| 4 | m6A RNA methylation-mediated HNF3γreduction renders hepatocellular carcinoma dedifferentiation and sorafenib resistance显示文摘Hepatocyte nuclear factor 3γ(HNF3γ)is a hepatocyte nuclear factor,but its role and clinical significance in hepatocellular carcinoma(HCC)remain unclear.Herein,we report that HNF3γexpression is downregulated in patient HCC and inversely correlated with HCC malignancy and patient survival.Moreover,our data suggested that the HNF3γreduction in HCC could be mediated by METTL14-dependent m6A methylation of HNF3γmRNA.HNF3γexpression was increased during hepatic differentiation and decreased in dedifferentiated HCC cells.Interestingly,HNF3γdelivery promoted differentiation of not only HCC cells but also liver CSCs,which led to suppression of HCC growth.Mechanistic analysis suggested an HNF3γ-centered regulatory network that includes essential liver differentiation-associated transcription factors and functional molecules,which could synergistically facilitate HCC cell differentiation.More importantly,enforced HNF3γexpression sensitized HCC cells to sorafenib-induced growth inhibition and cell apoptosis through transactivation of OATP1B1 and OATP1B3 expression,which are major membrane transporters for sorafenib uptake.Clinical investigation showed that patient-derived HCC xenografts with high HNF3γexpression exhibited a sorafenib response and patients with high HCC HNF3γlevels benefited from sorafenib therapy.Together,these results suggest that HNF3γplays an essential role in HCC differentiation and may serve as a therapeutic target and predictor of sorafenib benefit in patients. | Tengfei Zhou Shichao Li Daimin Xiang Junyu Liu Wen Sun Xiuliang Cui Beifang Ning Xiao Li Zhuo Cheng Weiqi Jiang Cheng Zhang Xijun Liang Liang Li Xin Cheng Liu Hui Hongyang Wang Jin Ding | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 4 |
| 5 | Conversion of hepatoma cells to hepatocyte-like cells by defined hepatocyte nuclear factors显示文摘Normal cells become cancer cells after a malignant transformation,but whether cancer cells can be reversed to normal status remains elusive.Here,we report that the combination of hepatocyte nuclear factor IA (HNFIA),HNF4A and forkhead box protein A3 (FOXA3)synergistically reprograms hepatocellular carcinoma (HCC)cells to hepatocyte-like cells (reprogrammed hepatocytes, rHeps).Our results show that rHeps lose the malignant phenotypes of cancer cells and retrieve hepatocyte-specific characteristics including hepatocyte-like morphology;global expression pattern of genes and specific biomarkers of hepatocytes;and the unique hepatic functions of albumin (ALB)secretion,glycogen synthesis,low-density lipoprotein (LDL)uptake,urea production, cytochrome P450 enzymes induction and drug metabolism.Intratumoral injection of these three factors efficiently shrank patientderived tumor xenografts and reprogrammed HCC cells in vivo.Most importantly,transplantation of rHeps in the liver of fumarylacetoacetate hydrolase-deficient (Fah^-/-)mice led to the reconstruction of hepatic lobules and the restoration of hepatic function.Mechanistically,exogenous expression of HNF1A,HNF4A and FOXA3 in HCC cells initiated the endogenous expression of numerous hepatocyte nuclear factors,which promoted the conversion of HCC cells to hepatocyte-like cells.Collectively,our results indicate the successful conversion of hepatoma cells to hepatocyte-like cells,not only extending our current knowledge of cell reprogramming but also providing a route towards a novel therapeutic strategy for cancer. | Zhuo Cheng Zhiying He Yongchao Cai Cheng Zhang Gongbo Fu Hengyu Li Wen Sun Changcheng Liu Xiuliang Cui Beifang Ning Daimin Xiang Tengfei Zhou Xiaofeng Li Weifen Xie Hongyang Wang Jin Ding | 2019 | Cell Research2019,29,2: | 3 |

| 7 | Effects of Acupuncture on Endometrium and Pregnancy Outcomes in Patients with Polycystic Ovarian Syndrome Undergoing in vitro Fertilization-Embryo Transfer:A Randomized Clinical Trial显示文摘Objective:To observe the effect of acupuncture on endometrium and pregnancy outcomes in patients with polycystic ovary syndrome(PCOS)infertility undergoing in vitro fertilization-embryo transfer(IVF-ET).Methods:Eighty-three patients were randomly assigned to observation group(40 cases)and control group(43 cases)according to the random numbers generated by SPSS software.The patients of the two groups received GnRH agonist long protocol as a routine treatment.In the observation group,acupuncture was given at two acupoint groups for 30 min once every other day.Group 1 included Guanyuan(CV 4),Qihai(CV 6),Zhongji(CV 3),bilateral acupoints Zigong(EX-CA1).Group 2 included Mingmen(GV 4),Yaoyangguan(GV 3),bilateral Shenshu(BL 23)and Ciliao(BL 32).The two groups of acupoints were used alternately.The whole needling process was performed at the time of ovulation induction until the transplantation day and consisted of3 courses,while the control group did not receive acupuncture interventions.The Gn dosage and Gn stimulation time,endometrial thickness and type(A,B,and C),serum oestradiol(E2)and progesterone(P)levels on the day of injection of human chorionic gonadotropin(hCG),clinical pregnancy rate,as well as live birth rate were observed.Adverse reactions were also be recorded.All patients were followed up for the pregnant rate 14 days after IVF-ET and live birth rate after pregnancy.All adverse reactions(AEs)of acupuncture were recorded during the trial.Ressults:The Gn dosage and Gn stimulation time in the observation group were lower in the observation group than in the control group(P<0.01).The proportion of type A endometrium in the observation group were higher than that in the control group(P<0.05).The serum E2 and P levels on the day of hCG injection was lower and the clinical pregnancy rate was higher in the observation group compared to those in the control group(P<0.05).There was no serious AEs during this trial.Conclusion:Acupuncture can improve the proportion of type A endometrium,regulate the levelse of serum Eand P on the day of hCG injection,and improve the pregnancy rate in patients with PCOS infertility undergoing IVF-ET. | WU Jia-man NING Yan YE Yang-yang LIU Yu-lei TANG Meng HU Shan ZHUO Yuan-yuan | 2022 | Chinese Journal of Integrative Medicine2022,28,8: | 3 |
| 8 | Numerical study on dynamic mechanism of brain volume and shear deformation under blast loading显示文摘Blast-induced traumatic brain injury(b-TBI)is a kind of significant injury to soldiers in the current military conflicts.However,the mechanism of b-TBI has not been well understood,and even there are some contradictory conclusions.It is crucial to reveal the dynamic mechanism of brain volume and shear deformations under blast loading for better understanding of b-TBI.In this paper,the numerical simulation method is adopted to carry out comprehensive and in-depth researches on this issue for the first time.Based on the coupled Eulerian-Lagrangian method,the fluid-structure coupling model of the blast wave and human head is developed to simulate two situations,namely the head subjected to the frontal and lateral impacts.The simulation results are analyzed to obtain the underlying dynamic mechanisms of brain deformation.The brain volume deformation is dominated by the local bending vibration of the skull,and the corresponding frequency for the forehead skull under the frontal impact and the lateral skull faced to the lateral impact is as high as 8 kHz and 5 kHz,respectively.This leads to the high-frequency fluctuation of brain pressure and the large pressure gradient along the skull,totally different from the dynamic response of brain under head collisions.While the brain shear deformation mainly depends on the relative tangential displacement between the skull and brain and the anatomical structure of inner skull,being not related to the brain pressure and its gradient.By further comparing the medical statistics,it is inferred that diffuse axonal injury and brain contusion,the two most common types of b-TBI,are mainly attributed to brain shear deformations.And the von Mises stress can be adopted as the indicator for these two brain injuries.This study can provide theoretical guidance for the diagnosis of b-TBI and the development of protective equipment. | Zhijie Li Zhibo Du Xiaochuan You Zhanli Liu Jian Cheng Chengcheng Luo Dongyang Chu Shaowu Ning Yue Kang Ce Yang Zhuo Zhuang | 2019 | Acta Mechanica Sinica2019,35,5: | 3 |

| 10 | Recursive adaptive filter using current innovation for celestial navigation during the Mars approach phase显示文摘Celestial navigation is a commonly used autonomous navigation technique for deep space navigation.A nonlinear filter such as the unscented Kalman filter(UKF) is typically applied in a celestial navigation system(CNS). However, on account of being subject to a number of factors, such as ephemeris errors and centroid determination, the measurement model error of a CNS cannot be accurately determined. The analysis conducted in this study also shows that the measurement model error is time-variant during the Mars approach phase.This implies that covariance matrix of the measurement error R is usually inaccurate, which may induce large estimation errors that even result in filter divergence. Some adaptive methods are able to address this issue.However, traditional adaptive filters, for scaling R, usually require a sequence of innovation and are affected by the statistic window size. A new recursive adaptive UKF(RAUKF) is proposed in this paper, which only uses current innovation to scale R. The navigation performance of the proposed RAUKF method is compared with some traditional adaptive filters through simulations. In this paper, we demonstrate a coupled ridge waveguide QCL array consisting of five elements with chirped geometry. In-phase mode operation is secured by managing supermode loss with properly designed geometries of ridges. A single-lobe lateral far-field with a near diffraction limited beam pattern was obtained in the whole current dynamic range. The devices were fabricated with the wet and dry etching method.
