| 1 | Artesunate synergizes with sorafenib to induce ferroptosis in hepatocellular carcinoma显示文摘Sorafenib is the first-line medication for advanced hepatocellular carcinoma(HCC),but it can only extend limited survival.It is imperative to find a combination strategy to increase sorafenib efficacy.Artesunate is such a preferred candidate,because artesunate is clinically well-tolerated and more importantly both drugs can induce ferroptosis through different mechanisms.In this study we investigated the combined effect of sorafenib and artesunate in inducing ferroptosis of HCC and elucidated the involved molecular mechanisms.We showed that artesunate greatly enhanced the anticancer effects of low dose of sorafenib against Huh7,SNU-449,and SNU-182 HCC cell lines in vitro and against Huh7 cell xenograft model in Balb/c nude mice.The combination index method confirmed that the combined effect of sorafenib and artesunate was synergistic.Compared with the treatment with artesunate or sorafenib alone,combined treatment induced significantly exacerbated lipid peroxidation and ferroptosis,which was blocked by N-acetyl cysteine and ferroptosis inhibitors liproxstatin-1 and deferoxamine mesylate,but not by inhibitors of other types of cell death(z-VAD,necrostatin-1 and belnacasan).In Huh7 cells,we demonstrated that the combined treatment induced oxidative stress and lysosome-mediated ferritinophagy,two essential aspects of ferroptosis.Sorafenib at low dose mainly caused oxidative stress through mitochondrial impairments and SLC7A11-invovled glutathione depletion.Artesunate-induced lysosome activation synergized with sorafenib-mediated pro-oxidative effects by promoting sequential reactions including lysosomal cathepsin B/L activation,ferritin degradation,lipid peroxidation,and consequent ferroptosis.Taken together,artesunate could be repurposed to sensitize sorafenib in HCC treatment.The combined treatment can be easily translated into clinical applications. | Zhong-jie Li Hui-qi Dai Xiao-wei Huang Ji Feng Jing-huan Deng Zi-xuan Wang Xiao-mei Yang Yu-jia Liu Yong Wu Pan-hong Chen Huan Shi Ji-gang Wang Jing Zhou Guo-dong Lu | 2021 | Acta Pharmacologica Sinica2021,42,2: | 31 |
| 2 | Morphological study on colonic pathology in ulcerative colitis treated by moxibustion显示文摘AIM To observe the therapeutic effect ofmoxibustion on ulcerative colitis and itsinfluence on the colonic mucosal morphology.METHODS Forty-six patients with ulcerativecolitis were randomly divided into themoxibustion with herbal medicine underneathgroup and the western medicine group.Thirtypatients were treated with the abovemoxibustion and 16 patients with Salicylayefapyridine(SASP).The colonic mucosa of 13patients in the moxibustion group was observedby colonoscopy before and after the treatment.Mucin was also analyzed by H.E and AB-PASstaining.RESULTS Seventeen patients were clinicallycured,12 were improved and 1 unchanged in themoxibustion group.In the control group,5patients were clinically cured,7 improved and 4unchanged.Thirteen patients with active UCwere taken as the subjects for histopathologicanalysis in this study.The colonic mucosallesions were remarkably improved and thecharacteristic of the mucin also changed.Inmost sections,the chronic inflammation ofmucosa was geatly ameliorated(P<0.01).Theinflammatory cell infiltratation much decreased and neutrophils,disapeared in most sections(P<0.001).The goblet cells significantlyincreased(P<0.001);crypt paracrypt abscessor mucosal ulceration was seen(P<0.001).CONCLUSION The rate of cure of ulcerative colitis by moxibustion with herbal medicine beneath is superior to that by SASP. | 3 | Epidemiology, causes, clinical manifestation and diagnosis, prevention and control of coronavirus disease (COVID-19) during the early outbreak period: a scoping review显示文摘Background:The coronavirus disease(COVID-19)has been identified as the cause of an outbreak of respiratory illness in Wuhan,Hubei Province,China beginning in December 2019.As of 31 January 2020,this epidemic had spread to 19 countries with 11791 confirmed cases,including 213 deaths.The World Health Organization has declared it a Public Health Emergency of International Concern.Methods:A scoping review was conducted following the methodological framework suggested by Arksey and O'Malley.In this scoping review,65 research articles published before 31 January 2020 were analyzed and discussed to better understand the epidemiology,causes,clinical diagnosis,prevention and control of this virus.The research domains,dates of publication,journal language,authors'affiliations,and methodological characteristics were included in the analysis.All the findings and statements in this review regarding the outbreak are based on published information as listed in the references.Results:Most of the publications were written using the English language(89.2%).The largest proportion of published articles were related to causes(38.5%)and a majority(67.7%)were published by Chinese scholars.Research articles initially focused on causes,but over time there was an increase of the articles related to prevention and control.Studies thus far have shown that the virus7 origination is in connection to a seafood market in Wuhan,but specific animal associations have not been confirmed.Reported symptoms include fever,cough,fatigue,pneumonia,headache,diarrhea,hemoptysis,and dyspnea.Preventive measures such as masks,hand hygiene practices,avoidance of public contact,case detection,contact tracing,and quarantines have been discussed as ways to reduce transmission.To date,no specific antiviral treatment has proven effective;hence,infected people primarily rely on symptomatic treatment and supportive care.Conclusions:There has been a rapid surge in research in response to the outbreak of COVID-19.During this early period,published research primarily explored the epidemiology,causes,clinical manifestation and diagnosis,as well as prevention and control of the novel coronavirus.Although these studies are relevant to control the current public emergency,more high-quality research is needed to provide valid and reliable ways to manage this kind of public health emergency in both the short-and long-term. | Sasmita Poudel Adhikari Sha Meng Yu-Ju Wu Yu-Ping Mao Rui-Xue Ye Qing-Zhi Wang Chang Sun Sean Sylvia Scott Rozelle Hein Raat Huan Zhou | 2020 | Infectious Diseases of Poverty2020,9,2: | 28 |

| 5 | Adjuvant sorafenib after heptectomy for Barcelona Clinic Liver Cancer-stage C hepatocellular carcinoma patients显示文摘AIM: To investigate the efficacy and safety of adjuvant sorafenib after curative resection for patients with Barcelona Clinic Liver Cancer(BCLC)-stage C hepatocellular carcinoma(HCC).METHODS: Thirty-four HCC patients, classified as BCLC-stage C, received adjuvant sorafenib for highrisk of tumor recurrence after curative hepatectomy at a tertiary care university hospital. The study group was compared with a case-matched control group of 68 patients who received curative hepatectomy for HCC during the study period in a 1:2 ratio.RESULTS: The tumor recurrence rate was markedly lower in the sorafenib group(15/34, 44.1%) than in the control group(51/68, 75%, P = 0.002). The median disease-free survival was 12 mo in the study group and 10 mo in the control group. Tumor number more than 3, macrovascular invasion, hilar lymph nodes metastasis, and treatment with sorafenib were significant factors of disease-free survival by univariate analysis. Tumor number more than 3 and treatment with sorafenib were significant risk factors of diseasefree survival by multivariate analysis in the Cox proportional hazards model. The disease-free survival and cumulative overall survival in the study group were significantly better than in the control group(P = 0.034 and 0.016, respectively). | 6 | The Prevalence of Nonalcoholic Fatty Liver Disease and its Association with Lifestyle/dietary Habits among University Faculty and Staff in Chengdu显示文摘Objective To investigate the prevalence of nonalcoholic fatty liver disease(NAFLD) in different university categories and its association with lifestyle/dietary habits.Methods A cross-sectional study was carried out on 9 378 faculty members and staff who participated in an annual health checkup at three universities selected by random cluster sampling.Demographic,anthropometric,biochemical indices and abdominal ultrasound measurements were collected.A nested case-control study was conducted with 200 NAFLD cases and 200 controls matched by gender,age(±3 years),and university.Results The overall prevalence of NAFLD was 10.3%(13.7% in males and 6.8% in females).The prevalence was significantly higher in the science and engineering university(22.1%) than in the comprehensive universities with(6.4%) and without(10.9%) medical colleges.Obesity/overweight,hyperlipidemia,diabetes mellitus,and family history of NAFLD were independently associated with higher risk of NAFLD,as were frequent consumption of desserts and salty/spicy foods.Using nutritional supplements was a protective factor against NAFLD.Intake of coarse cereals,potatoes,vegetables,fruits,and milk was significantly lower,and intake of red meat,viscera,candies and pastries,cooking oil,and total energy was significantly higher in participants with NAFLD than in controls.Conclusion Science and engineering university faculty and staff are key targets for NAFLD prevention.NAFLD is closely associated with age,gender,university type,metabolic diseases,and lifestyle/dietary habits. | SHI Lei LIU Zhong Wu LI Yun GONG Cai ZHANG Huan SONG Li Juan HUANG Cheng Yu LI Ming | 2012 | Biomedical and Environmental Sciences2012,25,4: | 16 |
| 7 | Genesis of the Zhaokalong Fe-Cu Polymetallic Deposit at Yushu, China: Evidence from Ore Geochemistry and Fluid Inclusions显示文摘The ore types of the Zhaokalong Fe-Cu deposit are divided into two categories: sulfide-type and oxide-type. The sulfide-type ore include siderite ore, galena-sphalerite ore and chalcopyrite ore, whereas the oxide-type ore include magnetite ore and hematite ore. The ore textures and structures indicate that the Zhaokalong deposit is of the sedimentary-exhalative mineralization type. Geochemical analyses show that the two ore types have a high As, Sb, Mn, Co and Ni content. The REE patterns reveal an enrichment of the LREE compared to the HREE. Isotopic analysis of siderite ore reveal that the δ13CPDB ranges from 2.01 to 3.34 (‰) whereas the δ18O SMOW ranges from 6.96 to 18.95 (‰). The fluid inclusion microthermometry results indicate that homogenization temperatures of fluid inclusions in quartz range from 131 to 181℃, with salinity values of 1.06 to 8.04 wt% NaCl eq. The mineralizing fluid therefore belongs to the low temperature - low salinity system, with a mineralizing solution of a CO2-Ca2+(Na+, K+)-SO42-(F-, Cl-)-H2O system. The geochemical results and fluid inclusion data provide additional evidence that the Zhaokalong deposit is a sedex-type deposit that experienced two stages of mineralization. The sulfide mineralization probably occurred first, during the sedimentary exhalative process, as exhibited by the abundance of marine materials associated with the sulfide ores, indicating a higher temperature and relatively deoxidized oceanic depositional environment. After the main exhalative stage, hydrothermal activity was superimposed to the sulfide mineralization. | 8 | Factors Affecting the Performance of Single-Chamber Soil Microbial Fuel Cells for Power Generation显示文摘There is limited information about the factors that affect the power generation of single-chamber microbial fuel cells(MFCs) using soil organic matter as a fuel source. We examined the effect of soil and water depths, and temperature on the performance of soil MFCs with anode being embedded in the flooded soil and cathode in the overlaying water. Results showed that the MFC with 5 cm deep soil and 3 cm overlaying water exhibited the highest open circuit voltage of 562 mV and a power density of 0.72 mW m-2. The ohmic resistance increased with more soil and water. The polarization resistance of cathode increased with more soil while that of anode increased with more water. During the 30 d operation, the cell voltage positively correlated with temperature and reached a maximum of 162 mV with a 500 ? external load. After the operation, the bacterial 16S rRNA gene from the soil and anode was sequenced. The bacteria in the soil were more diverse than those adhere to the anode where the bacteria were mainly affiliated to Escherichia coli and Deltaproteobacteria. Aggregates of a-synuclein,consisting of higher-level oligomers and insoluble fibrils,have been observed in many chronic neurological diseases and are implicated in neurotoxicity and neurodegeneration.a-Synuclein has recently been shown to aggregate following acute ischemic stroke, exacerbating neuronal damage.Propofol is an intravenous anesthetic that is commonly used during intravascular embolectomy following acute ischemic stroke. While propofol has demonstrated neuroprotective properties following brain injury, the mechanism of protection in the setting of ischemic stroke is unclear. In this study, propofol administration significantly reduced the neurotoxic aggregation of a-synuclein, decreased the infarct area, and attenuated the neurological deficits after ischemic stroke in a mouse model. | 13 | CO_2 gas emplacement age in the Songliao Basin: Insight from volcanic quartz ^(40)Ar-^(39)Ar stepwise crushing显示文摘Due to a lack of suitable minerals, the gas/oil emplacement ages have never been accurately obtained before. CH4-CO2-saline- bearing secondary inclusions are found in quartz from the volcanic rocks of the Yingcheng Formation, the container rocks of the deep CO2 gas reservoir in the Songliao Basin. The inclusion fluid was trapped into microcracks in quartz during the gas em- placement and accumulation, providing an optimal target for the 40Ar-39Ar stepwise crushing technique to determine the CO2 gas emplacement age. 40Ar-39Ar dating results of a quartz sample by stepwise crushing yield a highly linear-regression isochron with an age of 78.4±1.3 Ma, indicating that the accumulation of the deep CO2 gas reservoir in the Songliao Basin occurred in the late Cretaceous. | 14 | Monoclonal antibody-based serological methods for maize chlorotic mottle virus detection in China显示文摘Maize chlorotic mottle virus (MCMV) infects maize plants and causes significant losses in corn production worldwide. In this study, purified MCMV particles were used as the immunogen to produce monoclonal antibodies (MAbs) and polyclonal antibodies (PAbs). Four murine MAbs (4B8, 8C11, 6F4, and 9G1) against MCMV were obtained through the hybridoma technology. The triple antibody sandwich enzyme-linked immunosorbent assay (TAS-ELISA), dot-immunobinding assay (DIBA), and immunocapture reverse transcription-polymerase chain reaction (IC-RT-PCR) using the MAb 4B8 were then developed for sensitive, specific, and rapid detection of MCMV in fields. MCMV could be detected in infected leaf crude extracts at dilutions of 1:327 680, 1:64000, and 1:3276800 (w/v, g/ml) by TAS-ELISA, DIBA, and IC-RT-PCR, respectively. One hundred and sixty-one maize field samples showing virus-like symptoms and sixty-nine symptomless maize field samples from ten different provinces of China were collected and screened for the presence of MCMV using the established serological methods. A phylogenetic tree was constructed based on the full length CP genes and Chinese MCMV isolates formed one branch with Thailand isolates. | 15 | Joint contractures in severe burn patients with early rehabilitation intervention in one of the largest burn intensive care unit in China:a descriptive analysis显示文摘Background:Joint contracture is the major clinical complication in burn patients,especially,the severe burn patients.This study aimed to investigate the number and severity of joint contractures in patients with burns affecting greater than or equal to 50%of the total body surface area(TBSA)undergoing early rehabilitation in a burn intensive care unit(BICU).Methods:We analyzed burn patients with burns affecting greater than or equal to 50%of the TBSA admitted to a BICU who received early rehabilitation within 7 days post-injury from January 2011 to December 2015.Demographic and medical information was collected.The range of motion(ROM)of different joints was measured 1 month post-admission.Spearman’s correlation coefficient and logistic regression analysis was used to determine predictors of the presence and severity of contractures.Result:The average affected TBSA of the included burn patients was 67.4%,and the average length of stay in the BICU was 46.2±28.8 days.One hundred and one of 108 burn patients(93.5%)developed at least one joint contracture.The ROM in 67.9%of the affected joints was mildly limited.The majority of contractures in severe burn patients were mild(37.7%)or moderate(33.2%).The wrist was the most commonly affected joint(18.2%),followed by the shoulder,ankle,hip,knee,and elbow.A predictor of the presence of contractures was the length of hospital stay(p=0.049).The severe contracture was related to the area of full-thickness burns,the strict bed rest time,and the duration of rehabilitation in BICU.The length of rehabilitation stay(days)in patients with moderate contracture is 54.5%longer than that in severe contracture(p=0.024)Conclusion:During the long stay in BICU,the length of rehabilitation stay in a BICU could decrease the severity of contractures from severe to moderate in the patients with equal to 50%of the TBSA.Hence,this research reveals the important role of early rehabilitation interventions in severe burn patients. | Jianglin Tan Jian Chen Junyi Zhou Huapei Song Huan Deng Ming Ao Gaoxing Luo Jun Wu | 2019 | Burns & Trauma2019,7,1: | 9 |
| 16 | Intravenous tirofiban therapy for patients with capsular warning syndrome显示文摘Background Capsular warning syndrome(CWS)is defined as recurrent episodes of transient ischaemic attacks≥3 times during a short time frame.There is no effective therapy to stop these attacks.We,herein,report our experience of using intravenous tirofiban to treat CWS.Methods All patients with CWS in our hospital from January 2013 to September 2017 were reviewed.Patients in tirofiban group(T-group)were treated by intravenous tirofiban at 0.4μg/kg/min for 30 min followed by 0.1-0.15μg/kg/min infusion.Other treatments(non-T-group)included thrombolytic,oral antiplatelet agents and anticoagulant.Intracerebral haemorrhage(ICH),systematic bleeding,new attacks after treatment,National Institutes of Health Stroke Scale(NIHSS)scores at 24 hours and modified Rankin Scales(mRSs)at 3 months were recorded.Descriptive statistics were used for analysis.results Of 23 patients qualified(15 in T-group,8 in non-T-group),the duration of symptoms ranged from 2 to 100 min before treatments.After treatment,in T-group,four patients(26.7%)had recurrent attacks,and NIHSS scores were 0 in 11 patients(73.3%)at 24 hours.All patients reached a favourable outcome(mRS≤2) at 3 months.In non-T-group,five patients(62.5%)had new attacks.NIHSS scores were 0 in two patients(25%)at 24 hours.At 3 months,seven patients(87.5%)reached a favourable outcome.Neither ICH nor systematic bleeding or thrombocytopaenia occurred in both groups of patients.Conclusions Intravenous tirofiban can be a potentially effective and safe therapy to stop early symptomatic fluctuations and shorten the duration of functional deficits in patients with CWS. | Wei Li Ya Wu Xiao-Shu Li Cheng-Chun Liu Shu-Han Huang Chun-Rong Liang Huan Wang Li-Li Zhang Zhi-Qiang Xu Yan-Jiang Wang Meng Zhang | 2019 | Stroke & Vascular Neurology2019,4,1: | 9 |

| 20 | Regulating Effects of Novel CpG Chitosan-nanoparticles on Immune Responses of Mice to Porcine Paratyphoid Vaccines显示文摘Objective To study the regulating effects of a novel CpG oligodeoxynuleotide and the synergistic effect of chitosan-nanoparticles (CNP) with CpG on immune responses of mice, which were used to develop a novel immunoadjuvant to boost immune response to conventional vaccines. Methods A novel CpG ODN containing 11 CpG motifs was synthesized and its bioactivities to stimulate the proliferation of lymphocytes of pig in vitro were detected. Then it was entrapped with CNP prepared in our laboratory by the method of ionic cross linkage, and immunized Kunming mice were co-inoculated with paratyphoid vaccine. The peripheral blood was collected weekly from the tail vein of inoculated mice to detect the contents of IgG, IgA, IgM, and specific antibody against salmonella as well as the levels of interleukin-2 (IL2), IL-4, and IL-6 by SABC-ELISA assay. The numbers of leucocytes, monocytes, granuloytes, and lymphocytes were calculated separately using the routine method. The experimental mice were orally challenged with virulent salmonella 35 days after inoculation. Results This CpG ODN could remarkably provoke the proliferation of lymphocytes of pig in vitro in contrast with the control (P<0.05). Compared with those of the control, immunoglobulins, including IgG, IgA, IgM, and specific antibodies to paratyphoid vaccine, increased significantly in sera from the CpG or CpG-CNP-vaccinated mice (P<0.05). IL-2, IL-4, and IL-6 increased remarkably in sera from immunized mice (P<0.05). The leucocytes, monocytes, granuloytes, and lymphocytes of the mice immunized with CpG or CpG-CNP were also increased in number (P<0.05). After the challenge, these immunity values were elevated in the mice vaccinated with CpG or CpG-CNP. The immunized mice all survived, while the control mice fell ill with evident lesions with diffuse hemorrhage in stomach, small intestine, and peritoneum.
