| 2 | Efficacy and safety of trastuzumab combined with chemotherapy for first-line treatment and beyond progression of HER2-overexpressing advanced breast cancer显示文摘Objective: To observe the efficacy and safety of trastuzumab combined with chemotherapy in patients with human epidermal growth factor receptor 2(HER2)-overexpressing advanced breast cancer.Methods: A total of 90 patients with HER2-overexpressing advanced breast cancer were enrolled in this study. All patients were diagnosed with ductal invasive breast cancer by pathological analysis, and were aged between 31–73 years with a median of 51 years. HER2-positivity was defined as 3(+) staining in immunochemistry or amplification of fluorescence in situ hybridization(FISH, ratio ≥2.0). Trastuzumab was administered in combination with chemotherapy as first-line treatment and beyond progression as a secondline, third-line, and above treatment in 90, 34, 14, and 6 patients, respectively. The chemotherapy regimen was given according to normal clinical practice. The response rate was evaluated every two cycles, and the primary endpoints were progression-free survival(PFS) and overall survival(OS). Survival curves were estimated by using Kaplan-Meier graphs and were compared by using log-rank test statistics. Multivariate analysis was done using Cox's proportional hazards regression model, and the level of significance was P<0.05.Results: All 90 patients received at least one dose of trastuzumab, and efficacy could be evaluated in 85 patients. The median follow-up was 50 months. In total, 72(80.00%) patients had visceral metastasis, and 43(47.78%) patients had progressed after one or more extensive chemotherapy regimens for metastatic diseases. The median PFS for first-line trastuzumab was 10 months(range, 2–59 months), and the median OS after metastasis or initially local advanced disease was 22 months(range, 2–116 months).Conclusions: Trastuzumab combined with chemotherapy was active and well-tolerated as a first-line treatment and even beyond progression in HER2-overexpressing advanced breast cancer as a second-line or third-line treatment. | 3 | Ground Surface Ruptures and Near-Fault,Large-Scale Displacements Caused by the Wenchuan Ms8.0 Earthquake Derived from Pixel Offset Tracking on Synthetic Aperture Radar Images显示文摘The 12 May 2008 Wenchuan Ms8.0 earthquake produced surface displacements along the causative fault,the Yingxiu-Beichuan Fault,which are up to several meters near the fault.Because of the large gradient,satellite synthetic aperture radar(SAR) interferometric data are strongly incoherent;the usual SAR interferometry method does not allow such displacements to be measured.In the present study,we employed another approach,the technique based on pixel offset tracking,to solve this problem.The used image data of six tracks are from the Advanced Land Observing Satellite,Phased Array type L-band Synthetic Aperture Radar(ALOS/PALSAR) dataset of Japan.The results show that the entire surface rupture belt is 238 km long,extending almost linearly in a direction of 42° north-east.It is offset left laterally by a north-west-striking fault at Xiaoyudong,and turns at Gaochuan,where the rupture belt shifts toward the south by 5 km,largely keeping the original trend.In terms of the features of the rupture traces,the rupture belt can be divided into five sections and three types.Among them,the Beichuan-Chaping and Hongkou-Yingxiu sections are relatively complex,with large widths and variable traces along the trend.The Pingtong-Nanba and Qingping-Jingtang sections appear uniform,characterized by straight traces and small widths.West of Yingxiu,the rupture traces are not clear.North of the rupture belt,surface displacements are 2.95 m on average,mostly 2-3.5 m,with 7-9 m the maximum near Beichuan.South of the rupture belt,the average displacement is 1.75 m,dominated by 1-2 m,with 3-4 m at a few sites.In the north,the displacements in the radar line of sight are of subsidence,and in the south,they are uplifted,in accordance with a right-slip motion that moves the northern wall of the fault to the east,and the southern wall to the west,respectively.Along the Guanxian-Jiangyou Fault,there is a uplift zone in the radar line of sight,which is 66 km long,1.5-6 km wide,and has vertical displacements of approximately 2 m,but no observable rupture traces. | QU Chunyan SHAN Xinjian LIU Yunhua ZHANG Guohong SONG Xiaogang ZHANG Guifang GUO Liming HAN Yufei | 2012 | Acta Geologica Sinica(English Edition)2012，86，2： | 10 |
| 4 | Capecitabine maintenance therapy for XT chemotherapy-sensitive patients with metastatic triple-negative breast cancer显示文摘Objective: To investigate the efficacy and safety of capecitabine maintenance therapy(MT) after initial capecitabine plus docetaxel(XT) chemotherapy in patients with metastatic triple-negative breast cancer(m TNBC).Methods: Fifty-five m TNBC patients treated with XT chemotherapy between May 2007 and June 2013 were retrospectively analyzed. When initial disease control was achieved by the combination chemotherapy, capecitabine was continued for 32 patients(MT), while 23 patients remained without any treatment(nonMT). We compared progression-free survival(PFS) and safety of both groups.Results: The median PFS of 55 patients was 8.1 months, overall median PFS time of 32 patients in the capecitabine MT group and 23 in the non-MT group was 10.1 vs. 6.7 months(P=0.032), respectively. When compared PFS time of maintenance treatment, single-agent capecitabine prolonged PFS by 7.1 months, for non-MT patients, the PFS without any treatment was 3.1 months, and this between-group difference was statistically significant(P=0.003). | 5 | Efficacy of platinum in advanced triple-negative breast cancer with germline BRCA mutation determined by next generation sequencing显示文摘Objective:To compare the efficacy of platinum-and non-platinum-based regimens as first-line treatment for advanced triple-negative breast cancer(TNBC)and analyze the relationship between their efficacy and BRCA gene status.Methods:Retrospectively analyze clinical data of 220 patients diagnosed pathologically with advanced TNBC and treated at the Department of Breast Oncology,Peking University Cancer Hospital from 2013 to 2018 and evaluate the efficacy of chemotherapy.A total of 114 patients had BRCA1/2 gene tested by next generation sequencing(NGS)using peripheral blood,and we analyzed the correlation between their efficacy and BRCA1/2 gene status.Results:Non-platinum-based chemotherapy(NPCT)was administered to 129 and platinum-based chemotherapy(PBCT)to 91 study patients.The clinical benefit rate(CBR)and median progression-free survival(PFS)were not statistically different between NPCT and PBCT groups.The median overall survival(OS)was 30.0 and 22.5 months for PBCT and NPCT group,respectively[P=0.090,hazard ratios(HR)=0.703].BRCA status was assessed in 114 patients,14 of whom had deleterious germline BRCA1/2(g BRCA)mutations(seven in each group).In PBCT group,the CBR was 85.7%and 35.1%for patients with and without deleterious g BRCA mutations,respectively(P=0.039).The median PFS were 14.9 and 5.3 months and median OS were 26.5 and 15.5 months for patients with and without deleterious g BRCA mutations,respectively(P=0.001,P=0.161,respectively).Patients in PBCT group had significantly greater rates of grade 3-4 anemia(5.5%vs.0%)and thrombocytopenia(8.8%vs.0%),whereas palmar-plantar erythrodysesthesia(12.4%vs.0%)and peripheral neuropathy(8.6%vs.1.1%)occurred more frequently in NPCT group.Conclusions:Platinum-based regimens are more effective in patients with deleterious g BRCA mutations,but no difference in patients without BRCA gene mutations,so non-platinum is an option in patients without BRCA gene mutations considering the toxicity and side effect.And we recommend that patients with advanced TNBC should have BRCA gene test. | Nan Wang Kun Li Wenfa Huang Weiyao Kong Xiaoran Liu Weijie Shi Feng Xie Hanfang Jiang Guohong Song Lijun Di Quanren Wang Jianjun Yu Huiping Li | 2020 | Chinese Journal of Cancer Research2020，32，2： | 7 |
| 6 | Deformation of the Haiyuan-Liupanshan fault zone inferred from the denser GPS observations显示文摘The Haiyuan-Liupanshan fault,an active tectonic feature at the Tibetan Plateau's northeastern boundary,was ruptured by two MS earthquakes(1920 and 1927)bracketing an unbroken section(the Tianzhu seismic gap).A high seismic hazard is expected along the gap.To monitor deformation characteristics and do a seismic risk assessment,we made measurements at two newly built campaign-mode Global Positioning System(GPS) stations and 13 pre-existing stations in 2013 and 2014.Adding existing data from 1999 to 2014,we derived a new velocity field.Based on the horizontal velocity,we used three block models to invert the deformation of four crustal blocks.The results suggest non-uniform deformation in the interior of the Lanzhou block,the Ordos block and the Alaxan block,but uniform deformation in the Qilian block.Fault slip rates derived from block models show a decreasing trend from west to east,(2.0-3.2 mm/a on the Haiyuan fault to 0.9-1.5 mm/a on the Liupanshan fault).The Haiyuan fault evidences sinistral striking-slip movement,while the Liupanshan fault is primarily thrusting due to transformation of the displacement between the strike-slip and crustal shortening.The locking depth of each segment along the Haiyuan fault obtained by fitting the fault parallel velocities varies drastically from west to east(21.8-7.1 km).The moment accumulation rate,calculated using the slip rate and locking depth,is positively correlated with the locking depth.Given the paucity of large seismic events during the previous millennium,the Tuolaishan segment and the Maomaoshan segment have higher likelihood of nucleation for a future event. | Yanchuan Li Chunyan Qu Xinjian Shan Xiaogang Song Guohong Zhang Weijun Gan Shaoyan Wen Zhenjie Wang | 2015 | Earthquake Science2015，28，5： | 4 |
| 7 | Combined peripheral natural killer cell and circulating tumor cell enumeration enhance prognostic efficiency in patients with metastatic triple-negative breast cancer显示文摘Objective:Triple-negative breast cancer(TNBC)is a heterogeneous disease with poor prognosis.Circulating tumor cells(CTCs)are a promising predictor for breast cancer prognoses but their reliability regarding progression-free survival(PFS)is controversial.We aim to verify their predictive value in TNBC.Methods:In present prospective cohort study,we used the Pep@MNPs method to enumerate CTCs in baseline blood samples from 75 patients with TNBC(taken at inclusion in this study)and analyzed correlations between CTC numbers and outcomes and other clinical parameters.Results:Median PFS was 6.0(range:1.0–25.0)months for the entire cohort,in whom we found no correlations between baseline CTC status and initial tumor stage(P=0.167),tumor grade(P=0.783)or histological type(P=0.084).However,among those getting first-line treatment,baseline CTC status was positively correlated with ratio of peripheral natural killer(NK)cells(P=0.032),presence of lung metastasis(P=0.034)and number of visceral metastatic site(P=0.037).Baseline CTC status was predictive for PFS in first-line TNBC(P=0.033),but not for the cohort as a whole(P=0.118).This prognostic limitation of CTC could be ameliorated by combining CTC and NK cell enumeration(P=0.049).Conclusions:Baseline CTC status was predictive of lung metastasis,peripheral NK cell ratio and PFS in TNBC patients undergoing first-line treatment.We have developed a combined CTC-NK enumeration strategy that allows us to predict PFS in TNBC without any preconditions. | Xiaoran Liu Ran Ran Bin Shao Hope S.Rugo Yanlian Yang Zhiyuan Hu Zewen Wei Fengling Wan Weiyao Kong Guohong Song Hanfang Jiang Xu Liang Ruyan Zhang Ying Yan Guobing Xu Huiping Li | 2018 | Chinese Journal of Cancer Research2018，30，3： | 4 |
| 8 | PhaseⅠdose-escalation and expansion study of PARP inhibitor,fluzoparib(SHR3162),in patients with advanced solid tumors显示文摘Objective:Fluzoparib(SHR3162)is a novel,potent poly(ADP-ribose)polymerases(PARP)1,2 inhibitor that showed anti-tumor activity in xenograft models.We conducted a phaseⅠ,first-in-human,dose-escalation and expansion(D-Esc and D-Ex)trial in patients with advanced solid cancer.Methods:This was a 3+3 phaseⅠD-Esc trial with a 3-level D-Ex at 5 hospitals in China.Eligible patients for DEsc had advanced solid tumors refractory to standard therapies,and D-Ex enrolled patients with ovarian cancer(OC).Fluzoparib was administered orally once or twice daily(bid)at 11 dose levels from 10 to 400 mg/d.Endpoints included dose-finding,safety,pharmacokinetics,and antitumor activity.Results:Seventy-nine patients were enrolled from March,2015 to January,2018[OC(47,59.5%);breast cancer(BC)(16,20.3%);colorectal cancer(8,10.1%),other tumors(8,10.1%)];48 patients were treated in the D-Esc arm and 31 in the D-Ex arm.The maximum tolerated dose(MTD)was 150 mg bid,with a half-life of 9.14 h.Grade 3/4 adverse events included anemia(7.6%)and neutropenia(5.1%).The objective response rate(ORR)was 30%(3/10)in patients with platinum-sensitive OC and 7.7%(1/13)in patients with BC.Among patients treated with fluzoparib≥120 mg/d,median progression-free survival(m PFS)was 7.2[95%confidence interval(95%CI),1.8-9.3]months in OC,9.3(95%CI,7.2-9.3)months in platinum-sensitive OC,and 3.5(range,2.0-28.0)months in BC.In patients with germline BC susceptibility gene mutation(g BRCAMut)(11/43 OC;2/16 BC),m PFS was 8.9 months for OC(range,1.0-23.2;95%CI,1.0-16.8)and 14 and 28 months for BC(those two patients both also had somatic BRCAMut).Conclusions:The MTD of fluzoparib was 150 mg bid in advanced solid malignancies.Fluzoparib demonstrated single-agent antitumor activity in BC and OC,particularly in BRCAMut and platinum-sensitive OC. | Huiping Li Rongrui Liu Bin Shao Ran Ran Guohong Song Ke Wang Yehui Shi Jihong Liu Wenjing Hu Fu Chen Xiaoran Liu Gairong Zhang Chuanhua Zhao Ru Jia Quanren Wang Hope S.Rugo Yifan Zhang Guangze Li Jianming Xu | 2020 | Chinese Journal of Cancer Research2020，32，3： | 4 |
| 9 | Multicenter phaseⅡstudy of apatinib single or combination therapy in HER2-negative breast cancer involving chest wall metastasis显示文摘Objective:Breast cancer(BC)with chest wall metastasis(CWM)usually shows rich neovascularization.This trial explored the clinical effect of apatinib on human epidermal growth factor receptor 2(HER2)-negative advanced BC involving CWM.Methods:This trial involved four centers in China and was conducted from September 2016 to March 2020.Patients received apatinib 500 mg/d[either alone or with endocrine therapy if hormone receptor-positive(HR+)]until disease progression or unacceptable toxicity.Progression-free survival(PFS)was the primary endpoint.Results:We evaluated 26 patients for efficacy.The median PFS(mPFS)and median overall survival(mOS)were4.9[range:2.0-28.5;95%confidence interval(95%CI):2.1-8.3]months and 18(range:3-55;95%CI:12.9-23.1)months,respectively.The objective response rate(ORR)was 42.3%(11/26),and the disease-control rate was76.9%(20/26).In the subgroup analysis,HR+patients compared with HR-negative patients had significantly improved mPFS of 7.0(95%CI:2.2-11.8)months vs.2.3(95%CI:1.2-3.4)months,respectively(P=0.001);and mPFS in patients without or with chest wall radiotherapy was 6.4(95%CI:1.6-19.5)months vs.3.0(95%CI:1.3-4.6)months,respectively(P=0.041).In the multivariate analysis,HR+status was the only independent predictive factor for favorable PFS(P=0.014).Conclusions:Apatinib was highly effective for BC patients with CWM,especially when combined with endocrine therapy.PFS improved significantly in patients with HR+status who did not receive chest wall radiotherapy.However,adverse events were serious and should be carefully monitored from the beginning of apatinib treatment. | Huiping Li Cuizhi Geng Hongmei Zhao Hanfang Jiang Guohong Song Jiayang Zhang Yaxin Liu Xinyu Gui Jing Wang Kun Li Zhongsheng Tong Fangyuan Zhao Junlan Yang Guoliang Chen Qianyu Liu Xu Liang | 2021 | Chinese Journal of Cancer Research2021，33，2： | 4 |
| 10 | Experimental study of a micro-scale solar organic Rankine cycle system based on compound cylindrical Fresnel lens solar concentrator显示文摘In the present study, a micro-scale solar organic Rankine cycle power generation system was developed. The system comprises of a solar collection system based on compound cylindrical Fresnel lens concentrator and an organic Rankine cycle power generation system integrated with a scroll expander. YD320 and R245 fa were used as the heat transfer fluid and the working fluid, respectively. The effects of the evaporation pressure, the degree of superheat, and the mass flow rate of the working fluid were analyzed to evaluate the solar collection efficiency, the electric power output, the thermal efficiency and exergy efficiency of the system. The results illustrate that both the increasing evaporation pressure and decreasing superheat degree have positive impacts on solar collection efficiency. The electric power increases as the evaporation pressure increases, while the thermal efficiency and the exergy efficiency decrease. However, the system overall efficiency decreases slowly due to the increase of solar collection efficiency. The electric power increases with the increment of the working fluid mass flow rate. The increasing mass flow rate has no visible impact on the thermal and exergy efficiencies of organic Rankine cycle subsystem, whereas a slightly increase of the thermal and exergy efficiencies of the integrated system. The electric power decreases with the increase of the superheat degree, whereas the thermal and the exergy efficiencies of the system increase. However, the role of CYP11A1 in goose reproductive cycle remains largely unknown.Results: In this study, full-length CYP11A1 c DNA of Zhedong goose was obtained using reverse transcription polymerase chain reaction(RT-PCR) and rapid amplification of c DNA ends(RACE). The c DNA consisted of a 96-base pair(bp) 5′untranslated region(UTR), a 179-bp 3′UTR and a 1509-bp open reading frame. The open reading frame encodes a putative 503 amino acid protein that shares high homology with CYP11A1 of other birds. The amino acid sequence possesses conserved domains of the P450 superfamily, which include the steroid-binding domain and the heme-binding region. Real-time quantitative polymerase chain reaction(q PCR) analysis revealed CYP11A1 mR NA was expressed ubiquitously in every Zhedong goose tissue analyzed, including the heart, liver, glandular stomach,lung, spleen, kidney, intestinum tenue, intestinum crassum, cerebrum, cerebellum, muscle, oviduct, pituitary,hypothalamus and ovary.. The relatively low levels of CYP11A1 m RNA were detected in pituitary, ovary and oviduct tissues at ovulation when compared with levels at oviposition. Interestingly, higher expression was observed in ovary and oviduct tissues during brooding. | 14 | The Surface Rupture Zone and Coseismic Deformation Produced by the Yutian Ms7.3 Earthquake of 21 March 2008,Xinjiang显示文摘On 21 March 2008,a Ms7.3 earthquake occurred at Quickbird,Yutian County,Xinjiang.We attempt to reveal the features of the causative fault of this shock and its coseismic deformation field.Our work is based on analysis and interpretation to high-resolution satellite images as well as differential interferometric synthetic aperture radar(D-InSAR) data from the satellite Envisat SAR,coupled with seismicity,focal mechanism solutions and active tectonics in this region.The result shows that the 40km-long,nearly NS trending surface rupture zone by this event lies on a range-front alluvial platform in Qira County.It is characterized by distinct linear traces and simple structure with 1-3-m-wide individual seams and maximum 6.5m width of a collapse fracture.Along the rupture zone many secondary fractures and fault-bounded blocks are seen,exhibiting remarkable extension.The coseismic deformation affected a large area 100×100km2.D-InSAR analysis indicates that the interferometric deformation field is dominated by extensional faulting with a small strike-slip component.Along the causative fault,the western wall fell down and the eastern wall,that is the active unit,rose up,both with westerly vergence.Because of the big deformation gradients near the seismogenic fault,no interference fringes are seen on images,and what can be determined is a vertical displacement 70cm or more between the two fault walls.According to the epicenter and differential occurrence times from the National Earthquake Information Center,China Earthquake Network Center,Harvard and USGS,it is suggested that the seismic fault ruptured from north to south. | SHAN Xinjian QU Chunyan WANG Chisheng ZHANG Guifang ZHANG Guohong SONG Xiaogang GUO Liming LIU Yunhua | 2012 | Acta Geologica Sinica(English Edition)2012，86，1： | 2 |
| 15 | Effects of different Chinese herbal prescriptions on cytokines in autoimmune prostatitis rats显示文摘OBJECTIVE:To observe and compare the effects of Chinese herbal prescriptions for promoting blood circulation,clearing heat,removing toxicity,and dispersing stagnated liver-Qi on cytokines in model rats with experimental autoimmune prostatitis(EAP) to provide an experimental basis for the use of Chinese herbal prescriptions in the treatment of chronic prostatitis.METHODS:One-hundred and ten male Wistar rats were randomly divided into 11 groups:blank group;model group;Huoxuehuayu(promoting blood circulation to remove blood stasis) high,middle,and low dose groups;Qingrejiedu(clearing heat and removing toxicity) high,middle,and low dose groups;and Shuganliqi(dispersing stagnated liver-Qi) high,middle,and low dose groups.Except the blank group,rats in all groups were injected subcutaneously in multiple points on days 0 and 30 with prostatic protein extractive solution(60 mg/ml_),and intraperitoneally injected with diphtheria-pertussis and tetanus vaccine(DPT vaccine) to establish the EAP model.Model rats were administrated high,middle,and low doses of Chinese herbal prescriptions and were sacrificed after 4 weeks.Pathological changes in the prostate gland were observed with HE staining and changes in serum interleukin-6(IL-6),interleukin-8(IL-8),and prostaglandin E_2(PGE_2) levels were detected with enzyme-linked immunosorbent assay.RESULTS:Compared with the blank group,serum PGE_2,IL-6,and IL-8 levels in the model group were significantly higher(P < 0.05).Compared with the model group,serum PGE_2,IL-6,and IL-8 levels in the Qingrejiedu low dose and middle dose groups were significantly lower(P < 0.05),with the lower dose having a more obvious effect.Serum PGE_2,IL-6,and IL-8 levels in the Huoxuehuayu high dose group(P < 0.05),IL-6 and IL-8 levels in the Huoxuehuayu middle dose group(P < 0.05),and the IL-8level in the Huoxuehuayu low dose group were significantly lower than those in the model group(P < 0.05).There were significant differences in PGE_2 and IL-6 levels among the different dose groups of Shuganliqi drugs(P < 0.05).Compared with the model group,serum PGE_(2/) IL-6,and IL-8levels in the Shuganliqi high dose group(P <0.05) and IL-8 level in the Shuganliqi low dose group were significantly lower(P < 0.05),while the Shuganliqi middle dose group did not change significantly(P > 0.05).CONCLUSION:Therefore,in TCM treatment of autoimmune prostatitis,different treatment methods should select different doses.For prescriptions that clear heat and remove toxicity,low doses should be used.For prescriptions that promote blood circulation to remove blood stasis and for prescriptions that disperse stagnated liver-Qi,high doses should be used. | Song Guohong Zhang Qiumei Pang Baozhen He Lijuan Saimaiti Julaiti Tulahong Aisikeer Gao Xuan You Lina Wuxiuer Reyihan Zhou Wentao Pang Qingyang | 2015 | Journal of Traditional Chinese Medicine2015，35，2： | 2 |
