| 3 | Tenglong Buzhong granules(藤龙补中颗粒) inhibits the growth of SW620 human colon cancer in vivo显示文摘OBJECTIVE: To observe the anticancer effects of the granular preparation of Tenglong Buzhong decoction(藤龙补中汤,TBD), i.e Tenglong Buzhong granules(藤龙补中颗粒, TBG), in human SW620 colon cancer. METHODS: BALB/c nude mice were subcutaneously transplanted with SW620 cells, and treated with TBG(2.56 g/kg, once per day) and/or 5-Fu(104 mg/kg, once per week) for 21 d. Apoptosis, Caspase activities and cellular senescence were measured by commercial kits. The protein expression and phosphorylation were detected by Western blot or immunohistochemistry. RESULTS: TBG and 5-Fu inhibited tumor growth. The tumor inhibition rate of the TBG, 5-Fu, and TBG+5-Fu groups was 42.25%, 51.58%, and 76.08%, respectively. Combination of TBG and 5-Fu showed synergetic anticancer effects. TBG and 5-Fu induced apoptosis, activated caspase-3,-8, and-9, increased SMAC expression, inhibited XIAP expression. TBG induced cellular senescence, upregulated cyclin-dependent kinase inhibitor 1a(CDKN1a) and cyclin-dependent kinase inhibitor 2a(CDKN2a) expression, and inhibited phosphorylation of retinoblastoma-associated protein(RB) and expression of cyclin E1(CCNE1) and cyclindependent kinases(CDK) 2. TBG also inhibited angiogenesis accompanied by downregulation of vascular endothelial growth factor(VEGF) and hypoxiainducible factor-1α(HIF-1α). | 5 | Dedifferentiated human umbilical cord mesenchymal stem cell reprogramming of endogenous hSDF-1a expression participates in neural restoration in hypoxic-ischemic brain damagerats显示文摘The transplantation of human umbilical cord mesenchymal stem cells(hUC-MSCs)can promote hypoxic-ischemic brain damage(HIBD)nerve repair,but finding suitable seed cells to optimize transplantation and improve treatment efficiency is an urgent problem to be solved.In this study,we induced hUC-MSCs into dedifferentiated hUC-MSCs(De-hUC-MSCs),and the morphology,stem cell surface markers,proliferation and tri-directional differentiation ability of the De-hUC-MSCs and hUC-MSCs were detected.A whole-gene chip was utilized for genome cluster,gene ontology and KEGG pathway analyses of differentially expressed genes.De-hUC-MSCs were transplanted into HIBD rats,and behavioral experiments and immunofluorescence assays were used to assess the therapeutic effect.A lentivirus vector for human stromal cell-derived factor-1(hSDF-1a)was constructed,and the role of hSDF-1a in the neuroprotective effect and mechanism of De-hUC-MSCs was verified.De-hUC-MSCs displayed similar cell morphology,stem cell surface marker expression,cell proliferation and even three-dimensional differentiation ability as hUC-MSCs but exhibited greater treatment potential in vivo.The reprogramming mechanism of hSDF-1a participated in the dedifferentiation process.By successfully constructing a stable hSDF-1a cell line,we found that De-hUC-MSCs might participate in nerve repair through the hSDF-1a/CXCR4/PI3K/Akt pathway.De-hUC-MSCs reprogramming of endogenous hSDF-1a expression may mediate the hSDF-1a/CXCR4/PI3K/Akt pathway involved in nerve repair in HIBD rats. | Zhou Xiaoqin Liu Jia Dai Mengjie Gu Jialu Bi Yang Wang Yuting Hu Huajian Liu Bo Zhang Xiaojun Li Zhongyue Chen Jie Li Tingyu Zhan Xue | 2021 | Genes & Diseases2021,8,3: | 1 |

| 7 | Effect of Yanggan Jiedu Sanjie formula on human hepatocellular carcinoma Bel-7402 cells显示文摘OBJECTIVE: To observe the effect of Yanggan Jiedu Sanjie(YGJDSJ) formula on human hepatocellular carcinoma Bel-7402 cells.METHODS: Bel-7402 cells were treated with YGJDSJ. Cell proliferation was detected by cell counting kit-8 assay. Cell apoptosis was identified by Hoechst 33258 staining and flow cytometric analysis. Cell cycle distribution was quantified by flow cytometric analysis. Caspase activities were measured by commercial kit. Cell senescence was detected by senescence-associated β-galactosidase(SA-β-gal)staining. Protein expression and phosphorylation were identified by Western blot. Protein expression was knocked-down by siRNA.RESULTS: YGJDSJ inhibited proliferation of Bel-7402 cells in a dose-and time-dependent manner.YGJDSJ induced apoptosis and activated caspase-3, 8, and 9 in Bel-7402 cells. YGJDSJ-induced apoptosis was completely abrogated by a pan caspase inhibitor, Z-VAD-FMK. YGJDSJ also induced cell senescence, up-regulated cyclin-dependent kinase inhibitor 1 a(CDKN1 a) and CDKN2 a expression and down-regulated retinoblastoma protein(RB) phosphorylation in Bel-7402 cells. Specific knockdown of CDKN1 a and CDKN2 a significantly reduced YGJDSJ-induce cell senescence in Bel-7402 cells.CONCLUSION: YGJDSJ inhibited cell proliferation,induced caspase-dependent apoptosis and CDKN1 a/CDKN2 a-RB signalling mediated cell senescence in Bel-7402 cells. | 18 | Highly insulating phase of Bi_(2)O_(2)Se thin films with high electronic performance显示文摘Bi_(2)O_(2)Se is highly competitive as a candidate of next-generation high-performance semiconductors.Though dubbed as semiconductor,Bi_(2)O_(2)Se films exhibited high conductance,i.e.,metallic behavior,due to spontaneously ionized defects.Semiconducting/insulating films are of practical importance in broad applications based on low-power,high-performance electronics,the existence of which lacks firm evidence.Here,we synthesized highly insulating films in a controlled way,which exhibit semiconducting behavior with channel resistance up to 1 TΩ.The electron chemical potential lies within the band gap,in some cases,even below the charge neutrality level,signifying the trace of hole-type semiconducting.The performance of insulating devices remains high,comparable to high-quality devices previously.Especially,the threshold voltage(Vth)is positive,contrary to common negative values reported.Calculations indicate that our synthesis conditions suppress electron donors(Se vacancies(VSe))and promote the formation of compensating acceptors(Bi vacancies(VBi)),leading to insulating behaviors.Our work offers insights into electron dynamics of Bi_(2)O_(2)Se,moves one step further towards p-type transistors and provides a valuable playground for engineering ferroelectricity in high-performance semiconductors. | Tao Wang Zhuokai Xu Ziye Zhu Mengqi Wu Zhefeng Lou Jialu Wang Wanghua Hu Xiaohui Yang Tulai Sun Xiaorui Zheng Wenbin Li Xiao Lin | 2023 | Nano Research2023,16,2: | 0 |
