| 1 | Easily manageable prognostic factors in 152 Chinese elderly acute myeloid leukemia patients:a single-center retrospective study显示文摘We retrospectively investigated the prognostic factors of acute myeloid leukemia(AML) in 152 Chinese patients with de novo AML who were older than 60 years of age and who received treatment at our hospital.Log-rank test showed that 6 parameters including older age,higher white blood cell(WBC) counts,lactate dehydrogenase(LDH)and bone marrow(BM) blasts at diagnosis,unfavorable risk cytogenetics,and non-mutated CEBPα were significant adverse prognostic factors of overall survival(OS) for elderly AML patients(P = 0.0013,0.0358,0.0132,0.0242,0.0236 and 0.0130,respectively).Moreover,older age and higher LDH were significant adverse predictors for relapse-free survival(RFS)(P = 0.0447 and 0.0470,respectively).Univariate analysis revealed similar results for OS to those of the log-rank test and only higher LDH at diagnosis was a significant adverse predictor for RFS(P = 0.028,HR:1.979,95%CI:1.075-3.644).In multivariate analysis,we identified 2 trends towards independent prognostic factors for OS,including BM blasts at diagnosis(P = 0.057,HR:1.676,95%CI:0.984-2.854)and mutation status of CEBPα(P = 0.064,HR:4.173,95%CI:0.918-18.966).Our data indicated that older age,gender and a previous history of hematologic diseases resulted in lower complete remission rate(P = 0.012,0.051 and 0.086,respectively).We further developed an easy scoring system for predicting prognosis and response to induction therapy in older AML patients.Patients who had lower scores showed significantly longer OS and RFS(P = 0.0006 and 0.1001,respectively) and higher CR rate(P = 0.014).Our research is limited by its retrospective nature and the results from our study need to be further validated by prospective randomized clinical trials. | Jiadai Xu Tingmei Chen Yun Liu Huayuan Zhu Wei Wu WenYi Shen Bei Xu Sixuan Qian Jianyong Li Peng Liu | 2014 | The Journal of Biomedical Research2014,28,5: | 7 |
| 2 | Highly sensitive flexible capacitive pressure sensor with a broad linear response range and finite element analysis of micro-array electrode显示文摘With the continuous development of wearable electronics,health care and smart terminals,highly performance flexible pressure sensors present a huge application prospect.In this study,by introducing the micro-array structured electrodes and dielectric layers with high dielectric constant,capacitive pressure sensor fabricated with a brand new preparation strategy and highly sensitive is proposed.The prepared micro-array structure is the basis for sensors with high sensitivity.Besides,the contact area between the two electrodes changes from linear to planar with the increased loading,which result in a wider linear responding range.In addition,by introducing ceramic dielectric material-barium titanate(BT)fillers into the dielectric layer to increase it’s the dielectric constant,the sensitivity of the sensor shows two-fold increase.Moreover,the sensitivity gradients can be tuned by changing the loading contents of BT particles.Hence,compared with parallel board capacitive sensors with ordinary dielectric layer,these sensors exhibit excellent performance as follow,high sensitivity(up to 4.9 kPa^(-1))under low pressure range(0-2500 Pa),low detection limit(<1.7 Pa),short response time(<50 ms),a stable response over 5000 loading-unloading cycles,bending stability and an adjustable sensitivity.Further,the flexible pressure sensor can detect the pressure of the water droplets and monitor human movement behavior.With the facile design and excellent comprehensive properties,the flexible pressure sensor provides a new approach to improve the sensitivity and shows a broad application prospects in the wearable electronics,health care and smart terminals. | Longquan Ma Xuecheng Yu Yuanyuan Yang Yougen Hu Xinyu Zhang Huayuan Li Xing Ouyang Pengli Zhu Rong Sun Ching-ping Wong | 2020 | Journal of Materiomics2020,6,2: | 4 |
| 3 | Effect of different twirling and rotating acupuncture manipulation techniques on the blood flow perfusion at acupoints显示文摘OBJECTIVE: To analyze the effect of different twirling and rotating acupuncture manipulation techniques on the blood flow perfusion at acupoints to provide a reference for the study of acupoint specificity and the quantification and effectiveness of acupuncture methods. METHODS: Twenty healthy male or female college students each received four different acupuncture manipulation techniques, including simple acupuncture, uniform reinforcing-reducing needling, twirling reinforcing needling, and twisting reducing needling. The self-control method was applied. Acupuncture was performed by an acupuncture manipulation simulator at Neiguan (PC 6) and Zusanli (ST 36). The process of twirling and rotating was divided into seven timepoints. The PeriCam Perfusion Speckle Imager (PSI) System was used to collect the blood flow perfusion data at each acupoint. The specificity of twirling and rotating acupuncture manipulation was analyzed based on changes in the curve, video, and numerical blood flow perfusion data at each timepoint. RESULTS: There were two peaks in the blood flow perfusion curve of twirling and rotating acupuncture manipulation;one appeared after 3 min of needle retention, and the other appeared 3 min after needle removal. The blood flow perfusion parameters showed that the greatest differences between the four manipulation techniques occurred after 5 and 10 min of needle retention. The specificity of various manipulation techniques was most obvious at these two timepoints. There were significant differences between the four manipulation groups in the blood flow perfusion and the relative change rates of blood flow perfusion at each timepoint. CONCLUSION: Laser speckle imaging enables the real-time, non-invasive, rapid, and accurate collection of blood flow perfusion data during acupuncture. This imaging technique enables the easy attainment of various parameters such as visual images, two-dimensional curves, and data tables. At various timepoints, the four groups significantly differed regarding changes in blood flow perfusion and relative change rates of blood flow perfusion, which facilitated the differentiation of the four acupuncture manipulation methods. Based on this, further analysis could be conducted to study spatial distribution characteristics such as the influence area and flare area. | 5 | VvBBX44 and VvMYBA1 form a regulatory feedback loop to balance anthocyanin biosynthesis in grape显示文摘Anthocyanins are essential for the quality of perennial horticultural crops,such as grapes.In grapes,ELONGATED HYPOCOTYL 5(HY5)and MYBA1 are two critical transcription factors that regulate anthocyanin biosynthesis.Our previous work has shown that Vitis vinifera B-box protein 44(VvBBX44)inhibits anthocyanin synthesis and represses VvHY5 expression in grape calli.However,the regulatory mechanism underlying this regulation was unclear.In this study,we found that loss of VvBBX44 function resulted in increased anthocyanin accumulation in grapevine callus.VvBBX44 directly represses VvMYBA1,which activates VvBBX44.VvMYBA1,but not VvBBX44,directly modulates the expression of grape UDP flavonoid 3-O-glucosyltransferase(VvUFGT).We demonstrated that VvBBX44 represses the transcriptional activation of VvUFGT and VvBBX44 induced by VvMYBA1.However,VvBBX44 and VvMYBA1 did not physically interact in yeast.The application of exogenous anthocyanin stimulated VvBBX44 expression in grapevine suspension cells and tobacco leaves.These findings suggest that VvBBX44 and VvMYBA1 form a transcriptional feedback loop to prevent overaccumulation of anthocyanin and reduce metabolic costs.Our work sheds light on the complex regulatory network that controls anthocyanin biosynthesis in grapevine. | Wenwen Liu Huayuan Mu Ling Yuan Yang Li Yuting Li Shenchang Li Chong Ren Wei Duan Peige Fan Zhanwu Dai Yongfeng Zhou Zhenchang Liang Shaohua Li Lijun Wang | 2023 | Horticulture Research2023,10,10: | 0 |
| 6 | The class B heat shock factor HSFB1 regulates heat tolerance in grapevine显示文摘Grape is a widely cultivated crop with high economic value.Most cultivars derived from mild or cooler climates may not withstand increasing heat stress.Therefore,dissecting the mechanisms of heat tolerance in grapes is of particular significance.Here,we performed comparative transcriptome analysis of Vitis davidii‘Tangwei’(heat tolerant)and Vitis vinifera‘Jingxiu’(heat sensitive)grapevines after exposure to 25°C,40°C,or 45°C for 2 h.More differentially expressed genes(DEGs)were detected in‘Tangwei’than in‘Jingxiu’in response to heat stress,and the number of DEGs increased with increasing treatment temperatures.We identified a class B Heat Shock Factor,HSFB1,which was significantly upregulated in‘Tangwei’,but not in‘Jingxiu’,at high temperature.VdHSFB1 from‘Tangwei’and VvHSFB1 from‘Jingxiu’differ in only one amino acid,and both showed similar transcriptional repression activities.Overexpression and RNA interference of HSFB1 in grape indicated that HSFB1 positively regulates the heat tolerance.Moreover,the heat tolerance of HSFB1-overexpressing plants was positively correlated to HSFB1 expression level.The activity of the VdHSFB1 promoter is higher than that of VvHSFB1 under both normal and high temperatures.Promoter analysis showed that more TATA-box and AT∼TATA-box cis-elements are present in the VdHSFB1 promoter than the VvHSFB1 promoter.The promoter sequence variations between VdHSFB1 and VvHSFB1 likely determine the HSFB1 expression levels that inf luence heat tolerance of the two grape germplasms with contrasting thermotolerance.Collectively,we validated the role of HSFB1 in heat tolerance,and the knowledge gained will advance our ability to breed heat-tolerant grape cultivars. | Haiyang Chen Xinna Liu Shenchang Li Ling Yuan Huayuan Mu Yi Wang Yang Li Wei Duan Peige Fan Zhenchang Liang Lijun Wang | 2023 | Horticulture Research2023,10,3: | 0 |
| 7 | Phase Ⅰ study of CBM.CD19 chimeric antigen receptor T cell in the treatment of refractory diffuse large B-cell lymphoma in Chinese patients显示文摘Anti-CD 19 chimeric antigen receptor(CAR)T cell therapy has shown impressive efficacy in treating B-cell malignancies.A single-center phase I dose-escalation study was conducted to evaluate the safety and efficacy of T cells transduced with CBM.CD19 CAR,a second-generation anti-CD19 CAR bearing 4-1BB costimulatory molecule,for the treatment of patients with refractory diffuse large B-cell lymphoma(DLBCL).Ten heavily treated patients with refractory DLBCL were given CBM.CD19 CAR-T cell(C-CAR011)treatment.The overall response rate was 20%and 50%at 4 and 12 weeks after the infusion of C-CAR011,respectively,and the disease control rate was 60%at 12 weeks after infusion.Treatment-emergent adverse events occurred in all patients.The incidence of cytokine release syndrome in all grades and grade^3 was 90%and 0,respectively,which is consistent with the safety profile of axicabtagene ciloleucel and tisagenlecleucel.Neurotoxicity or other dose-limiting toxicides was not observed in any dose cohort of C-CAR011 therapy.Antitumor efficacy was apparent across dose cohorts.Therefore,C-CAR011 is a safe and effective therapeutic option for Chinese patients with refractory DLBCL,and further large-scale clinical trials are warranted. | Lei Fan Li Wang Lei Cao Huayuan Zhu Wei Xu Jianyong Li | 2022 | Frontiers of Medicine2022,16,2: | 0 |
| 8 | Overexpression of c-Myc-dependent heterogeneous nuclear ribonucleoprotein A1 promotes proliferation and inhibits apoptosis in NOTCH1-mutated chronic lymphocytic leukemia cells显示文摘Background: NOTCH1 mutation is an essential molecular biologic aberration in chronic lymphocytic leukemia (CLL). CLL patients withNOTCH1 mutation have shown an unfavorable survival and a poor response to chemoimmunotherapy. This study aims to present the mechanisms of adverse prognosis caused byNOTCH1 mutation from the perspective of the splicing factor heterogeneous nuclear ribonucleoprotein A1 (hnRNPA1).Methods: The microarray data in Gene Expression Omnibus datasets were analyzed by bioinformatics and the function of hnRNPA1 was checked by testing the proliferation and apoptosis of CLL-like cell lines. Afterward, quantitative reverse transcription-polymerase chain reaction and Western blotting were applied to explore the relationship among NOTCH1, c-Myc, and hnRNPA1.Results: RNA splicing was found to play a vital part inNOTCH1-mutated CLL cells;hence, hnRNPA1 was selected as the focus of this study. Higher expression of hnRNPA1 validated in primaryNOTCH1-mutated CLL samples could promote proliferation and inhibit apoptosis in CLL. The expression of hnRNPA1 increased when NOTCH1 signaling was activated by transfection with NOTCH1 intracellular domain (NICD)-overexpressed adenovirus vector and declined after NOTCH1 signaling was inhibited by NOTCH1-shRNA. Higher expression of c-Myc was observed in NICD-overexpressed cells and hnRNPA1 expression was downregulated after applying c-Myc inhibitor 10058-F4.
