| 1 | Hi-TOM: a platform for high-throughput tracking of mutations induced by CRISPR/Cas systems显示文摘The CRISPR/Cas system has been extensively applied to make precise genetic modifications in various organisms. Despite its importance and widespread use, large-scale mutation screening remains time-consuming, labour-intensive and costly. Here, we developed Hi-TOM(available at http://www.hi-tom.net/hi-tom/), an online tool to track the mutations with precise percentage for multiple samples and multiple target sites. We also described a corresponding next-generation sequencing(NGS) library construction strategy by fixing the bridge sequences and barcoding primers. Analysis of the samples from rice, hexaploid wheat and human cells reveals that the Hi-TOM tool has high reliability and sensitivity in tracking various mutations, especially complex chimeric mutations frequently induced by genome editing. Hi-TOM does not require special design of barcode primers,cumbersome parameter configuration or additional data analysis. Pre-treated shark skin was used as casting template to prepare a flexible female die of silicone rubber by soft die formation. A waterborne epoxy resin was then used to graft long-chains of drag reduction agent and prepare a synthetic drag reduction shark skin with nano-long chain drag reduction interface and lifelike micro-grooves. Replication precision analysis shows that this technology could replicate the complicated three-dimensional morphology of a biological drag reduction surface with high precision. | 5 | Mercury oxidation and adsorption characteristics of potassium permanganate modified lignite semi-coke显示文摘The adsorption characteristics of virgin and potassium permanganate modified lignite semi-coke(SC)for gaseous Hg0 were investigated in an attempt to produce more effective and lower price adsorbents for the control of elemental mercury emission.Brunauer-EmmettTeller(BET)measurements,X-ray powder diffraction(XRD)and X-ray photoelectron spectroscopy(XPS)were used to analyze the surface physical and chemical properties of SC,Mn-SC and Mn-H-SC before and after mercury adsorption.The results indicated that potassium permanganate modification had significant influence on the properties of semi-coke,such as the specific surface area,pore structure and surface chemical functional groups.The mercury adsorption efficiency of modified semi-coke was lower than that of SC at low temperature,but much higher at high temperature.Amorphous Mn7+,Mn6+ and Mn 4+ on the surface of Mn-SC and Mn-H-SC were the active sites for oxidation and adsorption of gaseous Hg0,which oxidized the elemental mercury into Hg2+ and captured it.Thermal treatment reduced the average oxidation degree of Mnx+ on the surface of Mn-SC from 3.80 to 3.46.However,due to the formation of amorphous MnOx,the surface oxidation active sites for gaseous Hg0 increased,which gave Mn-H-SC higher mercury adsorption efficiency than that of Mn-SC at high temperature. | Huawei Zhang Jitao Chen Peng Liang Li Wang | 2012 | Journal of Environmental Sciences2012,24,12: | 12 |

| 9 | Effectiveness and safety of Ginkgo biloba extract(GBE50)in the treatment of dizziness caused by cerebral arteriosclerosis:a multi-center,double-blind,randomized controlled trial显示文摘OBJECTIVE:To evaluate the effectiveness and safety of Ginkgo biloba extract(GBE50)in the treatment of dizziness caused by cerebral arteriosclerosis.METHODS:This was a multi-center,double-blind,double-dummy,positive-controlled,parallel randomized controlled clinical trial with 1∶1 allocation.We recruited 404 patients with dizziness caused by cerebral arteriosclerosis(blood stasis symptom pattern)in 10 hospitals in China.GBE50 group received GBE50 and Naoxinqing tablet(NXQ)of mimetic agent,control group received NXQ and GBE50 of mimetic agent.The main outcome was Traditional Chinese Medicine(TCM)symptom pattern score of blood stasis after 6 weeks.The secondary outcomes were changes in the dizziness handicap inventory(DHI)score,vertigo visual analogue scale(VAS)score,the university of California vertigo questionnaire(UCLA-DQ)score and single-item symptom score of TCM from baseline to 2,4 and 6 weeks.Safety indicators included the incidence of adverse events,severe adverse events and laboratory examination including blood routine,liver function,renal function,and so forth.RESULTS:The total effective rate of TCM symptom pattern score in the GBE50 group after 6 weeks of treatment was higher than that in the control group,the difference in rate was statistically significant(92.67%vs 83.07%,P=0.004).Compared with the control group,there was no difference in the incidence of adverse reactions(9.95%vs 14.85%,P=0.136).CONCLUSION:The treatment of dizziness caused by cerebral arteriosclerosis with GBE50 is effective,safe and reliable. | SHA Rina TANG Lu DU Yawei WU Shengxian SHI Huawei ZOU Hongxin ZHANG Xuran DONG Xinglu ZHOU Li | 2022 | Journal of Traditional Chinese Medicine2022,42,1: | 5 |

| 15 | Ambient TSP concentration and dustfall variation in Urumqi,China显示文摘Total suspended particulate(TSP, particle diameter≤100 μm) was the dominant air pollutant and significantly influenced local air quality. In this paper, we investigated the interannual and seasonal variations of TSP and dustfall of the atmosphere over Urumqi, the capital city of Xinjiang Uygur autonomous region, northwestern China, basing on environmental monitoring records and meteorological data from 1986–2012. The results showed that during the study period, annual average TSP concentration decreased from 716 to 260 μg/m3, with an average level of 422.9 μg/m3, while dustfall intensity reduced from 350.4 to 166.6 t/(km2?a), with an average level of 259 t/(km2?a). Over 50% of the annual pollution days were induced mainly by TSP. Spring and winter had relatively higher dustfall intensities, and dense traffic and residential areas had the highest dustfall intensities in the Urumqi metropolitan area. With an annual average precipitation of less than 300 mm, atmospheric particulates in Urumqi could be hardly removed through wet deposition. During spring and summertime prevailing winds from northwest and northeast could carry aeolian dust particles from sandy deserts to Urumqi. | 19 | Astaxanthine attenuates cisplatin ototoxicity in vitro and protects against cisplatin-induced hearing loss in vivo显示文摘Astaxanthine(AST) has important biological activities including antioxidant and antiinflammatory effects that could alleviate neurological and heart diseases, but its role in the prevention of cisplatin-induced hearing loss(CIHL) is not yet well understood. In our study, a steady interaction between AST and the E3 ligase adapter Kelch-like ECH-associated protein 1, a predominant repressor of nuclear factor erythroid 2-related factor 2(NRF2), was performed and tested via computer molecular docking and dynamics. AST protected against cisplatin-induced ototoxicity via NRF2 mediated pathwayusing quantitative PCR and Western blotting. The levels of reactive oxygen species(ROS) and mitochondrial membrane potential revealed that AST reduced ROS overexpression and mitochondrial dysfunction.Moreover, AST exerted anti-apoptosis effects in mouse cochlear explants using immunofluorescence staining and HEI-OC1 cell lines using quantitative PCR and Western blotting. Finally, AST combined with poloxamer was injected into the middle ear through the tympanum, and the protection against CIHL was evaluated using the acoustic brain stem test and immunofluorescent staining in adult mice. Our results suggest that AST reduced ROS overexpression, mitochondrial dysfunction, and apoptosis via NRF2-mediated pathway in cisplatin-exposed HEI-OC1 cell lines and mouse cochlear explants, finally promoting cell survival.
