3Recognition on the forming-vanishing process and underlying mechanisms of the hypoxia off the Yangtze River estuary显示文摘On the basis of compiled multidisciplinary historical data in 2006–2007 and incorporation of relevant simulation results and remote sensing data, we performed an in-depth study of the generation and dissipation process of the hypoxic zone and its distribution morphology and structure off the Yangtze River estuary. Based on the hydrological circulation dynamics, reproduction of phytoplankton(leading to the decomposition of organic matter), and other factors, we comprehensively and systematically investigated the generation and dissipation of the hypoxic zone and underlying mechanisms for the seasonal variation in its position, explored the multi-factorial synergistic reactions during the generation and dissipation process of the hypoxic zone, and revealed the controlled mechanism for the morphology and structure of the hypoxic zone's distribution. Our studies indicate that in the winter and spring seasons, the hydrological environment off the Yangtze River estuary provides a water body with relatively low contents of dissolved oxygen(DO), which is the background for the formation of a hypoxic zone. After entering into the summer season, the hypoxic zone gradually develops towards the north and becomes mature. Because of the impact of the terrain, local decomposition of organic matter, and upwelling of the Kuroshio subsurface water in July–August, the hypoxic zone off the Yangtze River estuary exhibits the characteristics of discontinuous distribution in space and has a south and north 'dual-core' structure in the inner continental shelf. In addition, there is a hypoxic core in the eastern outer continental shelf. The degrees of hypoxia vary for different areas; they are strongest overall in the north, next strongest in the south; they are weakest on the outer continental shelf. In summer, the hypoxic zone in the north is related to the northward differentiation of the southern hypoxic zone and results from local development and intensification. In August, the hypoxic zone in the north reaches its peak, and after September, it rapidly retreats southward and disappears because of weakening stratification. In the fall, there is hypoxic zone along the coast of Zhejiang in the south, and there is also a low-DO area to the southwest of Jeju Island, with both zones disappearing rapidly. In addition, the change of dynamic environment also causes the low-DO area of the outer continental shelf to move outward in the fall. The variation in the intensity of the stratification and its cumulative effects as a barrier of vertical DO transportation over long periods of time have a significant impact on the degree of hypoxia in the hypoxic zone. In addition, the seasonal variations in the size of the stratified region, intensity of each current system/water mass, upwelling, front, and high-value area of phytoplankton biomass jointly restrict the extension of the hypoxic zone in the inner continental shelf and latitudinal(south-north direction) movement of its location off the Yangtze River estuary. The combined effect of dynamic factors, such as that of the Kuroshio subsurface water, causes a low-DO core in the outer continental shelf. The bottom cold water to the north of the East China Sea is the dynamic basis for the formation of the low-DO area to the southwest of Jeju Island during the fall season. The special seabed topography and mud area distribution off the Yangtze River estuary have a certain degree of influence on the development of the hypoxic zone. 5Oceanic environmental changes of subarctic Bering Sea in recent 100 years: Evidence from molecular fossils显示文摘The core sample B2-9 from the seafloor of the subarctic Bering Sea was dated with 210 Pb to obtain a consecutive sequence of oceanic sedimentary environments at an interval of a decade during 1890—1999. A variety of molecular fossils were detected, including n-alkanes, isoprenoids, fatty acids, sterols, etc. By the characteristics of these fine molecules (C27, C28, and C29 sterols) and their molecular indices (Pr/Ph, ∑C+22/∑C?21, CPI and C18∶2/C18∶0) and in con- sideration of the variation of organic carbon content, the 100-year evolution history of subarctic sea paleoenvironment was reestablished. It is indicated that during the past 100 years in the Arctic, there were two events of strong climate warming (1920—1950 and 1980—1999), which resulted in an oxidated sediment environment owing to decreasing terrigenous organic matters and increasing marine-derived organic matters, and two events of transitory climate cooling (1910 and 1970—1980), which resulted in a slightly reduced sediment environment owing to increasing terrigenous organic matters and decreasing marine-derived organic matters. 6Quaternary biogenic opal records in the South China Sea: Linkages to East Asian monsoon, global ice volume and orbital forcing显示文摘Particulate fluxes investigated in the central South China Sea (SCS) during 1993―1996 indicate that opal flux can be used to show primary productivity change, which provides a foundation for tracing the evolutionary relationship between the surface productivity and East Asian monsoon in the SCS during the late Quaternary glacial and interglacial periods. Based on the studies of opal % and their mass accumulation rates (MAR) at the six sites recovered from the SCS during the “Resolution” ODP Leg 184 and “Sonne” 95 cruise of the Sino-Germany cooperation, opal % and their MARs increased evidently in the northern sites since 470―900 ka, and they enhanced and reduced, respectively, during the glacial and interglacial periods. Whereas they increased obviously in the southern sites since 420―450 ka, and they augmented and declined, respectively, during the interglacial and glacial periods. The vari- ability in opal % and their MARs in the late Quaternary glacial cyclicity indicate the “seesaw” pattern of surface productivity in the SCS. The winter monsoon intensified during the glacial periods, surface productivity increased and decreased, respectively, in the northern and southern SCS. The summer monsoon strengthened during the interglacial periods, surface productivity increased and decreased, respectively, in the southern and northern SCS. The cross spectral analyses between the opal % in the northern and southern SCS during the Quaternary and global ice volume (δ 18O) and orbital forcing (ETP) indicate that the East Asian winter and summer monsoons could be ascribed to the different drive mechanisms. On the orbital time scale, the global ice volume change could be a dominant factor for the winter monsoon intension and temporal variations. 7Variations in paleoproductivity and the environmental implications over the past six decades in the Changjiang Estuary显示文摘Total organic carbon and organic carbon stable isotope, biogenic silica, chlorin were measured from a high resolution sediment core to indicate the variation in paleoproductivity and the environmental implications over the past decades (1942 to 1997) in the Changjiang Estuary. Based on these biomarkers, the shift in the phytoplankton community structure in the past decades was discussed in response to the long-term variations in nutrient concentrations and nutrient ratios. The results showed that the δ 13 C values varied from –26.15‰ to –19.5‰, suggesting the combined organic carbon sources of riverine and marine organisms. Based on the biogenic proxies, paleoproductivity changes were categorized into three stages: low production before 1950; an increase in production with the dominance of diatoms during 1950 to 1980, consistent with increasing of nutrient concentrations; a decrease in production after 1980 when the diatom production decreased while the production of non-silicious phytoplankton increased due to high nutrient inputs, and high N/P and P/Si ratios in the Changjiang Estuary. 8Phytoplankton composition and its ecological effect in subsurface cold pool of the northern Bering Sea in summer as revealed by HPLC derived pigment signatures显示文摘CHEMTAX analysis of high-performance liquid chromatography(HPLC) pigment was conducted to study phytoplankton community structure in the northern Bering Sea shelf, where a seasonal subsurface cold pool emerges. The results showed that fucoxanthin(Fuco) and chlorophyll a(Chl a) were the most abundant diagnostic pigments, with the integrated water column values ranging from 141 to 2 160 μg/m2 and 477 to 5 535 μg/m2, respectively. Moreover, a diatom bloom was identified at Sta. BB06 with the standing stock of Fuco up to 9 214 μg/m3. The results of CHEMTAX suggested that the phytoplankton community in the northern Bering Sea shelf was dominated by diatoms and chrysophytes with an average relative contribution to Chl a of 80% and 12%, respectively, followed by chlorophytes, dinoflagellates, and cryptophytes. Diatoms were the absolutely dominant algae in the subsurface cold pool with a relative contribution exceeding 90%, while the contribution of chrysophytes was generally higher in oligotrophic upper water. 9Variations in organic carbon loading of surface sediments from the shelf to the slope of the Chukchi Sea,Arctic Ocean显示文摘The content of organic carbon(OC) normalized to the specific surface area(SSA) of sediment is widely used to trace variations in OC loading(OC/SSA).This study presents observations of OC/SSA of surface sediments collected in the Chukchi Sea,a typical Arctic marginal sea.Shelf sediments exhibit much higher OC/SSA values than slope sediments in the study area.Compared with OC/SSA values reported from the East Siberian Shelf and Mackenzie River,the slope sediments possess lower OC loading.This abrupt decrease in OC/SSA is mostly related to the lower primary production on slope as well as possible oxidization processes.The results of linear regression analysis between OC and SSA indicate a sedimentary source rock for the OC in the Chukchi Sea sediments.Moreover,shelf sediments with low SSA possess a larger rock OC fraction than slope sediments do.The dataset of the present study enables a more thorough understanding of regional OC cycling in the Chukchi Sea.LI Zhongqiao WANG Xinyi JIN Haiyan JI Zhongqiang BAI Youcheng CHEN Jianfang 2017Acta Oceanologica Sinica2017,36,8:6
10Inorganic Carbon Parameters Responding to Summer Hypoxia Outside the Changjiang Estuary and the Related Implications显示文摘The eutrophication,hypoxia and coastal acidification are attracting more and more attention.In this study,inorganic carbon parameters,including dissolved inorganic carbon (DIC) ,total alkalinity (TA) and calculated partial pressure of CO2 (pCO2) ,obtained from a summer cruise in August,2009,were used to investigate their integrated response to biological processes accompanying the oxygen depletion in the areas off the Changjiang Estuary.According to the observations,the typical hypoxia occurred in the bottom water just outside the Changjiang Estuary with Dissolved Oxygen (DO) lower than 2.00 mg L 1.The biological uptake in the surface water and the decomposition of organic matter in the bottom water were fully coupled with each other.The high concentration of Chl_a (Chl_a=10.9μg L 1) and DO (9.25 mg L 1) ,profoundly decreased DIC concentration (1828μmol kg 1) and elevated pH (8.42) was observed in the surface water.The correspondingly increased DIC and depletion of oxygen were observed in the bottom water.The semi-quantitative analysis proved that the locally-produced phytoplankton,determined by primary productivity,was deposited to the bottom and contributed about 76%of total amount of the organic carbon decomposition in the bottom.However,in the bottom hypoxia (DO=2.05 mg L 1) area observed in the Southern Zhejiang coastal water,the responding patterns of inorganic carbon parameters deviated from the previous one.The expanding of Changjiang Diluted Water (CDW) ,the adding of Hangzhou Bay water (with high DIC concentration) and Coastal Current together modify the DIC background value in this area,and the local degeneration and upwelling process may also help to offset the local DIC removed by net biological uptake in surface water.In addition,when the mixing occurring in autumn,which may break the summer stratification,the excess release of high DIC in the bottom water to the subsurface water could have an important influence on coastal acidification and the CO2uptake capacity in this area.WANG Bin CHEN Jianfang JIN Haiyan LI Hongliang XU Jie 2013Journal of Ocean University of China2013,12,4:6
11Activation of Toll-like receptors signaling in non-small cell lung cancer cell line induced by tumor-associated macrophages显示文摘Background:Inflammation is often linked with the progress and poor outcome of lung cancer.The understanding of the relationship between tumor-associated macrophages(TAMs) and lung cancer cells involves in the underlying mechanism of inflammatory cytokine production.Toll-like receptors(TLRs) are engaged in promoting the production of pro-inflammatory cytokines and play an important role in tumor immunology.Methods:To investigate the mechanisms by which TAMs influence the production of pro-inflammatory cytokines in lung cancer cells,we established an in vitro coculture system using TAMs and human nonsmall cell lung cancer(NSCLC) cell line SPC-A1.Levels of interleukin(IL)-1β,IL-6 and IL-8 in SPC-A1 were evaluated by RT-PCR and cytometric bead array assay after being cocultured with TAMs.Expression changes of TLRs and TLRs signaling pathway proteins in SPC-A1 were further confirmed by RT-PCR and western blot.The level changes of IL-1β,IL-6 and IL-8 in SPC-A1 were also detected after the stimulation of TLRs agonists.Results:We found that the phenotype markers of TAMs were highly expressed after stimulating human monocyte cell line THP-1 by phorbol-12-myristate-13-acetate(PMA).Higher mRNA and supernate secretion levels of IL-1β,IL-6 and IL-8 were detected in SPC-A1 after being cocultured with TAMs.We also found that TLR1,TLR6 and TLR7 were up-regulated in SPC-A1 in the coculture system with TAMs.Meanwhile,TLRs signaling pathway proteins were also significantly activated.Moreover,pre-treatment with agonist ligands for TLR1,TLR6 and TLR7 could dramatically promote inductions of IL-1β,IL-6 and IL-8.Conclusions:These findings demonstrated that TAMs may enhance IL-1β,IL-6 and IL-8 expressions via TLRs signaling pathway.We conclude that TAMs contribute to maintain the inflammation microenvironment and ultimately promote the development and progression of lung cancer.Xing Ke Meng Wu Jianfang Lou Shuping Zhang Peijun Huang Ruihong Sun Lei Huang Erfu Xie Fang Wang Bing Gu 2015Chinese Journal of Cancer Research2015,27,2:6
12Mechanisms leading to the frequent occurrences of hypoxia and a preliminary analysis of the associated acidification off the Changiiang estuary in summer显示文摘Based on literature data and shipboard observations,this study investigated the main environmental characteristics of the seafloor topography,current field,front,and upwelling that are closely related to hypoxia occurrence off the Changjiang estuary.The physical processes of the plume front and upwelling off the Changjiang estuary in summer were coupled.The vertical distribution pattern of the plume front was closely related to the upwelling.By reviewing and analyzing the historical summer hypoxia events off the Changjiang estuary,we statistically demonstrated the spatial structure of the horizontal distribution of the hypoxic zone and investigated the location of occurrence zone of the hypoxia.We found that the dissolved oxygen(DO)concentration on the inner continental shelf off the estuary showed a'V'shape in relation to station depth.Therefore,we noted that the hypoxic water on the inner continental shelf mostly occurred on the slopes with steep seafloor topography.Base on the current understanding of the hypoxic mechanisms off the Changjiang estuary,we analyzed the biogeochemical mechanisms that could cause the steep terrain off the Changjiang estuary to become the main areas prone to summer hypoxia and explained the internal relations between the location of the hypoxic zone on the slopes and the plume front and upwelling.The plume front and upwelling off the Changjiang estuary and their coupling were important driving forces of summer hypoxia.The continuous supply of nutrients affected by the interaction of the plume front extension of the Changjiang Diluted Water(CDW)and upwelling and the favorable light conditions were important mechanisms causing the phytoplankton blooms and benthic hypoxia off the Changjiang estuary in summer.By analyzing oxygen utilization,organic carbon mineralization,and nutrient regeneration in the hypoxic zone,we observed that the significant oxygen utilization process off the Changjiang estuary in summer also mainly occurred near the steep slopes with front and upwelling features and confirmed the apparent nutrient loss in the benthic hypoxic zone.Meanwhile,our analysis revealed that the sediment resuspension in the benthic boundary layer in the mud areas off the Changjiang estuary could also affect the oxygen utilization and mineralization of organic carbon and nutrient recycling and regeneration.This study also demonstrated that the steep terrain off the Changjiang estuary was the main location for summer acidification,and the coupling between the plume front and upwelling on the steep slopes was an important physical driving force inducing summer benthic acidification.Finally,we discussed issues to address in future studies of the hypoxic zone and water acidification off the Changjiang estuary.WEI QinSheng WANG BaoDong YU ZhiGang CHEN JianFang XUE Liang 2017Science China Earth Sciences2017,60,2:5
13Dissecting human embryonic skeletal stem cell ontogeny by single-cell transcriptomic and functional analyses显示文摘Human skeletal stem cells(SSCs)have been discovered in fetal and adult long bones.However,the spatiotemporal ontogeny of human embryonic SSCs during early skeletogenesis remains elusive.Here we map the transcriptional landscape of human limb buds and embryonic long bones at single-cell resolution to address this fundamental question.We found remarkable heterogeneity within human limb bud mesenchyme and epithelium,and aligned them along the proximal-distal and anterior-posterior axes using known marker genes.Osteo-chondrogenic progenitors first appeared in the core limb bud mesenchyme,which give rise to multiple populations of stem/progenitor cells in embryonic long bones undergoing endochondral ossification.Importantly,a perichondrial embryonic skeletal stem/progenitor cell(eSSPC)subset was identified,which could self-renew and generate the osteochondral lineage cells,but not adipocytes or hematopoietic stroma.eSSPCs are marked by the adhesion molecule CADM1 and highly enriched with FOXP1/2 transcriptional network.Interestingly,neural crest-derived cells with similar phenotypic markers and transcriptional networks were also found in the sagittal suture of human embryonic calvaria.Taken together,this study revealed the cellular heterogeneity and lineage hierarchy during human embryonic skeletogenesis,and identified distinct skeletal stem/progenitor cells that orchestrate endochondral and intramembranous ossification.Jian He Jing Yan Jianfang Wang Liangyu Zhao Qian Xin Yang Zeng Yuxi Sun Han Zhang Zhijie Bai Zongcheng Li Yanli Ni Yandong Gong Yunqiao Li Han He Zhilei Bian Yu Lan Chunyu Ma Lihong Bian Heng Zhu Bing Liu Rui Yue 2021Cell Research2021,31,7:5

15Response of nutrients and the surface phytoplankton community to ice melting in the central Arctic Ocean显示文摘During the fourth Chinese National Arctic Research Expedition cruise in summer 2010, a time-series observation was carried out to examine the response of nutrients and phytoplankton community in the ice-water interface to the ice melting ice in the central Arctic Ocean. Phosphate and silicate in the ice-water interface were rich relative to dissolved inorganic nitrogen (DIN), based on the Redfield ratio (16N:1P:16Si), suggesting that DIN was the potential limiting nutrient. DIN concentrations in the sea ice were about 3-4 times that in the surface seawater, indicating that melting ice delivered DIN to the surface water. Pigment analysis showed that fucoxanthin and chlorophyll a contribute to carotenoids and chlorophylls in particles. The mean concentrations of chlorophyll c,diatoxanthin, diadinoxanthin and fucoxanthin from 15 August to 18 August were 6 μgm-3, 22 μgm-3, 73 μgm-3 and 922 μg·m-3, respectively, suggesting that diatoms dominated in the phytoplankton community composition.Furthermore, a notable enhancement in fucoxanthin and chlorophyll a during a large-scale ice melting was likely attributed to senescent diatoms released from the bottom sea-ice as well as phytoplankton diatoms growth in the water column due to the input of nutrients (i.e., DIN) and reducing light limitation from melting ice. The analysis result indicates that C37:3 methylketone is predominate in the long-chain alkenones from the Chukchi and Bering Sea sediments. The abundance of C37 to C39 unsaturated alkenones changes in an order of C37 > C38 > C39. Based on C37 /C38 ratio, the detected organism precursors of the long-chain alkenones are mainly coccolithophrid (Emiliania huxleyi). By the calibration relationship between U37k and U37k indices, the sea surface paleotemperature in these seas is estimated. 17Climatic significance of n-alkanes and their compound-specific dD values from lake surface sediments on the Southwestern Tibetan Plateau显示文摘The content and hydrogen isotope ratios of n-alkanes extracted from recent lake surface sediments sampled from nine lakes in three different climate zones on the Southwestern Tibetan Plateau were analyzed.Values were compared with n-alkanes in plants from lake drainages,and dD values of meteoric water,lake water,and mean annual precipitation.The results showed that n-C23was predominantly derived from aquatic plants,and n-C27–n-C33from terrestrial higher plants.The average carbon chain length of n-C27–n-C33(ACL27–33)was positively correlated with the mean annual precipitation.dD values of the long-chain n-alkanes n-C29and n-C31of terrestrial origin(varying between-214%and-169%,and-226%and-185%,respectively)were inversely correlated with mean annual precipitation;but in accordance with the average annual variations in dD(OIPC),dD values of n-C31were strongly related to the dD values of growing season meteoric water(R2=0.74).The large difference between dD values of n-C23of aquatic origin and n-C31(an average of about 27%)demonstrates the enrichment of the lake water dD compared to precipitation,caused by strong evaporation in the semiarid–arid areas of the southwestern Tibetan Plateau.Average value of en C25 31=p(-95%)is evidently higher than the value observed in European wet regions(-128%);besides,en C31(about-116%)is constant along the study transect(SD=9),which indicates that n-C31is a useful proxy for the environment.Xing Hu Liping Zhu Yong Wang Junbo Wang Ping Peng Qingfeng Ma Jianfang Hu Xiao Lin 2014Chinese Science Bulletin2014,59,24:3

19Mechanism on minimization of excess sludge in oxic-settling- anaerobic (OSA) process显示文摘The oxic-settling-anaerobic(OSA)process is a promising wastewater treatment technique for efficiently reducing sludge production and improving the stability of process operation.In this paper,the possible factors of sludge reduction such as sludge decay,uncoupled metabol-ism,and anaerobic oxidation with low sludge production were discussed in the OSA process.It has been confirmed that sludge decay is the decisive cause in the OSA process,accounting for 66.7%of sludge production reduction.Sludge decay includes hydrolysis and acidogenesis of dead microorganisms and particle organic carbon adsorbed in sludge floc and endogenous metabolism.By batch experi-ments,it has been proven that there is energetic uncoupling in the OSA system since microorganisms were exposed to alternative anaerobic and aerobic environment.It accounts for about 7.5%of sludge production reduction.Soluble chemical oxygen demand(SCOD)released from the anaerobic sludge tank in the OSA process was used as the substrate for cryptic growth.The substrate was used for anoxic denitrifying,anaerobic phosphorus release,sulfate reduction,and methane production.These anaerobic reactions in the sludge anaerobic tank have lower sludge production than in the aerobic oxidation when equivalent SCOD is consumed,which may lead to approximately 23%of sludge reduction in the OSA process.It has been concluded that multiple causes resulted in the minimization of excess sludge in the OSA system.The microbial community structure and diversity of sludge samples from the CAS(conventional activated sludge)and OSA systems were investigated by 16 SrDNA PCR-DG-DGGE(poly-merase chain reaction-double gradient-denaturing gradient gel electrophoresis).DGGE profile and cluster analysis showed more abundant species in the OSA system contrasting to microbial communities in the CAS system.Jianfang WANG Qingliang ZHAO Wenbiao JIN Jikan LIN 2008Frontiers of Environmental Science & Engineering2008,2,1:3
