| 1 | Responses of Soil Microbial Community Structure and Diversity to Agricultural Deintensification显示文摘Using a scheme of agricultural fields with progressively less intensive management (deintensification), different manage- ment practices in six agroecosystems located near Goldsboro, NC, USA were tested in a large-scale experiment, including two cash-grain cropping systems employing either tillage (CT) or no-tillage (NT), an organic farming system (OR), an integrated cropping system with animals (IN), a successional field (SU), and a plantation woodlot (WO). Microbial phos- pholipid fatty acid (PLFA) profiles and substrate utilization patterns (BIOLOG ECO plates) were measured to examine the effects of deintensification on the structure and diversity of soil microbial communities. Principle component analyses of PLFA and BIOLOG data showed that the microbial community structure diverged among the soils of the six systems. Lower microbial diversity was found in lowly managed ecosystem than that in intensive and moderately managed agro- ecosystems, and both fungal contribution to the total identified PLFAs and the ratio of microbial biomass C/N increased along with agricultural deintensification. Significantly higher ratios of C/N (P < 0.05) were found in the WO and SU systems, and for fungal/bacterial PLFAs in the WO system (P < 0.05). There were also significant decreases (P < 0.05) along with agricultural deintensification for contributions of total bacterial and gram positive (G+) bacterial PLFAs. | 3 | Decomposition of Different Litter Fractions in a Subtropical Bamboo Ecosystem as Affected by Experimental Nitrogen Deposition显示文摘As an important component of the global carbon (C) budget, litter decomposition in terrestrial ecosystems is greatly affected by the increasing nitrogen (N) deposition observed globally. We hypothesized that different litter fractions derived from a single tree species may respond to N deposition differently depending on the quality of the litter substrate. To test the hypothesis, a two-year field experiment was conducted using the litterbag method in a Pleioblastus amarus plantation in the rainy region of Southwest China. Four N treatment levels were applied: control (no N added), low-N (50 kg N ha-1 year-1), medium-N (150 kg N ha-1 year-1), and high-N (300 kg N ha-1 year-1). We observed different patterns of mass loss for the three P. amarus litter fractions (leaves, sheaths, and twigs) of varying substrate quality in the control plots. There were two decomposition stages with different decay rates (fast rate in early stages and slow rate in the later stages) for leaves and sheaths, while we did not observe a slower phase for the decay of twigs during the 2-year study period. The annual decomposition rate (k) of twigs was significantly lower than that of leaves or sheaths. Addition of N slowed the decomposition of leaves and twigs in the later stages of decomposition by inhibiting the decay of lignin and cellulose, while addition of N did not affect the mass loss of sheaths during the study period. In the decomposition of all three litter fractions, experimental N deposition reduced the net N accumulation in the early stages and also decreased the net N release in the later stages. | 4 | Cardioprotective mechanism of SGLT2 inhibitor against myocardial infarction is through reduction of autosis显示文摘Sodium-glucose cotransporter 2(SGLT2)inhibitors reduce cardiovascular mortality in patients with diabetes mellitus but the protective mechanism remains elusive.Here we demonstrated that the SGLT2 inhibitor,Empagliflozin(EMPA),suppresses cardiomyocytes autosis(autophagic cell death)to confer cardioprotective effects.Using myocardial infarction(Ml)mouse models with and without diabetes mellitus,EMPA treatment significantly reduced infarct size,and myocardial fibrosis,thereby leading to improved cardiac function and survival.In the context of ischemia and nutritional glucose deprivation where autosis is already highly stimulated,EMPA directly inhibits the activity of the Na^(+)/H^(+)exchanger 1(NHE1)in the cardiomyocytes to regulate excessive autophagy.Knockdown of NHE1 significantly rescued glucose deprivation-induced autosis.In contrast,overexpression of NHE1 aggravated the cardiomyocytes death in response to starvation,which was effectively rescued by EMPA treatment.Furthermore,in vitro and in vivo analysis of NHE1 and Beclin 1 knockout mice validated that EMPA s cardioprotective effects are at least in part through downregulation of autophagic flux.These findings provide new insights for drug development,specifically targeting NHE1 and autosis for ventricular remodeling and heart failure after Ml in both diabetic and non-diabetic patients. | Kai Jiang Yue Xu Dandan Wang Feng Chen Zizhuo Tu Jie Qian Sheng Xu Yixiang Xu John Hwa Jian Li Hongcai Shang Yaozu Xiang | 2022 | Protein & Cell2022,13,5: | 13 |

| 6 | Comparison of amplicon-sequencing, pyrosequencing and real-time PCR for detection of YMDD mutants in patients with chronic hepatitis B显示文摘AIM: To compare the sequencing of PCR products, pyro- sequencing, and real-time PCR for detection of Tyrosine- methionine-aspartate-aspartate (YMDD) mutants in patients with chronic hepatitis B. METHODS: Mixtures of plasmids and serum samples from 69 chronic hepatitis B patients treated with lamivu- dine were tested for YMDD mutations by sequencing of PCR products, pyrosequencing, and real-time PCR, re- spectively. Time required and reagent costs of the three assays were evaluated. RESULTS: Real-time PCR detected 100%, 50%, 10%, 1% and 0.1% of YVDD plasmid in mixtures with 106 copies/mL of YMDD plasmid, whereas sequencing and pyrosequencing only detected 100% and 50% of YVDD plasmid in aliquots of the corresponding mixtures. Com- pletely concordant results were obtained from 60 (87%) out of the 69 clinical serum samples by the three assays. Mutants were detected by real-time PCR in less than 20% of the total virus population, but no mutant was de- tected by sequencing and pyrosequencing. In addition, real-time PCR required less time and was more cost-ef- fective than the other two assays. However, throughput of pyrosequencing was the highest. | 8 | Outcomes in children with biliary atresia following liver transplantation显示文摘BACKGROUND: Congenital biliary atresia is a rare condition characterized by idiopathic dysgenesis of the bile ducts. If untreated, congenital biliary atresia leads to liver cirrhosis, liver failure and premature death. The present study aimed to evaluate the outcomes of orthotopic liver transplantation in children with biliary atresia. METHOD: We retrospectively analyzed 45 patients with biliary atresia who had undergone orthotopic liver transplantation from September 2006 to August 2012. RESULTS: The median age of the patients was 11.0 months (5-102). Of the 45 patients, 41 were younger than 3 years old. Their median weight was 9.0 kg (4.5-29.0), 34 of the 45 patients were less than 10 kg. Thirty-one patients had undergone Kasai portoenterostomy prior to orthotopic liver transplantation. We performed 30 living donor liver transplants and 15 split liver transplants. Six patients died during a follow-up. The median follow-up time of surviving patients was 11.4 months (1.4-73.7). The overall 1-, 2- and 3-year survival rates were 88.9%, 84.4% and 84.4%, respectively. | 9 | Litterfall,Litter Decomposition,and Nutrient Dynamics in Two Subtropical Bamboo Plantations of China显示文摘Bamboos are one of the fast-growing and multiple use species in the world,and thus bamboo forests/plantations play an important role in C sequestration at regional and global levels.We studied aboveground litterfall,litter decomposition and nutrient dynamics for two years in two subtropical bamboo ecosystems in Southwest China so as to test the hypothesis that litter quality determine the rate and nutrient dynamics during decomposition of different litter fractions.Mean annual total aboveground litter production ranged from 494 to 434 g m^(-2)in two bamboo stands(P stand,dominated by Pleioblastus amarus and H stand,hybrid bamboo dominated by Bambusa pervariabilis x Dendrocalamopsis daii).Bulk(~80%)of litter production was contributed by leaf litter in two stands followed by twigs and sheathes.Different litter fractions represented considerable variations in the rates of mass loss and nutrient release.Variation of the mass remaining after 2 years of decomposition was significantly explained by initial C/N ratio and initial P concentration.Initial concentrations of N,P,Ca,and Mg explained 57.9%,95.0%,99.8%and 98.1%,respectively,of the variations of these elements mass remaining after 2 years of decomposition.The patterns of nutrient dynamics and the final amount remaining were mainly determined by their initial litter substrate quality in these two subtropical bamboo plantations. | TU Li-Hua HU Hong-Ling HU Ting-Xing ZHANG Jian LI Xian-Wei LIU Li XIAO Yin-Long CHEN Gang LI Ren-Hong | 2014 | Pedosphere2014,24,1: | 8 |
| 10 | Design,methodology,and baseline of whole city‑million scale children and adolescents myopia survey(CAMS)in Wenzhou,China显示文摘Background:Myopia is the most common visual impairment in children and adolescents worldwide.This study described an economical and effective population-based screening pipeline and performed the project of a million scale children and adolescents myopia survey(CAMS),which will shed light on the further study of myopia from the level of epidemiology and precision medicine.Methods:We developed a novel population-based screening pattern,an intelligent screening process and internetbased information transmission and analysis system to carry out the survey consisting of school children in Wenzhou,China.The examination items include unaided distance visual acuity,presenting distance visual acuity,and noncycloplegic autorefraction.Myopia and high myopia were defined as spherical equivalent(SE)≤−1.00 diopters(D)and SE≤−6.00 D,respectively.Next,the reports of the vision checking were automatically sent to parents and the related departments.The CAMS project will be done two to four times annually with the support of the government.An online eyesight status information management system(OESIMS)was developed to construct comprehensive and efficient electronic vision health records(EVHRs)for myopia information inquiry,risk pre-warning,and further study.Results:The CAMS completed the first-round of screening within 30 days for 99.41%of Wenzhou students from districts and counties,in June 2019.A total of 1,060,925 participants were eligible for CAMS and 1,054,251(99.37%participation rate)were selected through data quality control,which comprised 1305 schools,and 580,609,251,050 and 170,967 elementary,middle,and high school students.The mean age of participants was 12.21±3.32 years(6-20 years),the female-to-male ratio was 0.82.The prevalence of myopia in elementary,middle,and high school students was 38.16%,77.52%,and 84.00%,respectively,and the high myopia incidence was 0.95%,6.90%,and 12.98%.Conclusions:The CAMS standardized myopia screening model involves automating large-scale information collection,data transmission,data analysis and early warning,thereby supporting myopia prevention and control.The entire survey reduced 90%of staff,cost,and time consumption compared with previous surveys.This will provide new insights for decision support for public health intervention. | Liangde Xu Youyuan Zhuang Guosi Zhang Yunlong Ma Jian Yuan Changseng Tu MiaoMiao Li Wencan Wang Yaru Zhang Xiaoyan Lu Jing Li Xinting Liu Zhengbo Xue Meng Zhou Jie Sun Jinhua Bao Ming Li Fan Lu Hong Wang Jianzhong Su Jia Qu | 2021 | Eye and Vision2021,8,1: | 7 |

| 12 | Uncompromised NK cell activation is essential for virus-specific CTL activity during acute influenza virus infection显示文摘Natural killer (NK) cells are indispensable components of both the innate and adaptive immune response. However,their precise roles in the cross-talk between innate and adaptive immunity during influenza virus infection remaincontroversial. By comparing NK cell dynamics and activity under a sub-lethal dose and high dose of influenza virusinfection, we showed that influenza virus PR8 directly infected NK cells during natural infection, which wasconsistent with our previous findings obtained from an in vitro investigation of human NK cells. The impairments incytotoxicity and IFN-γ production by spleen NK cells following high-dose infection were accompanied by decreasedvirus-specific killing mediated by cytotoxic T lymphocytes (CTLs). Importantly, the weakened CTL activity could bereversed by adoptive transfer of spleen NK cells harvested from low-dose-infected mice but not healthy donors.Taken together, our data provide direct evidence supporting the contribution of NK cells to antiviral T-cellresponses. | 14 | Nano-cerium-element-doped titanium dioxide induces apoptosis of Bel 7402 human hepatoma cells in the presence of visible light显示文摘AIM: To investigate the apoptotic effect of photoexcited titanium dioxide (TiO2) nanoparticles in the presence of visible light on human hepatoma cell line (Bel 7402) and to study the underlying mechanism. METHODS: Cerium-element-doped titanium dioxide nanoparticles were prepared by impregnation method. Bel 7402 human hepatoma cells were cultured in RPMI 1640 medium in a humidified incubator with 50 mL/L CO2 at 37℃. A 15 W fluorescent lamp with continuous wavelength light was used as light source in the photocatalytic test. Fluorescence morphology and agarose gel eletrophoresis pattern were performed to analyze apoptotic cells. RESULTS: The Ce (Ⅳ)-doped TiO2 nanoparticles displayed their superiority. The adsorption edge shifted to the 400-450 nm region. With visible light illuminated for 10 min, 10 μg/cm3 Ce (Ⅳ)-doped TiO2 induced micronuclei and significant apoptosis in 4 and 24 h, respectively. Hochest 33 258 staining of the fixed cells revealed typical apoptotic structures (apoptotic bodies), agarose gel electrophoresis showed typical DNA ladder pattern in treated cells but not in untreated ones. | 16 | CD40-activated B cells are more potent than immature dendritic cells to induce and expand CD41 regulatory T cells显示文摘CD4^(+)regulatory T cells(Tregs)play an important role in maintaining immune tolerance by suppressing pathologic immune responses.,The generation of large numbers of antigen-specific Tregs ex vivo is critical for the development of clinical immunotherapy based on the adoptive transfer of Tregs.Both CD40-activated B cells(CD40-B)and immature dendritic cells(imDCs)have been used as professional antigen-presenting cells(APCs)to generate antigen-specific Tregs.However,the efficiencies of CD40-B and imDCs to generate CD4^(+)Tregs have not been compared directly and the mechanism driving the generation of these Tregs remains largely unknown.In this study,we found that CD40-B exhibited mature phenotypes and were more able to induce and expand CD4^(high)'CD25^(+)Tregs than imDCs.Moreover,Tregs induced by CD40-B had greater suppressive capacity than those induced by imDCs.The generation of CD4^(high)CD25^(+)Tregs by CD40-B and imDCs is cell-cell contact dependent and partially relies on the expression of human leukocyte antigen(HLA)-DR and CD80/86.Differences in CD4^(high)CD25^(+)Treg generation efficiency were largely explained by the production of endogenous IL-2 by CD40-B.Our results suggest that CD40-B is better able to generate large numbers of antigen-specific Tregs than imDCs.Additionally,using CD40-B to generate Tregs may accelerate the clinical use of Treg-based immunotherapy in the treatment of allograft rejection,graft versus host disease(GVHD)and autoimmune diseases. | Jian Zheng Yinping Liu Yu-Lung Lau Wenwei Tu | 2010 | Cellular & Molecular Immunology2010,7,1: | 5 |

| 20 | Evolution of surface microstructure of Cu-50Cr alloy treated by high current pulsed electron beam显示文摘A Cu-50Cr alloy was treated by the high current pulsed electron beam(HCPEB)at 20 and 30 ke V with pulse numbers ranging from 1 to 100.Surface morphologies and microstructures of specimens before and after the treatments were investigated by employing scanning electron microscopy and X-ray diffraction.Results show that the HCPEB technique is able to induce remarkable surface modifications for the Cu-50Cr alloy.Cracks in Cr phases appear even after one-pulse treatment and their density always increases with the pulse number.Formation reason for these cracks is attributed to quasi-static thermal stresses accumulated along the specimen surface.Craters with typical morphologies are formed due to the dynamic thermal field induced by the HCPEB and they are found to prefer the sites near cracks or boundaries between neighboring Cr phases.Another microstructural characteristic produced by the HCPEB is the fine Cr spheroids,which are determined to be due to occurrence of liquid phase separation in the Cu-50Cr alloy.Finally,a general microstructural evolution profile that incorporates various HCPEB-induced surface features is tentatively outlined. | CHAI LinJiang ZHOU ZhiMing XIAO ZhiPei TU Jian WANG YaPing HUANG WeiJiu | 2015 | Science China(Technological Sciences)2015,58,3: | 5 |
