1Upreguiation of human telomerase reverse transcriptase mRNA expression by in vitro transfection of hepatitis B virus X gene into human hepatocarcinoma and cholangiocarcinoma cells显示文摘AIM: To study the changes of human telomerase reverse transcriptase (hTERT) mRNA expression in human hepatocarcinoma cell lines (HepG2) and cholangiocarcinoma cell lines (QBC939) after HBx gene transfection and to illustrate the significance of transcriptional regulation of hTERT gene by HBx gene in the carcinogenesis.METHODS: HepG2 and QBC939 cell lines were cultured and co-transfected with eukaryotic expression vector containing the HBx coding region and cloning vector containing enhanced green fluorescent protein (EGFP) coding sequence using lipid-mediated gene transduction technique. Thirty-six hours after transfection, EGFP expression in cells was used as the indicator of successful transfection. Flow cytometry was performed to determine the transfection efficiency.Cells were harvested and total RNA was extracted using TRIzol() reagent. The expression of hTERT mRNA in HepG2and QBC939 cell lines was assayed by reverse transcriptionpolymerase chain reaction. The expression of HBx protein in both cell lines was detected by immunocytochemical staining and Western blotting.RESULTS: Flow cytometry showed that the transfection efficiency was 46.4% in HepG2 cells and 29.6% in QBC939cells for both HBx gene expression vector and blank vector. The expression of hTERT mRNA was meaningfully increased in HepG2 and QBC939 cell lines when transfected with HBx gene expression vector compared to those transfected with OPTI-MEM medium and blank vector.Immunocytochemical staining and Western blotting revealed HBx protein expression in HepG2 and QBC939cells only when transfected with HBx gene.CONCLUSION: HBx gene transfection can upregulate the transcriptional expression of hTERT mRNA. 2Anti-proliferation effects of Twist gene silencing in gastric cancer SGC7901 cells显示文摘AIM:To study the role of Twist gene in gastric cancer by gene silencing,including the potential of induction of apoptosis,cell cycle arrest,and proliferation inhibition in human malignant gastric SGC7901 cells.METHODS:The expression level of Twist in gastric cancer samples was measured by immunohistochemistry.The effects of Twist gene silencing were detected at both m RNA and protein levels by RT-PCR and Western blot.We also evaluated the cell proliferation and apoptosis by CCK-8 assay and flow cytometry.We determined the activity of caspase-3 and caspase-9 with a caspase activity assay kit.Cell cycle distribution was analyzed by flow cytometry.Cell migration and invasion ability was evaluated by wound scratch assay and Boyden chamber assay.RESULTS:Twist protein was highly expressed in gastric cancer samples.Twist gene silencing significantly induced apoptosis,cell cycle arrest at G0/G1 phase,proliferation inhibition,and reduced the ability of migration and invasion in human gastric cancer SGC7901 cells.Meanwhile,both caspase-3 and caspase-9 were activated.CONCLUSION:The Twist gene could serve as a potential molecular target for gene therapy of gastric cancer with targeted small interfering RNA.Hui Zhang Jian Gong Di Kong Hong-Yi Liu 2015World Journal of Gastroenterology2015,21,10:11
3Computational and experimental analysis of Mach 2 air flow over a blunt body with plasma aerodynamic actuation显示文摘According to the mechanism of the arc plasma heating effect,and from a phenomenological perspective of view,the plasma actuation was simplified as heating energy injected into the supersonic flow field for the numerical research on controlling detached shock of the blunt body in non-center symmetrical positions.Besides,experimental research on the form and strength of detached shock wave control by plasma aerodynamic actuation in non-center symmetrical positions was conducted in a high-speed shock tunnel(M=2).The results showed that the detached distance of shock wave increased and the strength of normal shock wave ahead of the detached shock wave reduced when plasma actuation was applied.The control effect was greatly improved after the magnetic field was applied and the effect of upwind-direction flow was the best one.When the upwind-direction flow was applied with 1000 V voltage actuation,the distance of detached shock wave would increase from 3.4 to 7.6 mm and the time average strength of normal shock wave was weaken by 5.5%.At last,the mechanism of plasma actuation on controlling the detached shock wave was briefly analyzed.SUN Quan CHENG BangQin LI YingHong KONG WeiSong LI Jun ZHU YiFei JIN Di 2013Science China(Technological Sciences)2013,56,4:9

5Efficacy of platinum in advanced triple-negative breast cancer with germline BRCA mutation determined by next generation sequencing显示文摘Objective:To compare the efficacy of platinum-and non-platinum-based regimens as first-line treatment for advanced triple-negative breast cancer(TNBC)and analyze the relationship between their efficacy and BRCA gene status.Methods:Retrospectively analyze clinical data of 220 patients diagnosed pathologically with advanced TNBC and treated at the Department of Breast Oncology,Peking University Cancer Hospital from 2013 to 2018 and evaluate the efficacy of chemotherapy.A total of 114 patients had BRCA1/2 gene tested by next generation sequencing(NGS)using peripheral blood,and we analyzed the correlation between their efficacy and BRCA1/2 gene status.Results:Non-platinum-based chemotherapy(NPCT)was administered to 129 and platinum-based chemotherapy(PBCT)to 91 study patients.The clinical benefit rate(CBR)and median progression-free survival(PFS)were not statistically different between NPCT and PBCT groups.The median overall survival(OS)was 30.0 and 22.5 months for PBCT and NPCT group,respectively[P=0.090,hazard ratios(HR)=0.703].BRCA status was assessed in 114 patients,14 of whom had deleterious germline BRCA1/2(g BRCA)mutations(seven in each group).In PBCT group,the CBR was 85.7%and 35.1%for patients with and without deleterious g BRCA mutations,respectively(P=0.039).The median PFS were 14.9 and 5.3 months and median OS were 26.5 and 15.5 months for patients with and without deleterious g BRCA mutations,respectively(P=0.001,P=0.161,respectively).Patients in PBCT group had significantly greater rates of grade 3-4 anemia(5.5%vs.0%)and thrombocytopenia(8.8%vs.0%),whereas palmar-plantar erythrodysesthesia(12.4%vs.0%)and peripheral neuropathy(8.6%vs.1.1%)occurred more frequently in NPCT group.Conclusions:Platinum-based regimens are more effective in patients with deleterious g BRCA mutations,but no difference in patients without BRCA gene mutations,so non-platinum is an option in patients without BRCA gene mutations considering the toxicity and side effect.And we recommend that patients with advanced TNBC should have BRCA gene test.Nan Wang Kun Li Wenfa Huang Weiyao Kong Xiaoran Liu Weijie Shi Feng Xie Hanfang Jiang Guohong Song Lijun Di Quanren Wang Jianjun Yu Huiping Li 2020Chinese Journal of Cancer Research2020,32,2:7
6The grafts modified by heparinization and catalytic nitric oxide generation used for vascular implantation in rats显示文摘Small-diameter(<6mm)vascular grafts are increasingly needed in peripheral vascular surgery but have few successes because of acute thrombosis,incomplete endothelialization and intimal hyperplasia after implantation.This study used electrospun poly(ε-caprolactone)as the matrix material.Heparin and selenium-containing catalyst-organoselenium modified polyethyleneimine were introduced through layer-by-layer assembly in order to build a vascular graft with in situ nitric oxide(NO)generation.The aim of this study was to explore the application of the graft with improved histocompatibility and biological function for vascular implantation in rats.After implantation in rats,compared to poly(ε-caprolactone),the modified grafts could promote the adhesion and proliferation of endothelial cells,and inhibit the adhesion of smooth muscle cells.The modified grafts remarkably promoted endothelialization,inhibited intimal hyperplasia and increased the ratio of alternatively activated macrophages(M2)to classical activated macrophages(M1).This work constructed a vascular graft with heparinization and catalytic NO generation for improving the vascularization,and accelerating the tissue regeneration by regulating the inflammatory response.The present study indicates that it is a promising method for regulating response and tissue regeneration of small diameter vascular grafts by a novel approach of combining heparinization and catalytic NO generation.Jingchen Gao Li Jiang Qinge Liang Jie Shi Ding Hou Di Tang Siyuan Chen Deling Kong hufangWang 2018Regenerative Biomaterials2018,5,2:4

8Photocatalytic activity of Lu^(3+)/TiO_2 prepared by ball milling method显示文摘Ball milling method was applied to prepare Lu^(3+)/TiO_2 photocatalysts. The catalysts were characterized with X-ray powder diffraction(XRD), X-ray photoelectron spectroscopy(XPS), UV-visible diffuse reflectance spectra(UV-vis DRS), energy dispersive X-ray spectrometer(EDS), transmission electron microscopy(TEM) and Brunauer-Emmett-Teller(BET) method. The photocatalytic activities were determined by the degradation of methylene blue(MB) equipped with a 300 W medium pressure mercury lamp. Results show that the first order reaction rate constants of Lu^(3+)/TiO_2 and pure TiO_2 are0.0565 and 0.0263 min-1, respectively, which both were evaluated under the condition of catalysts loading of 0.2 g/L,initial concentration of 25 mg/L for MB, mole ratio of Lu^(3+)/TiO_2 of 1.5% and milling time of 4 h. 9Self-immolative micellar drug delivery:The linker matters显示文摘Redox-responsive polymer-drug conjugate micelles are excellent nanoscale vehicles for self-immolative intracellular drug delivery.To covalently connect the polymer and drug,disulfide-bearing linkers,such as 3,3'-dithiodipropionic acid (DDPA) and 4,4'-dithiodibutyric add (DDBA),are used.In this paper,we report the influence of linker length on the therapeutic outcome of redox-sensitive conjugate micelles.Curcumin was selected as the model drug and it was conjugated to a multivalent methoxy poly(ethylene glycol)-polylysine copolymer with DDPA or DDBA as the linker.The obtained two polymer-curcumin conjugates were amphiphilic and could self-assemble into micelles that have a hydrodynamic diameter less than 100 nm.The loading of curcumin in both micelles was above 20% (w/w).Irrespective of the linker type,micelle disassembly was observed due to the collapse of the disulfide bond in a reducing environment.However,the rate of curcumin release was much faster with the DDBA linker than with the DDPA linker as the side product was a 5-membered ring with a low ring strain.The linker length-induced variation of curcumin release kinetics caused a significant difference in the intracellular drug concentration and a higher cytotoxicity was witnessed in three model cell lines (HeLa,PC3,and 4T1) for the micelles with a DDBA linker compared to those containing a DDPA linker.As expected,this phenomenon was also observed in HeLa tumor-bearing nude mice in vivo.The current work highlights the significance of linker length in engineering redox-responsive on-demand delivery systems.Xuan Meng Min Gao Jian Deng Di Lu Aiping Fan Dan Ding Deling Kong Zheng Wang Yanjun Zhao 2018Nano Research2018,11,12:2
10Crystal Structure and Thermodynamic Study of Ephedrine Hydrochloride C_(10)H_(16)NOCl(s)显示文摘The crystal structure of ephedrine hydrochloride was determined by means of X-ray crystallography.The crystal system of the compound is monoclinic,and the space group is P21.Unit cell parameters are a=0.7308(6) nm,b=0.6124(5) nm,and c=1.2618(11) nm;α=90°,β=102°,and γ=90°;Z=2.Low-temperature heat capacities of the title compound were measured with an improved precision automated adiabatic calorimeter over a temperature range from 77 K to 396 K.A polynomial equation of the heat capacities as a function of temperature in the temperature region was fitted by the least-squares.Based on the fitted polynomial equation,the smoothed heat capacities and thermodynamic functions of the compound relative to the standard reference temperature 298.15 K were calculated and tabulated at the intervals of 5 K.YANG Wei-wei DI You-ying LI Jing KONG Yu-xia 2010Chemical Research in Chinese Universities2010,26,2:2

12Stability of HIV-1 subtype B and C Tat is associated with variation in the carboxyl-terminal region显示文摘The multifunctional trans-activator Tat is an essential regulatory protein for HIV-1 replication and is characterized by high sequence diversity. Numerous experimental studies have examined Tat in HIV-1 subtype B, but research on subtype C Tat is lacking, despite the high prevalence of infections caused by subtype C worldwide. We hypothesized that amino acid differences contribute to functional differences among Tat proteins. In the present study, we found that subtype B NL4-3Tat and subtype C isolate HIV1084 i Tat exhibited differences in stability by overexpressing the fusion protein Tat-Flag. In addition, 1084 i Tat can activate LTR and NF-κB more efficiently than NL4-3 Tat. In analyses of the activities of the truncated forms of Tat, we found that the carboxylterminal region of Tat regulates its stability and transactivity. 17Computational cytometer based on magnetically modulated coherent imaging and deep learning显示文摘Detecting rare cells within blood has numerous applications in disease diagnostics.Existing rare cell detection techniques are typically hindered by their high cost and low throughput.Here,we present a computational cytometer based on magnetically modulated lensless speckle imaging,which introduces oscillatory motion to the magneticbead-conjugated rare cells of interest through a periodic magnetic force and uses lensless time-resolved holographic speckle imaging to rapidly detect the target cells in three dimensions(3D).In addition to using cell-specific antibodies to magnetically label target cells,detection specificity is further enhanced through a deep-learning-based classifier that is based on a densely connected pseudo-3D convolutional neural network(P3D CNN),which automatically detects rare cells of interest based on their spatio-temporal features under a controlled magnetic force.To demonstrate the performance of this technique,we built a high-throughput,compact and cost-effective prototype for detecting MCF7 cancer cells spiked in whole blood samples.Through serial dilution experiments,we quantified the limit of detection(LoD)as 10 cells per millilitre of whole blood,which could be further improved through multiplexing parallel imaging channels within the same instrument.This compact,cost-effective and high-throughput computational cytometer can potentially be used for rare cell detection and quantification in bodily fluids for a variety of biomedical applications.Yibo Zhang Mengxing Ouyang Aniruddha Ray Tairan Liu Janay Kong Bijie Bai Donghyuk Kim Alexander Guziak Yi Luo Alborz Feizi Katherine Tsai Zhuoran Duan Xuewei Liu Danny Kim Chloe Cheung Sener Yalcin Hatice Ceylan Koydemir Omai B.Garner Dino Di Carlo Aydogan Ozcan 2019Light(Science & Applications)2019,8,1:1
