| 2 | Zircon SHRIMP U-Pb geochronology of the Neoproterozoic Chengjiang Formation in central Yunnan Province(SW China) and its geological significance显示文摘The Chengjiang Formation is the earliest continental clastic deposit after the Jinning Orogeny in central Yunnan Province,and therefore its accurate depositional age is significant for understanding the formation and evolution of the Neoproterozoic rift basins in southern China.However,hampered by accuracy of the dating technique,the existing age data for the Chengjiang Formation are not very reliable.A large number of magmatic zircons were obtained from the tuff interbed in the lower part of the Chengjiang Formation in Jinyang area and the bottom part of the Chengjiang Formation in Dongchuan area,central Yunnan Province,and high-precision SHRIMP U-Pb dating was carried out on these zircons.The results show that the weighted mean 206 Pb/238 U ages are 797.8±8.2 and 803.1±8.7 Ma respectively.In combination with related geological evidence,it has been demonstrated that the bottom boundary age of the Chengjiang Formation should be 800±5 Ma,whereas the top boundary age could be ca 725 Ma.By synthetically analyzing the latest age data for the relevant strata,it has been confirmed that the Chengjiang Formation should be correlated with the Suxiong Formation and the Kaijianqiao Formation in western Sichuan Province,the Liantuo Formation in the middle and lower Yangtze,the Hongchicun Formation and the Shangshu Formation in northern Zhejiang Province,the Puling Formation in southern Anhui Province,the Luokedong Formation and the Majianqiao Formation in northwestern Jiangxi Province,the Wuqiangxi Formation in northwestern Hunan Province,the Fanzhao Formation and the Qingshuijiang Formation in southeastern Guizhou Province,and the Sanmenjie Formation and the Gongdong Formation in northern Guangxi Province.Sedimentary cycle analysis shows that the sedimentary filling sequence of the Neoproterozoic rift basins in southern China can be divided into four cycles.Among them,Cycle II began at ca.800 Ma,accompanied by intensive tectonic-thermal events.The zircon U-Pb ages from the bottom of the Chengjiang Formation reported in this paper indicate that the Neoproterozoic Kangdian rift subbasin probably started to develop at ca.800 Ma and therefore missed Cycle I of the Neoproterozoic sedimentary filling sequence in southern China. | JIANG XinSheng WANG Jian CUI XiaoZhuang ZHUO JieWen XIONG GuoQing LU JunZe LIU JianHui | 2012 | Science China Earth Sciences2012,55,11: | 25 |
| 3 | Wnt5a participates in hepatic stellate cell activation observed by gene expression profile and functional assays显示文摘AIM:To identify differentially expressed genes in quiescent and activated hepatic stellate cells(HSCs)and explore their functions.METHODS:HSCs were isolated from the normal Sprague Dawley rats by in suit perfusion of collagenase and pronase and density Nycodenz gradient centrifugation.Total RNA and mRNA of quiescent HSCs,and cultureactivated HSCs were extracted,quantified and reversely transcripted into cDNA.The global gene expression profile was analyzed by microarray with Affymetrix rat genechip.Differentially expressed genes were annotated with Gene Ontology(GO)and analyzed with Kyoto encyclopedia of genes and genomes(KEGG)pathway using the Database for Annotation,Visualization and Integrated Discovery.Microarray data were validated by quantitative real-time polymerase chain reaction(qRTPCR).The function of Wnt5a on human HSCs line LX-2 was assessed with lentivirus-mediated Wnt5a RNAi.The expression of Wnt5a in fibrotic liver of a carbon tetrachloride(CCl4)-induced fibrosis rat model was also analyzed with Western blotting.RESULTS:Of the 28 700 genes represented on this chip,2566 genes displayed at least a 2-fold increase or decrease in expression at a P<0.01 level with a false discovery rate.Of these,1396 genes were upregulated,while 1170 genes were downregulated in culture-activated HSCs.These differentially expressed transcripts were grouped into 545 GO based on biological process GO terms.The most enriched GO terms included response to wounding,wound healing,regulation of cell growth,vasculature development and actin cytoskeleton organization.KEGG pathway analysis revealed that Wnt5a signaling pathway participated in the activation of HSCs.Wnt5a was significantly increased in cultureactivated HSCs as compared with quiescent HSCs.qRTPCR validated the microarray data.Lentivirus-mediated suppression of Wnt5a expression in activated LX-2 resulted in significantly impaired proliferation,downregulated expressions of typeⅠcollagen and transforming growth factor-β1.Wnt5a was upregulated in the fibrotic liver of a CCl4-induced fibrosis rat model.CONCLUSION:Wnt5a is involved in the activation of HSCs,and it may serve as a novel therapeutic target in the treatment of liver fibrosis. | Wu-Jun Xiong Li-Juan Hu Yi-Cheng Jian Li-Jing Wang Ming Jiang Wei Li Yi He | 2012 | World Journal of Gastroenterology2012,18,15: | 17 |
| 4 | Opening time and filling pattern of the Neoproterozoic Kangdian Rift Basin,western Yangtze Continent,South China显示文摘The Neoproterozoic Kangdian Rift Basin is a continental rift basin in the western Yangtze Continent.Determining its time of opening and subsequent filling pattern is an important aspect of understanding the formation and evolution of the Yangtze Continent,South China.The Luliang Formation is the early filling in the eastern part of this rift basin,and its deposition age and filling sequence are significant for studies of the regional stratigraphic correlation,opening time,and filling pattern of this basin.Having been correlated to the upper part of the Mesoproterozoic Kunyang Group or to the Neoproterozoic Chengjiang Formation,the age and regional stratigraphic correlation of the Luliang Formation have long been uncertain.This is due to a lack of reliable absolute age constraints.To address this issue,the first zircon SHRIMP U-Pb geochronology has been established for this formation,yielding two high-precision isotopic ages of 818.6±9.2 and 805±14 Ma for the tuff layers at the bottoms of the lower and upper members of the Luliang Formation,respectively.Given the error factor,the bottom age of the lower member of the Luliang Formation can be interpreted as ca.820 Ma,corresponding to the bottom age of the lower part of the Banxi Group,which is the early filling of the Neoproterozoic Xianggui Rift Basin,a representative basin of the Neoproterozoic rift basins in the Yangtze Continent,South China.The bottom age of the upper member of the Luliang Formation can be interpreted as ca.800±5 Ma,corresponding to the bottom age of the Chengjiang Formation in the western part of the Neoproterozoic Kangdian Rift Basin and also corresponding to the bottom age of the upper part of the Banxi Group in the Neoproterozoic Xianggui Rift Basin.These ages indicate that the Neoproterozoic Kangdian Rift Basin shares the same opening time and filling sequences as those of the other Neoproterozoic rift basins in South China.Basin analysis shows that the Neoproterozoic Kangdian Rift Basin is a typical half-graben basin,with its main boundary fault in the west and basin center in the east.This basin consisted of mini unidirectional half-graben basins in its juvenile stage and simplified to become a large united half-graben basin in its mature stage,i.e.,a supradetachment basin. | ZHUO JieWen JIANG XinSheng WANG Jian CUI XiaoZhuang XIONG GuoQing LU JunZe LIU JianHui MA MingZhu | 2013 | Science China Earth Sciences2013,56,10: | 12 |

| 6 | Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. | 10 | Comparison of two methods used to culture and purify rat retinal Mller cells显示文摘AIM:To study two methods for culturing and purifying Sprague-Dawley(SD)rat retinal Muller cells and determine which one is better.METHODS:The passage culture method of Muller cells was respectively carried out by complete pancreatic enzyme digestion method and repeated incomplete pancreatic enzyme digestion method.After culturing retinal cells for one month through these two methods,fluorescence-activated cell sorter(FACS),RT-PCR,and immunohistochemistry technology were performed to examine the enrichment and purity of Muller glial cells,and carried out two-sample approximate t test using SSPS 13.0 to further compare the Muller cell positive rate in both methods.RESULTS:The statistical results showed that the purity of Muller cells was 83.2%±5.16%in group A,and the purity was 98.5%±1.08%in group B.The two-sample approximate t test analysis demonstrated that the difference between group A and group B was statistically significant(t=-9.178,P<0.005).The results clearly exhibited a difference between the purity of Muller cells cultured by the complete pancreatic enzyme digestion method(group A)and the repeated incomplete pancreatic enzyme digestion method(group B).CONCLUSION:Compared with the complete pancreatic enzyme digestion method,this novel method was more efficient and a higher purity of Muller cells could be obtained using this approach. | Wei-Tao Song Xue-Yong Zhang Si-Qi Xiong Dan Wen Jian Jiang Xiao-Bo Xia | 2013 | International Journal of Ophthalmology(English edition)2013,6,6: | 2 |
| 11 | Biomedical polymers: synthesis, properties, and applications显示文摘Biomedical polymers have been extensively developed for promising applications in a lot of biomedical fields, such as therapeutic medicine delivery, disease detection and diagnosis, biosensing, regenerative medicine, and disease treatment. In this review, we summarize the most recent advances in the synthesis and application of biomedical polymers, and discuss the comprehensive understanding of their property-function relationship for corresponding biomedical applications. In particular, a few burgeoning bioactive polymers, such as peptide/biomembrane/microorganism/cell-based biomedical polymers, are also introduced and highlighted as the emerging biomaterials for cancer precision therapy. Furthermore, the foreseeable challenges and outlook of the development of more efficient, healthier and safer biomedical polymers are discussed. | 12 | Improved Mechanical Properties of Poly(butylene succinate) Membrane by Co-electrospinning with Gelatin显示文摘Gelatin, a natural proteinous polymer, was used to co-electrospin with poly(butylene succinate)(PBS) in order to improve the mechanical properties of PBS membrane and facilitate its applications in biomedical field. The PBS/gelatin blend membranes have narrower distribution of fiber diameter and smoother surface than neat PBS membrane. The contact angles, water absorption rates and water uptakes of the PBS/gelatin blend membranes were measured, showing increased hydrophilicity. The interaction between PBS and gelatin was investigated by attenuated total reflectance Fourier transform infrared spectroscopy(ATR-FTIR) and differential scanning calorimetry(DSC). The mechanical properties of PBS/gelatin blend membranes in both dry and wet states were evaluated by uniaxial tensile tests. In the dry state, the PBS/gelatin blend membrane containing 10% gelatin has a 3-times increase in tensile strength without any adverse effect on ductility because of the existence of interaction between the two blend components, little change in crystallinity of PBS, and possible interaction between any adjacent fibers; the tensile strength and elongation at break are even better in the wet state attributed to some gelatin on fiber surfaces, which act as a binder in the presence of water. The potential applications of PBS/gelatin blend membranes were demonstrated by successful immobilization of thrombin, a clinically-used hemostatic drug. | 16 | Early life migration and population discrimination of the small yellow croaker Larimichthys polyactis from the Yellow Sea: inferences from otolith Sr/Ca ratios显示文摘The small yellow croaker Larimichthys polyactis is a benthic marine fish species of high ecological and commercial importance and is widely distributed in the northwestern Pacifi c Ocean,especially in the Chinese coastal waters of the Bohai,Yellow,and East China Seas.As a highly migratory species,the whole life migration of L.polyactis has been intensively studied.However,knowledge about its early life migration is scarce,and population divisions are inconsistent,limiting the ability of fishery scientists and administrators to evaluate the design and potential benefits of thorough conservation and resource-management strategies.In the present study,otolith Sr/Ca was analyzed to investigate the early migratory patterns and discriminate the populations of L.polyactis in the Yellow Sea,including two spawning groups and one overwintering group.The variation in Sr/Ca ratios of ontogenetic growth zones,including the nucleus(N),larval(L),metamorphosis(M),juvenile(J),and edge(E)zones,was measured by electron probe microanalysis.The variation in Sr/Ca ratios in early developmental growth zones was generally characterized by an evident downward trend from the N to J zone,which suggests that the early migratory pattern of L.polyactis might be from inshore to nearshore water.Canonical discriminant analysis,based on the otolith Sr/Ca ratios of the N,L,M,and J zones,allowed the successful discrimination of the two populations,namely,the northern and southern Yellow Sea groups,whose diff erences were mainly reflected in the L and J zones.Compared with previous studies,the traditional geographic boundaries(34°N)separating these two populations might be moving northward.The application of otolith Sr/Ca ratios based on ontogenetic stage could improve our understanding of the migration and population discrimination of L.polyactis from the Yellow Sea. | Dade SONG Ying XIONG Tao JIANG Jian YANG Xiaming ZHONG Jianhua TANG | 2022 | Journal of Oceanology and Limnology2022,40,2: | 1 |

| 20 | Mapping of de novo mutations in primary biliary cholangitis to a disease-specific co-expression network underlying homeostasis and metabolism显示文摘Primary biliary cholangitis(PBC) is an autoimmune disease involving dysregulation of a broad array of homeostatic and metabolic processes. Although considerable single-nucleotide polymorphisms have been unveiled, a large fraction of risk factors remains enigmatic. Candidate genes with rare mutations that tend to confer more deleterious effects need to be identified. To help pinpoint cellular and developmental mechanisms beyond common noncoding variants, we integrate whole exome sequencing with integrative network analysis to investigate genes harboring de novo mutations. Prominent convergence has been revealed on a network of disease-specific co-expression comprised of 55 genes associated with homeostasis and metabolism. The transcription factor gene MEF2 D and the DNA repair gene PARP2 are highlighted as hub genes and identified to be up-and down-regulated, respectively, in peripheral blood data set. Enrichment analysis demonstrates that altered expression of MEF2 D and PARP2 may trigger a series of molecular and cellular processes with pivotal roles in PBC pathophysiology.
