| 1 | ACBD3 is required for FAPP2 transferring glucosylceramide through maintaining the Golgi integrity显示文摘Glycosphingolipid (GSL) metabolism is involved in various physiological processes, including all major cell signaling pathways, and its dysregulation is linked to some diseases. The four-phosphate adaptor protein FAPP2-mediated glucosylceramide (GlcCer) transport for complex GSL synthesis has been studied extensively. However, the molecular machinery of FAPP2 as a GlcCer-transferring protein remains poorly defined. Here, we identify a Golgi-resident protein, acyl-coenzyme A binding domain containing 3 (ACBD3), as an interacting partner of FAPP2. We find that ACBD3 knockdown leads to dramatic Golgi fragmentation, which subsequently causes FAPP2 dispersal throughout the cytoplasm and a decreased localization at trans-Golgi network. The further quantitative Upidomic analysis indicates that ACBD3 knockdown triggers abnormal sphingolipid metabolism. Interestingly, the expression of siRNA-resistant full-length ACBD3 can rescue these defects caused by ACBD3 knockdown. | 2 | Molecular cloning and expression analysis of Crustin-like gene from Chinese shrimp Fenneropenaeus chinensis显示文摘A new member of antimicrobial protein genes of the Crustin family was cloned from haemocytes of the Chinese shrimp Fenneropenaeus chinensis by 3′and 5′ RACE. The full-length cDNA of Crustin-like gene contains a 390 bp open reading frame, encoding 130 amino acids. The deduced peptide contains a putative signal peptide of 17 amino acids and mature peptide of 113 amino acids. The molecular mass of the deduced mature peptide is 12.3 ku. It is highly cationic with a theoretical isoelectric point of 8.5. The deduced amino acids sequence of this Crustin showed high homology with those of Penaeus (Litopenaeus) setferus. Northern blotting showed that the cloned Crustin gene was mainly expressed in haemocytes, gill, intestine, and RNA in situ hybridization indicated that the Crustin gene was constitutively expressed exclusively in haemocytes of these tissues. Capillary electrophoresis RT-PCR analysis showed that Crustin was up-regulated dramatically from 12 to 48 h after a brief decrease of mRNA during first 6 h in response to microbe infection. The level of Crustin mRNA began to restore at 72 h post-challenge. | 12 | Clinical Study on the Anti-Aging Effect of a Cream Containing Ginseng Root Extract and L-Carnosine显示文摘The anti-aging effects of a cream containing L-carnosine and ginseng root extract to provide a reference for the evaluation and development of anti-aging cosmetics were tested. Sixty healthy females, aged 35-60,were grouped into test sample and placebo groups. Skin elasticity, gloss, and wrinkles of the participants were analyzed before use and after use for 8 weeks. Instrument measurement results showed that after 8 weeks,skin elasticity, gloss, and wrinkles in the members of the test sample group obviously improved relative to those of the placebo group. Clinical effect assessment results showed that subjects did not experience adverse skin reactions, and their skin gloss showed obvious improvement. Self-assessment results showed that 87%subjects are satisfied with the test sample. | 17 | Effects of long-term fast charging on a layered cathode for lithium-ion batteries显示文摘Fast charging, which aims to shorten recharge times to 10–15 min, is crucial for electric vehicles(EVs),but battery capacity usually decays rapidly if batteries are charged under such severe conditions.Revealing the failure mechanism is a prerequisite to improving the charging performance of lithium(Li)-ion batteries. Previous studies have focused less on cathode materials while also mostly focusing on their early changes. Thus, the cumulative effect of long-term fast charging on cathode materials has not been fully studied. Here, we study the changes in a layered cathode material during 1000 cycles of 6 C charging based on 1.6 Ah LiCoO_(2)/graphite pouch cells. Postmortem analysis reveals that the surface structure, charge transfer resistance and Li-ion diffusion coefficient of the cathode degenerate during repeated fast charging, causing a large increase in polarization. This polarization-induced poor utilization of the Li inventory is an important reason for the rapid capacity fading of batteries.
