3Role of prophylactic hyperthermic intraperitoneal chemotherapy in patients with locally advanced gastric cancer显示文摘BACKGROUND Gastric cancer is the second most common malignant tumor in China,ranking third among all malignant tumor mortality rates.Hyperthermic intraperitoneal chemotherapy(HIPEC)has been shown to increase significantly the effectiveness of intraperitoneal chemotherapeutic drugs,prolong the action time of these drugs on intraperitoneal tumor cells,and enhance their diffusion in tumor tissues.HIPEC may be one of the best choices for the eradication of residual cancer cells in the abdominal cavity.AIM The aim of this study was to study the role of preventive HIPEC after radical gastrectomy.METHODS A prospective analysis was performed with patients with c T4 N0-3 M0 gastric cancer to compare the effects of postoperative prophylactic HIPEC plus intravenous chemotherapy with those of routine adjuvant chemotherapy.Patients’medical records were analyzed,and differences in the peritoneal recurrence rate,diseasefree survival time,and total survival time between groups were examined.RESULTS The first site of tumor recurrence was the peritoneum in 11 cases in the conventional adjuvant chemotherapy group and in 2 cases in the HIPEC group(P=0.020).The 1-year and 3-year disease-free survival rates were 91.9%and 60.4%,respectively,in the conventional adjuvant chemotherapy group and 92.1%and 63.0%,respectively,in the HIPEC group.The 1-year and 3-year overall survival rates were 95.2%and 66.3%,respectively,in the conventional adjuvant chemotherapy group and 96.1%and 68.6%,respectively,in the HIPEC group.No significant difference in postoperative or chemotherapy complications was observed between groups.CONCLUSION In patients with c T4 N0-3 M0 gastric cancer,prophylactic HIPEC after radical tumor surgery is beneficial to reduce peritoneal tumor recurrence and prolong survival.Tian-Yu Xie Di Wu Shuo Li Zhao-Yan Qiu Qi-Ying Song Da Guan Li-Peng Wang Xiong-Guang Li Feng Duan Xin-Xin Wang 2020World Journal of Gastrointestinal Oncology2020,12,7:7
4PCDH17 increases the sensitivity of colorectal cancer to 5-fluorouracil treatment by inducing apoptosis and autophagic cell death显示文摘5-Fluorouracil(5-FU)is known as a first-line chemotherapeutic agent against colorectal cancer(CRC),but drug resistance occurs frequently and significantly limits its clinical success.Our previous study showed that the protocadherin 17(PCDH17)gene was frequently methylated and functioned as a tumor suppressor in CRC.However,the relationship between PCDH17 and 5-FU resistance in CRC remains unclear.Here,we revealed that PCDH17 was more highly expressed in 5-FU-sensitive CRC tissues than in 5-FU-resistant CRC tissues,and high expression of PCDH17 was correlated with high BECN1 expression.Moreover,this expression profile contributed to superior prognosis and increased survival in CRC patients.Restoring PCDH17 expression augmented the 5-FU sensitivity of CRC in vitro and in vivo by promoting apoptosis and autophagic cell death.Furthermore,autophagy played a dominant role in PCDH17-induced cell death,as an autophagy inhibitor blocked cell death to a greater extent than the pancaspase inhibitor Z-VAD-FMK.PCDH17 inhibition by siRNA decreased the autophagy response and 5-FU sensitivity.Mechanistically,we showed that c-Jun NH2-terminal kinase(JNK)activation was a key determinant in PCDH17-induced autophagy.The compound SP600125,an inhibitor of JNK,suppressed autophagy and 5-FU-induced cell death in PCDH17-reexpressing CRC cells.Taken together,our findings suggest for the first time that PCDH17 increases the sensitivity of CRC to 5-FU treatment by inducing apoptosis and JNK-dependent autophagic cell death.PCDH17 may be a potential prognostic marker for predicting 5-FU sensitivity in CRC patients.Shuiping Liu Haoming Lin Da Wang Qiang Li Hong Luo Guoxiong Li Xiaohui Chen Yongqiang Li Peng Chen Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Mingming Zhang Xuemeng Han Qiujie Li Liuxi Chen Ying Liu Xiaying Chen Guohua Li Yu Xiang Ting Duan Jiao Feng Jianshu Lou Xingxing Huang Qin Zhang Ting Pan Lili Yan Ting Jin Wenzheng Zhang Lvjia Zhuo Yitian Sun Tian Xie Xinbing Sui 2019Signal Transduction and Targeted Therapy2019,4,1:4
5Development of Cueing Algorithm Based on “Closed-Loop” Control for Flight Simulator Motion System显示文摘The classical washout algorithm had fixed gains and manually constructed filters, so that it led to poor adaptability. Furthermore, it lost the sustained acceleration cues of high-and mid-frequency in cross-over(tilt-coordination) channel, and the acceleration of cross-over frequency was also limited by angular velocity limiter, so the false cues in flight simulation process were clearly perceived by pilots. The paper studied the characteristics of the classical washout algorithm and flight simulator motion platform, tried to redesign the source of cross-over acceleration channel and translation acceleration channel, and transferred the part of cross-over acceleration that was unsimulated sustained acceleration to translation acceleration channel. Comparisons were mainly made between classical washout algorithm and revised algorithm in a longitudinal/pitch direction. The evaluation was based on the implementation of human vestibular perception system. The results demonstrated that the revised algorithm could significantly reduce the phase lag, and improved the spikes tracking performance. Furthermore, sensory angular velocity and the error of sensory acceleration were strictly controlled within the threshold of human perception system, and the displacement was a little broader than the classical washout algorithm. Therefore, it was proved that the new algorithm could diminish the filters parameters and heighten the self-adaptability for the washout algorithm. 7Differentiation of human olfactory mucosa mesenchymal stem cells into photoreceptor cells in vitro显示文摘AIM:To investigate whether the human olfactory mucosa mesenchymal stem cells(OM-MSCs) can differentiate into photoreceptor cells in vitro.METHODS:Through the olfactory mucosa adherent method,olfactory mucosa was isolated,cultured and identified in vitro among mesenchymal stem cells.The cell surface markers were analyzed by flow cytometry,induced to differentiate into retinal photoreceptor cells in vitro,and the expression of rhodopsin was observed and identified by Immunofluorescence and Western blot methods.RESULTS:OM-MSCs from human were spindle cellbased,and showing radial colony arrangement.OM-MSCs were negative for CD34,CD45 and CD105,but positive for CD73 and CD90.Following induction,a strong positive reaction was produced by photoreceptor specific marker rhodopsin in the cells.CONSLUSION:This novel finding demonstrates that OM-MSCs can be cultured and expanded in vitro.They possess biological characteristics of mesenchymal stem cells,and have the ability to be induced into retinal cells.Wen Lu Da Duan Zacharia Ackbarkhan Ming Lu Min-Li Huang 2017International Journal of Ophthalmology(English edition)2017,10,10:2

19Reductive damage induced autophagy inhibition for tumor therapy显示文摘Numerous therapeutic anti-tumor strategies have been developed in recent decades.However,their therapeutic efficacy is reduced by the intrinsic protective autophagy of tumors.Autophagy plays a key role in tumorigenesis and tumor treatment,in which the overproduction of reactive oxygen species(ROS)is recognized as the direct cause of protective autophagy.Only a few molecules have been employed as autophagy inhibitors in tumor therapy to reduce protective autophagy.Among them,hydroxychloroquine is the most commonly used autophagy inhibitor in clinics,but it is severely limited by its high therapeutic dose,significant toxicity,poor reversal efficacy,and nonspecific action.Herein,we demonstrate a reductive-damage strategy to enable tumor therapy by the inhibition of protective autophagy via the catalytic scavenging of ROS using porous nanorods of ceria(PN-CeO_(2))nanozymes as autophagy inhibitor.The antineoplastic effects of PN-CeO_(2)were mediated by its high reductive activity for intratumoral ROS degradation,thereby inhibiting protective autophagy and activating apoptosis by suppressing the activities of phosphatidylinositide 3-kinase/protein kinase B and p38 mitogen-activated protein kinase pathways in human cutaneous squamous cell carcinoma.Further investigation highlighted PN-CeO_(2)as a safe and efficient anti-tumor autophagy inhibitor.Overall,this study presents a reductive-damage strategy as a promising anti-tumor approach that catalytically inhibits autophagy and activates the intrinsic antioxidant pathways of tumor cells and also shows its potential for the therapy of other autophagy-related diseases.Yuqian Wang Yingjian Huang Yu Fu Zhixiong Guo Da Chen Fangxian Cao Qi Ye Qiqi Duan Meng Liu Ning Wang Dan Han Chaoyi Qu Zhimin Tian Yongquan Qu Yan Zheng 2023Nano Research2023,16,4:1
