1Expression profiles of microRNAs after focal cerebral ischemia/reperfusion injury in rats显示文摘Rat models of focal cerebral ischemia/reperfusion injury were established by occlusion of the middle cerebral artery.Microarray analysis showed that 24 hours after cerebral ischemia,there were nine up-regulated and 27 down-regulated microRNA genes in cortical tissue.Bioinformatic analysis showed that bcl-2 was the target gene of microRNA-384-5p and microRNA-494,and caspase-3 was the target gene of microRNA-129,microRNA-320 and microRNA-326.Real-time PCR and western blot analyses showed that 24 hours after cerebral ischemia,bcl-2 mRNA and protein levels in brain tissue were significantly decreased,while caspase-3 mRNA and protein levels were significantly increased.This suggests that following cerebral ischemia,differentially expressed microRNA-384-5p,microRNA-494,microRNA-320,microRNA-129 and microRNA-326 can regulate bcl-2 and caspase-3 expression in brain tissue.Fengguo Zhai Xiuping Zhang Yue Guan Xudong Yang Yang Li Gaochen Song Lixin Guan 2012Neural Regeneration Research2012,7,12:13
2Pharmacometabolomic prediction of individual differences of gastrointestinal toxicity complicating myelosuppression in rats induced by irinotecan显示文摘Pharmacometabolomics has been already successfully used in toxicity prediction for one specific adverse effect. However in clinical practice, two or more different toxicities are always accompanied with each other, which puts forward new challenges for pharmacometabolomics. Gastrointestinal toxicity and myelosuppression are two major adverse effects induced by Irinotecan(CPT-11),and often show large individual differences. In the current study, a pharmacometabolomic study was performed to screen the exclusive biomarkers in predose serums which could predict late-onset diarrhea and myelosuppression of CPT-11 simultaneously. The severity and sensitivity differences in gastrointestinal toxicity and myelosuppression were judged by delayed-onset diarrhea symptoms, histopathology examination, relative cytokines and blood cell counts. Mass spectrometry-based non-targeted and targeted metabolomics were conducted in sequence to dissect metabolite signatures in predose serums. Eventually,two groups of metabolites were screened out as predictors for individual differences in late-onset diarrhea and myelosuppression using binary logistic regression, respectively. This result was compared with existing predictors and validated by another independent external validation set. Our study indicates the prediction of toxicity could be possible upon predose metabolic profile. Pharmacometabolomics can be a potentially useful tool for complicating toxicity prediction. 3Functional metabolomics reveal the role of AHR/GPR35 mediated kynurenic acid gradient sensing in chemotherapy-induced intestinal damage显示文摘Intestinal toxicity induced by chemotherapeutics has become an important reason for the interruption of therapy and withdrawal of approved agents. In this study, we demonstrated that chemotherapeutics-induced intestinal damage were commonly characterized by the sharp upregulation of tryptophan(Trp)àkynurenine(KYN)àkynurenic acid(KA) axis metabolism. Mechanistically,chemotherapy-induced intestinal damage triggered the formation of an interleukin-6(IL-6)àindoleamine2,3-dioxygenase 1(IDO1)àaryl hydrocarbon receptor(AHR) positive feedback loop, which accelerated kynurenine pathway metabolism in gut. Besides, AHR and G protein-coupled receptor 35(GPR35) negative feedback regulates intestinal damage and inflammation to maintain intestinal integrity and homeostasis through gradually sensing kynurenic acid level in gut and macrophage, respectively. Moreover, based on virtual screening and biological verification, vardenafil and linagliptin as GPR35 and AHR agonists respectively were discovered from 2388 approved drugs. 4The influence of nanosized precipitates on Portevin-Le Chatelier bands and surface roughness in AlMgScZr alloy显示文摘The influence of different precipitate-dislocation interactions,namely dislocation shearing and bypassing mechanisms,on PLC bands and the resultant surface roughness in AlMgScZr alloy was investigated.Three-dimensional surface roughness was quantitatively measured by confocal microscopy.We find that the introduction of shearable precipitates increases the stress amplitude,decreases the PLC bands number and surface roughness.However,the stress amplitude decreases,the PLC bands number and surface roughness increase with shearable precipitates turning to nonshearable precipitates.By analyzing the precipitation strengthening mechanisms quantitatively,the influence of precipitates on PLC bands and the resultant surface roughness was explored.Furthermore,our study demonstrates that the shearable precipitates can decrease the surface roughness by decreasing the number of PLC bands,which is instructive for designing structural materials with desirable mechanical property and surface quality.Yaoxiang Duan Han Chen Zhe Chen Lei Wang Mingliang Wang Jun Liu Fengguo Zhang Haowei Wang 2021Journal of Materials Science & Technology2021,,28:3
5Predicting the grades of Astragali radix using mass spectrometrybased metabolomics and machine learning显示文摘Astragali radix(AR,the dried root of Astragalus)is a popular herbal remedy in both China and the United States.The commercially available AR is commonly classified into premium graded(PG)and ungraded(UG)ones only according to the appearance.To uncover novel sensitive and specific markers for AR grading,we took the integrated mass spectrometry-based untargeted and targeted metabolomics approaches to characterize chemical features of PG and UG samples in a discovery set(n=16 batches).A series of five differential compounds were screened out by univariate statistical analysis,including arginine,calycosin,ononin,formononetin,and astragalosideⅣ,most of which were observed to be accumulated in PG samples except for astragalosideⅣ.Then,we performed machine learning on the quantification data of five compounds and constructed a logistic regression prediction model.Finally,the external validation in an independent validation set of AR(n=20 batches)verified that the five compounds,as well as the model,had strong capability to distinguish the two grades of AR,with the prediction accuracy>90%.Our findings present a panel of meaningful candidate markers that would significantly catalyze the innovation in AR grading.Xinyue Yu Jingxue Nai Huimin Guo Xuping Yang Xiaoying Deng Xia Yuan Yunfei Hua Yuan Tian Fengguo Xu Zunjian Zhang Yin Huang 2021Journal of Pharmaceutical Analysis2021,11,5:2
6An Improved Whale Algorithm and Its Application in Truss Optimization显示文摘The current Whale Optimization Algorithm(WOA)has several drawbacks,such as slow convergence,low solution accuracy and easy to fall into the local optimal solution.To overcome these drawbacks,an improved Whale Optimization Algorithm(IWOA)is proposed in this study.IWOA can enhance the global search capability by two measures.First,the crossover and mutation operations in Differential Evolutionary algorithm(DE)are combined with the whale optimization algorithm.Second,the cloud adaptive inertia weight is introduced in the position update phase of WOA to divide the population into two subgroups,so as to balance the global search ability and local development ability.ANSYS and Matlab are used to establish the structure model.To demonstrate the application of the IWOA,truss structural optimizations on 52-bar plane truss and 25-bar space truss were performed,and the results were are compared with that obtained by other optimization algorithm.It is verified that,compared with WOA,the IWOA has higher efficiency,fast convergence speed,better solution accuracy and stability.So IWOA can be used in the optimization design of large truss structures.Fengguo Jiang Lutong Wang Lili Bai 2021Journal of Bionic Engineering2021,18,3:2
7Spatiotemporal variation in forest fire danger from 1996 to 2010 in Jilin Province,China显示文摘We evaluated the spatial and temporal patterns of forest fires in two fire seasons(March to June and September to November) from 1996 to 2010 in Jilin Province,China,using the Canadian Forest Fire Weather Index System.Fire data were obtained from the Provincial Fire Agency,and historical climate records of daily weather observations were collected from 36 weather stations in Jilin and its neighboring provinces.A linear regression model was used to analyze linear trends between climate and fire weather indices with time treated as an independent variable.Correlation analysis was used to detect correlations between fire frequency,areas burned,and fire weather indices.A thin-plate smooth spline model was used to interpolate the point data of 36 weather stations to generate a surface covering the whole province.Our analyses indicated fire frequency and areas burned were significantly correlated with fire weather indices.Overall,the Canadian Forest Fire Weather Index System appeared to be work well for determining the fire danger rating in Jilin Province.Also,our analyses indicated that in the forthcoming decades,the overall fire danger in March and April should decrease across the province,but the chance of a large fire in these months would increase.The fire danger in the fall fire season would increase in the future,and the chance of large fire would also increase.Historically,because most fires have occurred in the spring in Jilin Province,such a shift in the future fire danger between the two fire seasons would be beneficial for the province's fire management.Yanlong Shan Yonghe Wang Mike Flannigan Shuyuan Tang Pingyan Sun Fengguo Du 2017Journal of Forestry Research2017,28,5:2

11Long-Term Bending Behaviour of Prestressed Glulam Bamboo-Wood Beam Based on Creep Effect显示文摘Creep is an important characteristic of bamboo and wood materials under long-term loading.This paper aims to study the long-term bending beha-viour of prestressed glulam bamboo-wood beam(GBWB).For this,14 pre-stressed GBWBs were selected and subjected to a long-term loading test for 60 days.Then,a comparative analysis was performed for the effects of pre-tension values,the number of pre-stressed wires,and long-term load on the stress variation of the steel wire and the long-term deflection of the beam midspan.The test results showed that with the number of prestressed wires increasing,the total stress of the steel wire in the beam midspan and the ratio of the long-term deflec-tion to the total deflection decreases decreased,but when the number of steel wires exceeded 4,the total stress and long-term deflection was less infuenced;with the pre-tension value increasing,the ratio of the total stress of the steel wire in the beam midspan and the ratio of the long-temm deflection to the total deflec-tion also decreased,but when the prestress force was greater than 3.975 kN,the:total stress and long-term deflection were less affected;with the other parameters unchanged,when the value of the long-term load increased,the total stress of the steel wire decreased,and the long-temm deflection of the beam midspan increased,which shall be more significant with the long-term load greater than 30%of the standard ultimate bearing capacity.After the test,the experimental data were fitted,and the creep coefficient was given.Finally,the long-term stiffness calcula-tion fommula of the pre-stressed GBWB based on creep effect was proposed.The research findings have certain theoretical significance and engineering value.Nan Guo Huajing Xiong Mingtao Wu Hongliang Zuo Fengguo Jiang Xiaofeng Hou Dabo Xin 2020Structural Durability & Health Monitoring2020,14,3:1
