| 1 | Long-term antiviral efficacy of entecavir and liver histology improvement in Chinese patients with hepatitis B virus-related cirrhosis显示文摘AIM: To evaluate the clinical outcomes of 240-wk treatment with entecavir(0.5 mg) in Chinese nucleosidenaive patients with cirrhosis.METHODS: A total of 204 nucleoside-naive patients with compensated(n = 96) or decompensated(n = 108) hepatitis B virus(HBV)-induced cirrhosis at the Department of Gastroenterology of the China-Japan Union Hospital(Jilin University, Changchun, China) who were treated with entecavir(0.5 mg) for 240 wk were enrolled in this study. Liver biopsy samples obtained from 38 patients prior to treatment(baseline) and at week 240 were evaluated by different independent histopathologists. Efficacy assessments included the proportions of patients who achieved an HBV DNA level < 500 copies/m L, the association of interleukin-28 B genetic variation with antivirus therapy, clinical outcomes, and histologic improvement. Changes in liver disease severity were analyzed, and liver histologic evaluation was performed in 38 patients with paired biopsies. Student t tests were used to compare the means of continuous variables between the groups, and the proportions of patients who achieved the endpoints were compared using the χ2 test.RESULTS: At week 240, 87.5% of the patients with compensated cirrhosis and 92.6% of the patients with decompensated cirrhosis achieved a HBV DNA level < 500 copies/m L. Three patients had genotypic entecavir resistance within the 240-wk period. No significant association was observed between virologic response and interleukin-28 genotype(CT, 88.2% vs CC, 90.6%). The proportion of patients with Child-Pughclass A disease was significantly increased at week 240(68%) from the baseline(47%; P < 0.01). The proportion of patients with Child-Pugh class B disease was significantly decreased at week 240(25%) from the baseline(39%; P = 0.02). In the patients with paired liver biopsies, the mean reduction in the Knodell necroinflammatory score from the baseline was 3.58 ± 1.03 points(7.11 ± 1.80 vs 3.53 ± 1.35, P < 0.01). | 3 | A hybrid dynamic programming-rule based algorithm for real-time energy optimization of plug-in hybrid electric bus显示文摘The optimization of the control strategy of a plug-in hybrid electric bus(PHEB) for the repeatedly driven bus route is a key technique to improve the fuel economy. The widely used rule-based(RB) control strategy is lacking in the global optimization property, while the global optimization algorithms have an unacceptable computation complexity for real-time application. Therefore, a novel hybrid dynamic programming-rule based(DPRB) algorithm is brought forward to solve the global energy optimization problem in a real-time controller of PHEB. Firstly, a control grid is built up for a given typical city bus route, according to the station locations and discrete levels of battery state of charge(SOC). Moreover, the decision variables for the energy optimization at each point of the control grid might be deduced from an off-line dynamic programming(DP) with the historical running information of the driving cycle. Meanwhile, the genetic algorithm(GA) is adopted to replace the quantization process of DP permissible control set to reduce the computation burden. Secondly, with the optimized decision variables as control parameters according to the position and battery SOC of a PHEB, a RB control is used as an implementable controller for the energy management. Simulation results demonstrate that the proposed DPRB might distribute electric energy more reasonably throughout the bus route, compared with the optimized RB. | 5 | Sophocarpine attenuates liver fibrosis by inhibiting the TLR4 signaling pathway in rats显示文摘AIM:To explore the effect of sophocarpine on experimental liver fibrosis and the potential mechanism involved.METHODS:Sophocarpine was injected intraperitoneally in two distinct rat hepatic fibrosis models induced either by dimethylnitrosamine or bile duct ligation.Masson’s trichrome staining,Sirius red staining and hepatic hydroxyproline level were used for collagen determination.Primary hepatic stellate cells(HSCs)were isolated and treated with different concentrations of sophocarpine.Real-time reverse transcription-polymerase chain reaction was used to detect the mRNA levels of fibrotic markers and cytokines.The expression of pathway proteins was measured by Western blot.The Cell Counting Kit-8 test was used to detect the proliferation rate of activated HSCs treated with a gradient concentration of sophocarpine.RESULTS:Sophocarpine decreased serum levels of aminotransferases and total bilirubin in rats under chronic insult.Moreover,administration of sophocarpine suppressed extracellular matrix deposition and prevented the development of hepatic fibrosis.Furthermore,sophocarpine inhibited the expression ofα-smooth muscle actin(SMA),interleukin(IL)-6,transforming growth factor-β1(TGF-β1),Toll-like receptor 4(TLR4),and extracellular-related kinase(ERK)in rats.Sophocarpine also down-regulated the mRNA expression ofα-SMA,collagenⅠ,collagenⅢ,TGF-β1,IL-6,tumor necrosis factor-αand monocyte chemoattractant protein-1,and decreased protein levels of TLR4,p-ERK,p-JNK,p-P38 and p-IKK in vitro after Lipopolysaccharide induction.In addition,sophocarpine inhibited the proliferation of HSCs accompanied by a decrease in the expression of Cyclin D1.The protein level of proliferating cell nuclear antigen was decreased in activated HSCs following a gradient concentration of sophocarpine.CONCLUSION:Sophocarpine can alleviate liver fibrosis mainly by inhibiting the TLR4 pathway.Sophocarpine may be a potential chemotherapeutic agent for chronic liver diseases. | Hui Qian Jian Shi Ting-Ting Fan Jiao Lv Si-Wen Chen Chun-Yan Song Zhi-Wu Zheng Wei-Fen Xie Yue-Xiang Chen | 2014 | World Journal of Gastroenterology2014,20,7: | 19 |

| 7 | Geothermal data analysis at the high-temperature hydrothermal area in Western Sichuan显示文摘The western Sichuan hydrothermal area is located at the northeastern margin of the eastern syntaxis of the Qinghai-Tibet Plateau, which is also the eastern end of the Mediterranean-Himalayan geothermal activity zone. There are 248 warm or hot springs in this area, and 11 have temperatures beyond the local boiling temperature. Most of these hot springs are distributed along the Jinshajiang, Dege-Xiangcheng, Ganzi-Litang, and Xianshuihe faults, forming a NW-SE hydrothermal belt. A geothermal analysis of this high-temperature hydrothermal area is an important basis for understanding the deep geodynamic process of the eastern syntaxis of the Qinghai-Tibet Plateau. In addition, this study offers an a priori view to utilize geothermal resources, which is important in both scientific research and application. We use gravity, magnetic, seismic, and helium isotope data to analyze the crust-mantle heat flow ratio and deep geothermal structure. The results show that the background terrestrial heat flow descends from southwest to northeast. The crustal heat ratio is not more than 60%. The high temperature hydrothermal active is related to crustal dynamics processes. Along the Batang-Litang-Kangding line, the Moho depth increases eastward, which is consistent with the changing Qc/Qm(crustal/mantle heat flow) ratio trend. The geoid in the hydrothermal zone is 4–6 km higher than the surroundings, forming a local 'platform'. The NW-SE striking local tensile stress zone and uplift structure in the upper and middle crust corresponds with the surface hydrothermal active zone. There is an average Curie Point Depth(CPD) of 19.5–22.5 km in Batang, Litang, and Kangding. The local shear-wave(S-wave) velocity is relatively low in the middle and lower crust. The S-wave shows a low velocity trap(Vs<3.2 km s.1) at 15–30 km, which is considered a high-temperature partial melting magma, the crustal source of the hydrothermal active zone. We conclude that the hydrothermal system in this area can be divided into Batang-type and Kangding-type, both of which rely on a crustal heating cycle of atmospheric precipitation and surface water along the fracture zone. | 8 | Dual-targeting and microenvironment-responsive micelles as a gene delivery system to improve the sensitivity of glioma to radiotherapy显示文摘Dbait is a small double-stranded DNA molecule that has been utilized as a radiosensitizer to enhance the sensitivity of glioma to radiotherapy(RT). However, there is no effective drug delivery system to effectively overcome the blood–brain barrier(BBB). The aim of this study was to develop a gene delivery system by using the BBB and glioma dual-targeting and microenvironment-responsive micelles(ch-Kn(s-s)R8-An) to deliver Dbait into glioma for RT. Angiopep-2 can target the low-density lipoprotein receptor-related protein-1(LRP1) that is overexpressed on brain capillary endothelial cells(BCECs) and glioma cells. In particular, due to upregulated matrix metalloproteinase 2(MMP-2) in the tumor microenvironment, we utilized MMP-2-responsive peptides as the enzymatically degradable linkers to conjugate angiopep-2. The results showed that ch-Kn(s-s)R8-An micelles maintained a reasonable size(80–160 nm) with a moderate distribution and a decreased mean diameter from the cross-linking as well as exhibited low critical micelle concentration(CMC) with positive surface charge, ranging from 15 to40 mV. The ch-K5(s-s)R8-An/pEGFP showed high gene transfection efficiency in vitro, improved uptake in glioma cells and good biocompatibility in vitro and in vivo. In addition, the combination of ch-K5(s-s)R8-An/Dbait with RT significantly inhibited the growth of U251 cells in vitro. The effect of silicon on the carbothermal reduction process has been investigated in detail. Compared with traditional reduction, the introduction of silicon can change the reaction path and further increase the conversion of silicon carbide at a lower temperature. It is considered that the assistant reduction consists of three steps: vaporizing and melting of silicon, formation of silicon monoxide, and synthesis of silicon carbide. | 11 | Discrete element modeling on the crack evolution behavior of brittle sandstone containing three fissures under uniaxial compression显示文摘Based on experimental results of brittle, intact sandstone under uniaxial compression, the micro-parameters were firstly confirmed by adopting particle flow code(PFC2D). Then, the validation of the simulated models were cross checked with the experimental results of brittle sandstone containing three parallel fissures under uniaxial compression. The simulated results agreed very well with the experimental results, including the peak strength, peak axial strain, and ultimate failure mode. Using the same microparameters, the numerical models containing a new geometry of three fissures are constructed to investigate the fissure angle on the fracture mechanical behavior of brittle sandstone under uniaxial compression. The strength and deformation parameters of brittle sandstone containing new three fissures are dependent to the fissure angle. With the increase of the fissure angle, the elastic modulus, the crack damage threshold,and the peak strength of brittle sandstone containing three fissures firstly increase and secondly decrease. But the peak axial strain is nonlinearly related to the fissure angle. In the entire process of deformation, the crack initiation and propagation behavior of brittle sandstone containing three fissures under uniaxial compression are investigated with respect tothe fissure angle. Six different crack coalescence modes are identified for brittle sandstone containing three fissures under uniaxial compression. The influence of the fissure angle on the length of crack propagation and crack coalescence stress is evaluated. | 12 | Rapid generation of genetic diversity by multiplex CRISPR/Cas9 genome editing in rice显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated endonuclease 9(CRISPR/Cas9) system has emerged as a promising technology for specific genome editing in many species. Here we constructed one vector targeting eight agronomic genes in rice using the CRISPR/Cas9 multiplex genome editing system. By subsequent genetic transformation and DNA sequencing, we found that the eight target genes have high mutation efficiencies in the T_0 generation. Both heterozygous and homozygous mutations of all editing genes were obtained in T_0 plants. In addition, homozygous sextuple, septuple, and octuple mutants were identified. As the abundant genotypes in T_0 transgenic plants, various phenotypes related to the editing genes were observed. | 14 | Resequencing of 145 Landmark Cultivars Reveals Asymmetric Sub-genome Selection and Strong Founder Genotype Effects on Wheat Breeding in China显示文摘Controlled pedigrees and the multi-decade timescale of national crop plant breeding programs offer a unique experimental context for examining how selection affects plant genomes.More than 3000 wheat cultivars have been registered,released,and documented since 1949 in China.In this study,a set of 145 elite cultivars selected from historical points of wheat breeding in China were re-sequenced.A total of 43.75 Tb of sequence data were generated with an average read depth of 17.94x for each cultivar,and more than 60.92 million SNPs and 2.54 million InDels were captured,based on the Chinese Spring RefSeq genome v1.0.Seventy years of breeder-driven selection led to dramatic changes in grain yield and related phenotypes,with distinct genomic regions and phenotypes tar-geted by different breeders across the decades.There are very clear instances illustrating how introduced Italian and otherforeign germplasm was integrated into Chinese wheat programs and reshaped the genomic landscape of local modern cultivars.Importantly,the resequencing data also highlighted significant asymmetric breeding selec-tion among the three sub-genomes:this was evident in both the collinear blocks for homeologous chromosomes and among sets of three homeologous genes.Accumulation of more newly assembled genes in newer cultivars implied the potential value of these genes in breeding.Conserved and extended sharing of linkage disequilibrium(LD)blocks was highlighted among pedigree-related cultivars,in which fewer haplotype differences were detected.Fixation or replacement of haplotypes from founder genotypes after generations of breeding was related to their breeding value.Based on the haplotype frequency changes in LD blocks of pedigree-related cultivars,we propose a strategy for evaluating the breeding value of any given line on the basis of the accumulation(pyramiding)of bene-ficial haplotypes.Collectively,our study demonstrates the influence of 'founder genotypes' on the output of breeding efforts over many decades and also suggests that founder genotype perspectives are in fact more dy-namic when applied in the context of modern genomics-informed breeding. | Chenyang Hao Chengzhi Jiao Jian Hou Tian Li Hongxia Liu Yuquan Wang Jun Zheng Hong Liu Zhihong Bi Fengfeng Xu Jing Zhao Lin Ma Yamei Wang Uzma Majeed Xu Liu Rudi Appels Marco Maccaferri Roberto Tuberosa Hongfeng Lu Xueyong Zhang | 2020 | Molecular Plant2020,13,12: | 9 |

| 16 | Fabrication of kW-level chirped and tilted fiber Bragg gratings and filtering of stimulated Raman scattering in high-power CW oscillators显示文摘Suppression of stimulated Raman scattering(SRS)by means of chirped and tilted fiber Bragg gratings(CTFBGs)has become a key topic.However,research on high-power systems is still lacking due to two problems.Firstly,after the inscription,there are a large number of hydroxyl compounds and hydrogen molecules in CTFBGs that cause significant heating due to their strong infrared absorption.Secondly,CTFBGs can couple Stokes light from the core to the cladding and the coating,which causes serious heating in the coating of the CTFBG.Aimed at overcoming these bottlenecks,a process that combines constant-low-temperature and variable-high-temperature annealing is used to reduce the thermal slope of the CTFBG.Also,a segmented-corrosion cladding power stripping technology is used on the CTFBG to remove the Stokes light which is coupled to the cladding,which solves the problem of overheating in the coating of the CTFBG.Thereby,a CTFBG with both a kilowatt-level power-carrying load and the ability to suppress SRS in a fiber laser has been developed.Further,we establish a kW-level CW oscillator to test the CTFBG.Experimental results demonstrate that the power-carrying load of the CTFBG is close to 1 kW,the thermal slope is lower than 0.015 ℃/W,and the SRS suppression ratio is nearly 23 dB. | Kerong Jiao Jian Shu Hua Shen Zhiwen Guan Feiyan Yang Rihong Zhu | 2019 | High Power Laser Science and Engineering2019,7,2: | 9 |

| 19 | Status and development of high-power laser facilities at the NLHPLP显示文摘In this paper, we review the status of the multifunctional experimental platform at the National Laboratory of High Power Laser and Physics(NLHPLP). The platform, including the SG-II laser facility, SG-II 9th beam, SG-II upgrade(SG-II UP) facility, and SG-II 5 PW facility, is operational and available for interested scientists studying inertial confinement fusion(ICF) and a broad range of high-energy-density physics. These facilities can provide important experimental capabilities by combining different pulse widths of nanosecond, picosecond, and femtosecond scales. In addition, the SG-II UP facility, consisting of a single petawatt system and an eight-beam nanosecond system, is introduced including several laser technologies that have been developed to ensure the performance of the facility.
