Science Letters:A synthetic Toll-like receptor 2 ligand decreases allergic immune responses in a mouse rhinitis model sensitized to mite allergen显示文摘It has been proposed that activation of Toll-like receptors (TLRs) plays crucial roles in the polarization of adaptive immune responses. A synthetic Toll-like receptor 2 (TLR2) ligand, Pam3CSK4, has been reported to modulate the balance of Th1/Th2 responses. We evaluated the modulation effect of Pam3CSK4 on allergic immune response in a mouse rhinitis model sensitized to house dust mite allergen (HDM). Mice were sensitized and challenged with Dermatophagoides farinae allergen (Der f), and then the allergic mice were treated by Pam3CSK4. Nasal allergic symptoms and eosinophils were scored. Der f-specific cytokine responses were examined in the splenocytes and bronchoalveolar lavage fluid (BALF). Serum level of total IgE was also detected. After establishing a mouse allergic rhinitis model with HDM, we have showed that Pam3CSK4 treatment not only ameliorated the nasal allergic symptoms remarkably but also decreased the eosinophils and total inflammation cells in BALF significantly. Analysis of cytokine profile found that IFN-γ released from either BALF or stimulated splenocytes increased markedly in Pam3CSK4-treated mice, while IL-13 decreased significantly. Moreover, serum level of total IgE was significantly lower in Pam3CSK4-treated mice than in the untreated. Thus, in an allergic rhinitis mouse model developed with HDM, Pam3CSK4 was shown to exhibit an antiallergic effect, indicating its potential application in allergic diseases. | Cheng ZHOU Xiao-dong KANG Zhi CHEN | 2008 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2008,9,4: | 6
Effect of Simulated Microgravity and its Associated Mechanism on Pulmonary Circulation in Rats显示文摘Objective To study the effect of Simulated Microgravity and its Associated Mechanism on Pulmonary Circulation in Rats. Methods Rat tail-suspension model was used to simulate the physiological effects of microgravity and changes in pulmonary blood vessel morphology, pulmonary arterial and venous blood pressure, pulmonary vascular resistance, pulmonary vasomotoricity, as well as the regulation of pulmonary circulation by cytokines produced and released by the lung of rats were measured. Results The walls of pulmonary blood vessels of rats were thickened, and the pulmonary artery was reconstructed with increased pulmonary vascular resistance. The pulmonary blood vessels of rats became more prone to dilation as contractions increased. Rat epithelial Adrenomedulin gene transcription and protein expression were upregulated. The level of basic fibroblast growth Factor of rat was also elevated. Conclusion Findings from the present study on rats revealed that the microgravity can affect pulmonary blood vessel structure, pulmonary arterial pressure, and pulmonary blood vessel self-regulation and cytokine production. | LI Tian Zhi YUAN Ming CHEN Zhen Hong GUO Ying Hua KANG Chun Yan WANG Jing Yu LI Zhi Li WANG De Sheng WANG Hui Juan YUAN Min LIU Chang Ting | 2013 | Biomedical and Environmental Sciences2013,26,2: | 6
Aberrant translation regulated by METTL1/WDR4-mediated tRNA N7-methylguanosine modification drives head and neck squamous cell carcinoma progression显示文摘Background:Cancer cells selectively promote the translation of oncogenic tran-scripts to stimulate cancer progression.Although growing evidence has revealed that tRNA modifications and related genes participate in this process, their roles in head and neck squamous cell carcinoma(HNSCC)remain largely unchar-acterized.Here,we sought to investigate the function and mechanisms of the transfer RNA(tRNA)N7-methylguanosine(m'G)modification in regulating the occurrence and development of HNSCC.Methods:Cell lost of-function and gain-of function assays, xenograft models, conditional knockout and knockin mouse models were used to study the physi-ological functions of tRNA m'G modification in HNSCC tumorigenesis.tRNA modification and expression profiling,mRNA translation profiling and res-cue assays were performed to uncover the underlying molecular mechanisms.Single-cell RNA sequencing(scRNA seq)was conducted to explore the tumor microenvironment changes. Results:The tRNA.m7G methyltransferase complex components Methyltransferase-like 1(METTL1)/WD repeat domain 4(WDR4)were upregulated in HNSCC and associated with a poor prognosis.Functionally,METTL1/WDR4 promoted HNSCC progression and metastasis in cell-based and transgenic mouse models.Mechanistically, ablation of METTL1 reduced the m'G levels of 16 tRNAS,inhibiting the translation of a subset of oncogenic transcripts, including genes related to the phosphatidylinositol-3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/AKT/mTOR)signaling pathway. In addition, chemical modulators of the PI3K/Akt/mTOR signaling pathway reversed the effects of Mettll in mouse HNSCC.Furthermore,scRNA-seq results revealed that Mettll knockout in mouse tumor cells altered the immune landscape and cell-cell interaction between the tumor and stromal compartment. Conclusions:The tRNA m?G methyltransferase METTLI was found to promote the development and malignancy of HNSCC through regulating global mRNA translation, including the PI3K/AKT/mTOR signaling pathway, and found to alter immune landscape.METTLI could be a promising treatment target for HNSCC patients. | Jie Chen Kang Li Jianwen Chen Xiaochen Wang Rongsong Ling Maosheng Cheng Zhi Chen Fangfang Chen Qianting He Shuai Li Caihua Zhang Yizhou Jiang Qianming Chen Anxun Wang Demeng Chen | 2022 | Cancer Communications2022,42,3: | 6
Induction of Endoplasmic Reticulum Stress by Cadmium and Its Regulation on Nrf2 Signaling Pathway in Kidneys of Rats显示文摘Objective This study was conducted to investigate the regulation of endoplasmic reticulum stress on Nrf2 signaling pathway in the kidneys of rats. Methods Rats were divided into twelve groups of six animals each. Some groups were pre-administered with bacitracin or tauroursodeoxycholic acid(TUDCA), and all of them were treated with 5-20 μmol/kg cadmium(Cd) for 48 h. The oxidative stress levels were analyzed using kits. The mRNA and protein expression levels of endoplasmic reticulum stress-related factors and Nrf2 signaling pathway-related factors were determined using RT-PCR and western blot. Results Cd exposure resulted in oxidative stress in the kidneys of rats and upregulated the expression of endoplasmic reticulum stress(ERS)-related factors and Nrf2 signaling pathway-related factors, especially at doses of 10 and 20 μmol/kg Cd, and the expression changes were particularly obvious. Moreover, after pretreatment with bacitracin, Cd upregulated the expression of ERS-related factors to a certain extent and, at higher doses, increased the mRNA expression of Nrf2. After pretreatment with TUDCA, Cd reduced the level of ERS to a certain extent; however, at these doses, there were no significant changes in the expression of Nrf2. Conclusion Cadmium can result in ERS and oxidative stress in the kidneys of rats, activate Nrf2, and upregulate the transcriptional expression of phase II detoxification enzymes under these experimental conditions. ERS has a positive regulation effect on Nrf2 signaling pathway but has little effect on the negative regulation of Nrf2 signaling pathway in cadmium toxicity. | CHEN Zhi Jian CHEN Jia Xing WU Li Kang LI Bi Yun TIAN Ya Feng XIAN Min HUANG Zi Pei YU Ri An | 2019 | Biomedical and Environmental Sciences2019,32,1: | 6

Clinical practice guideline for transurethral plasmakinetic resection of prostate for benign prostatic hyperplasia(2021 Edition)显示文摘Benign prostatic hyperplasia (BPH) is highly prevalent among older men, impacting on their quality of life, sexual function, and genitourinary health, and has become an important global burden of disease. Transurethral plasmakinetic resection of prostate (TUPKP) is one of the foremost surgical procedures for the treatment of BPH. It has become well established in clinical practice with good efficacy and safety. In 2018, we issued the guideline “2018 Standard Edition”. However much new direct evidence has now emerged and this may change some of previous recommendations. The time is ripe to develop new evidence-based guidelines, so we formed a working group of clinical experts and methodologists. The steering group members posed 31 questions relevant to the management of TUPKP for BPH covering the following areas: questions relevant to the perioperative period (preoperative, intraoperative, and postoperative) of TUPKP in the treatment of BPH, postoperative complications and the level of surgeons’ surgical skill. We searched the literature for direct evidence on the management of TUPKP for BPH, and assessed its certainty generated recommendations using the grade criteria by the European Association of Urology. Recommendations were either strong or weak, or in the form of an ungraded consensus-based statement. Finally, we issued 36 statements. Among them, 23 carried strong recommendations, and 13 carried weak recommendations for the stated procedure. They covered questions relevant to the aforementioned three areas. The preoperative period for TUPKP in the treatment of BPH included indications and contraindications for TUPKP, precautions for preoperative preparation in patients with renal impairment and urinary tract infection due to urinary retention, and preoperative prophylactic use of antibiotics. Questions relevant to the intraoperative period incorporated surgical operation techniques and prevention and management of bladder explosion. The application to different populations incorporating the efficacy and safety of TUPKP in the treatment of normal volume (< 80 ml) and large-volume (≥ 80 ml) BPH compared with transurethral urethral resection prostate, transurethral plasmakinetic enucleation of prostate and open prostatectomy;the efficacy and safety of TUPKP in high-risk populations and among people taking anticoagulant (antithrombotic) drugs. Questions relevant to the postoperative period incorporated the time and speed of flushing, the time indwelling catheters are needed, principles of postoperative therapeutic use of antibiotics, follow-up time and follow-up content. Questions related to complications incorporated types of complications and their incidence, postoperative leukocyturia, the treatment measures for the perforation and extravasation of the capsule, transurethral resection syndrome, postoperative bleeding, urinary catheter blockage, bladder spasm, overactive bladder, urinary incontinence, urethral stricture, rectal injury during surgery, postoperative erectile dysfunction and retrograde ejaculation. Final questions were related to surgeons’ skills when performing TUPKP for the treatment of BPH. We hope these recommendations can help support healthcare workers caring for patients having TUPKP for the treatment of BPH. | Xian-Tao Zeng Ying-Hui Jin Tong-Zu Liu Fang-Ming Chen De-Gang Ding Meng Fu Xin-Quan Gu Bang-Min Han Xing Huang Zhi Hou Wan-Li Hu Xin-Li Kang Gong-Hui Li Jian-Xing Li Pei-Jun Li Chao-Zhao Liang Xiu-Heng Liu Zhi-Yu Liu Chun-Xiao Liu Jiu-Min Liu Guang-Heng Luo Yi Luo Wei-Jun Qin Jian-Hong Qiu Jian-Xin Qiu Xue-Jun Shang Ben-Kang Shi Fa Sun Guo-Xiang Tian Ye Tian Feng Wang Feng Wang Yin-Huai Wang Yu-Jie Wang Zhi-Ping Wang Zhong Wang Qiang Wei Min-Hui Xiao Wan-Hai Xu Fa-Xian Yi Chao-Yang Zhu Qian-Yuan Zhuang Li-Qun Zhou Xiao-Feng Zou Nian-Zeng Xing Da-Lin He Xing-Huan Wang 无 | 2022 | Military Medical Research2022,9,5: | 4
First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. We for the first time extended the limit to the m_(χ)<6 GeV/c^(2) region. | Yi Wang Zhi Zeng Qian Yue LiTao Yang KeJun Kang YuanJing Li Mehment Agartioglu HaiPeng An JianPing Chang JingHan Chen YunHua Chen JianPing Cheng Cheng Yi Chiang WenHan Dai Zhi Deng ChangHao Fang XinPing Geng Hui Gong QiuJu Guo XuYuan Guo HongJian He Li He ShengMing He JinWei Hu TuChen Huang HanXiong Huang HaiTao Jia LiPing Jia Xi Jiang HauBin Li JianMin Li Jin Li MingXuan Li RenMingJie Li Xia Li YuLan Li Bin Liao FongKay Lin ShinTed Lin ShuKui Liu YanDong Liu Yu Liu YuanYuan Liu ZhongZhi Liu Hao Ma YuCai Mao QiYuan Nie JinHua Ning Hui Pan NingChun Qi Jie Ren XiChao Ruan ChangSong Shang Vivek Sharma Ze She Lakhwinder Singh Monoj Kumar Singh TianXi Sun ChangJian Tang WeiYou Tang Yang Tian GuangFu Wang Li Wang Qing Wang Yu Chen Wang YunXiang Wang Zhen Wang Henry Tsz-King Wong ShiYong Wu YuCheng Wu HaoYang Xing Yin Xu Tao Xue YuLu Yan Nan Yi ChunXu Yu HaiJun Yu JianFeng Yue Ming Zeng BingTao Zhang Lei Zhang FengShou Zhang ZhenYu Zhang KangKang Zhao MingGang Zhao JiFang Zhou ZuYing Zhou JingJun Zhu | 2021 | Science China(Physics,Mechanics & Astronomy)2021,64,8: | 3
A CRISPR/Cas12a-empowered surface plasmon resonance platform for rapid and specific diagnosis of the Omicron variant of SARS-CoV-2显示文摘The outbreak of the COVID-19 pandemic was partially due to the challenge of identifying asymptomatic and presymptomatic carriers of the virus, and thus highlights a strong motivation for diagnostics with high sensitivity that can be rapidly deployed. On the other hand, several concerning SARS-CoV-2 variants, including Omicron, are required to be identified as soon as the samples are identified as‘positive’.Unfortunately,a traditional PCR test does not allow their specific identification.Herein, for the first time,we have developed MOPCS(Methodologies of Photonic CRISPR Sensing),which combines an optical sensing technology-surface plasmon resonance(SPR)with the‘gene scissors’clustered regularly interspaced short palindromic repeat(CRISPR)technique to achieve both high sensitivity and specificity when it comes to measurement of viral variants.MOPCS is a low-cost,CRISPR/Cas12a-systemempowered SPR gene-detecting platform that can analyze viral RNA,without the need for amplification, within 38 min from sample input to results output, and achieve a limit of detection of 15 fM.MOPCS achieves a highly sensitive analysis of SARS-CoV-2,and mutations appear in variants B.1.617.2(Delta),B.1.1.529(Omicron)and BA.1(a subtype of Omicron). This platform was also used to analyze some recently collected patient samples from a local outbreak in China, identified by the Centers for Disease Control and Prevention. This innovative CRISPR-empowered SPR platform will further contribute to the fast, sensitive and accurate detection of target nucleic acid sequences with single-base mutations. | Zhi Chen Jingfeng Li Tianzhong Li Taojian Fan Changle Meng Chaozhou Li Jianlong Kang Luxiao Chai Yabin Hao Yuxuan Tang Omar AAl-Hartomy Swelm Wageh Abdullah GAl-Sehemi Zhiguang Luo Jiangtian Yu Yonghong Shao Defa Li Shuai Feng William J.Liu Yaqing He Xiaopeng Ma Zhongjian Xie Han Zhang | 2022 | National Science Review2022,9,8: | 3

Agricultural Risk Factors Influence Microbial Ecology in Honghu Lake显示文摘Agricultural activities, including stock-farming, planting industry, and fish aquaculture, can affect the physicochemical and biological characters of freshwater lakes. However, the effects of pollution producing by agricultural activities on microbial ecosystem of lakes remain unclear.Hence, in this work, we selected Honghu Lake as a typical lake that is influenced by agriculture activities. We collected water and sediment samples from 18 sites, which span a wide range of areas from impacted and less-impacted areas. We performed a geospatial analysis on the composition of microbial communities associated with physicochemical properties and antibiotic pollution of samples. The co-occurrence networks of water and sediment were also built and analyzed. Our results showed that the microbial communities of impacted and less-impacted samples of water were largely driven by the concentrations of TN, TP, NO_3^--N, and NO_2^--N, while those of sediment were affected by the concentrations of Sed-OM and Sed-TN. Antibiotics have also played important roles in shaping these microbial communities: the concentrations of oxytetracycline and tetracycline clearly reflected the variance in taxonomic diversity and predicted functional diversity between impacted and less-impacted sites in water and sediment samples, respectively. Furthermore, for samples from both water and sediment, large differences of network topology structures between impacted and less-impacted were also observed. Our results provide compelling evidence that the microbial community can be used as a sentinel of eutrophication and antibiotics pollution risk associated with agricultural activity; and that proper monitoring of this environment is vital to maintain a sustainable environment in Honghu Lake. | Maozhen Han Melissa Dsouza Chunyu Zhou Hongjun Li Junqian Zhang Chaoyun Chen Qi Yao Chaofang Zhong Hao Zhou Jack A Gilbert Zhi Wang Kang Ning | 2019 | Genomics, Proteomics & Bioinformatics2019,17,1: | 3
Methyltransferase like 13 mediates the translation of Snail in head and neck squamous cell carcinoma显示文摘Methyltransferase like 13(METTL13),a kind of methyltransferase,is implicated in protein binding and synthesis. The upregulation of METTL13 has been reported in a variety of tumors. However, little was known about its potential function in head and neck squamous cell carcinoma(HNSCC)so far.In this study,we found that METTL13 was significantly upregulated in HNSCC at both mRNA and protein level.Increased METTL13 was negatively associated with clinical prognosis. And METTL13 markedly affected HNSCC cellular phenotypes in vivo and vitro.Further mechanism study revealed that METTL13 could regulate EMT signaling pathway by mediating enhancing translation efficiency of Snail, the key transcription factor in EMT,hence regulating the progression of EMT.Furthermore,Snail was verified to mediate METTL13-induced HNSCC cell malignant phenotypes.Altogether, our study had revealed the oncogenic role of METTL13 in HNSCC,and provided a potential therapeutic strategy. | Xiaochen Wang Kang Li Yuehan Wan Fangfang Chen Maosheng Cheng Gan Xiong Ganping Wang Shuang Chen Zhi Chen Jianwen Chen Xiuyun Xu Cheng Wang Liang Peng Demeng Chen | 2021 | International Journal of Oral Science2021,13,3: | 3
New constructions of quaternary low correlation zone sequence sets显示文摘Sequence sets with low correlation have very important applications in modern communication systems. As in quasi-synchronous code-division multiple access(QS-CDMA) system, sequence sets with low correlation zone(LCZ) perform better than other well-known sequence sets. Furthermore, binary or quaternary sequence sets are preferred because of their easy implementation. In this paper, based on the inverse Gray mapping and special binary sequence pairs, new quaternary LCZ sequence sets were constructed. In the LCZ, the maximum of the nontrivial autocorrelation and crosscorrelation values is 1 which show that the QS-CDMA system used the new sequences sets can control the interference in a very low level. | Ma Xiuwen Kang Baoyuan Chen Zhi Li Yuelong | 2015 | The Journal of China Universities of Posts and Telecommunications2015,22,5: | 2
PINK1 gene mutation by pair truncated sgRNA/Cas9-D10A in cynomolgus monkeys显示文摘Mutations of PTEN-induced kinase I(PINK1)cause early-onset Parkinson’s disease(PD)with selective neurodegeneration in humans. However, current PINK1 knockout mouse and pig models are unable to recapitulate the typical neurodegenerative phenotypes observed in PD patients. This suggests that generating PINK1 disease models in non-human primates(NHPs)that are close to humans is essential to investigate the unique function of PINK1 in primate brains.Paired single guide RNA(sgRNA)/Cas9-D10A nickases and truncated sgRNA/Cas9,both of which can reduce off-target effects without compromising on-target editing, are two optimized strategies in the CRISPR/Cas9 system for establishing disease animal models. Here,we combined the two strategies and injected Cas9-D10A mRNA and two truncated sgRNAs into one-cell-stage cynomolgus zygotes to target the PINK1 gene.We achieved precise and efficient gene editing of the target site in three newborn cynomolgus monkeys. The frame shift mutations of PINK1 in mutant fibroblasts led to a reduction in mRNA.However, western blotting and immunofluorescence staining confirmed the PINK1 protein levels were comparable to that in wild-type fibroblasts. We further reprogramed mutant fibroblasts into induced pluripotent stem cells(iPSCs),which showed similar ability to differentiate into dopamine(DA)neurons.Taken together, our results showed that co-injection of Cas9-D10A nickase mRNA and sgRNA into one-cell-stage cynomolgus embryos enabled the generation of human disease models in NHPs and target editing by pair truncated sgRNA/Cas9-D10A in PINK1 gene exon 2 did not impact protein expression. | Zhen-Zhen Chen Jian-Ying Wang Yu Kang Qiao-Yan Yang Xue-Ying Gu Da-Long Zhi Li Yan Cheng-Zu Long Bin Shen Yu-Yu Niu | 2021 | Zoological Research2021,42,4: | 2

Highly selective metal recovery from spent lithium-ion batteries through stoichiometric hydrogen ion replacement显示文摘Spent lithium-ion battery recycling has attracted significant attention because of its importance in regard to the environment and resource importance.Traditional hydrometallurgical methods usually leach all valuable metals and subsequently extract target meals to prepare corresponding materials. However,Li recovery in these processes requires lengthy operational procedures, and the recovery efficiency is low.In this research,we demonstrate a method to selectively recover lithium before the leaching of other elements by introducing a hydrothermal treatment.Approximately 90%of Li is leached from high-Ni layered oxide cathode powders, while consuming a nearly stoichiometric amount of hydrogen ions. With this selective recovery of Li,the transition metals remain as solid residue hydroxides or oxides.Furthermore, the extraction of Li is found to be highly dependent on the content of transition metals in the cathode materials.A high leaching selectivity of Li(>98%)and nearly 95%leaching efficiency of Li can be reached with LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2).In this case, both the energy and material consumption during the proposed Li recovery is significantly decreased compared to traditional methods;furthermore, the proposed method makes full use of H+to leach Li+.This research is expected to provide new understanding for selectively recovering metal from secondary resources. | Weiguang Lv Xiaohong Zheng Li Li Hongbin Cao Yi Zhang Renjie Chen Hancheng Ou Fei Kang Zhi Sun | 2021 | Frontiers of Chemical Science and Engineering2021,15,5: | 1
Arabidopsis transcriptional response to extracellular Ca^(2+) depletion involves a transient rise in cytosolic Ca^(2+)显示文摘Ecological evidence indicates a worldwide trend of dramatically decreased soil Ca2 tlevels caused by increased acid deposition and massive timber harvesting. Little is known about the genetic and cellular mechanism of plants' responses to Ca2 t depletion. In this study, transcriptional profiling analysis helped identify multiple extracellular Ca^(2+)([Ca^(2+)]ext) depletion-responsive genes in Arabidopsis thaliana L., many of which are involved in response to other environmental stresses. Interestingly, a group of genes encoding putative cytosolic Ca^(2+)([Ca^(2+)]cyt)sensors were significantly upregulated, implying that [Ca^(2+)]cyt has a role in sensing [Ca^(2+)]extdepletion. Consistent with this observation, [Ca^(2+)]extdepletion stimulated a transient rise in[Ca^(2+)]cytthat was negatively influenced by [Kt]ext, suggesting the involvement of a membrane potential-sensitive component. The [Ca^(2+)]cytresponse to [Ca^(2+)]extdepletion was significantly desensitized after the initial treatment, which is typical of a receptor-mediated signaling event. The response was insensiResearch Ative to an animal Ca2 tsensor antagonist, but was suppressed by neomycin, an inhibitor of phospholipase C. Gd3 t, an inhibitor of Ca2 tchannels, suppressed the [Ca^(2+)]ext-triggered rise in[Ca^(2+)]cytand downstream changes in gene expression. Taken together, this study demonstrates that [Ca^(2+)]cytplays an important role in the putative receptor-mediated cellular and transcriptional response to [Ca^(2+)]extdepletion of plant cells. | Jing Wang Tergel Tergel Jianhua Chen Ju Yang Yan Kang Zhi Qi | 2015 | Journal of Integrative Plant Biology2015,57,2: | 1
