1Salivary mycobiome dysbiosis and its potential impact on bacteriome shifts and host immunity in oral lichen planus显示文摘The biodiversity of the mycobiome,an important component of the oral microbial community,and the roles of fungal–bacterial and fungal–immune system interactions in the pathogenesis of oral lichen planus (OLP) remain largely uncharacterized.In this study,we sequenced the salivary mycobiome and bacteriome associated with OLP.First,we described the dysbiosis of the microbiome in OLP patients,which exhibits lower levels of fungi and higher levels of bacteria.Significantly higher abundances of the fungi Candida and Aspergillus in patients with reticular OLP and of Alternaria and Sclerotiniaceae_unidentified in patients with erosive OLP were observed compared to the healthy controls.Aspergillus was identified as an “OLP-associated” fungus because of its detection at a higher frequency than in the healthy controls.Second,the co-occurrence patterns of the salivary mycobiome–bacteriome demonstrated negative associations between specific fungal and bacterial taxa identified in the healthy controls,which diminished in the reticular OLP group and even became positive in the erosive OLP group.Moreover,the oral cavities of OLP patients were colonized by dysbiotic oral flora with lower ecological network complexity and decreased fungal–Firmicutes and increased fungal–Bacteroidetes sub-networks.Third,several keystone fungal genera (Bovista,Erysiphe,Psathyrella,etc.) demonstrated significant correlations with clinical scores and IL-17 levels.Thus,we established that fungal dysbiosis is associated with the aggravation of OLP.Fungal dysbiosis could alter the salivary bacteriome or may reflect a direct effect of host immunity,which participates in OLP pathogenesis.Yan Li Kun Wang Bo Zhang Qichao Tu Yufei Yao Bomiao Cui Biao Ren Jinzhi He Xin Shen Joy D.Van Nostrand Jizhong Zhou Wenyuan Shi Liying Xiao Changqing Lu Xuedong Zhou 2019International Journal of Oral Science2019,11,3:8
2Markov repairable systems with stochastic regimes switching显示文摘Compared with the classical Markov repairable system,the Markov repairable system with stochastic regimes switching introduced in the paper provides a more realistic description of the practical system.The system can be used to model the dynamics of a repairable system whose performance regimes switch according to the external conditions.For example,to satisfy the demand variation that is typical for the power and communication systems and reduce the cost,these systems usually adjust their operating regimes.The transition rate matrices under distinct operating regimes are assumed to be different and the sojourn times in distinct regimes are governed by a finite state Markov chain.By using the theory of Markov process,Ion channel theory,and Laplace transforms,the up time of the system are studied.A numerical example is given to illustrate the obtained results.The effect of sojourn times in distinct regimes on the availability and the up time are also discussed in the numerical example.Liying Wang Lirong Cui Mingli Yu 2011Journal of Systems Engineering and Electronics2011,22,5:5
3Study on the Anti-Liver Fibrosis Effect of Benefit Liver Granule (益肝冲剂) and Its Mechanism in Rats显示文摘Objective:To explore the effect of Benefit Liver Granule (BLG), a traditional Chinese medical preparation, in antagonizing liver fibrosis and its possible mechanism. Methods:Carbon tetrachloride was used to establish the experimental liver fibrosis model of rat. The model rats were randomly divided into 4 groups and BLG, Astragalus (As), Salvia (Sa) and normal saline (for control) respectively were given to them by gastrogavage. Changes of serum alanine transaminase (ALT), aspartate aminotransferase (AST), transforming growth factor α(TGF α), interleukine 6 (IL 6), liver content of hydroxyproline (Hyp), superoxide dismutase (SOD), malonyldialdehyde (MDA) as well as liver pathology after treatment were observed. A normal control group was also established for control.Results: Pathological examination showed that in the saline group, the structure of normal hepatic lobuli was destroyed with swelled liver cells, focal necrosis, extensive fatty degeneration, and focal inflammatory cell infiltration. Small amounts of proliferated fiber tissue were found in the intra lobular area, peri central vein area, portal area and limiting plate area, and formation of pseudolobuli was also seen. In the 3 treated groups, the serum ALT, AST, TGF α and IL 6 as well as liver content of Hyp, and MDA were lower and SOD were higher than those in the control group significantly, P <0.05 or P <0.01.Conclusion: BLG, As and Sa have the action of anti liver fibrosis, while BLG has the best effect. 4Effectiveness of intravenous r- tPA versus UK for acute ischaemic stroke: a nationwide prospective Chinese registry study显示文摘Background Intravenous recombinant tissue plasminogen activator(r-tPA)and urokinase(UK)are both recommended for the treatment of acute ischaemic stroke(AIS)in China,but with few comparative outcome data being available.We aimed to compare the outcomes of these two thrombolytic agents for the treatment of patients within 4.5 hours of onset of AIS in routine clinical practice in China.Methods A pre-planned,prospective,nationwide,multicentre,real-world registry of consecutive patients with AIS(age≥18 years)who received r-tPA or UK within 4.5 hours of symptom onset according to local decision-making and guideline recommendations during 2017-2019.The primary effectiveness outcome was the proportion of patients with an excellent functional outcome(defined by modified Rankin scale scores 0 to 1)at 90 days.The key safety endpoint was symptomatic intracranial haemorrhage according to standard definitions.Multivariable logistic regression was used for comparative analysis,with adjustment according to propensity scores to ensure balance in baseline characteristics.Results Overall,4130 patients with AIS were registered but 320 had incomplete or missing data,leaving 3810 with available data for analysis of whom 2666 received r-tPA(median dose 0.88(IQR 0.78-0.90)mg/kg)and 1144 received UK(1.71(1.43-2.00)×104 international unit per kilogram).There were several significant intergroup differences in patient characteristics:r-tPA patients were more educated,had less history of stroke,lower systolic blood pressure,greater neurological impairment and shorter treatment times from symptom onset than UK patients.However,in adjusted analysis,the frequency of excellent outcome(OR 1.18,95%CI 1.00 to 1.40,p=0.052)and symptomatic intracranial haemorrhage(OR 0.70,95%CI 0.33 to 1.47,p=0.344)were similar between groups.Conclusions UK may be as effective and carry a similar safety profile as r-tPA in treating mild to moderate AIS within guidelines in China.Registration http://www.clinicaltrials.gov.unique identifier:NCT02854592.Xinhong Wang Xiaoqiu Li Yuming Xu Runhui Li Qingcheng Yang Yong Zhao Fengyun Wang Baoying Sheng Runqing Wang Shaoyuan Chen Lihua Wang Liying Shen Xiaowen Hou Yu Cui Duolao Wang Bin Peng Craig S Anderson Huisheng Chen On behalf of INTRECIS Investigators 2021Stroke & Vascular Neurology2021,6,4:2
5Effect of carbon and nitrogen ratio control on Artemia growth, water quality, biofloc microbial diversity under high salinity and zero-water exchange culture condition显示文摘Biofloc technology has been applied successfully in the intensive aquaculture of several fish and shrimp species. The growth of heterotrophic microorganisms can be stimulated through adding extra carbon, which reduces the nitrogen level in the water and provides microbial protein to the animals. However, most of the studies and practical applications have been conducted in freshwater and marine environment. This paper focused on brine shrimp Artemia that lives in high salinity environment together with other halophilic or halotolerant microorganisms. The effect of carbon supplementation on Artemia growth, water quality, and microbial diversity of biofl ocs was studied in the closed culture condition without any water exchange. The salinity of the culture medium was 100. A 24-d culture trial was conducted through supplementing sucrose at carbon/nitrogen (C/N) ratio of 5, 15, and 30 (Su5, Su15, and Su30), respectively. The culture without adding sucrose was used as a control. Artemia was fed formulated feed at a feeding ration of 60% recommended feeding level. The results showed that sucrose supplementation at higher C/N ratio (15 and 30) signifi cantly improved the Artemia survival, growth and water quality ( P <0.05). Addition of sucrose at C/N ratio of 15 and 30 significantly increased biofloc volume (BFV)( P <0.05). The Illumina MiSeq sequencing analysis showed that supplementing carbon at C/N ratio of 15 had a better total bacterial diversity and richness, and shaped the microbial composition at genera level. 6Conditioned medium-electrospun fiber biomaterials for skin regeneration显示文摘Conditioned medium(CM)contains variety of factors secreted by cells,which directly regulate cellular processes,showing tremendous potential in regenerative medicine.Here,for the first time,we proposed a novel regenerative therapy mediated by biodegradable micro-nano electrospun fibers loaded with highly active conditioned medium of adipose-derived stem cells(ADSC-CM).ADSC-CM was successfully loaded into the nanofibers with biological protection and controllable sustained-release properties by emulsion electrospinning and protein freeze-drying technologies.In vitro,ADSC-CM released by the fibers accelerated the migration rate of fibroblasts;inhibited the over proliferation of fibroblasts by inducing apoptosis and damaging cell membrane;in addition,ADSC-CM inhibited the transformation of fibroblasts into myofibroblasts and suppressed excessive production of extracellular matrix(ECM).In vivo,the application of CM-biomaterials significantly accelerated wound closure and improved regeneration outcome,showing superior pro-regenerative performance.This study pioneered the application of CM-biomaterials in regenerative medicine,and confirmed the practicability and significant biological effects of this innovative biomaterials.Lu Chen Liying Cheng Zhen Wang Jianming Zhang Xiyuan Mao Zhimo Liu Yuguang Zhang Wenguo Cui Xiaoming Sun 2021Bioactive Materials2021,6,2:2

10The Gold Coast criteria increases the diagnostic sensitivity for amyotrophic lateral sclerosis in a Chinese population显示文摘Objectives:The aim of this study was to assess and compare the diagnostic utility of a new diagnostic criteria for amyotrophic lateral sclerosis(ALS),abbreviated as the‘Gold Coast Criteria’,with the revised El Escorial(rEEC)and Awaji criteria.Methods:Clinical and electrophysiological data of 1185 patients from January 2014 to December 2019 in the Peking Union Medical College Hospital ALS database were reviewed.The sensitivity of the Gold Coast criteria was compared to that of the possible rEEC and Awaji criteria(defined by the proportion of patients categorized as definite,probable,or possible ALS).Results:A final diagnosis of ALS was recorded in 1162 patients.The sensitivity of the Gold Coast criteria(96.6%,95%confidence interval[CI]=95.3%-97.5%)was greater than that of the rEEC(85.1%,95%CI=82.9%-87.1%)and Awaji(85.3%,95%CI=83.2%-87.3%).In addition,the sensitivity of the novel criteria maintained robust across subgroups,and the advantage was more prominent in limb-onset ALS patients and those who completed electromyographic tests.In those who did not achieve any of the rEEC diagnostic categories,the sensitivity of Gold Coast criteria was 84.4%.Conclusions:The current study demonstrated that the Gold Coast criteria exhibited greater diagnostic sensitivity than the rEEC and Awaji criteria in a Chinese ALS population.The application of the Gold Coast criteria should be considered in clinical practice and future therapeutic trials.Dongchao Shen Xunzhe Yang Yanying Wang Di He Xiaohan Sun Zhengyi Cai Jinyue Li Mingsheng Liu Liying Cui 2021Translational Neurodegeneration2021,10,3:1

12Molecular Simulation of Transesterification of Ethylene Carbonate and Methanol Catalyzed by Ionic Liquids显示文摘Four ionic liquids [BMIM]OH, [BMIM]IM, [BMIM]Br, and [BMIM]PF6 were synthesized and characterized by infrared spectroscopy. Then the effects of ionic liquids(ILs), cocatalysts, and reaction temperature on the catalytic performance for transesterification of ethylene carbonate and methanol were investigated with orthogonal experiments. The influence of cations and anions of ILs on catalytic activity was revealed by the density functional theory(DFT). The reaction mechanism was proposed based on the experimental results and DFT. The results demonstrated that the optimal catalyst was [Bmim]PF6/CaO, which exhibited the advantages of high activity, excellent stability, and easy recycling. Under the optimized conditions covering a catalytic temperature of 130 °C, an ionic liquid/cocatalyst mass ratio of 5:1, and a catalyst dosage of 4.0%, the conversion rate could reach 65.23% with a dimethyl carbonate selectivity of 98.95%. 13Programmable immune activating electrospun fibers for skin regeneration显示文摘Immune cells play a crucial regulatory role in inflammatory phase and proliferative phase during skin healing.How to programmatically activate sequential immune responses is the key for scarless skin regeneration.In this study,an“Inner-Outer”IL-10-loaded electrospun fiber with cascade release behavior was constructed.During the inflammatory phase,the electrospun fiber released a lower concentration of IL-10 within the wound,inhibiting excessive recruitment of inflammatory cells and polarizing macrophages into anti-inflammatory phenotype“M2c”to suppress excessive inflammation response.During the proliferative phase,a higher concentration of IL-10 released by the fiber and the anti-fibrotic cytokines secreted by polarized“M2c”directly acted on dermal fibroblasts to simultaneously inhibit extracellular matrix overdeposition and promote fibroblast migration.The“Inner-Outer”IL-10-loaded electrospun fiber programmatically activated the sequential immune responses during wound healing and led to scarless skin regeneration,which is a promising immunomodulatory biomaterial with great potential for promoting complete tissue regeneration.Lu Chen Liucheng Zhang Hongbo Zhang Xiaoming Sun Dan Liu Jianming Zhang Yuguang Zhang Liying Cheng Hélder A.Santos Wenguo Cui 2021Bioactive Materials2021,6,10:1

17CsTRM5 regulates fruit shape via mediating cell division direction and cell expansion in cucumber显示文摘Fruit shape and size are important appearance and yield traits in cucumber,but the underlying genes and their regulatory mechanisms remain poorly understood.Here we identified a mutant with spherical fruits from an Ethyl Methane Sulfonate(EMS)-mutagenized library,named the qiu mutant.Compared with the cylindrical fruit shape in 32X(wild type),the fruit shape in qiu was round due to reduced fruit length and increased fruit diameter.MutMap analysis narrowed the candidate gene in the 6.47 MB range on Chr2,harboring the FS2.1 locus reported previously.A single-nucleotide polymorphism(SNP)(11359603)causing a truncated protein of CsaV3_2G013800,the homolog of tomato fruit shape gene SlTRM5,may underlie the fruit shape variation in the qiu mutant.Knockout of CsTRM5 by the CRISPR-Cas9 system confirmed that CsaV3_2G013800/CsTRM5 was the causal gene responsible for qiu.Sectioning analysis showed that the spherical fruit in qiu resulted mainly from increased and reduced cell division along the transverse and longitudinal directions,respectively.Meanwhile,the repressed cell expansion contributed to the decreased fruit length in qiu.Transcriptome profiling showed that the expression levels of cell-wall-related genes and abscisic acid(ABA)pathway genes were significantly upregulated in qiu.Hormone measurements indicated that ABA content was greatly increased in the qiu mutant.Exogenous ABA application reduced fruit elongation by inhibiting cell expansion in cucumber.Taken together,these data suggest that CsTRM5 regulates fruit shape by affecting cell division direction and cell expansion,and that ABA participates in the CsTRM5-mediated cell expansion during fruit elongation in cucumber.Yang Xie Xiaofeng Liu Chengzhen Sun Xiaofei Song Xiaoli Li Haonan Cui Jingyu Guo Liu Liu Ao Ying Zeqin Zhang Xueyun Zhu Liying Yan Xiaolan Zhang 2023Horticulture Research2023,10,3:1

19The remedial effect of soluble interleukin-1 receptor type Ⅱ on endometriosis in the nude mouse model显示文摘Objective:Recent studies have shown that the local expression of soluble interleukin(IL)-1 receptor typeⅡ (sIL-1 RⅡ)in endometrial tissue of women with endometriosis is decreased,and the depression of IL-1 RⅡwas more significant in infertile women than that in fertile women with endometriosis.In this research,we investigated the remedial effect of sIL-1-RⅡadministration on endometriosis in the nude mouse model.Methods: Nineteen nude model mice with endometriosis were randomly divided into three groups:group A was treated by intraperitoneal administration with only sIL-1 RⅡfor two weeks,group B was similarly treated with only IL- 1,and group C(control)was administered saline.After 2 weeks,the size of the ectopic endometrial lesions was calculated,and the expression of vascular endothelial growth factor(VEGF)and B-cell lymphoma leukemia-2(Bcl- 2)were detected by immunohistochemistry.The IL-8 and VEGF levels in the peritoneal fluid(PF)and serum were also measured by enzyme-linked immunosorbent assay(ELISA).Results:The mean size of ectopic endometrial lesion did not differ between the three groups(P>0.05).Compared with the control,the expression of VEGF and Bcl-2 was significantly lower in group A,and higher in group B.In the three groups,the levels of IL-8 in the PF and serum were highest in group A,and lowest in group B.Conclusion:sIL-1 RⅡmay suppresse hyperplasia of ectopic endometriosis,perhaps by reducing the expression of certain cytokines,such as VEGF,IL-8,and Bcl-2, which could provide a new clinical strategy for the treatment of endometriosis.Liying Gao Liang Sun Yugui Cui Zhen Hou Li Gao Jing Zhou Yundong Mao Suping Han Jiayin Liu 2010The Journal of Biomedical Research2010,24,1:1
