1Human carboxylesterases:a comprehensive review显示文摘Mammalian carboxylesterases(CEs) are key enzymes from the serine hydrolase superfamily.In the human body, two predominant carboxylesterases(CES1 and CES2) have been identified and extensively studied over the past decade. These two enzymes play crucial roles in the metabolism of a wide variety of endogenous esters, ester-containing drugs and environmental toxicants. The key roles of CES in both human health and xenobiotic metabolism arouse great interest in the discovery of potent CES modulators to regulate endobiotic metabolism or to improve the efficacy of ester drugs. This review covers the structural and catalytic features of CES, tissue distributions, biological functions, genetic polymorphisms, substrate specificities and inhibitor properties of CES1 and CES2, as well as the significance and recent progress on the discovery of CES modulators. The information presented here will help pharmacologists explore the relevance of CES to human diseases or to assign the contribution of certain CES in xenobiotic metabolism. 2Recent progress and challenges in screening and characterization of UGT1A1 inhibitors显示文摘Uridine-diphosphate glucuronosyltransferase 1 A1(UGT1 A1) is an important conjugative enzyme in mammals that is responsible for the conjugation and detoxification of both endogenous and xenobiotic compounds. Strong inhibition of UGT1 A1 may trigger adverse drug/herb–drug interactions, or result in metabolic disorders of endobiotic metabolism. Therefore, both the US Food and Drug Administration(FDA)and the European Medicines Agency(EMA) have recommended assaying the inhibitory potential of drugs under development on the human UGT1 A1 prior to approval. This review focuses on the significance,progress and challenges in discovery and characterization of UGT1 A1 inhibitors. Recent advances in the development of UGT1 A1 probes and their application for screening UGT1 A1 inhibitors are summarized and discussed in this review for the first time. Furthermore, a long list of UGT1 A1 inhibitors, including information on their inhibition potency, inhibition mode, and affinity, has been prepared and analyzed. Challenges and future directions in this field are highlighted in the final section. 3The ex vivo and in vivo biological performances of graphene oxide and the impact of surfactant on graphene oxide's biocompatibility显示文摘Graphene oxide(GO)displays promising properties for biomedical applications including drug delivery and cancer therapeutics.However,GO exposure also raises safety concerns such as potential side effects on health.Here,the biological effects of GO suspended in phosphate buffered saline(PBS)with or without 1% nonionic surfactant Tween 80 were investigated.Based on the ex vivo experiments,Tween 80 significantly affected the interaction between GO and peripheral blood from mice.GO suspension in PBS tended to provoke the aggregation of diluted blood cells,which could be prevented by the addition of Tween 80.After intravenous administration,GO suspension with or without 1% Tween 80 was quickly eliminated by the mononuclear phagocyte system.Nevertheless,GO suspension without Tween 80 showed greater accumulation in lungs than that containing 1% Tween 80.In contrast,less GO was found in livers for GO suspension compared to Tween 80 assisted GO suspension.Organs including hearts,livers,lungs,spleens,kidneys,brains,and testes did not reveal histological alterations.The indexes of peripheral blood showed no change upon GO exposure.Our results together demonstrated that Tween 80 could greatly alter GO's biological performance and determine the pattern of its biodistribution in mice.Guangbo Qu Xiaoyan Wang Qian Liu Rui Liu Nuoya Yin Juan Ma Liqun Chen Jiuyang He Sijin Liu Guibin Jiang 2013Journal of Environmental Sciences2013,25,5:7
4B7-H3:Another Molecule Marker for Mo-DCs?显示文摘Using a newly generated monoclonal antibody (2E6) against human B7-H3, we explored the expression of the molecule on dendritic cells derived from monocytes (Mo-DCs). Its expression was examined by means of immunostaining and flow cytometric (FCM) analysis. The results showed that B7-H3 was expressed in the course of Mo-DC maturation induced with interleukin 4 (IL-4) and granulocyte/macrophage colony-stimulating factor (GM-CSF). The expression could be detected at all the stages of Mo-DC differentiation, and remained at a quite stable level. Interestingly, B7-H3 was not expressed by T cells and B cells, even these cells were activated respectively by PHA or PWM. A weak expression could be detected on resting monocytes. These data showed that constitutive expression of B7-H3 at a high level was found on imDCs and mDCs derived from monocytes. 5Comparison of the inhibition potentials of icotinib and erlotinib against human UDP-glucuronosyltransferase 1A1显示文摘UDP-glucuronosyltransferase 1A1(UGT1A1) plays a key role in detoxification of many potentially harmful compounds and drugs. UGT1A1 inhibition may bring risks of drug–drug interactions(DDIs), hyperbilirubinemia and drug-induced liver injury. This study aimed to investigate and compare the inhibitory effects of icotinib and erlotinib against UGT1A1, as well as to evaluate their potential DDI risks via UGT1A1 inhibition. The results demonstrated that both icotinib and erlotinib are UGT1A1 inhibitors, but the inhibitory effect of icotinib on UGT1A1 is weaker than that of erlotinib. The IC_(50) values of icotinib and erlotinib against UGT1A1-mediated NCHN-O-glucuronidation in human liver microsomes(HLMs) were 5.15 and 0.68 μmol/L, respectively. Inhibition kinetic analyses demonstrated that both icotinib and erlotinib were non-competitive inhibitors against UGT1A1-mediated glucuronidation of NCHN in HLMs, with the Kivalues of 8.55 and 1.23 μmol/L, respectively. Furthermore, their potential DDI risks via UGT1A1 inhibition were quantitatively predicted by the ratio of the areas under the concentration–time curve(AUC) of NCHN. 6Multimodal hyperspectral remote sensing:an overview and perspective显示文摘Since the advent of hyperspectral remote sensing in the 1980 s,it has made important achievements in aerospace and aviation field and been applied in many fields.Conventional hyperspectral imaging spectrometer extends the number of spectral bands to dozens or hundreds,and provides spatial distribution of the reflected solar radiation from the scene of observation at the same time.Nowadays,with the fast development of new technology in the fields of information and photoelectricity sensing,and the popularity of unmanned aerial vehicle,hyperspectral remote sensing imaging presents the new trends of multimodality and acquires integration information while keeping high or very-high spectral resolution,especially,high temporal even real time sensing and stereo sensing.Therefore,three important modes of hyperspectral imaging come into existence:(1)multitemporal hyperspectral imaging,which refers to the observation of same region at different dates;(2)hyperspectral video imaging,which captures full frame spectral images in real-time;(3)hyperspectral stereo imaging,which obtains the full dimension information(including 2D image,elevation,and spectra)of observed scene.Along this perspective,firstly,the current researches on hyperspectral remote sensing and image processing are briefly reviewed,and then,comprehensive descriptions of the aforementioned three main hyperspectral imaging modes are carried out from the following four aspects:fundamental principle of new mode of hyperspectral imaging,corresponding scientific data acquisition,data processing and application,and potential challenges in data representation,feature learning and interpretation.Through the analysis of development trend of hyperspectral imaging and current research situation,we hope to provide a direction for future research on multimodal hyperspectral remote sensing.Yanfeng GU Tianzhu LIU Guoming GAO Guangbo REN Yi MA Jocelyn CHANUSSOT Xiuping JIA 2021Science China(Information Sciences)2021,64,2:6
7Practical Structural Design Approach of Multiconfiguration Planar Single‑Loop Metamorphic Mechanism with a Single Actuator显示文摘As a type of multiconfiguration mechanism that can operate in an under-actuated state,metamorphic mechanisms were proposed more than two decades ago and attracted significant interest.Studies on structural synthesis of metamorphic mechanisms tend to focus more on metamorphic techniques and the structural synthesis of source mechanisms for metamorphic mechanisms.By designing different constraint architectures of metamorphic joints,multistructures can be obtained from the same source metamorphic mechanism.To determine the constraint architectures of metamorphic joints and their different assembly combinations,a kinematic status matrix and a corresponding constraint status matrix are constructed based on the metamorphic cyclogram of a source mechanism.According to the equivalent resistance gradient model and the constraint status matrix,an equivalent resistance matrix for the metamorphic joints is proposed.A structural synthesis matrix of the metamorphic mechanism is then obtained from the equivalent resistance matrix by deducing the constraint form vectors of the metamorphic joints.Furthermore,a kinematic diagram synthesis of the source metamorphic mechanism of a planar single-loop metamorphic mechanism is proposed,which is based on only the 14 one-or zero-degrees-of-freedom linkage groups.The entire structural design method of a metamorphic mechanism is based on the structural synthesis matrix and is presented as a systematic process.Finally,the proposed structural design approach is illustrated by two examples to verify its feasibility and practicality.This study provides an effective method for designing a practical multi-mobility and multiconfiguration planar single-loop metamorphic mechanism with a single actuator.Qiang Yang Guangbo Hao Shujun Li Hongguang Wang Haiyang Li 2020Chinese Journal of Mechanical Engineering2020,33,5:5
8Experimental investigation on flow asymmetry in solid entrance region of a square circulating fluidized bed显示文摘To study the influence of back feeding particles on gas-solid flow in the riser, this paper investigated the flow asymmetry in the solid entrance region of a fluidized bed by particle concentration/velocity measurements in a cold square circulating fluidized beds (CFB). The pressure drop distribution along the riser and the saturation carrying capacity of gas for Geldart-B type particles were first analyzed. Under the condition of u0=4 m/s and Gs=21 kg/(m2 s), the back feeding particles were found to penetrate the lean gas-solid flow near the entrance (rear) wall before reaching the opposite (front) wall, thus leading to a relatively denser region near the front wall in the bottom bed. Higher solid circulation rate (u0=4 m/s, Gs=33 kg/(m2 s)) resulted in a higher particle concentration in the riser. However the back feeding particles with higher momentum increased the asymmetry of the particle concentration/velocity profile in the solid entrance region. Lower air velocity (u0=3.2 m/s) and Gs=21 kg/(m2 s), beyond the saturation carrying capacity of gas, induced an S-shaped axial solid distribution with a denser bottom zone. This limited the penetration of the back feeding particles and forced the fluidizing air to flow in the central region, thus leading to a higher solid holdup near the rear wall. 9Vapor deposition of aluminium oxide into N-rich mesoporous carbon framework as a reversible sulfur host for lithium-sulfur battery cathode显示文摘Restraining the shuttle effects of lithium polysulfides is the key to improve the cycling reversibility and stability of lithium-sulfur(Li-S)batteries for which design of robust sulfur hosts has been regarded as the most effective strategy.In this work,we report a new type of hybrid sulfur host which is composed of Al_(2)O_(3) homogenously decorated in nitrogen-rich mesoporous carbon framework(NMC-Al_(2)O_(3)).The NMC-Al_(2)O_(3) hybrid host features a poly-dispersed spherical morphology and a mesoporous configuration with high surface area and large pore volume that can accommodate a high sulfur content up to 73.5 wt.%.As a result,the fabricated NMC-Al_(2)O_(3)-S cathode exhibits all-round improvements in electrochemical properties in term of capacities(1,212 mAh·g^(-1)at 0.2 C;755 mAh·g^(-1)at 2 C),cycling charge-discharge reversibility(sustainably 100%efficiencies)and stability(1,000 cycles with only 0.023%capacity decay per cycle at 0.5 C).By contrast,the Al_(2)O_(3)-free NMC-S cathode shows both decreased capacities and rapidly descending Coulombic efficiencies during cycling.Density functional theory(DFH')calculations further reveal that the implanted Al_(2)O_(3) can greatly enhance the chemical adsorption and catalytic conversion for various lithium polysulfides and thereby effectively prevent the polysulfide shuttling and significantly improve the utilizability,reversibility and stability of sulfur cathode.Fei Sun Zhibin Qu Hua Wang Xiaoyan Liu Tong Pei Rui Han Jihui Gao Guangbo Zhao Yunfeng Lu 2021Nano Research2021,14,1:3

11Detection of tris-(2, 3-dibromopropyl) isocyanurate as a neuronal toxicant in environmental samples using neuronal toxicity-directed analysis显示文摘Neuronal toxic pollutants in environment possess hazards to human health. It is essential to determine the causative neuronal toxicants in environmental samples. In the present study, viability of primary cultured cerebellar granule neurons (CGNs), combined with sample extraction, chemical fractionation and identification, was applied for screening acid-resistant neuronal toxic substances in environmental samples. River sediments and agricultural soils along the river near a brominated flame retardant (BFR) manufacturing plant in South China were collected to screen the key neuronal toxicants. The results indicated that the manufacturing plant was a source of neuronal toxicity risks. In the sediment and soil near the plant, one of the causative toxicants was identified as tris-(2,3-dibromopropyl) isocyanurate (TBC) using HPLC-MS/MS. 12Arenobufagin is a novel isoform-specific probe for sensing human sulfotransferase 2A1显示文摘Human cytosolic sulfotransferase 2 A1(SULT2A1) is an important phase II metabolic enzyme. The detection of SULT2A1 is helpful for the functional characterization of SULT2A1 and diagnosis of its related diseases. However, due to the overlapping substrate specificity among members of the sulfotransferase family, it is difficult to develop a probe substrate for selective detection of SULT2A1. In the present study, through characterization of the sulfation of series of bufadienolides, arenobufagin(AB) was proved as a potential probe substrate for SULT2A1 with high sensitivity and specificity. Subsequently, the sulfation of AB was characterized by experimental and molecular docking studies. The sulfate-conjugated metabolite was identified as AB-3-sulfate.The sulfation of AB displayed a high selectivity for SULT2A1 which was confirmed by in vitro reaction phenotyping assays. The sulfation of AB by human liver cytosols and recombinant SULT2A1 both obeyed Michaelis-Menten kinetics, with similar kinetic parameters. Molecular docking was performed to understand the interaction between AB and SULT2A1, in which the lack of interaction with Met-137 and Tyr-238 of SULT2A1 made it possible to eliminate substrate inhibition of AB sulfation. 16Synthesis and Biological Evaluation of Hydroxylcoumarin Derivatives as Antioxidant Agents显示文摘In this study, esculetin(1) was chosen as a lead compound and some structural modifications weredesigned to explore the antioxidant activities of esculetin derivatives. Meanwhile, a convenient method for selectivemethylation of catechol cournarins with different bases was developed. Furthermore, a few 5,7-dihydroxylcoumarinswere synthesized and 7-hydroxylcoumarins were employed in order to explore the potential structure-antioxidantactivity relationships. The antioxidant activities of these compounds were evaluated and compared with standardantioxidant Trolox by the 22'-diphenyl-l-picrylhydrazyl(DPPH) assay, 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulfonate)cation(ABTS+) assay and ferric reducing antioxidant power(FRAP) assay. The results show that the catechol group isthe key pharmacophore. 18Experimental research on oil fi lm thickness and its microwave scattering during emulsifi cation显示文摘Synthetic Aperture Radar(SAR) plays a major role in identifying oil spills on the sea surface.However,obtaining information of oil spill thickness(volume) is still a challenge.Emulsification is an important process affecting the thickness and normalized radar cross section(NRCS) of oil film.Experiments of crude oil emulsification with C-band fully-polarized scatterometer were conducted combining airborne hyperspectral imaging spectrometer and 3 D laser scanner observation data,to provide experimental parameters and method to support accurate remote sensing monitoring on marine oil spill.It is further proved that through quantitative homogeneous emulsified oil spill experiments,to a certain extent,the NRCS of oil film increased during the emulsification process of crude oil.The backscattering mechanism of crude oil emulsification was explored using a semi-empirical model(SEM);the change of oil film NRCS was modulated by its dielectric constant and surface roughness,in which the dielectric constant showed a dominant effect.The relationship between thickness and NRCS of oil film was studied under two experimental conditions.The differences of NRCS between oil film and adjacent seawater(Δσ~0) and the damping ratio(DR) were found to have a linear relationship with oil thickness,which were best in the vertical polarization mode(VV) at 45° incident angle during the quantitative crude oil homogeneous emulsification process.In the natural emulsification process of continuous oil spill in which oil film was mixed with both crude oil and emulsified oil,an empirical equation of oil film thickness is preliminarily established.The Δσ~0,DR,and the empirical equation of oil film thickness were applied to the marine continuous oil spill incident on a 19-3 oil platform with spaceborne SAR image and successfully explained the distribution of the relative thickness of the oil film.Jie GUO Chenqi XU Genwang LIU Xi ZHANG Junmin MENG Guangbo REN 2022Journal of Oceanology and Limnology2022,40,4:2
19Sensing carboxylesterase 1 in living systems by a practical and isoformspecific fluorescent probe显示文摘Carboxylesterase 1(CES1), one of the most abundant serine hydrolases in mammals, has drawn much attentions in recent years, owing to this enzyme involves in many physiological processes via hydrolysis of both endogenous esters and xenobiotic esters. Herein, to real-time monitor the activities of CES1 in various biological systems, a practical and iso form-specific fluorescent probe was developed on the basis of the substrate preference of CES1, as well as the structural and optical properties of BODIPY dyes. After screening of a panel of BODIPY ester derivatives, probe 1 displayed the best combination of specificity,sensitivity, enzymatic kinetics and applicability for monitoring CES1 activities in real samples. This probe was successfully used to detect CESl activities in several biological systems including tissue preparations,living cells, tissue slices and zebrafish. Furthermore, the biomedical applications of probe 1 for screening of CES1 inhibitors were also demonstrated using tissue preparations or living cells as enzyme sources.
