1Electrospinning of ceramic nanofibers:Fabrication,assembly and applications显示文摘This paper provides a brief review of current research activities that focus on the synthesis and controlled assembly of inorganic nano-bers by electrospinning,their electrical,optical and magnetic properties,as well as their applications in various areas including sensors,catalysts,batteries,filters and separators.We begin with a brief introduction to electrospinning technology and a brief method to produce ceramic nanofibers from electrospinning.We then discuss approaches to the controlled assembly and patterning of electrospun ceramic nanofibers.We continue with a highlight of some recent applications enabled by electrospun ceramic nano-bers,with a focus on the physical properties of functional ceramic nanofibers as well as their applications in energy and environmental technologies.In the end,we conclude this review with some perspectives on the future directions and implications for this new class of functional nanomaterials.It is expected that this review paper can help the readers quickly become acquainted with the basic principles and particularly the experimental procedure for preparing and assembly of 1D ceramic nanofiber and its arrays.Hui WU Wei PAN Dandan LIN Heping LI 2012Journal of Advanced Ceramics2012,1,1:9
2Expression of reconstructive hF Ⅷ in the hrDNA by using hrDNA targeting vector显示文摘Our lab has constructed a new nonviral vec-tor—hrDNA targeting vector(pHrneo). pHrneo is a human derived vector that can target gene into human ribosomal DNA(hrDNA) locus. In this study, we inserted expression cassette of reconstructive hF Ⅷ (hFVIII-BDDAK39) to pHrneo to construct targeting vector: pHrneo-BDDAK39. Through electroporation of pHrneo-BDDAK39 into HT1080 cells, we identified the homologous recombinants by PCR and Southen blotting, and tested the expression of hFVIII- BDDAK39 in the hrDNA locus. The hFⅧ-BDDAK39 was successfully targeted into hrDNA locus of HT-1080 by pHrneo-BDDAK39, and the efficiency of site-specific inte-gration was 2.0×10?5. hFⅧ-BDDAK39 in hrDNA locus of HT-1080 is found to be able to express efficiently (32±5 ng·106 cells?1·24 h?1). 3Investigation of hrDNA targeting vector-mediated tumor-specific suicide gene therapy for hepatocellular carcinoma显示文摘Human ribosomal DNA (hrDNA) target- ing vector (pHrn) is one of the human derived vectors, which was devised by our lab and has got patent authority. To investigate its effect on gene therapy for hepatocellular carcinoma, a double suicide fusion gene expression cassette, CDUPRT/GFP controlled by a synthetic CMV enhancer-enhanced hTERT promoter (CeTp) which was determined by luciferase assays was constructed in pHrn backbone, creating an expression vector pHr-CeTpCDUPRT/GFP. After transfer of plasmid to hepatocellular carcinoma cell line Bel7402 in vitro, the transfection efficiency reached 30%―50% by using flow cytometer. The expression of CDUPRT/GFP was detected by RT-PCR and Western Blotting. After the administra- tion of 5-FC, high performance liquid chromatography (HPLC) was applied to examine the level of 5-FU in supernatant, resulting in a concentration of 60.15 μg/mL. The Methylthiazolyl tetrazolium (MTT) assay was then utilized to investigate the antitumor effect of pHr-CeTpCDUPRT/GFP in Bel7402 cells, and rela- tive cell survival of 60%-35% was observed after 5-FC treatment. In vivo experiments, the nude mouse model of hepatocellular carcinoma was constructed and in situ gene therapy was performed. The results indicated the tumor growth of treatment group was obviously suppressed, and some even shrank, whenthe vectors and prodrugs were injected continuously. The expression of CDUPRT in tumor tissues was also identified by RT-PCR, and the concentration of 5-FU was 7.694 μg/mL in blood serum using HPLC detection. Then the pathological section of tumor tissues revealed significant tumor cell necrosis. 4Functional analysis of three genetic disorder related PITX2 mutants显示文摘The autosome dominant disorders, ring dermoid of the cornea (RDC), iris hypolasia (IH) and Axenfeld-Rieger syndrome (ARS), are allelic disor- ders, as all three can result from mutations of the transcriptional factor PITX2. Among three disorder phenotypes, ARS is the most severe, IH is milder than ARS, and RDC is the mildest. Missense muta- tions of the PITX2 homeodomain identified in RDC (R62H), IH (R84W) and ARS patients (T68P) were introduced into PITX2 cDNA by site-directed mutagenesis. PITX2 mutant proteins expressed in eucaryotic cells were stable and localized to the nu- cleus. Analysis of these mutant PITX2 proteins by DNA-binding shift and transactivation studies dem- onstrated that the R62H had the most activity in both studies, and the R84W still retained somewhat func- tions, whereas the T68P proved to be non-functional. 5Silica nanoparticle is a possible safe carrier for gene therapy显示文摘In order to develop a safe and effective gene therapy carrier, some toxicological and biodynamical ex- periments were carried out on silica nanoparticles (SiNPs). First we prepared SiNPs with appropriate portions of cyclo- hexane, deionized water and ethyl silicate, and then trans- fected the modified SiNPs and GFP plasmid DNA complex into the HT1080 cells to test the effectiveness of transfection for gene therapy. At the same time, we injected the SiNPs into a number of mice through tail vein. Then we made the mice crossed to evaluate the acute, long-term and reproduc- tive toxicity. In vivo distribution analysis and pathological examination were made on both adult mice and their off- spring. SiNPs were uniform and had an average diameter of 40 nm, and the modified SiNPs carried exogenous DNA molecules into target cells and the transferred GFP fusion gene was effectively expressed in the cells. The SiNPs injected via tail vein were widely distributed in almost all of tissues, and the injected mice had the ability to reproduce normally. 6Radiative heat transfer and thermal characteristics of Fe-based oxides coated SiC and Alumina RPC structures as integrated solar thermochemical reactor显示文摘This paper investigated radiative heat transfer and thermal characteristics of Fe-based oxides coated SiC and Alumina reticulated porous ceramic structures as integrated solar thermochemical reactor.High-flux solar radiation absorption and axial temperature distribution in the ceramic foams reactor were analyzed by adopting surface-to-surface radiation model coupled to the P1approximation for radiation heat transfer.The radiative heat transfer and thermal characteristics of different foam-type RPC structures,including SiC,CeO_2,FeAl_2O_4,NiFeAlO_4,Fe_3O_4/SiC,and NiFe_2O_4/SiC were evaluated.The mass flow rate and foam structural parameters,including the permeability,pore mean cell size,and extinction coefficients have significantly affected the axial temperature distribution,pressure drop,heat transfer,and fluid flow.Integrated porous structure to the solar receiver could maximize the incorporation of redox powder in the reacting medium,lower the pressure drop,and enhance the thermal performance of the thermochemical reacting system.SiC structure was the candidate materials in the case where more heat flux and high axial temperature distribution is needed.However,Fe-based oxide coated Al_2O_3structure could be considered regarding the heat transfer enhancement along with the catalyst activity of oxygen carriers for solar thermochemical reacting system performance.GUENE LOUGOU Bachirou SHUAI Yong PAN RuMing CHAFFA Gédéon AHOUANNOU Clément ZHANG Hao TAN HePing 2018Science China(Technological Sciences)2018,61,12:2
7Identification and characterization of human neuronal voltage-gated calcium channel gamma 3 subunit gene显示文摘By homologous expressed sequence tag (EST) searching, one EST (GenBank: W29095) was obtained, which shows 75% identity in 435 bp overlap with the coding sequence of mouse Cacng2 gene. A 1 545 bp cDNA fragment was obtained from the nested polymerase chain reaction (PCR) and rapid applification of cDNA end (RACE) reaction in the human brain prefrontal cortex cDNA library and the human brain Ready cDNA with the primers designed on W29095. The fragment contained a 948-bp open reading frame (ORF) encoding 315 amino acids, and was named CACNG3. As it was identical to a BAC clone (GenBank: AC004125) from chromosome 16p12-p13.1, the CACNG3 gene was mapped to human chromosome 16p12-p13.1, and the coding region was composed of 4 exons. Reverse transcription PCR (RT-PCR) analysis showed that the CACNG3 gene expressed in human adult brain and fetal brain. 8Exogenous glutamic acid effectively involved in N-(1-deoxy-D-galulos-1-yl)-glutamic acid degradation for simultaneous improvement of both milk-like and baking flavor显示文摘This research elucidated the reason of flavor deficiency during the thermal degradation of glutamic acid(Glu)-galactose(Gal)Amadori rearrangement product(GG-ARP)and the solution.The flavor formation from GG-ARP during the thermal degradation was investigated.Compared to the volatile compounds in Glu-Gal Maillard reaction products(GG-MRP),pyrazines in the volatile compounds derived from GG-ARP were restrained.The disadvantage in evolution asynchrony and content sufficiency ofα-dicarbonyl compounds and Glu in the GG-ARP model limited the pyrazines formation.With the addition of Gal,the asynchronous effect was further exacerbated,and the furans in the GG-ARP-Gal model(956.81μg/L,120℃,60 min)were more significantly dominant among the volatile flavor compounds than in the GG-ARP model(454.98μg/L,120℃,60 min),while the pyrazines were still missing.Whereas,the extra-added Glu in the GG-ARP model could offset the deficiency of amino acids growth afterα-dicarbonyl formation and promote the formation of pyrazines(74.47μg/L,120℃,60 min).Meanwhile,the formation of diketones(92.96μg/L,120℃,60 min)was promoted.The addition of Glu enhanced both milk-like and baking flavor of the GG-ARP system simultaneously.Formulated GG-ARP-Glu mixture was proposed to promisingly achieve the controlled formation of process flavors for baked foods with both desirable milk-like and baking scent.Caixing Pan Heping Cui Khizar Hayat Xiaoming Zhang Chi-Tang Ho 2022Food Bioscience2022,47,3:1

12Multiphysics Field Co-simulation Research Based on Electric Drive System for a Specific Type of Car显示文摘Electric drive systems for new energy cars are complex systems that should have multivariate,strong coupling,and non-linear characteristics and should also involve the multiphysics field.The singular simulation software used at present in the modeling of electric drive systems cannot simulate the influences of all the physics fields on the operating system.The co-simulation model used in this paper was based on a specific type of car.The motor control algorithm model was built in MATLAB/Simulink,the electromagnetic finite element model of the motor was built in ANSYS EM-Maxwell,and the motor controller hardware circuit was built in ANSYS EM-Simplorer.To make real-time connections among these software platforms,a multi-software co-simulation platform was built,and the co-simulation platform’s simulation results were input into STAR CCM+software to enable finite element modeling of the motor and running of thermal analysis.When compared with the electric drive system model built using single Simulink software,the simulation results from this co-simulation platform were more realistic and were shown to be closer to reality when the dynamic characteristics of the electric drive system’s power semiconductor switching devices and the motor’s electromagnetic characteristics were considered.Finally,by benchmarking the multiphysics field co-simulation platform simulation results using dyno bench test results,the validity of the co-simulation platform was verified and the development of the multiphysics field co-simulation of the basic electric drive system was complete.Chuanfu Wang Yubo Lian Heping Ling Hua Pan Yuxin Zhang 2019Automotive Innovation2019,2,3:1

14Molecular wakes for ultrashort laser pulses显示文摘The molecular wake-assisted interaction between two collinear femotosecond laser pulses is investigated in air,which leads to the generation of a controllable 1.8 mJ super-continuum pulse with an elongated self-guided channel due to the cross-phase modulation of the impulsively aligned diatomic molecules in air. 18DOCK2 regulates antifungal immunity by regulating RAC GTPase activity显示文摘Fungal infections cause~1.5 million deaths each year worldwide,and the mortality rate of disseminated candidiasis currently exceeds that of breast cancer and malaria.The major reasons for the high mortality of candidiasis are the limited number of antifungal drugs and the emergence of drug-resistant species.Therefore,a better understanding of antifungal host defense mechanisms is crucial for the development of effective preventive and therapeutic strategies.Here,we report that DOCK2(dedicator of cytokinesis 2)promotes indispensable antifungal innate immune signaling and proinflammatory gene expression in macrophages.DOCK2-deficient macrophages exhibit decreased RAC GTPase(Rac family small GTPase)activation and ROS(reactive oxygen species)production,which in turn attenuates the killing of intracellular fungi and the activation of downstream signaling pathways.Mechanistically,after fungal stimulation,activated SYK(spleen-associated tyrosine kinase)phosphorylates DOCK2 at tyrosine 985 and 1405,which promotes the recruitment and activation of RAC GTPases and then increases ROS production and downstream signaling activation.Importantly,nanoparticle-mediated delivery of in vitro transcribed(IVT)Rac1 mRNA promotes the activity of Rac1 and helps to eliminate fungal infection in vivo.Taken together,this study not only identifies a critical role of DOCK2 in antifungal immunity via regulation of RAC GTPase activity but also provides proof of concept for the treatment of invasive fungal infections by using IVT mRNA.Xiaojian Ma Xi Tan Bingbing Yu Wanwei Sun Heping Wang Huijun Hu Yanyun Du Ruirui He Ru Gao Qianwen Peng Zhihui Cui Ting Pan Xiong Feng Junhan Wang Chengqi Xu Bin Zhu Wei Liu Chenhui Wang 2022Cellular & Molecular Immunology2022,19,5:1
19N-doped interconnected carbon aerogels as an efficient SeS2 host for long life Na-SeS2 batteries显示文摘Selenium sulfide(SeS2)cathodes have attracted much concern as an optimized choice comparing to sulfur and selenium for lithium and sodium storage.However,it also suffers from poor cycling stability due to the dissolution of reaction intermediate products.In this study,N-doped Interconnected carbon aerogels was applied as an efficient SeS2 host by infiltrating selenium sulfide into its microporous structure(denoted as SeS2@NCAs),which could effectively accommodate the volume change of SeS2 during cycling and alleviate the dissolution of reaction intermediate products.Therefore,as for Na storage,the SeS2@NCAs cathode delivers a superior long-term cycling performance of 536 mA·h·g^-1 at a current density of 0.5 A·g^-1 after 1,000 cycles with only 0.04%capacity decline per cycle and a high rate performance(524 mA·h·g^-1 at 2 A·g^-1 and 745 mA·h·g^-1 at 0.1 A·g^-1 retained),indicating the remarkable cycling stability of SeS2@NCAs cathodes.Yurui Deng Lunlun Gong Hoda Ahmed Yuelei Pan Xudong Cheng Siyu Zhu Heping Zhang 2020Nano Research2020,13,4:1
