4A 3-year clinical trial of lamivudine in treatment of patients with chronic hepatitis B显示文摘BACKGROUND: Lamivudine was approved for the treat- ment of chronic hepatitis B in China in 1999; however the long-term result has not yet been reported in detail. This clinical trial was to evaluate the long-term efficacy and safe- ty of 3-year lamivudine treatment for chronic hepatitis B and the impact of emergence of YMDD mutation of hepa- titis B virus (HBV). METHODS: This multi-center, randomized, double-blind, placebo controlled trial began from 1996 to 1999. A total of 429 patients with serum HBsAg, HBeAg and HBV DNA positive were randomized to receive either lamivudine 100 mg daily (322 patients) or placebo (107) for the first 12 weeks. All patients were given subsequently open labelled lamivudine 100 mg/d for a total of 156 weeks. RESULTS: After 12-week lamivudine therapy, the levels of serum HBV DNA decreased rapidly. The negativity of HBV DNA (<1.6 pg/ml) at week 12 was 92.2% in the lamivudine group, whereas it was only 14.1% in the place- bo group (P<0.01). After 1-year lamivudine treatment, 72.7% of the patients showed undetectable serum HBV DNA (<1.6 pg/ml). At the end of 3 years, serum HBV DNA continued to be substantially suppressed with a me- dian level below a detectable level in patients with non-YM- DD variant HBV, which was increased to 86 mEq/ml (bD- NA method, equivalent hybridization method 10 pg/ml) in patients with YMDD mutation. At the end of 1, 2 and 3 years, the rates of HBeAg loss were 9.5%, 16.8% and 20.0% respectively and the rates of HBeAg/anti-HBe sero- conversion were 8.3%, 11.5% and 17.3%. The rates of HBeAg loss and seroconversion were correlated with the baseline level of ALT. In patients with a baseline level of alanine transaminase (ALT)>2 × upper limit of normal (ULN) and ALT >5×ULN, the rates of HBeAg loss were 42.2% and 66.7%, and the rates of seroconversion were34.4% and 61.1% respectively (P<0.01) at the end of year 3. The levels of ALT at year 3 remained normal in 58.8% of patients whose baseline level of ALT was elevated, and in 79.1% of patients whose level of ALT was normal before treatment. YMDD mutations occurred in 12.1%, 49.7% and 70.5% of patients respectively at year 1, 2 and 3. In pa- tients with YMDD mutation, the levels of HBV DNA were increased slightly with mild to moderate elevation of ALT level. HBeAg loss and seroconversion were 20.0% and 15.1% in patients with YMDD mutation at the end of year 3, which were lower than those in non-variant patients (P<0.01). Adverse drug reactions or events varied gene- rally from mild to moderate. In 2 patients serious adverse events (fatigue and abdominal distension) were related to medication. ALT flares (ALT>5×ULN) occurred in 17 patients: 10 were YMDD mutants and 7 were non-mutants; all of them were relieved. No death occurred in the period of 3 years. 5Contribution of elF-4E inhibition to the expression and activity of heparanase in human colon adenocarcinoma cell line:LS-174T显示文摘AIM: Heparanase degrades heparan sulfate proteoglycans (HSPGs) and is a critical mediator of tumor metastasis and angiogenesis. Recently, it has been cloned as a single gene family and found to be a potential target for antimetastasis drugs. However, the molecular basis for the regulation of heparanase expression is still not quite clear. The aim of this study was to determine whether the expression of eukaryotic initiation factor 4E (eIF-4E) correlated with the heparanase level in tumor cells and to explore the correlation between heparanase expression and metastatic potential of LS- 174T cells.METHODS: A 20-met antisense s-oligodeoxynucleotide (asODN) targeted against the translation start site of eIF-4E mRNA was introduced into LS-174T cells by lipid-mediated DNA-transfection. eIF-4E protein and mRNA levels were detected by Western blot analysis and RT-PCR, respectively.Heparanase activity was defined as the ability to degrade high molecular weight (40-100 kDa) radiolabeled HS (heparan sulfate) substrate into low molecular weight (5-15 kDa) HS fragments that could be differentiated by gel filtration chromatography. The invasive potential of tumor cell in vitro was observed by using a Matrigel invasion assay system.RESULTS: The 20-mer asODN against eIF-4E specifically and significantly inhibited eIF-4E expression at both transcriptional and translational levels. As a result, the expression and activity of heparanase were effectively retarded and the decreased activity of heparanase resulted in the decreased invasive potential of LS-174T.CONCLUSION: eIF-4E is involved in the regulation of heparanase production in colon adenocarcinoma cell line LS-174T, and its critical function makes it a particularly interesting target for heparanase regulation. 7Putative epidermal stem cell convert into corneal epithelium-like cell under corneal tissue in vitro显示文摘Rhesus putative epidermal stem cells are being investigated for their potential use in regenerative corneal epithelium-like cells, which may provide a practical source of autologous seed cells for the construction of bioengineered corneas. The goal of this study was to investigate the potential of epi-dermal stem cells for trans-differentiation into corneal epithelium-like cells. Rhesus putative epidermal stem cells were isolated by type IV collagen attachment method. Flow cytometry analysis, immuno-histology and RT-PCR were conducted to identify the expression of specific markers (β1, α6 integrin, K15, K1/K10, K3/K12 and CD71) on the isolated rapid attaching cells. The isolated cells were cocultured with human corneal limbal stroma and corneal epithelial cells. After coculture, the expression of the same specific markers was evaluated in order to identify expression difference caused by the coculture conditions. K3/K12 expression was analyzed in coculture cells on day 2, 4, 6, 8 and 10. Putative epi-dermal stem cells in conditioned culture media were used as control. Putative epidermal stem cells were predominant in rapid attaching cells by type IV collagen attachment isolation. Before being co-cultured, the rhesus putative epidermal stem cells expressed K15, α6 and β1 integrin, but no CD71, K1/K10 and K3/K12. After coculture, these cells expressed K3/K12 (a marker of corneal epithelial cells), K15 and β 1 integrin, but no K1/K10. Cells being not coculture converted into terminally differentiated cells expressing K1/K10. 10Stable isotopic analysis on sus bones from the Wanfabozi site, Tonghua, Jilin显示文摘In order to study the sus diets and explore the possibility to distinguish domestic pig from wild boar through dietary analysis, stable carbon and nitrogen isotopes of 28 pig bones from archaeological site of Wanfabozi in Tonghua City, Jilin Province were analyzed. The δ 13C and δ 15N values of uncontaminated bones show that the overall pigs in the site were generally herbivores and ate mainly C3 plants. Significant difference of δ15N values was observed between wild boar and domestic pigs, which may result from the higher consumption of animal protein in domestic pigs other than from that in wild boar, possibly from human leftover or waste,. 11Changes of Coastal Wetland Ecosystems in the Yellow River Delta and Protection Countermeasures to Them显示文摘Coastal wetlands in the Yellow River Delta are typical new wetland ecosystems in warm temperate zone. In recent years, influenced by natural and human factors, these coastal wetlands in the Yellow River Delta have undergone changes of landscape fragmentation, vegetation degradation, pollution, species reduction, and harmful exotic species invasion. These changes have influenced sustainable and healthy development of marine economy of the Yellow River Delta. 13The relationship between point mutation and abnormal expression of c-fms oncogene in hepatocellular carcinoma显示文摘BACKGROUND: Recent research found abnormal expres- sion of the c-fms oncogene, which encodes the macro- phage colony-stimulating factor receptor (CSF-1R), in several human carcinomas including hepatocellular carcino- ma (HCC). But the relationship between the point muta- tion and abnormal expressing of c-fms oncogene in HCC was not clear. This study is to investigate the relationship between point mutation and abnormal expression of c-fms oncogene in hepatocellular carcinoma (HCC) and to clari- fy the mechanism of HCC. METHODS: The expression of c-fms oncogene at different levels of cell, protein and transcription was observed using immune histological ABC, Western blot and Northern blot. PCR-single strand conformation polymorphism and gene sequencing were used to detect the mutation of c-fms in HCC tissues and their surrounding tissues of 30 patients. RESULTS: The expression of c-fms was significantly higher in HCC tissues than in their surrounding tissues (P <0.01). Point mutation of Leu (TTG)—>Ser (TCG) at codon 301 of c-fms amino acids was observed in 21.4% (3/14) HCC tissues. No mutation of c-fms oncogene was detected in the surrounding cancerous tissues. 15THRUST NAPPE STRUTRUE IN THE JINGGANG MOUNTAIN: CHARACTERISTICS AND SIGNIFICANCE FOR PROSPECTION OF ORE DEPOSITS显示文摘Nappe structure, as was first discovered by the authors during the regional geological survey at the scale of 1:50,000 in The Jinggang Mountain, is mainly comprised of a series of NNE-NE-striking thrust fault zones and thrust sheets among them. Sinian, Cambrian, Ordovician, Devonian, Carboniferous,Triassic, Jurassic and Cretaceous strata are involved in the thrust nappe system. The nappe structure is of the type of duplex structures formed as a result of the earlier stage migration from SE to NW and late stage migration from E to W of sedimentary cover or basement strata. Formation of the nappe structure in the studied area involves two main epochs: Early Yanshanian and Late Yanshanian to Early Himalayan.
