1Increased Tim-3 expression alleviates liver injury by regulating macrophage activation in MCD-induced NASH mice显示文摘As an immune checkpoint,Tim-3 plays roles in the regulation of both adaptive and innate immune cells including macrophages and is greatly involved in chronic liver diseases.However,the precise roles of Tim-3 in nonalcoholic steatohepatitis(NASH)remain unstated.In the current study,we analyzed Tim-3 expression on different subpopulations of liver macrophages and further investigated the potential roles of Tim-3 on hepatic macrophages in methionine and choline-deficient diet(MCD)-induced NASH mice.The results of flow cytometry demonstrated the significantly increased expression of Tim-3 on all detected liver macrophage subsets in MCD mice,including F4/80^(+)CD11b^(+),F4/80^(+)CD68^(+),and F4/80^(+)CD169^(+)macrophages.Remarkably,Tim-3 knockout(KO)significantly accelerated MCD-induced liver steatosis,displaying higher serum ALT,larger hepatic vacuolation,more liver lipid deposition,and more severe liver fibrosis.Moreover,compared with wild-type C57BL/6 mice,Tim-3 KO MCD mice demonstrated an enhanced expression of NOX2,NLRP3,and caspase-1 p20 together with increased generation of IL-1βand IL-18 in livers.In vitro studies demonstrated that Tim-3 negatively regulated the production of reactive oxygen species(ROS)and related downstream pro-inflammatory cytokine secretion of IL-1βand IL-18 in macrophages.Exogenous administration of N-Acetyl-L-cysteine(NAC),a small molecular inhibitor of ROS,remarkably suppressed caspase-1 p20 expression and IL-1βand IL-18 production in livers of Tim-3 KO mice,thus significantly reducing the severity of steatohepatitis induced by MCD.In conclusion,Tim-3 is a promising protector in MCD-induced steatohepatitis by controlling ROS and the associated pro-inflammatory cytokine production in macrophages.Xianhong Du Zhuanchang Wu Yong Xu Yuan Liu Wen Liu Tixiao Wang Chunyang Li Cuijuan Zhang Fan Yi Lifen Gao Xiaohong Liang Chunhong Ma 2019Cellular & Molecular Immunology2019,16,11:9
2LncRNA PU.1 AS regulates arsenic-induced lipid metabolism through EZH2/Sirt6/SREBP-1c pathway显示文摘Arsenic (As) is an omnipresent metalloid toxicant,which has elicited serious environmental pollution and health risky problems.Previous studies have uncovered that the As exposure could also cause markedly reduction of serum triglycerides in mice.However,the regulation mechanisms are still largely unknown.The present study is aimed to elucidate the molecular mechanisms of lncRNAs in As-induced lipid metabolic disequilibrium.We demonstrated that lncRNA PU.1 AS was significantly induced in the liver of As-feed mice companied with lower serum triglycerides contents;further in vitro experiment confirmed that PU.1 AS regulated liver cells lipid accumulation by nile red fluorescence staining.Intensive mechanistic investigations illustrated that PU.1 AS could interact with EZH2 protein to regulate its downstream target gene expression,and Asinduced PU.1 AS attenuated EZH2-supppressed Sirt6 expression,thereafter leading to a decreased SREBP-1c protein expression,as well as the diminished synthesis of triglycerides in hepatocytes.In conclusion,this study provided a new lncRNA-related regulatory signaling pathway participating in As-induced abnormal lipid metabolism.Zheng Dong Changying Li Chunyang Yin Ming Xu Sijin Liu Ming Gao 2019Journal of Environmental Sciences2019,31,11:8
3Expressed sequence tags analysis revealing the taxonomic position and fatty acid biosynthesis in an oleaginous green microalga,Myrmecia incisa Reisigl(Trebouxiophyceae,Chlorophyta)显示文摘cDNA library of Myrmecia incisa H4301 was constructed using λ phage vectors.The library consisted of 1.5×10 6 clones with a recombination rate of 90%,and 1942 clones were randomly sequenced.All 1854 readable expressed sequence tags(ESTs) were clustered into 596 non-redundant sequences(NRSs),among which 126 NRSs were from 1384 ESTs,showing a high redundancy.Among the 596 NRSs,30 were ribosomal RNA,and 152 significantly matched with those available in NCBI database and JGI Genome Portal,the latter were divided into nine subcategories.Overall,59 NRSs were involved in photosynthesis,the respiratory electron transport chain,ATP synthesis,oxidation reduction,fatty acid biosynthesis,glucose metabolism,protein metabolism,and small molecular metabolism,suggesting that these genes were abundantly transcribed during energy and substance metabolism.Acyl-carrier protein,ferrodoxin and fatty acid elongase genes obtained from this cDNA library enabled presumption of a possible biosynthesis pathway of ArA in M.incisa.Codon usage analysis of 142 NRSs with 17798 codons in the predicted coding regions showed that the average G+C content level of the total codons was 55.39%,and that of the third position in base trimers was 66.42%,indicating a strong bias toward cytosine and/or guanosine in this algal genome.Among all synonymous codons,NAG was most favored,while NUA was most avoided.Phylogenic trees inferred from ribulose-1,5-bisphosphate carboxylase/oxygenase small subunit genes and the extra partial sequences of 18S rRNA obtained from this library demonstrated that M.incisa belonged to Trebouxiophyceae and was significantly clustered with M.incisa SAG 2007,Lobosphaera tirolensis,M.bisecta,and Parietochloris incisa,but was clearly distant from P.pseudoalveolaris and P.alveolar.Transmission electron microscopy revealed pyrenoids traversed by many parallel thylakoids membranes,while starch grains were only clearly observed when cells were grown under nitrogen stress.Based on combined investigation of the phylogeny and morphological characteristics,it is proposed that M.incisa be kept in the genus Myrmecia in which there might be two parallel groups,one living freely and another symbiotic species.OUYANG LongLing DU DaoHai YU ShuiYan LI ChunYang ZHANG ChengWu GAO HongJian ZHOU ZhiGang 2012Chinese Science Bulletin2012,57,25:4
4Hepatitis B virus evades immune recognition via RNA adenosine deaminase ADAR1-mediated viral RNA editing in hepatocytes显示文摘HBV is considered as a“stealth”virus that does not invoke interferon(IFN)responses;however,the mechanisms by which HBV bypasses innate immune recognition are poorly understood.In this study,we identified adenosine deaminases acting on RNA 1(ADAR1),which is a key factor in HBV evasion from IFN responses in hepatocytes.Mechanically,ADAR1 interacted with HBV RNAs and deaminated adenosine(A)to generate inosine(I),which disrupted host immune recognition and thus promoted HBV replication.Loss of ADAR1 or its deficient deaminase activity promoted IFN responses and inhibited HBV replication in hepatocytes,and blocking the IFN signaling pathways released the inhibition of HBV replication caused by ADAR1 deficiency.Notably,the HBV X protein(HBx)transcriptionally promoted ADAR1 expression to increase the threshold required to trigger intrinsic immune activation,which in turn enhanced HBV escape from immune recognition,leading to persistent infection.Supplementation with 8-azaadenosine,an ADAR1 inhibitor,efficiently enhanced liver immune activation to promote HBV clearance in vivo and in vitro.Taken together,our results delineate a molecular mechanism by which HBx promotes ADAR1-derived HBV immune escape and suggest a targeted therapeutic intervention for HBV infection.Liyuan Wang Yang Sun Xiaojia Song Zehua Wang Yankun Zhang Ying Zhao Xueqi Peng Xiaodong Zhang Chunyang Li Chengjiang Gao Nailin Li Lifen Gao Xiaohong Liang Zhuanchang Wu Chunhong Ma 2021Cellular & Molecular Immunology2021,18,8:2
5Experimental and Numerical Analysis of High-Strength Concrete Beams Including Steel Fibers and Large-Particle Recycled Coarse Aggregates显示文摘In order to study the performances of high-strength concrete beams including steel fibers and large-particle recycled aggregates,four different beams have been designed,tested experimentally and simulated numerically.As varying parameters,the replacement rates of recycled coarse aggregates and CFRP(carbon fiber reinforced polymer)sheets have been considered.The failure mode of these beams,related load deflection curves,stirrup strain and shear capacity have been determined through monotonic loading tests.The simulations have been conducted using the ABAQUS finite element software.The results show that the shear failure mode of recycled concrete beams is similar to that of ordinary concrete beams.The shear carrying capacity of high-strength concrete beams including steel fibers and large-particle recycled coarse aggregates grows with an increase in the replacement rate of recycled coarse aggregates.Reinforcement with CFRP sheets can significantly improve the beam’s shear carrying capacity and overall resistance to deformation.Chunyang Liu Yangyang Wu Yingqi Gao Zhenyun Tang 2021Fluid Dynamics & Materials Processing2021,17,5:2
6Controlling mine pressure by subjecting high-level hard rock strata to ground fracturing显示文摘When mining extra-thick coal seams,the main cause of strong ground pressure are the high-level thick and hard strata,but as yet there is no active and effective control technology.This paper proposes the method of subjecting hard roofs to ground fracturing,and physical simulation is used to study the control effect of ground fracturing on the strata structure and energy release.The results show that ground fracturing changes the structural characteristics of the strata and reduces the energy release intensity and the spatial extent of overburden movement,thereby exerting significant control on the ground pressure.The Datong mining area in China is selected as the engineering background.An engineering test was conducted on site by ground horizontal well fracturing,and a 20-m-thick hard rock layer located 110 m vertically above the coal seam was targeted as the fracturing layer.On-site microseismic monitoring shows that the crack propagation length is up to 216 m and the height is up to 50 m.On-site mine pressure monitoring shows that(1)the roadway deformation is reduced to 100 mm,(2)the periodic weighting characteristics of the hydraulic supports are not obvious,and(3)the ground pressure in the working face is controlled significantly,thereby showing that the ground fracturing is successful.Ground fracturing changed the breaking characteristics of the high-level hard strata,thereby helping to ameliorate the stress concentration in the stope and providing an effective control approach for hard rock.Rui Gao Tiejun Kuang Yanqun Zhang Wenyang Zhang Chunyang Quan 2021International Journal of Coal Science & Technology2021,8,6:2
7Fenton Pre-Oxidation Followed by Microbial Degradation for Removing Crude Oil from Contaminated Soil显示文摘Through the Fenton pre-oxidation followed by microbial degradation,this study gave full play to its advantages while avoiding its shortcomings for the remediation of crude oil contaminated soil.The Fenton reagent coupled with different volumes of H2O2 was applied to the oil contaminated soil and then the microbial agents were introduced to biodegrade the residual oil for 15 days.The correlation between the characteristics of residual oil in soil,the changes in soil physical-chemical property after the Fenton pre-oxidation,and the biodegradation were analyzed in this paper.The results show that the above factors are strongly correlated with the subsequent biodegradation rate,and the order of correlation is as follows:the ratio of TOC to NH4+-N(R^2=0.9513)>the ratio of light oil components to the heavy oil components(R^2=0.9095)>the proportion of hydrocarbons with carbon chain number of less than C23(R^2=0.8259)>the crude oil content(R^2=0.7603)>the soil pH(R^2=0.7492)>the number of microorganisms(R^2=0.6506).During the biodegradation and pre-oxidation reactions of heavy oil components,an appropriate C:N ratio turns out to be the most critical factor in this study.Gao Chunyang Zhang Yu Han Zhantao Song Quanwei Chen Changzhao Chen Hongkun Zheng Jin 2019China Petroleum Processing & Petrochemical Technology2019,21,3:1

10Pyridoxine regulates hair follicle development via the PI3K/Akt, Wnt and Notch signalling pathways in rex rabbits显示文摘This study was conducted to evaluate the effect of pyridoxine on the development of hair follicles in Rex rabbits and the underlying molecular mechanism.Two hundred 3-month-old Rex rabbits were randomly divided into 5 groups and fed diets supplemented with 0,5,10,20,or 40 mg/kg pyridoxine.The hair follicle density on the dorsal skin and the gene and protein expression levels of components of the phosphoinositide 3-kinase(PI3 K)/protein kinase B(PKB or Akt),Wnt,Notch and bone morphogenetic protein(BMP)signalling pathways were measured.In addition,free hair follicles were isolated from Rex rabbits and cultured with pyridoxine in vitro to measure hair shaft growth.Furthermore,dermal papilla cells(DPC)were isolated from the skin of Rex rabbits and cultured with pyridoxine in vitro to measure the gene and protein expression levels of components of the PI3 K/Akt,Wnt,Notch and BMP signalling pathways.The results showed that the addition of dietary pyridoxine significantly increased the total follicle density,secondary follicle density,and secondary-to-primary ratio(S/P,P<0.05),that the growth ratio of hair stems was promoted by pyridoxine in basic culture medium,and that the growth length of tentacle hair follicles cultured in the pyridoxine group was longer than that in the control group(P<0.05).In addition,pyridoxine changed the DPC cycle progression and promoted cell proliferation,and appropriate concentrations of pyridoxine(10 and 20μmol/L)significantly inhibited cell apoptosis(P<0.05).Pyridoxine significantly affected the gene expression of components of the PI3 K/Akt,Wnt and Notch signalling pathways in the skin and DPC of Rex rabbits(P<0.05),increased the levels of phosphorylated catenin beta 1(CTNNB1)and Akt,and decreased the level of phosphorylated glycogen synthase kinase 3 beta(GSK-3β)(P<0.05).Therefore,the molecular mechanism by which pyridoxine promotes hair follicle density in Rex rabbits probably occurs through activation of the PI3 K/Akt,Wnt and Notch signalling pathways,prolonging hair follicle growth and delaying the onset of telogen.Gongyan Liu Guangmin Cheng Yongcui Zhang Shuxia Gao Haitao Sun Liya Bai Shu Li Yanli Zhu Chunyang Wang Fuchang Li 2021Animal Nutrition2021,,4:1
11Atomic origin of magnetic coupling of antiphase boundaries in magnetite thin films显示文摘Revealing the magnetic coupling nature of boundary defects is crucial for in-depth understanding of the behavior and properties of magnetic materials and devices.Here,magnetite(i.e.,Fe_(3)O_(4))thin films were grown epitaxially on(100)SrTiOsingle-crystal substrates by pulsed laser deposition.Atomic-scale scanning transmission electron microscopy characterizations reveal that three types of antiphase boundaries(APBs)are formed in the Fe_(3)O_(4)thin film.They are the(100)APB that is formed on the(100)plane with a crystal translation of(1/4)a[011^(-)],the typeⅠand typeⅡ(110)APBs that are both formed on the(110)plane with the same crystal translation of(1/4)a[101]but different terminated atomic planes.The typeⅠ(110)APB is terminated at the atomic plane with mixed tetrahedral-and octahedral-sites Fe atoms,the typeⅡ(110)APB is terminated at the octahedral-site Fe plane.First-principles calculations reveal that the(100)APB and the typeⅠ(110)APB tend to form the ferromagnetic coupling that will not decrease the spin polarization of Fe_(3)O_(4)films,while the typeⅡ(110)APB prefers to form the antiferromagnetic coupling that will degrade the magnetic properties.The magnetic coupling modes of the APBs are closely related to the Fe-O-Fe bond angles across the boundaries.Chunyang Gao Yixiao Jiang Tingting Yao Ang Tao Xuexi Yan Xiang Li Chunlin Chen Xiu-Liang Ma Hengqiang Ye 2022Journal of Materials Science & Technology2022,,12:1
12Bioinspired porous microspheres for sustained hypoxic exosomes release and vascularized bone regeneration显示文摘Exosomes derived from mesenchymal stem cells(MSCs)have demonstrated regenerative potential for cell-free bone tissue engineering,nevertheless,certain challenges,including the confined therapeutic potency of exosomes and ineffective delivery method,are still persisted.Here,we confirmed that hypoxic precondition could induce enhanced secretion of exosomes from stem cells from human exfoliated deciduous teeth(SHEDs)via comprehensive proteomics analysis,and the corresponding hypoxic exosomes(H-Exo)exhibited superior potential in promoting cellular angiogenesis and osteogenesis via the significant up-regulation in focal adhesion,VEGF signaling pathway,and thyroid hormone synthesis.Then,we developed a platform technology enabling the effective delivery of hypoxic exosomes with sustained release kinetics to irregular-shaped bone defects via injection.This platform is based on a simple adsorbing technique,where exosomes are adsorbed onto the surface of injectable porous poly(lactide-co-glycolide)(PLGA)microspheres with bioinspired polydopamine(PDA)coating(PMS-PDA microspheres).The PMS-PDA microspheres could effectively adsorb exosomes,show sustained release of H-Exo for 21 days with high bioactivity,and induce vascularized bone regeneration in 5-mm rat calvarial defect.These findings indicate that the hypoxic precondition and PMS-PDA porous microsphere-based exosome delivery are efficient in inducing tissue regeneration,hence facilitating the clinical translation of exosome-based therapy.Yike Gao Zuoying Yuan Xiaojing Yuan Zhuo Wan Yingjie Yu Qi Zhan Yuming Zhao Jianmin Han Jianyong Huang Chunyang Xiong Qing Cai 2022Bioactive Materials2022,7,8:1

14Study on Functional Mechanical Performance of Honeycomb Array Structures Inspired by Gideon Beetle显示文摘To obtain bio-inspired structures with superior biological function,four bio-inspired structures named regular arrangement honeycomb structure(RAHS),staggered arrangement honeycomb structure(SAHS),floral arrangement honeycomb structure(FLAHS)and functional arrangement honeycomb structure(FUAHS)are designed by observing the microstructure of the Gideon beetle,based on the optimal size bio-inspired cells by response surface method(RSM)and particle swarm optimization(PSO)algorithm.According to Euler theory and buckling failure theory,compression deformation properties of bio-inspired structures are explained.Experiments and simulations further verify the accuracy of theoretical analysis results.The results show that energy absorption of FLAHS is,respectively,increased by 26.95%,22.85%,and 121.45%,compared with RAHS,SAHS,and FUAHS.Elastic modulus of FLAHS is 110.37%,110.37%,and 230.56% of RAHS,SAHS,and FUAHS,respectively.FLAHS perfectly inherits crashworthiness and energy absorption properties of the Gideon beetle,and FLAHS has the most stable force.Similarly,RAHS,SAHS,and FUAHS,respectively,inherit mechanical properties of the Gideon beetle top horn,the Gideon beetle middle horn,and the abdomen of the beetle.This method,designing bio-inspired structures with biological functions,can be introduced into the engineering field requiring the special function.Ruiyao Liu Guofeng Yao Zezhou Xu Xue Guo Kuiyang Gao Qing Cao Zhenglei Yu Zhihui Zhang Chunyang Han Jiabao Liu 2022Journal of Bionic Engineering2022,19,4:0
15Diverse habitat preferences of two sea cucumber species and the seasonal change in a coral reef area显示文摘Diff erent sea cucumbers have diff erent preferred habitats and seasonal changes,which is still lack of detailed research.This study selects two common tropical sea cucumbers(Holothuria edulis and Stichopus chloronotus)to explore the reasons that aff ect their habitat selection and seasonal changes,so as to provide support for the spatial planning of sea cucumbers reserve in coral reef area.The study area is a compound distribution area of living coral reefs and sandy bottom in the north of Wuzhizhou Island.The survey time is January,April-September,2019.Three fi xed transects(G,H,and Q)were selected to reveal the population ecological characteristics of them in the typical dry season(January,April,May,and June)and rainy season(July,August,and September).The variation range of density for H.edulis and S.chloronotus were 2.0-8.9 inds./10 m^(2) and 0.2-0.7 inds./10 m^(2),respectively.The population density in rainy season was signifi cantly higher than that in dry season(P<0.05).H.edulis tends to be distributed in deeper water.The distribution of H.edulis was signifi cant positively correlated with the sand coverage and sand continuity in G transect(P<0.05),and signifi cant negatively correlated with live coral coverage in three transects(P<0.01).The shift from dry season to rainy season is a key factor aff ecting their habitat preference.In dry season,the preferred habitats of S.chloronotus are small rock(SR),sand(S),and small coral reef(SCR),while the H.edulis is S,SR,and dead coral rubbles(DCR).In rainy season,the preferred habitats of S.chloronotus are shift to SCR and SR,while the H.edulis is SCR,DCR,and sand.Chunyang SUN Duanjie HUANG Qiang XU Fei GAO Xiubao LI Aimin WANG 2022Journal of Oceanology and Limnology2022,40,4:0
16Recognition for avian influenza virus proteins based on support vector machine and linear discriminant analysis显示文摘Total 200 properties related to structural characteristics were employed to represent structures of 400 HA coded proteins of influenza virus as training samples. Some recognition models for HA proteins of avian influenza virus (AIV) were developed using support vector machine (SVM) and linear discriminant analysis (LDA). The results obtained from LDA are as follows: the identification accuracy (Ria) for training samples is 99.8% and Ria by leave one out cross validation is 99.5%. Both Ria of 99.8% for training samples and Ria of 99.3% by leave one out cross validation are obtained using SVM model, respectively. External 200 HA proteins of influenza virus were used to validate the external predictive power of the resulting model. 20A case of chronic pancreatitis treated by laparoscopic duodenum-preserving pancreatic head resection显示文摘Pancreaticoduodenectomy(PD) has long been used for chronic pancreatitis(CP), but greatly affects the postoperative quality of life. A new procedure called duodenum-preserving pancreatic head resection(DPPHR) has been introduced, and has little effect on the structure and function of the digestive system. With the development of minimally invasive surgical techniques, treatment of CP can be performed with laparoscopic DPPHR(LDPPHR). We present a case of CP that was successfully treated with LDPPHR. The postoperative pathological diagnosis was pancreatitis, demonstrating the feasibility of LDPPHR.
