| 1 | The PI3K/Akt/GSK-3β/ROS/eIF2B pathway promotes breast cancer growth and metastasis via suppression of NK cell cytotoxicity and tumor cell susceptibility显示文摘Objective: To examine the effect of pSer9-GSK-3β on breast cancer and to determine whether the underlying metabolic and immunological mechanism is associated with ROS/eIF2B and natural killer(NK) cells.Methods: We employed TWS119 to inactivate GSK-3β by phosphorylating Ser9 and explored its effect on breast cancer and NK cells. The expression of GSK-3β, natural killer group 2 member D(NKG2D) ligands, eIF2B was quantified by PCR and Western blot. We measured intracellular reactive oxygen species(ROS) and mitochondrial ROS using DCFH-DA and MitoSOX^(TM) probe,respectively, and conducted quantitative analysis of cellular respiration on 4T1 cells with mitochondrial respiratory chain complex Ⅰ/Ⅲ kits.Results: Our investigation revealed that TWS119 downregulated NKG2D ligands(H60 a and Rae1), suppressed the cytotoxicity of NK cells, and promoted the migration of 4T1 murine breast cancer cells. Nevertheless, LY290042, which attenuates p-GSK-3β formation by inhibiting the PI3K/Akt pathway, reversed these effects. We also found that higher expression of p Ser9-GSK-3β induced higher levels of ROS, and observed that abnormality of mitochondrial respiratory chain complex Ⅰ/Ⅲ function induced the dysfunction of GSK-3β-induced electron transport chain, naturally disturbing the ROS level. In addition, the expression of NOX3 and NOX4 was significantly up-regulated, which affected the generation of ROS and associated with the metastasis of breast cancer. Furthermore, we found that the expression of pSer535-eIF2B promoted the expression of NKG2D ligands(Mult-1 and Rae1) following by expression of pSer9-GSK-3β and generation of ROS.Conclusions: The PI3K/Akt/GSK-3β/ROS/eIF2B pathway could regulate NK cell activity and sensitivity of tumor cells to NK cells,which resulted in breast cancer growth and lung metastasis. | 3 | Human activities and elevational constraints restrict ranging patterns of snub-nosed monkeys in a mountainous refuge显示文摘Both natural conditions and anthropogenic factors affect the survivability,distribution,and population density of wildlife.To understand the extent and how these factors drive species distributions,a detailed description of animal movement patterns in natural habitats is needed.In this study,we used satellite telemetry to monitor elevational ranges favored by endangered golden snub-nosed monkeys(Rhinopithecus roxellana),in the Qinling Mountains,central China.We investigated the abundance and distribution of food resources through sampling vegetation quadrats at different elevations and sampled anthropogenic activities using field surveys.Our results indicated that although there was no significant variation in food resources between low-(<1500 m)and middle-elevations(1500–2200 m),monkeys were found most often in areas above 1500 m,where there was less anthropogenic development(e.g.houses and roads);however,monkeys rarely ranged above 2200 m and had limited food availability at this altitude.There was limited human disturbance at this elevation.We suggest that both human activity and ecological constraints(i.e.food resources)have considerable effects on elevational use of R.roxellana in the Qinling Mountains.This study highlights the critical roles these factors can play in shaping the vertical distribution of high-altitude primates.This research provides useful insights for habitat-based conservation plans in which human disturbance management and habitat restoration should be prioritized. | Pengzhen HUANG Kun BIAN Zhipang HUANG Qi Li Derek WDUNN Gu FANG Jiahui LIU Mengyao WANG Xianfeng YANG Ruliang PAN Cunlao GAO Kaichuang SI Baoguo LI Xiaoguang QI | 2021 | Integrative Zoology2021,16,2: | 3 |

| 5 | Extra low friction coefficient caused by the formation of a solid-like layer:A new lubrication mechanism found through molecular simulation of the lubrication of MoS_2 nanoslits显示文摘Monolayer molybdenum disulfide(MoS2) is a novel two-dimensional material that exhibits potential application in lubrication technology. In this work, molecular dynamics was used to investigate the lubrication behaviour of different polar fluid molecules(i.e., water, methanol and decane) confined in monolayer Mo S2 nanoslits. The pore width effect(i.e., 1.2, 1.6 and 2.0 nm) was also evaluated. Results revealed that decane molecules exhibited good lubricating performance compared to the other two kinds of molecules. The friction coefficient followed the order of decane b methanol b water, and decreased evidently as the slit width increased, except for decane. Analysis of the spatial distribution and mobility of different confined fluid molecules showed that a solid-like layer was formed near the slit wall. | 6 | Myristica fragrans promotes ABCA1 expression and cholesterol efflux in THP-1-derived macrophages显示文摘Myristica fragrans is a traditional herbal medicine and has been shown to alleviate the development of atherosclerosis.However,the anti-atherogenic mechanisms of M.fragrans are still to be addressed.In this study,we explored the effect of M.fragrans on lipid metabolism and inflammation and its mechanisms in THP-1-derived macrophages.The quantitative polymerase chain reaction and western blot analysis results showed that M.fragrans promotes cholesterol efflux from THP-1-derived macrophages and reduces intracellular total cholesterol,cholesterol ester,and free cholesterol contents in a dose-and a time-dependent manner.Further study found that liver X receptor alpha(LXRα)antagonist GGPP significantly blocked the upregulation of ABCA1 expression with M.fragrans treatment.In addition,chromatin immunoprecipitation assay confirmed that GATA binding protein 3(GATA3)can bind to the LXRαpromoter,and inhibition of GATA3 led to the downregulation of LXRαand ATP-binding cassette subfamily A member 1 expression.Furthermore,M.fragrans reduced lipid accumulation,followed by decreasing tumor necrosis factor-α,interleukin(IL)-6,and IL-1βand increasing IL-10 produced by THP-1-derived macrophages.Therefore,M.fragrans is identified as a valuable therapeutic medicine for atherosclerotic cardiovascular disease. | Shangming Liu Jiahui Gao Linhao He Zhenwang Zhao Gang Wang Jin Zou Li Zhou Xiangjun Wan Shilin Tang Chaoke Tang | 2021 | Acta Biochimica et Biophysica Sinica2021,53,1: | 2 |

| 16 | Biomass accumulation and organic carbon stocks of Kandelia obovata mangrove vegetation under different simulated sea levels显示文摘Mangrove forests are vulnerably threatened by sea level rise(SLR).Vegetation organic carbon(OC)stocks are important for mangrove ecosystem carbon cycle.It is critical to understand how SLR affects vegetation OC stocks for evaluating mangrove blue carbon budget and global climate change.In this study,biomass accumulation and OC stocks of mangrove vegetation were compared among three 10 year-old Kandelia obovata(a common species in China)mangrove forests under three intertidal elevations through species-specific allometric equations.This study simulated mangrove forests with SLR values of 0 cm,40 cm and 80 cm,respectively,representing for the current,future~100 a and future~200 a SLR of mangrove forests along the Jiulong River Estuary,China.SLR directly decreased mangrove individual density and inhibited the growth of mangrove vegetation.The total vegetation biomasses were(12.86±0.95)kg/m^2,(7.97±0.90)kg/m^2 and(3.89±0.63)kg/m^2 at Sites SLR 0 cm,SLR40 cm and SLR 80 cm,respectively.The total vegetation OC stock decreased by approximately 3.85 kg/m^2(in terms of C)from Site SLR 0 cm to Site SLR 80 cm.Significantly lower vegetation biomass and OC stock of various components(stem,branch,leaf and root)were found at Site SLR 80 cm.Annual increments of vegetation biomass and OC stock also decreased with SLR increase.Moreover,significant lower sedimentation rate was found at Site SLR 80 cm.These indicated that SLR will decrease mangrove vegetation biomass and OC stock,which may reduce global blue carbon sink by mangroves,exacerbate global warming and give positive feedback to SLR. | Jiahui Chen Min Gao Guangcheng Chen Heng Zhu Yong Ye | 2022 | Acta Oceanologica Sinica2022,41,8: | 1 |
| 17 | CCGD-ESCC: A Comprehensive Database for Genetic Variants Associated with Esophageal Squamous Cell Carcinoma in Chinese Population显示文摘Esophageal squamous-cell carcinoma(ESCC) is one of the most lethal malignancies in the world and occurs at particularly higher frequency in China. While several genome-wide association studies(GWAS) of germline variants and whole-genome or whole-exome sequencing studies of somatic mutations in ESCC have been published, there is no comprehensive database publically available for this cancer. Here, we developed the Chinese Cancer Genomic Database-Esophageal Squamous Cell Carcinoma(CCGD-ESCC) database, which contains the associations of 69,593 single nucleotide polymorphisms(SNPs) with ESCC risk in 2022 cases and 2039 controls, survival time of 1006 ESCC patients(survival GWAS) and gene expression(expression quantitative trait loci,eQTL) in 94 ESCC patients. Moreover, this database also provides the associations between8833 somatic mutations and survival time in 675 ESCC patients. Our user-friendly database is a resource useful for biologists and oncologists not only in identifying the associations of genetic variants or somatic mutations with the development and progression of ESCC but also in studying the underlying mechanisms for tumorigenesis of the cancer. | 19 | Extracellular RNAs-TLR3 signaling contributes to cognitive impairment after chronic neuropathic pain in mice显示文摘Chronic pain is often associated with cognitive decline,which could influence the quality of the patient’s life.Recent studies have suggested that Toll-like receptor 3(TLR3)is crucial for memory and learning.Nonetheless,the contribution of TLR3 to the pathogenesis of cognitive decline after chronic pain remains unclear.The level of TLR3 in hippocampal neurons increased in the chronic constriction injury(CCI)group than in the sham group in this study.Importantly,compared to the wild-type(WT)mice,TLR3 knockout(KO)mice and TLR3-specific neuronal knockdown mice both displayed improved cognitive function,reduced levels of inflammatory cytokines and neuronal apoptosis and attenuated injury to hippocampal neuroplasticity.Notably,extracellular RNAs(exRNAs),specifically double-stranded RNAs(dsRNAs),were increased in the sciatic nerve,serum,and hippocampus after CCI.The co-localization of dsRNA with TLR3 was also increased in hippocampal neurons.And the administration of poly(I:C),a dsRNA analog,elevated the levels of dsRNAs and TLR3 in the hippocampus,exacerbating hippocampus-dependent memory.In additon,the dsRNA/TLR3 inhibitor improved cognitive function after CCI.Together,our findings suggested that exRNAs,particularly dsRNAs,that were present in the condition of chronic neuropathic pain,activated TLR3,initiated downstream inflammatory and apoptotic signaling,caused damage to synaptic plasticity,and contributed to the etiology of cognitive impairment after chronic neuropathic pain. | Xueying Zhang Rui Gao Changteng Zhang Yi Teng Hai Chen Qi Li Changliang Liu Jiahui Wu Liuxing Wei Liyun Deng Lining Wu Shixin Ye-Lehmann Xiaobo Mao Jin Liu Tao Zhu Chan Chen | 2023 | Signal Transduction and Targeted Therapy2023,8,9: | 0 |
| 20 | Calcium carbonate promotes the formation and stability of soil macroaggregates in mining areas of China显示文摘We studied changes in the concentrations of aggregate-cementing agents after different reclamation times and with different fertilization regimes,as well as the formation mechanism of aggregates in reclaimed soil,to provide a theoretical basis for rapid reclamation of soil fertility in the subsidence area of coal mines in Shanxi Province,China.In this study,soil samples of 0–20 cm depth were collected from four fertilization treatments of a longterm experiment started in 2008:no fertilizer (CK),inorganic fertilizer (NPK),chicken manure compost (M),and50%inorganic fertilizer plus 50%chicken manure compost (MNPK).The concentrations of cementing agents and changes in soil aggregate size distribution and stability were analysed.The results showed that the formation of>2 mm aggregates,the aggregate mean weight diameter (MWD),and the proportion of>0.25 mm water-stable aggregates (WR_(0.25)) increased significantly after 6 and 11 years of reclamation.The concentration of organic cementing agents tended to increase with reclamation time,whereas free iron oxide (Fed) and free aluminium oxide(Ald) concentrations initially increased but then decreased.In general,the MNPK treatment signi?cantly increased the concentrations of organic cementing agents and CaCO_(3),and CaCO_(3) increased by 60.4%at 11 years after reclamation.Additionally,CaCO_(3) had the greatest effect on the stability of aggregates,promoting the formation of>0.25 mm aggregates and accounting for 54.4%of the variance in the proportion and stability of the aggregates.It was concluded that long-term reclamation is bene?cial for improving soil structure.The MNPK treatment was the most effective measure for increasing maize grain yield and concentration of organic cementing agents and CaCO_(3). | Junyu Xie Jianyong Gao Hanbing Cao Jiahui Li Xiang Wang Jie Zhang Huisheng Meng Jianping Hong Tingliang Li Minggang Xu | 2024 | Journal of Integrative Agriculture2024,23,3: | 0 |
