5Exploring and exploiting genetics and genomicsfor sweetpotato improvement: Status andperspectives显示文摘Sweetpotato(Ipomoea batatas(L.)Lam.)is one of the most important root crops cultivated worldwide.Because of its adaptability,high yield potential,and nutritional value,sweetpotato has become an important food crop,particularly in developing countries.To ensure adequate crop yields to meet increasing demand,it is essential to enhance the tolerance of sweetpotato to environmental stresses and other yield-limiting factors.The highly heterozygous hexaploid genome of I.batatas complicates genetic studies and limits improvement of sweetpotato through traditional breeding.However,application of next-generation sequencing and high-throughput genotyping and phenotyping technologies to sweetpotato genetics and genomics research has provided new tools and resources for crop improvement.In this review,we discuss the genomics resources that are available for sweetpotato,including the current reference genome,databases,and available bioinformatics tools.We systematically review the current state of knowledge on the polyploid genetics of sweetpotato,including studies of its origin and germplasm diversity and the associated mapping of important agricultural traits.We then outline the conventional and molecular breeding approaches that have been applied to sweetpotato.Finally,we discuss future goals for genetic studies of sweetpotato and crop improvement via breeding in combination with state-of-the-art multi-omics approaches such as genomic selection and gene editing.These approaches will advance and accelerate genetic improvement of this important root crop and facilitate its sustainable global production.Mengxiao Yan Haozhen Nie Yunze Wang Xinyi Wang Robert Jarret Jiamin Zhao Hongxia Wang Jun Yang 2022Plant Communications2022,3,5:2
6Transplantation of Mesenchymal Stem Cells Attenuates Acute Liver Failure in Mice via an Interleukin-4-dependent Switch to the M2 Macrophage Anti-inflammatory Phenotype显示文摘Background and Aims:Transplantation of mesenchymal stem cells(MSCs)derived from bone marrow(BM)is an alternative treatment of acute liver failure(ALF)mainly be-cause of the resulting anti-inflammatory activity.It is not known how MSCs regulate local immune responses and liver regeneration.This study explored the effects of MSCs on hepatic macrophages and the Wnt signaling pathway in ALF.Methods:MSCs were isolated from BM aspirates of C57BL/6J mice,and transplanted in mice with ALF induced by D-galactosamine(D-Gal).The proliferation of hepato-cytes was assayed by immunohistochemical(IHC)staining of Ki-67 and proliferating cell nuclear antigen(PCNA).The levels of key proteins in the Wnt signaling pathway were assayed by western blotting and cytokines were determined enzyme-linked immunosorbent assays(ELISAs).A mac-rophage polarization assay characterized the M1/M2 ratio.The potential role of interleukin-4(IL-4)in the biological ac-tivity of MSCs was determined by silencing of IL-4.Results:Transplantation of allogeneic MSCs significantly attenuated D-Gal-induced hepatic inflammation and promoted liver re-generation.MSC transplantation significantly promoted a phenotypic switch from proinflamatory M1 macrophages to anti-inflammatory M2 macrophages,leading to significant Wnt-3a induction and activation of the Wnt signaling path-way in mice with D-Gal-induced ALF.Of the paracrine fac-tors secreted by MSCs(G-CSF,IL-6,IL-1 beta,IL-4,and IL-17A),IL-4 was specifically induced following transplantation in the ALF model mice.The silencing of IL-4 significantly ab-rogated the phenotypic switch to M2 macrophages and the protective effects of MSCs in both the ALF model mice and a co-culture model in an IL-4 dependent manner.Conclu-sions:In vivo and in vitro studies showed that MSCs ame-liorated ALF through an IL-4-dependent macrophage switch toward the M2 anti-inflammatory phenotype.The findings may have clinical implications in that overexpression of IL-4 may enhance the therapeutic effects of allogeneic MSC transplantation in the treatment of ALF.Jinglin Wang Haoran Ding Jingchao Zhou Senzhe Xia Xiaolei Shi Haozhen Ren 2022Journal of Clinical and Translational Hepatology2022,10,4:2
7Self-assembled manganese phthalocyanine nanoparticles with enhanced peroxidase-like activity for anti-tumor therapy显示文摘The use of functional nanoparticles as peroxidase-like(POD-like)catalyst has recently become a focus of research in cancer therapy.Phthalocyanine is a macrocyclic conjugated metal ligand,which is expected to achieve a high POD-like catalytic activity,generating free radicals and inhibiting the proliferation of cancer cells.In this paper,we synthesized phthalocyanine nanocrystals with different structures through noncovalent self-assembly confined within micro-emulsion droplets,and manganese phthalocyanine(MnPc)possessing a metal–N–C active center was used as the building block.These nano-assemblies exhibit shape-dependent POD-like catalytic activities,because the emulsifier and MnPc co-mixed assembly reduced the close packing between MnPc molecules and exposed more active sites.The assembly had a water-dispersed nanostructure,which is conducive to accumulation at tumor sites through the enhanced permeability and retention effect(EPR).Because of a highly efficient microenvironmental response,the assembly showed higher catalytic activity only emerged under the acidic tumor-like microenvironment,but caused less damage to normal tissues in biomedical applications.In vivo and in vitro catalytic therapy tests showed excellent anti-tumor effects.This work explored a new way for the application of metal–organic macromolecules such as MnPc as nanozymes for catalytic tumor therapy.Jinghan Wang Shanqing Gao Xiao Wang Haozhen Zhang Xitong Ren Juewen Liu Feng Bai 2022Nano Research2022,15,3:2

14Polysulfide regulation by defect-modulated Ta_(3)N_(5-x) electrocatalyst toward superior room-temperature sodium-sulfur batteries显示文摘Resolving low sulfur reaction activity and severe polysulfide dissolution remains challenging in metalsulfur batteries.Motivated by a theoretical prediction,herein,we strategically propose nitrogenvacancy tantalum nitride(Ta3N5-x)impregnated inside the interconnected nanopores of nitrogendecorated carbon matrix as a new electrocatalyst for regulating sulfur redox reactions in roomtemperature sodium-sulfur batteries.Through a pore-constriction mechanism,the nitrogen vacancies are controllably constructed during the nucleation of Ta3N5-x.The defect manipulation on the local environment enables well-regulated Ta 5d-orbital energy level,not only modulating band structure toward enhanced intrinsic conductivity of Ta-based materials,but also promoting polysulfide stabilization and achieving bifunctional catalytic capability toward completely reversible polysulfide conversion.Moreover,the interconnected continuous Ta3N5-x-in-pore structure facilitates electron and sodium-ion transport and accommodates volume expansion of sulfur species while suppressing their shuttle behavior.Due to these attributes,the as-developed Ta3N5-x-based electrode achieves superior rate capability of 730 mAh g-1 at 3.35 A g-1,long-term cycling stability over 2000 cycles,and high areal capacity over 6 mAh cm-2 under high sulfur loading of 6.2 mg cm-2.This work not only presents a new sulfur electrocatalyst candidate for metal-sulfur batteries,but also sheds light on the controllable material design of defect structure in hopes of inspiring new ideas and directions for future research.Zhen Zhang Dan Luo Jun Chen Chuyin Ma Matthew Li Haoze Zhang Renfei Feng Rui Gao Haozhen Dou Aiping Yu Xin Wang Zhongwei Chen 2024Science Bulletin2024,69,2:0
15Engineered stem cells by emerging biomedical stratagems显示文摘Stem cell therapy holds immense potential as a viable treatment for a widespread range of intractable disorders.As the safety of stem cell transplantation having been demonstrated in numerous clinical trials,various kinds of stem cells are currently utilized in medical applications.Despite the achievements,the therapeutic benefits of stem cells for diseases are limited,and the data of clinical researches are unstable.To optimize tthe effectiveness of stem cells,engineering approaches have been developed to enhance their inherent abilities and impart them with new functionalities,paving the way for the next generation of stem cell therapies.This review offers a detailed analysis of engineered stem cells,including their clinical applications and potential for future development.We begin by briefly introducing the recent advances in the production of stem cells(induced pluripotent stem cells(ipsCs),embryonic stem cells(ESCs),mesenchymal stem cells(MSCs)and hematopoietic stem cells(HSCs).Furthermore,we present the latest developments of engineered strategies in stem cells,including engineered methods in molecular biology and biomaterial fields,and their application in biomedical research.Finally,we summarize the current obstacles and suggest future prospects for engineered stem cells in clinical translations and biomedical applications.Jinglin Wang Xiaoxuan Zhang Hanxu Chen Haozhen Ren Min Zhou Yuanjin Zhao 2024Science Bulletin2024,69,2:0
16Incidence and risk factors of in-hospital venous thromboembolism in non-oncological urological inpatients:A single center experience显示文摘Objective To determine incidence and risk factors for venous thromboembolism(VTE)development of in-hospital VTE in urological inpatients who underwent non-oncological surgery in a tertiary hospital in China.Methods Consecutive 1453 inpatients who were admitted to a non-oncological urological ward in the tertiary hospital from January 1,2018 to December 31,2018 were enrolled in the study,and the VTE events were diagnosed by ultrasound or computed tomographic pulmonary angiography.Patients’occurrence of VTE and characteristics which may contribute to the development of VTE were collected and analyzed as incidence and risk factors.Results The incidence of VTE in non-oncological urological inpatients is 2.3%.In our cohort,patients who experienced previous VTE(adjusted odds ratios[aOR]14.272,95%CI 3.620-56.275),taking anticoagulants or antiplatelet agents before admission(aOR 10.181,95%CI 2.453-42.256),D-dimer(max)≥1μg/mL(aOR 22.456,95%CI 6.468-77.967),lower extremity swelling(aOR 10.264,95%CI 2.242-46.994),chest symptoms(aOR 79.182,95%CI 7.132-879.076),operation time of more than or equal to 180 min(aOR 10.690,95%CI 1.356-84.300),and Caprini score(max)of more than or equal to 5(aOR 34.241,95%CI 1.831-640.235)were considered as risk factors for VTE.Conclusion In this study,we found that the incidence of VTE in non-oncological surgery was about 2.3%,which was higher than some previous studies.Risk factors could be used for early detection and diagnosis of VTE.Zhao Wang Kaixuan Li Quan Zhu Haozhen Li Ziqiang Wu Xuesong Liu Zhengyan Tang 2023Asian Journal of Urology2023,10,4:0
17Infection fatality rate and infection attack rate of COVID-19 in South American countries显示文摘Background:The ongoing COVID-19 pandemic hit South America badly with multiple waves.Different COVID-19 variants have been storming across the region,leading to more severe infections and deaths even in places with high vaccination coverage.This study aims to assess the spatiotemporal variability of the COVID-19 pandemic and estimate the infection fatality rate(IFR),infection attack rate(IAR)and reproduction number(R0)for twelve most affected South American countries.Methods:We fit a susceptible-exposed-infectious-recovered(SEIR)-based model with a time-varying transmission rate to the reported COVID-19 deaths for the twelve South American countries with the highest mortalities.Most of the epidemiological datasets analysed in this work are retrieved from the disease surveillance systems by the World Health Organization,Johns Hopkins Coronavirus Resource Center and Our World in Data.We investigate the COVID-19 mortalities in these countries,which could represent the situation for the overall South American region.We employ COVID-19 dynamic model with-and-without vaccination considering time-varying flexible transmission rate to estimate IFR,IAR and R0 of COVID-19 for the South American countries.Results:We simulate the model in each scenario under suitable parameter settings and yield biologically reasonable estimates for IFR(varies between 0.303% and 0.723%),IAR(varies between 0.03 and 0.784)and R0(varies between 0.7 and 2.5)for the 12 South American countries.We observe that the severity,dynamical patterns of deaths and time-varying transmission rates among the countries are highly heterogeneous.Further analysis of the model with the effect of vaccination highlights that increasing the vaccination rate could help suppress the pandemic in South America.Conclusions:This study reveals possible reasons for the two waves of COVID-19 outbreaks in South America.We observed reductions in the transmission rate corresponding to each wave plausibly due to improvement in nonpharmaceutical interventions measures and human protective behavioral reaction to recent deaths.Thus,strategies coupling social distancing and vaccination could substantially suppress the mortality rate of COVID-19 in South America.Salihu Sabiu Musa Amna Tariq Liu Yuan Wei Haozhen Daihai He 2022Infectious Diseases of Poverty2022,11,2:0
18Human endoderm stem cells reverse inflammation-related acute liver failure through cystatin SN-mediated inhibition of interferon signaling显示文摘Acute liver failure(ALF)is a life-threatening disease that occurs secondary to drug toxicity,infection or a devastating immune response.Orthotopic liver transplantation is an effective treatment but limited by the shortage of donor organs,the requirement for life-long immune suppression and surgical challenges.Stem cell transplantation is a promising alternative therapy for fulminant liver failure owing to the immunomodulatory abilities of stem cells.Here,we report that when transplanted into the liver,human endoderm stem cells(hEnSCs)that are germ layer-specific and nontumorigenic cells derived from pluripotent stem cells are able to effectively ameliorate hepatic injury in multiple rodent and swine drug-induced ALF models.We demonstrate that hEnSCs tune the local immune microenvironment by skewing macrophages/Kupffer cells towards an anti-inflammatory state and by reducing the infiltrating monocytes/macrophages and inflammatory T helper cells.Single-cell transcriptomic analyses of infiltrating and resident monocytes/macrophages isolated from animal livers revealed dramatic changes,including changes in gene expression that correlated with the change of activation states,and dynamic population heterogeneity among these cells after hEnSC transplantation.We further demonstrate that hEnSCs modulate the activation state of macrophages/Kupffer cells via cystatin SN(CST1)-mediated inhibition of interferon signaling and therefore highlight CST1 as a candidate therapeutic agent for diseases that involve over-activation of interferons.We propose that hEnSC transplantation represents a novel and powerful cell therapeutic treatment for ALF.Yilin Xu Jinglin Wang Haozhen Ren Hao Dai Ying Zhou Xiongzhao Ren Yang Wang Sisi Feng Xiaogang Deng Jiaying Wu Tianlong Fu Tengfei Nie Haifeng He Tongkun Wei Bing Zhu Lijian Hui Bin Li Jing Wang Hongyan Wang Luonan Chen Xiaolei Shi Xin Cheng 2023Cell Research2023,33,2:0
19Eco-friendly physical blowing agent mass loss of bio-based polyurethane rigid foam materials显示文摘Through systematical experiment design, the physical blowing agent(PBA) mass loss of bio-based polyurethane rigid foam(PURF)in the foaming process was measured and calculated in this study, and different eco-friendly PBA mass losses were measured quantitatively for the first time. The core of the proposed method is to add water to replace the difference, and this method has a high fault tolerance rate for different foaming forms of foams. The method was proved to be stable and reliable through the standard deviations σ1and σ2for R1(ratio of the PBA mass loss to the material total mass except the PBA) and R2(ratio of the PBA mass loss to the PBA mass in the material total mass) in parallel experiments. It can be used to measure and calculate the actual PBA mass loss in the foaming process of both bio-based and petroleumbased PURF. The results show that the PBA mass loss in PURF with different PBA systems is controlled by its initial mass content of PBA in PU materials ω. The main way for PBA to dissipate into the air is evaporation/escape along the upper surface of foam. This study further reveals the mechanism of PBA mass loss: the evaporation/escape of PBA along the upper surface of foam is a typical diffusion behavior. Its spread power comes from the difference between the chemical potential of PBA in the interface layer and that in the outside air. 20Stimuli-Responsive Gene Delivery Nanocarriers for Cancer Therapy显示文摘Gene therapy provides a promising approach in treating cancers with high efficacy and selectivity and few adverse effects.Currently,the development of functional vectors with safety and effectiveness is the intense focus for improving the delivery of nucleic acid drugs for gene therapy.For this purpose,stimuli-responsive nanocarriers displayed strong potential in improving the overall efficiencies of gene therapy and reducing adverse effects via effective protection,prolonged blood circulation,specific tumor accumulation,and controlled release profile of nucleic acid drugs.Besides,synergistic therapy could be achieved when combined with other therapeutic regimens.This review summarizes recent advances in various stimuliresponsive nanocarriers for gene delivery.Particularly,the nanocarriers responding to endogenous stimuli including pH,reactive oxygen species,glutathione,and enzyme,etc.,and exogenous stimuli including light,thermo,ultrasound,magnetic field,etc.,are introduced.Finally,the future challenges and prospects of stimuli-responsive gene delivery nanocarriers toward potential clinical translation are well discussed.The major objective of this review is to present the biomedical potential of stimuli-responsive gene delivery nanocarriers for cancer therapy and provide guidance for developing novel nanoplatforms that are clinically applicable.Qingfei Zhang Gaizhen Kuang Wenzhao Li Jinglin Wang Haozhen Ren Yuanjin Zhao 2023Nano-Micro Letters2023,15,3:0
