprehensive investigation aimed at identifying the transmission sources in this dengue epidemic. To analyze the phylogenetics of the 2014 dengue strains, the envelope(E) gene sequences from 17 viral strains isolated from 168 dengue patient serum samples were sequenced and a phylogenetic tree was reconstructed. All 17 strains were serotype Ⅰ strains, including 8genotype Ⅰ and 9 genotype V strains. Additionally, 6 genotype Ⅰ strains that were probably introduced to China from Thailand before 2009 were widely transmitted in the 2013 and 2014 epidemics, and they continued to circulate until 2015, with one affinis strain being found in Singapore. The other 2 genotype Ⅰ strains were introduced from the Malaya Peninsula in 2014. The transmission source of the 9 genotype Ⅴ strains was from Malaysia in 2014. DENVs of different serotypes and genotypes co-circulated in the 2014 dengue outbreak in Guangzhou. | 4 | Differential SW16.1 allelic effects and genetic backgrounds contributed to increased seed weight after soybean domestication显示文摘Although seed weight has increased following domestication from wild soybean(Glycine soja) to cultivated soybean(Glycine max), the genetic basis underlying this change is unclear. Using mapping populations derived from chromosome segment substitution lines of wild soybean, we identified SW16.1 as the causative gene underlying a major quantitative trait locus controlling seed weight.SW16.1 encodes a nucleus-localized LIM domaincontaining protein. Importantly, the GsSW16.1 allele from wild soybean accession N24852 had a negative effect on seed weight, whereas the GmSW16.1 allele from cultivar NN1138-2 had a positive effect. Gene expression network analysis,reverse-transcription quantitative polymerase chain reaction, and promoter-luciferase reporter transient expression assays suggested that SW16.1 regulates the transcription of MT4, a positive regulator of seed weight. The natural variations in SW16.1 and other known seed weight genes were analyzed in soybean germplasm. The SW16.1 polymorphism was associated with seed weight in 247 soybean accessions, showing much higher frequency of positive-effect alleles in cultivated soybean than in wild soybean. Interestingly,gene allele matrix analysis of the known seed weight genes revealed that G. max has lost 38.5%of the G. soja alleles and that most of the lost alleles had negative effects on seed weight. | 6 | Tailoring MgH_(2) for hydrogen storage through nanoengineering and catalysis显示文摘Hydrogen energy has been recognized as “Ultimate Power Source” in the 21st century, which could be the best solution to the looming energy crisis and climate degeneration in the near future. Due to its high safety, low price, abundant resources and decent hydrogen storage density, magnesium based solid-state hydrogen storage materials are becoming the leading candidate for onboard hydrogen storage. However,the high operation temperature and slow reaction rate of MgH_(2), as a result of the large formation enthalpy and high reaction activation energy,respectively, are the first and most difficult problems we need to face and overcome to realize its industrialization. Herein, a state-of-the-art review on tailoring the stable thermodynamics and sluggish kinetics of hydrogen storage in MgH_(2), particularly through nanoengnieering and catalysis is presented, aiming to provide references and solutions for its promotion and application. | 12 | Identification of two glycerophosphodiester phosphodiesterase genes in maize leaf phosphorus remobilization显示文摘Phosphate deficiency is one of the leading causes of crop productivity loss.Phospholipid degradation liberates phosphate to cope with phosphate deficiency.Glycerophosphodiester phosphodiesterases(GPX-PDEs)hydrolyse the intermediate products of phospholipid catabolism glycerophosphodiesters into glycerol-3-phosphate,a precursor of phosphate.However,the function of GPX-PDEs in phosphate remobilization in maize remains unclear.In the present study,we characterized two phosphate deficiency-inducible GPX-PDE genes,ZmGPX-PDE1 and ZmGPX-PDE5,in maize leaves.ZmGPX-PDE1 and ZmGPX-PDE5 were transcriptionally regulated by ZmPHR1,a well-described phosphate starvation-responsive transcription factor of the MYB family.Complementation of the yeast GPX-PDE mutant gde1Δindicated that ZmGPX-PDE1 and ZmGPX-PDE5 functioned as GPX-PDEs,suggesting their roles in phosphate recycling from glycerophosphodiesters.In vitro enzyme assays showed that ZmGPX-PDE1 and ZmGPX-PDE5 catalysed glycerophosphodiester degradation with different substrate preferences for glycerophosphoinositol and glycerophosphocholine,respectively.ZmGPX-PDE1 was upregulated during leaf senescence,and more remarkably,loss of ZmGPXPDE1 inmaize compromised the remobilization of phosphorus fromsenescing leaves to young leaves,resulting in a stay-green phenotype under phosphate starvation.These results suggest that ZmGPX-PDE1 catalyses the degradation of glycerophosphodiesters in maize,promoting phosphate recycling from senescing leaves to new leaves.This mechanism is crucial for improving phosphorus utilization efficiency in crops. | Jingxin Wang Wenbo Pan Alexiy Nikiforov William King Wanting Hong Weiwei Li Yang Han Jana Patton-Vogt Jianbo Shen Lingyun Cheng | 2021 | The Crop Journal2021,9,1: | 1 |

| 15 | A novel in vitro prognostic model of bladder cancer based on urine-derived living tumor cells显示文摘Bladder cancer(BLCA)remains a difficult malignancy to manage because of its high recurrence,intense follow-up,and invasive diagnostic and treatment techniques.Immune checkpoint inhibitors(ICIs)have forged a new direction for the treatment of BLCA,but it is currently challenging to predict whether an individual patient will be sensitive to ICIs.We collected 43 urine/tumor samples from BLCA patients for primary bladder cancer cells(BCCs)culturing using our previously reported BCC culture platform.We used flow cytometry(FCM)to measure the expression levels of Programmed Death-Ligand 1(PD-L1)on BCCs before and after interferon-gamma(IFN-γ)treatment and found that PD-L1 expression and the sensitivities to IFN-γvaried among patients.RNA-sequencing,western blotting,and programmed death-1(PD-1)binding assays confirmed that the BCC FCM-based PD-L1 detection platform(BC-PD-L1)was reliable and was not hindered by the glycosylation of PD-L1.In the subsequent retrospective study,we found that IFN-γ-stimulated PD-L1(sPD-L1)expression on BCCs detected by BC-PD-L1 could predict the prognosis of BLCA patients.Importantly,the prognostic value was similar or even better in urine-derived BC-PD-L1(UBC-PD-L1).Transcriptome analysis showed that BCCs with high sPD-L1 tended to enrich genes associated with the collagen-containing extracellular matrix,cell–cell adhesion,and positive regulation of the immune system.In addition,the UBC-PD-L1 also exhibited predictive value for ICI response in BLCA patients.In conclusion,as a novel personalized urine-detection method,UBC-PD-L1 may provide a rapid,accurate,and non-invasive tool for monitoring tumor progression,predicting therapeutic responses,and helping improve BLCA clinical treatment in future. | Jiaqi Wang Jiying Zhu Junchi Hu Ziruoyu Wang Xiaobo Wang Jianbo Pan Yiwei Chu Zengxia Li Wei Jiang Chunmin Liang Jun Hou Jianming Guo Yongjun Dang Shuai Jiang | 2023 | Genes & Diseases2023,10,6: | 0 |
