1Enhanced Vitamin C Production Mediated by an ABA-Induced PTP-like Nucleotidase Improves Plant Drought Tolerance in Arabidopsis and Maize显示文摘Abscisic acid(ABA)is a key phytohormone that mediates environmental stress responses.Vitamin C,or L-ascorbic acid(AsA),is the most abundant antioxidant protecting against stress damage in plants.How the ABA and AsA signaling pathways interact in stress responses remains elusive.In this study,we characterized the role of a previously unidentified gene,PTPN(PTP-like Nucleotidase)in plant drought tolerance.In Arabidopsis,(AtPTPN was expressed in multiple tissues and upregulated by ABA and drought treatments.Loss-of-function mutants oiAtPTPN were hyposensitive to ABA but hypersensitive to drought stresses,whereas plants with enhanced expression AtPTPN showed opposite phenotypes to.Overexpression of maize PTPN(ZmPTPN)promoted,while knockdown oiZmPTPN inhibited plant drought tolerance,indicating conserved and positive roles of PTPN in plant drought tolerance.We found that both AtPTPN and ZmPTPN release Pi by hydrolyzing GDP/GMP/dGMP/IMP/dIMP,and that AtPTPN positively regulated AsA production via endogenous Pi content control.Consistently,overexpression of VTC2,the rate-limiting synthetic enzyme in AsA biosynthesis,promoted AsA production and plant drought tolerance,and these effects were largely dependent on AtPTPN activity.Furthermore,we demonstrated that the heat shock transcription factor HSFA6a directly binds the AtPTPN promoter and activates AtPTPN expression.Genetic analyses showed that AtPTPN is required for HSFA6a to regulate ABA and drought responses.Taken together,our data indicate that PTPN-mediated crosstalk between the ABA signaling and AsA biosynthesis pathways positively controls plant drought tolerance.Hui Zhang Yanli Xiang Neng He Xiangguo Liu Hongbo Liu Liping Fang Fei Zhang Xiaopeng Sun Delin Zhang Xingwang Li William Terzaghi Jianbing Yan Mingqiu Dai 2020Molecular Plant2020,13,5:7
2Molecular characterization and expression analysis of caveolin-1 in pig tissues显示文摘Members of the caveolin family played important roles during fundamental cellular processes,such as regulation of cell morphology,migration,and gene expression in muscle cells.In this study,caveolin-1 (Cav-1),one of the caveolins,was identified from longissimus dorsi muscle of Large Yorkshire pig and Chinese indigenous Lantang pig based on the results of mRNA differential display analysis.The deduced amino acids sequence of the porcine Cav-1 contained a caveolin domain,and was very conservative among different species.The Cav-1 mRNA was widely expressed in the eight tissues in this study,including heart,liver,kidney,encephalon,spleen,lung,longissimus dorsi muscle,and back fat, and the highest expression quantity was found in back fat of the two pig breeds.The expression quantity of porcine Cav-1 in back fat and longissimus dorsi muscle of Lantang pig was significantly higher than that of Large Yorkshire(P<0.01,and P<0.05,respectively).These results suggested that the Cav-1 might be a candidate gene for carcass traits,and might provide valuable information for understanding the mechanism of caveolae signaling in fat deposition by using the animal model of pig.WANG Chong1,MEI YingJie 1,LI Li 1,MO DeLin 2,LI JiaQi 1,ZHANG Hao 1,TIAN XingGuo 1 &CHEN YaoSheng2 1College of Animal Science,South China Agricultural University,Guangzhou 510642,China 2School of Life Science,Sun Yat-Sen University,Guangzhou 510006,China 2008Science China(Life Sciences)2008,51,7:2
3Molecular simulations of charged complex fluids: A review显示文摘Molecular simulation plays an increasingly important role in studying the properties of complex fluid systems containing charges,such as ions,piezoelectric materials,ionic liquids,ionic surfactants,polyelectrolytes,zwitterionic materials,nucleic acids,proteins,biomembranes and etc.,where the electrostatic interactions are of special significance.Several methods have been available for treating the electrostatic interactions in explicit and implicit solvent models.Accurate and efficient treatment of such interactions has therefore always been one of the most challenging issues in classical molecular dynamics simulations due to their inhomogeneity and long-range characteristics.Currently,two major challenges remain in the application field of electrostatic interactions in molecular simulations;(i)improving the representation of electrostatic interactions while reducing the computational costs in molecular simulations;(ii)revealing the role of electrostatic interactions in regulating the specific properties of complex fluids.In this review,the calculation methods of electrostatic interactions,including basic principles,applicable conditions,advantages and disadvantages are summarized and compared.Subsequently,the specific role of electrostatic interactions in governing the properties and behaviors of different complex fluids is emphasized and explained.Finally,challenges and perspective on the computational study of charged systems are given.Zhiyong Xu Zhongjin He Xuebo Quan Delin Sun Zhaohong Miao Hai Yu Shengjiang Yang Zheng Chen Jinxiang Zeng Jian Zhou 2021Chinese Journal of Chemical Engineering2021,34,3:2

16Parameter Identification Based on Force-Displacement Curves for a Bolted Joint Finite Element Model显示文摘In order to identify the uncertain parameters of a bolted joint finite element model,a simple and applicable way of parameter identification is introduced.By utilizing numerical simulation with the Abaqus software and experimental investigation with the MTS material testing system,the tangential force-displacement curves that reflect the characteristics of the bolted joint were acquired.On the basis of this,by employing the response surface methodology(RSM)and genetic algorithms(GAs),parameters in the FEM model were identified.The force-displacement curves by both virtual and experimental approaches are well correlated at the end.This phenomenon-based parameter identification method may help facilitate precise prediction of complex jointed connection structures.Delin Sun Ridong Liao Dexing Qian Zhengkun Cheng 2017Journal of Beijing Institute of Technology2017,26,1:0
17Kinetic Characteristic of Hydrogenation Zr-Ti-Cu-Ni-Be Bulk Amorphous Alloys显示文摘The relationship between the hydrogen content and the microhardness and the charging period, the effect of hydrogen on the activation energy, the kinetics of glass transition and crystallization of Zr-Ti-Cu-Ni-Be bulk amorphous alloy were studied by differential scanning calorimetry (DSC) and the Kissinger equation. It shows that both of the hydrogen content and the microhardness are related to the charging period, and that the glass transition and crystallization behavior are associated with the heating rate, and possess the kinetic effect. Hydrogen increases the glass transition temperature and the crystallization temperature, decreasing the enthalpies in the different stages of crystallization.Hydrogen increases the activation energies of the glass transition and the crystallization and changes the kinetic effect. 18Progress of Brain Network Studies on Anesthesia and Consciousness: Framework and Clinical Applications显示文摘Although the relationship between anesthesia and consciousness has been investigated for decades, our understanding of the underlying neural mechanisms of anesthesia and consciousness remains rudimentary, which limits the development of systems for anesthesia monitoring and consciousness evaluation. Moreover, the current practices for anesthesia monitoring are mainly based on methods that do not provide adequate information and may present obstacles to the precise application of anesthesia. Most recently, there has been a growing trend to utilize brain network analysis to reveal the mechanisms of anesthesia, with the aim of providing novel insights to promote practical application. This review summarizes recent research on brain network studies of anesthesia, and compares the underlying neural mechanisms of consciousness and anesthesia along with the neural signs and measures of the distinct aspects of neural activity. Using the theory of cortical fragmentation as a starting point, we introduce important methods and research involving connectivity and network analysis. We demonstrate that whole-brain multimodal network data can provide important supplementary clinical information. 19Discovery of a first-in-class ANXA3 degrader for the treatment of triple-negative breast cancer显示文摘Triple-negative breast cancer(TNBC)is a nasty disease with extremely high malignancy and poor prognosis.Annexin A3(ANXA3)is a potential prognosis biomarker,displaying an excellent correlation of ANXA3 overexpression with patients'poor prognosis.Silencing the expression of ANXA3effectively inhibits the proliferation and metastasis of TNBC,suggesting that ANXA3 can be a promising therapeutic target to treat TNBC.Herein,we report a first-in-class ANXA3-targeted small molecule(R)-SL18,which demonstrated excellent anti-proliferative and anti-invasive activities to TNBC cells.(R)-SL18 directly bound to ANXA3 and increased its ubiquitination,thereby inducing ANXA3 degradation with moderate family selectivity.Importantly,(R)-SL18 showed a safe and effective therapeutic potency in a high ANXA3-expressing TNBC patient-derived xenograft model.Furthermore,(R)-SL18 could reduce theβ-catenin level,and accordingly inhibit the Wnt/β-catenin signaling pathway in TNBC cells.Collectively,our data suggested that targeting degradation of ANXA3 by(R)-SL18 possesses the potential to treat TNBC.Yongxi Liang Delin Min Hulin Fan Kunlin Liu Juchuanli Tu Xueyan He Bingjie Liu Lu Zhou Suling Liu Xun Sun 2023Acta Pharmaceutica Sinica B2023,13,4:0
20Strategic design of lysine-targeted irreversible covalent NDM-1 inhibitors显示文摘New Delhi metallo-β-lactamase 1(NDM-1) can hydrolyze most β-lactam antibiotics, which is the major factor for drug resistance of Gram-negative bacteria. The binding of most reversible inhibitors to NDM-1 is relatively weak due to the shallow active pocket of NDM-1. Alternatively, irreversible covalent inhibitors can prevent their dissociation from the target, leading to permanent inactivation of the protein.Herein, we report a series of irreversible covalent inhibitors of NDM-1 targeting the conserved Lys211 in the active pocket. Several methods, including mass spectrometry, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, fluorescent labeling, and coumarin probe were used to demonstrate that pentafluorophenyl ester formed a covalent bond with Lys211. Moreover, our target inhibitor, in combination with meropenem, achieved an antibacterial effect on drug-resistant bacteria, along with an excellent safety profile.
