1Multi-omics analyses of 398 foxtail millet accessions reveal genomic regions associated with domestication,metabolite traits,and antiinflammatory effects显示文摘Foxtail millet(Setaria italica),which was domesticatedfromthewild speciesgreenfoxtail(Setaria viridis),isa richsource of phytonutrientsfor humans.To evaluate how breeding changed themetabolome offoxtail millet grains,we generated and analyzed the datasets encompassing the genomes,transcriptomes,metabolomes,and anti-inflammatory indices from 398 foxtail millet accessions.We identified hundreds of common variants that influence numerous secondary metabolites.We observed tremendous differences in natural variations of the metabolites and their underlying genetic architectures between distinct sub-groups of foxtail millet.Furthermore,we found that the selection of the gene alleles associated with yellow grains led to altered profiles of metabolites such as carotenoids and endogenous phytohormones.Using CRiSPR-mediated genome editing wevalidated the function of PHYTOENE SYNTHASE1(PSY1)gene in affecting milletgrain colorand quality.Interestingly,our in vitro cell inflammation assays showed that 83 metabolites in millet grains have anti-inflammatory effects.Taken together,ourmulti-omics study illustrates how the breeding history of foxtail millet has shaped its metabolite profile.The datasets we generated in this study also provide important resources for further understanding how millet grain quality is affected by different metabolites,laying the foundations for future millet genetic research and metabolome-assisted improvement.Xukai Li Jianhua Gao Jingyi Song Kai Guo Siyu Hou Xingchun Wang Qiang He Yanyan Zhang Yakun Zhang Yulu Yang Jiaoyan Tang Hailang Wang Staffan Persson Mingquan Huang Lishuai Xu Linlin Zhong Dongqin Li Yongming Liu Hua Wu Xianmin Diao Peng Chen Xiaowen Wang Yuanhuai Han 2022Molecular Plant2022,15,8:8
2Magnitude of modulation of gene expression in aneuploid maize depends on the extent of genomic imbalance显示文摘Aneuploidy has profound effects on an organism,typically more so than polyploidy,and the basis of this contrast is not fully understood.A dosage series of the maize long arm of chromosome 1(1L)was used to compa re relative global gene expression in diffe rent types and degrees of aneuploidy to gain insights into how the magnitude of genomic imbalance as well as hypoploidy affects global gene expression.While previously available methods require a selective examination of specific genes,RNA sequencing provides a whole-genome view of gene expression in aneuploids.Most studies of global aneuploidy effects have concentrated on individual types of aneuploids because multiple dose aneuploidies of the same genomic region are difficult to produce in most model genetic organisms.The genetic toolkit of maize allows the examination of multiple ploidies and 1-4 doses of chromosome arms.Thus,a detailed examination of expression changes both on the varied chromosome arms and elsewhere in the genome is possible,in both hypoploids and hyperploids,compared with euploid controls.Previous studies observed the inverse trans effect,in which genes not varied in DNA dosage were expressed in a negative relationship to the varied chromosomal region.This response was also the major type of changes found globally in this study.Many genes varied in dosage showed proportional expression changes,though some were seen to be partly or fully dosage compensated.It was also found that the effects of aneuploidy were progressive,with more severe aneuploids producing effects of greater magnitude.Adam F·Johnson Jie Hou Hua Yang Xiaowen Shi Chen Chen Md Soliman Islam Tieming Ji Jianlin Cheng James A·Birchler 2020Journal of Genetics and Genomics2020,47,2:3
3Antiplatelet Therapy in Acute Mild-Moderate Ischemic Stroke(ATAMIS):a parallel,randomised,open-label,multicentre,prospective study显示文摘Background A recent study shows that dual antiplatelet therapy with clopidogrel plus aspirin is superior to aspirin monotherapy for minor stroke,which is defined as a National Institutes of Health Stroke Scale(NIHSS)score of≤3.However,acute mild-moderate ischaemic stroke(4≤NIHSS≤10)still needs aggressive antiplatelet intervention to prevent deterioration and recurrence of stroke.The efficacy and safety of dual antiplatelet therapy versus aspirin monotherapy in the population are not clear.A multicentre clinical trial is designed to evaluate the efficacy and safety of clopidogrel plus aspirin therapy versus aspirin monotherapy within 48 hours of symptom onset of mild-moderate ischaemic stroke.Methods/Design The study is a randomised,open-label,multicentre,prospective trial with a target enrolment of 2700 patients from 60 centres in Northeast China.A treatment allocation identification number to each enrolled patient will be provided by a random number generator.The follow-up time for the clopidogrel plus aspirin and aspirin monotherapy groups is 90 days.The primary efficacy endpoint is a stroke progression event,which is defined as≥4 point increase in the NIHSS score in 48 hours.The second efficacy endpoints include new ischaemic stroke within 90 days,change in the NIHSS score within 14 days,modified Rankin Scale score on day 90 and other vascular or death events within 90 days.The safety endpoints include mucocutaneous haemorrhage,organ haemorrhage and intracranial haemorrhage,adverse events and severe adverse events.χ^(2)test,t-test(or Mann-Whitney test),survival analysis and Cox proportional hazards models will be conducted.The findings of the study may provide an important evidence for clinical practice for these patients.Discussion The trial will be conducted under a rational design and will provide valuable evidence on the appropriate treatment for this population.Ethics and dissemination The study was reviewed and approved by the Ethics Committee of the General Hospital of Shen-Yang Military Region(no K(2016)6).Trial registration number NCT02869009;Pre-results.Xiaowen Hou Xiaoqiu Li Xinhong Wang Huisheng Chen 2018Stroke & Vascular Neurology2018,3,4:3
4Effectiveness of intravenous r- tPA versus UK for acute ischaemic stroke: a nationwide prospective Chinese registry study显示文摘Background Intravenous recombinant tissue plasminogen activator(r-tPA)and urokinase(UK)are both recommended for the treatment of acute ischaemic stroke(AIS)in China,but with few comparative outcome data being available.We aimed to compare the outcomes of these two thrombolytic agents for the treatment of patients within 4.5 hours of onset of AIS in routine clinical practice in China.Methods A pre-planned,prospective,nationwide,multicentre,real-world registry of consecutive patients with AIS(age≥18 years)who received r-tPA or UK within 4.5 hours of symptom onset according to local decision-making and guideline recommendations during 2017-2019.The primary effectiveness outcome was the proportion of patients with an excellent functional outcome(defined by modified Rankin scale scores 0 to 1)at 90 days.The key safety endpoint was symptomatic intracranial haemorrhage according to standard definitions.Multivariable logistic regression was used for comparative analysis,with adjustment according to propensity scores to ensure balance in baseline characteristics.Results Overall,4130 patients with AIS were registered but 320 had incomplete or missing data,leaving 3810 with available data for analysis of whom 2666 received r-tPA(median dose 0.88(IQR 0.78-0.90)mg/kg)and 1144 received UK(1.71(1.43-2.00)×104 international unit per kilogram).There were several significant intergroup differences in patient characteristics:r-tPA patients were more educated,had less history of stroke,lower systolic blood pressure,greater neurological impairment and shorter treatment times from symptom onset than UK patients.However,in adjusted analysis,the frequency of excellent outcome(OR 1.18,95%CI 1.00 to 1.40,p=0.052)and symptomatic intracranial haemorrhage(OR 0.70,95%CI 0.33 to 1.47,p=0.344)were similar between groups.Conclusions UK may be as effective and carry a similar safety profile as r-tPA in treating mild to moderate AIS within guidelines in China.Registration http://www.clinicaltrials.gov.unique identifier:NCT02854592.Xinhong Wang Xiaoqiu Li Yuming Xu Runhui Li Qingcheng Yang Yong Zhao Fengyun Wang Baoying Sheng Runqing Wang Shaoyuan Chen Lihua Wang Liying Shen Xiaowen Hou Yu Cui Duolao Wang Bin Peng Craig S Anderson Huisheng Chen On behalf of INTRECIS Investigators 2021Stroke & Vascular Neurology2021,6,4:2
5Surface patterning and modification of polyurethane biomaterials using silsesquioxane-gelatin additives for improved endothelial affinity显示文摘Polyurethanes(PUs) are well-known for their biocompatibility but their intrinsic inert property hampers cell-matrix interactions. Surface modifications are thus necessary to widen their use for biomedical applications. In this work, surface modifications of PU were achieved first by incorporating polyhedral oligomeric silsesquioxane(POSS), followed by alteration of the surface topography via the breath figures method. Subsequently, surface chemistry was also modified by immobilization of gelatin molecules through grafting, for the enhancement of the surface cytocompatibility. Scanning electron microscopy(SEM) was used to verify the formation of highly ordered microstructures while static contact angle, FTIR and XPS confirmed the successful grafting of gelatin molecules onto the surfaces. In vitro culture of human umbilical vein endothelial cells(HUVECs) revealed that endothelial cell adhesion and proliferation were significantly enhanced on the gelatin-modified surfaces, as shown by live/dead staining and WST-1 proliferation assay. The results indicated that the combination of the strategies yielded an interface that improves cell attachment and subsequent growth. 8Facile fabrication of branched-chain carbohydrate chips for studying carbohydrate-protein interactions by QCM biosensor显示文摘A novel approach for fabricating branched-chain(BC) carbohydrate chips to study carbohydrate-protein interactions using Quartz Crystal Microbalance(QCM) biosensor was developed. This approach utilizes functional alkynyl-branch molecule modified chip surfaces, which through the terminal alkynyl group for covalent linking of unprotected azide-carbohydrates. The unprotected azide-carbohydrates were syhthesized in one-step using 2-azido-1,3-dimethyl-imidazolinium as catalyst, avoiding complex chemical modifications. Additionally, the branch surface modified with the carbohydrates not only supplies more specific binding site but also reveals significant cluster effect. To exemplify the sugar cluster effect on BC carbohydrate chips, BC Galactose and Mannose chips prepared in this work were used to determine carbohydrate-lectin interactions using QCM biosensor. The results clearly showed that BC chip significantly improves the detection sensitivity compared with the straight-chain(SC) chip. 11Empirical Study on Customer Satisfaction Influencing Factors of Industry Application Products Based on Experiential Level Theory显示文摘The rapid development of industry application products has greatly influenced society. It has become essential for some telecommunication companies to focus on customer satisfaction with industrial products used by the government and enterprise customers. Following the review of a previous study of customer satisfaction and with the help of experiential level theory,this article attempts to establish a Satisfaction Factors Model for researching customer satisfaction with industry applications. We chose a product named 'Mobile Monitor' and investigated the emotional satisfaction of 127 users and company leaders from different walks of life,and showed that 7 of the product's features affected the customers' overall emotional satisfaction. Lastly,this paper points out management implications of this application based on quantitative results. It presents a preliminary exploration program for customer satisfaction research in this specific field. 12Small-Molecule Fluorophores for Near-Infrared IIb Imaging and ImageGuided Therapy of Vascular Diseases显示文摘Accurate and dynamic visualization of vascular diseases can contribute to restraining further deterioration from diseases in a timely manner.However,visualization is still unable to precisely determine whether and to what extent blood vessels or brain tissues are damaged.Here,we report novel benzobis(1,2,5-thiadiazole)-based second near-infrared region(NIR-II)fluorophores HY1-HY4 with highly twisted structures(55°at the S_(0) state),extremely strong aggregation-induced emission(AIE)characteristics(I/I_(0)>13),and remarkably high fluorescence quantum yields(QYs)(up to 14.45%)in the NIR-II region(>1000 nm)and∼0.27%in the nearinfrared IIb window(NIR-IIb,>1500 nm)in aqueous solution.Using NIR-IIb AIE HY4 dots,high-resolution NIR-IIb fluorescence imaging of revascularization and thrombolysis,and real-time feedback of the therapeutic efficacy of Chinese medicine Dengzhan Xixin injection(DXI)on ischemic stroke,were achieved for the first time.In addition,results showed that DXI conferred neuroprotection against cerebral ischemia injury mediated via the angiogenesis pathway.These attractive results provide a new perspective for designing ultrabright NIR-IIb probes for vascular-related phenomena,disease assessment,and precise intraoperative imageguided therapy with a deeper tissue penetration depth and higher resolution.Yang Li Hua Zhu Xiaobo Wang Yan Cui Lijuan Gu Xiaowen Hou Mengting Guan Junzhu Wu Yuling Xiao Xiaoxing Xiong Xianli Meng Xuechuan Hong 2022CCS Chemistry2022,4,12:0
13Fluorescence and sensitization performance of phenylene-vinylene-substituted polythiophene显示文摘A new kind of polythiophene derivative, Poly(3-{2-[4-(2-ethylhexyloxy)-phenyl]-vinyl}-2,2′-bithiophene) (PTh), was applied in dye-sensitized solar cell to extend the light response of nanocrystalline TiO2 electrode. UV-vis absorption and fluorescence spectra were employed to investigate the interaction of PTh with nanocrystalline TiO2. The absorption coefficient of the PTh was high in visible part of spec- trum, and the fluorescence emission of the PTh can be efficiently quenched by TiO2 nanoparticles ow- ing to charge injection from the excited singlet state of PTh to the conduction band of the TiO2 particles. Cyclic voltammetry measurements were performed to study the dye regeneration reaction at the na- nocrystalline TiO2/electrolyte interface. 14Genomic imbalance modulates transposable element expression in maize显示文摘Genomic imbalance refers to the more severe phenotypic consequences of changing part of a chromosome compared with the whole genome set.Previous genome imbalance studies in maize have identified prevalent inverse modulation of genes on the unvaried chromosomes(trans)with both the addition or subtraction of chromosome arms.Transposable elements(TEs)comprise a substantial fraction of the genome,and their reaction to genomic imbalance is therefore of interest.Here,we analyzed TE expression using RNA-seq data of aneuploidy and ploidy series and found that most aneuploidies showed an inverse modulation of TEs,but reductions in monosomy and increases in disomy and trisomy were also common.By contrast,the ploidy series showed little TE modulation.The modulation of TEs and genes in the same experimental group were compared,and TEs showed greater modulation than genes,especially in disomy.Class Ⅰ and Ⅱ TEs were differentially modulated in most aneuploidies,and some superfamilies in each TE class also showed differential modulation.Finally,the significantly upregulated TEs in three disomies(TB-7Lb,TB9Lc,and TB-10L19)did not increase the proportion of adjacent gene expression when compared with non-differentially expressed TEs,indicating that modulations of TEs do not compound the effect on genes.These results suggest that the prevalent inverse TE modulation in aneuploidy results from stoichiometric upset of the regulatory machinery used by TEs,similar to the response of core genes to genomic imbalance.Hua Yang Xiaowen Shi Chen Chen Jie Hou Tieming Ji Jianlin Cheng James A.Birchler 2023Plant Communications2023,4,2:0
15CSI-Model-Based Research on Factors Affecting Rate Satisfaction of Government and Corporate CustomersTaking Special-Line Products as an Example显示文摘The rate satisfaction of government and corporate customers,to which telecom companies pay special attention,is an important part of the overall satisfaction of the market segment.Focusing on specialline products and based on interview with experts and literature review,this article builds an effect model comprising four factors affecting the major government and corporate customers' satisfaction with the rate,including cost performance,discount degree,rate communication and bill composition.Besides,employing AMOS and other analytical tools,it reveals significant positive effects of the aforementioned four factors on the customer's rate satisfaction,and arranges them in sequence according to the path coefficient of each influence.Based on quantified results,this article proposes management recommendations to improve the rate satisfaction,providing basis for research on the overall rate satisfaction optimization of government and corporate customers.LI Xiaowen TONG Lili HU Zuohao HOU Xiaoge 2015China Communications2015,12,8:0
