| 1 | Phase 1 clinical trial demonstrated that MUC1 positive metastatic seminal vesicle cancer can be effectively eradicated by modified Anti-MUC1 chimeric antigen receptor transduced T cells显示文摘Recent progress in chimeric antigen receptor-modified T-cell(CAR-T cell) technology in cancer therapy is extremely promising, especially in the treatment of patients with B-cell acute lymphoblastic leukemia. In contrast, due to the hostile immunosuppressive microenvironment of a solid tumor, CAR T-cell accessibility and survival continue to pose a considerable challenge, which leads to their limited therapeutic efficacy. In this study, we constructed two anti-MUC1 CAR-T cell lines. One set of CAR-T cells contained SM3 single chain variable fragment(sc Fv) sequence specifically targeting the MUC1 antigen and co-expressing interleukin(IL) 12(named SM3-CAR). The other CAR-T cell line carried the SM3 sc Fv sequence modified to improve its binding to MUC1 antigen(named p SM3-CAR) but did not co-express IL-12. When those two types of CAR-T cells were injected intratumorally into two independent metastatic lesions of the same MUC1+ seminal vesicle cancer patient as part of an interventional treatment strategy, the initial results indicated no side-effects of the MUC1 targeting CAR-T cell approach, and patient serum cytokines responses were positive. | 3 | Gelation of Hole Transport Layer to Improve the Stability of Perovskite Solar Cells显示文摘To achieve high power conversion efficiency(PCE) and long-term stability of perovskite solar cells(PSCs), a hole transport layer(HTL) with persistently high conductivity, good moisture/oxygen barrier ability, and adequate passivation capability is important. To achieve enough conductivity and effective hole extraction, spiro-OMe TAD, one of the most frequently used HTL in optoelectronic devices, often needs chemical doping with a lithium compound(LiTFSI). However, the lithium salt dopant induces crystallization and has a negative impact on the performance and lifetime of the device due to its hygroscopic nature. Here, we provide an easy method for creating a gel by mixing a natural small molecule additive(thioctic acid, TA) with spiro-OMe TAD. We discover that gelation effectively improves the compactness of resultant HTL and prevents moisture and oxygen infiltration. Moreover, the gelation of HTL improves not only the conductivity of spiro-OMe TAD, but also the operational robustness of the devices in the atmospheric environment. In addition, TA passivates the perovskite defects and facilitates the charge transfer from the perovskite layer to HTL. | 4 | Inhibition of aberrant Hif1αactivation delays intervertebral disc degeneration in adult mice显示文摘The intervertebral disc(IVD) is the largest avascular tissue. Hypoxia-inducible factors(HIFs) play essential roles in regulating cellular adaptation in the IVD under physiological conditions. Disc degeneration disease(DDD) is one of the leading causes of disability, and current therapies are ineffective. This study sought to explore the role of HIFs in DDD pathogenesis in mice. The findings of this study showed that among HIF family members, Hif1α was significantly upregulated in cartilaginous endplate(EP) and annulus fibrosus(AF) tissues from human DDD patients and two mouse models of DDD compared with controls. Conditional deletion of the E3 ubiquitin ligase Vhl in EP and AF tissues of adult mice resulted in upregulated Hif1α expression and age-dependent IVD degeneration. Aberrant Hif1α activation enhanced glycolytic metabolism and suppressed mitochondrial function. On the other hand, genetic ablation of the Hif1α gene delayed DDD pathogenesis in Vhl-deficient mice. Administration of 2-methoxyestradiol(2ME2), a selective Hif1α inhibitor, attenuated experimental IVD degeneration in mice. | 5 | Control method of unfavorable speed interval for high-speed trains显示文摘In tests on dynamic performance of high-speed trains, it is found that there is an unfavorable speed interval for some vehicles, which would be harmful to the daily operation of the vehicle. By analyzing the relationship of vibration frequencies of the vehicle and its running speed, this paper finds that the unfavorable speed interval is caused by the vibration superposition of the natural frequency of the vehicle system with the frequency of external excitation. Taking some electric multiple units (EMUs) as examples, we proposed an approach to obtaining the unfavorable speed interval of vehicles. By analyzing the relation between vibration frequencies and the running speed of the vehicle, the natural frequencies of the vehicle system and the external excitations are distinguished. | 6 | Multi-Branch High-Dimensional Guided Transformer-Based 3D Human Posture Estimation显示文摘The human pose paradigm is estimated using a transformer-based multi-branch multidimensional directed the three-dimensional(3D)method that takes into account self-occlusion,badly posedness,and a lack of depth data in the per-frame 3D posture estimation from two-dimensional(2D)mapping to 3D mapping.Firstly,by examining the relationship between the movements of different bones in the human body,four virtual skeletons are proposed to enhance the cyclic constraints of limb joints.Then,multiple parameters describing the skeleton are fused and projected into a high-dimensional space.Utilizing a multi-branch network,motion features between bones and overall motion features are extracted to mitigate the drift error in the estimation results.Furthermore,the estimated relative depth is projected into 3D space,and the error is calculated against real 3D data,forming a loss function along with the relative depth error.This article adopts the average joint pixel error as the primary performance metric.Compared to the benchmark approach,the estimation findings indicate an increase in average precision of 1.8 mm within the Human3.6M sample. | Xianhua Li Haohao Yu Shuoyu Tian Fengtao Lin Usama Masood | 2024 | Computers, Materials & Continua2024,78,3: | 0 |
| 7 | Facile fabrication of superior antibacterial cotton fabric based on ZnO nanoparticles/quaternary ammonium salts hybrid composites and mechanism study显示文摘In the research for the safe and efficiently antibacterial cotton fabrics to minimize risk for human health,an organic–inorganic hybrid material of ZnO nanoparticles(NPs)and quaternary ammonium salt(QAS)was employed to modify cotton fabrics by a dipping–padding–drying method.The synergistic effects of ZnO NPs and QAS on the structure and antibacterial properties of cotton fabrics were studied in detail.Results displayed that the QAS and ZnO NPs were immobilized firmly in cotton fabric by the formation of chemical covalent bonds and silica gel structure.ZnO/QAS/cotton had a good inhibitory effect on the growth of E.coli and S.aureus,with superior antibacterial efficiency of>99.99%.ZnO/QAS/cotton preserved good mechanical property,water absorbability,and limpness.We also provided a detailed analysis of antibacterial mechanism for the hybrid materials.The contact mechanism and the Zn2+release were considered as the main mechanisms for the ZnO/QAS/cotton,while the reactive oxygen species(ROS)generation only had a little contribution to the antibacterial activity.In short,the excellent integrated properties endowed the hybrid cotton fabrics as potential application in many fields,like healthcare,food packaging. | Yechen Hu Lin Zhang Yafeng Huang Xiufang Chen Fengtao Chen Wangyang Lu | 2023 | Frontiers of Materials Science2023,17,2: | 0 |
