| 2 | Uncommon EGFR mutations in a cohort of Chinese NSCLC patients and outcomes of first-line EGFR-TKIs and platinum-based chemotherapy显示文摘Objective: Data on the clinical activity of epidermal growth factor receptor(EGFR) tyrosine kinase inhibitors(TKIs) in patients with non-small-cell lung cancer(NSCLC) and uncommon EGFR mutations remain insufficient.This study aimed to investigate the effect of first-line EGFR-TKIs or platinum-based chemotherapy in NSCLC patients with uncommon EGFR mutations.Methods: We retrospectively enrolled 504 patients with EGFR-mutant NSCLC.The clinical characteristics and treatment outcomes were collected and compared between patients with common and uncommon EGFR-mutant NSCLC.Results: Seventy patients(13.9%) harboring uncommon EGFR mutations were included.Thirty of these patients received EGFR-TKIs and 40 received platinum-based chemotherapy as first-line therapy.The objective response rate(ORR) and median progression-free survival(m PFS) of patients treated with TKIs in the uncommon mutation group was significantly inferior to that in the common mutation group(ORR: 23.3% vs.51.8%,P=0.003; m PFS:7.1 vs.10.9 months,P<0.001).In the uncommon group,m PFS was similar between first-line EGFR-TKIs treatment and platinum-based chemotherapy(7.1 vs.6.1 months,P=0.893).In patients with EGFR G719 X or L861 Q mutations,the m PFS was longer in the first-line EGFR-TKIs treatment group than in the chemotherapy group,but the difference was not statistically significant(G719 X: 8.2 vs.5.8 months,P=0.061; L861 Q: 7.6 vs.4.1 months,P=0.872).Multivariate analyses identified adenocarcinoma(P=0.003) as the independent predictive factor for PFS in patients with uncommon EGFR mutations who were treated with first-line EGFR-TKIs.Conclusions: The current study demonstrated that the effect of first-line EGFR-TKIs was similar to that of platinum-based chemotherapy in patients with uncommon EGFR-mutant NSCLC.Adenocarcinoma was the independent predictive factor for PFS in uncommon EGFR-mutant NSCLC patients treated with first-line EGFRTKIs. | Jinpeng Shi Hui Yang Tao Jiang Xuefei Li Chao Zhao Limin Zhang Sha Zhao Xiaozhen Liu Yijun Jia Yan Wang Lei Xi Shijia Zhang Chunxia Su Shengxiang Ren Caicun Zhou | 2017 | Chinese Journal of Cancer Research2017,29,6: | 6 |
| 3 | Selection and characterization of the novel anti-human PD-1 FV78 antibody from a targeted epitope mammalian cell-displayed antibody library显示文摘Currently,display-based methods are well established and widely used in antibody engineering for affinity maturation and structural stability improvement.We obtained a novel anti-human programmed death 1(PD-1)antibody using computer-aided design and a mammalian cell display technology platform.We used computer-aided modeling and distance geometry methods to predict and assign the key residues that contributed to the binding of human PD-L1 to PD-1.Then,we analyzed the sequence of nivolumab(an anti-human PD-1 antibody,referred to as MIL75 in the article)to determine the template for antibody design and library construction.We identified a series of potential substitutions on the obtained template and constructed a virtual epitope-targeted antibody library based on the physicochemical properties and each possible location of the assigned key residues.The virtual antibody libraries were displayed on the surface of mammalian cells as the antigen-binding fragments of full-length immunoglobulin G.Then,we used flow cytometry and sequencing approaches to sort and screen the candidates.Finally,we obtained a novel anti-human PD-1 antibody named FV78.FV78 competitively recognized the PD-1 epitopes that interacted with MIL75 and possessed an affinity comparable to MIL75.Our results implied that FV78 possessed equivalent bioactivity in vitro and in vivo compared with MIL75,which highlighted the probability and prospect of FV78 becoming a new potential antibody therapy. | Longlong Luo Shi Wang Xiaoling Lang Tingting Zhou Jing Geng Xinying Li Chunxia Qiao Jiannan Feng Beifen Shen Ming Lv Yan Li | 2018 | Cellular & Molecular Immunology2018,15,2: | 4 |

| 6 | Bilateral papilledema caused by chronic infantile neurological cutaneous and articular syndrome in a child with a novel (p. D305N) mutation in NLRP3 gene: a case report显示文摘The rare disease of chronic infantile neurological cutaneous and articular(CINCA)syndrome,is caused by the over-secretion of interleukin(IL)-1βdue to a gain-of-function NLRP3 gene mutation in the autosomal chromosome which often involves in eyes.In this report,we studied a 9-year-old girl with CINCA.The eyes were also involved and presented bilateral papilledema.Genetic testing revealed that the symptoms were caused by a novel gene mutation site(c.913G>A,p.D305N)in conservative domain exon-3 of NLRP3 which is gain-function gene of CINCA.The patient had the characteristic facial features,frontal fossa and saddle nose,manifested the generalized urticaria-like skin rash at two weeks after birth,periodic fever 6 months after birth,sensorineural deafness at 7 years old,and bilateral papilledema,aseptic meningitis and knee arthropathy at 9 years old.White cell counts,C-reactive protein increased and intracranial pressure raised to 300 mmH2O.The meningeal thickening enhanced by gadolinium in magnetic resonance imaging(MRI).Based on clinical features and genetic test,the girl was diagnosed bilateral papilledema secondary to CINCA and administered prednisone and lowered intracranial pressure medicine to resolve symptoms.With 3-year follow-up,patient had no inflammatory flare-up with visual acuity improvement.The finding of novel genetic mutation site(p.D305N)in NLRP3 gene expanded genotype spectrum associated with CINCA.This case also expanded the cause spectrum of papilledema and it highlighted systemic disease history for patients with bilateral papilledema. | Li Li Huanfen Zhou Wei Shi Xiaotun Ren Xiaohua Tan Lirong Tian Chunxia Peng | 2021 | Annals of Eye Science2021,,3: | 1 |

| 9 | Brasenia-inspired hydrogel with sustained and sequential release of BMP and WNT activators for improved bone regeneration显示文摘Although bone morphogenetic protein(BMP) and WNT signaling play pivotal roles in bone development,homeostasis, and regeneration, the applications of proteins to stimulate corresponding signaling pathways showed limited outcomes in the repair and regeneration of bone defects that might be attributed to the reciprocal interventions of these pathways. In order to satisfy the combinational and sequential activation of BMP and WNT pathways, inspired by the heterogeneous hydrogel-liked structures of Brasenia, heterogeneous alginate/chitosan hydrogels were fabricated and spatially loaded with FK506 and BIO to achieve sustained and sequential release of the activators. Alkaline phosphatase staining, alizarin red staining and q RT-PCR results suggested that FK506 and BIO enhanced osteoblastic differentiation in vitro when used separately. Besides, by mixing and matching the activators and the hydrogel layers, a superior releasing mode that a combination of early FK506 release and following BIO release was identified via both in vitro and in vivo explorations for most efficient bone regeneration. | 12 | Intra-Patient and Inter-Patient Multi-Classification of Severe Cardiovascular Diseases Based on CResFormer显示文摘Severe cardiovascular diseases can rapidly lead to death.At present,most studies in the deep learning field using electrocardiogram(ECG)are performed on intra-patient experiments for the classification of coronary artery disease(CAD),myocardial infarction,and congestive heart failure(CHF).By contrast,actual conditions are inter-patient experiments.In this study,we proposed a deep learning network,namely,CResFormer,with dual feature extraction to improve accuracy in classifying such diseases.First,fixed segmentation of dual-lead ECG signals without preprocessing was used as input data.Second,one-dimensional convolutional layers performed moderate dimensionality reduction to accommodate subsequent feature extraction.Then,ResNet residual network block layers and transformer encoder layers sequentially performed feature extraction to obtain key associated abstract features.Finally,the Softmax function was used for classifications.Notably,the focal loss function is used when dealing with unbalanced datasets.The average accuracy,sensitivity,positive predictive value,and specificity of four classifications of severe cardiovascular diseases are 99.84%,99.68%,99.71%,and 99.90%in intra-patient experiments,respectively,and 97.48%,93.54%,96.30%,and 97.89%in inter-patient experiments,respectively.In addition,the model performs well in unbalanced datasets and shows good noise robustness.Therefore,the model has great application potential in diagnosing CAD,MI,and CHF in the actual clinical environment. | Dengao Li Changcheng Shi Jumin Zhao Yi Liu Chunxia Li | 2023 | Tsinghua Science and Technology2023,28,2: | 0 |
| 13 | Bio-Adhesives Combined with Lotus Leaf Fiber to Prepare Bio-Composites for Substituting the Plastic Packaging Materials显示文摘This work was aim to prepare a packing material from natural resources to reduce the environment pollution caused by plastics.Four bio-adhesives(guar gum,sodium alginate,agar and chitosan)were combined with lotus leaf fibers to prepare degradable composites,respectively.The mechanical properties,moisture absorption profiles and the thermal conductivity of the composites were studied and the cross section morphology and the thermal properties of the composites were analyzed.The Fourier-transform infrared spectroscopy(FTIR)results showed that the polar groups such as–OH and–COO^(–)in bio-adhesives can form hydrogen bond with–OH in lotus leaf fibers to connect the two components.The combination of agar and lotus leaf fiber was good,and their composite had the best mechanical properties,with the tensile strength,flexural strength and impact strength of 2.05,5.9 MPa and 4.29 kJ·m_(−2),respectively,and the composite had a low moisture absorption profile,and the equilibrium moisture absorption rate was 32.32%.The lotus leaf fiber/agar composite(LAC)had an excellent comprehensive performance and it was non-toxic,degradable and thermal insulating,which indicated that it had the potential to use in packaging field to substitute plastics. | Ke Shi Luyang Wang Ruige Qi Chunxia He | 2022 | Journal of Renewable Materials2022,10,5: | 0 |
