1Reversal effect of bufalin on multidrug resistance in K562/VCR vincristine-resistant leukemia cell line显示文摘OBJECTIVE: To probe insights into the reversal effect of bufalin on vincristine-acquired multidrug resistance(MDR) in human leukemia cell line K562/VCR.METHODS: Proliferative inhibition rate and the reversal index(RI) of bufalin were determined by Methyl thiazolyl tetrazolium assay. The uptake of Adriamycin(ADM) in K562/VCR cells, cell cycle and apoptosis rate were determined by flow cytometry(FCM). Cell morphologic changes were observed with Wright-Giemsa staining. The expression of P-glycoprotein(P-gp), multidrug-associated protein-1(MRP1), Bcl-x L and Bax protein were measured by immunocytochemistry.RESULTS: The human leukemia multidrug resistant K562/VCR cells showed no cross-resistance to bufalin. The RIs of bufalin at concentrations of 0.0002,0.001 and 0.005 μmol/L were 4.85, 6.94 and 14.77,respectively. Preincubation of 0.001 μmol/L bufalin for 2 h could increase intracellular ADM fluorescence intensity to 28.07%(P<0.05) and down-regulate MRP1 expression simultaneously, but no remarkable effect was found on P-gp protein. Cell cycle analysis indicated increased apoptosis rate and apparent decreased G2/M phase proportion after treatment with bufalin. When exposed to 0.01μmol/L bufalin, typical morphological changes of apoptosis could be observed. 2Simulation of micro-behaviors including nucleation,growth,and aggregation in particle system显示文摘A new method for the solution of population balance equations(PBE) describing the micro-processes such as nucleation,growth,aggregation of particle swarms in a multiphase system is proposed.The method is based on the fixed pivot moment and allows arbitrary number of moments to be tracked si-multaneously.By expressing PBEs for both batch and continuous operations in a general form,and using weighted residual method to derive the moment equations,different moments can be tracked directly.The numerical density function is assumed to be a summation of several weighted Dirac Delta functions,and the integral and derivative terms in PBEs are transformed to a summation in order to reduce computational cost.Simulations of a batch nucleation-growth process and a continuous ag-gregation-growth process have demonstrated good agreement with the corresponding analytical solu-tions,with relative errors less than 108%.Simulation of a combined nucleation-growth-aggregation process,which does not have an analytical solution,is also included,so as to reproduce the mi-cro-behaviors of such a complex system,demonstrating the feasibility and reliability of this method.GU ZhaoLin SU JunWei JIAO JianYing XU X Yun 2009Science China Chemistry2009,52,2:4
3Connections between Evaporite Deposition and Glacial Periods since the Middle Pleistocene in Salt Lakes in the Western Qaidam Basin, China显示文摘The Quaternary was the main evaporite deposition period in the Qaidam Basin(QB), but the correlation between the evaporite deposition period and the glacial period is still unclear. In this study, the research objects are primarily evaporite-bearing strata in a 461.58 m-long drill core in the QB. X-ray powder diffraction(XRD) and scanning electron microscopy(SEM) were applied to study the evaporite minerals, U-Th dating being applied to construct the ~(230)Th geochronological framework. Evaporite deposition from Marine Isotope Stage(MIS) 15 to MIS 4 in the borehole was reconstructed via mineralogical and geochronological data. The evaporite minerals are mainly halite(NaCl), mirabilite(Na_(2)SO_(4)·10H_(2)O), thenardite(Na_(2)SO_(4)) and gypsum(CaSO_(4)·2H_(2)O). A total of 9 effective ~(230)Th data points, ranging from 492.5 ± 43.0 ka to 62.0 ± 11.9 ka, were obtained. The depositional age of the earliest halite layer in the borehole is 592.5–563.0 ka. There were mirabilite deposits in the QB during the cold glacial environment of MIS 6. During MIS 4, the study area desiccated, with mirabilite and halite being deposited. 4Construction of nanofibrous scaffolds with interconnected perfusable microchannel networks for engineering of vascularized bone tissue显示文摘Vascularization and bone regeneration are two closely related processes during bone reconstruction.A three-dimensional(3D)scaffold with porous architecture provides a suitable microenvironment for vascular growth and bone formation.Here,we present a simple and general strategy to construct a nanofibrous poly(L-lactide)/poly(ε-caprolactone)(PLLA/PCL)scaffold with interconnected perfusable microchannel networks(IPMs)based on 3D printing technology by combining the phase separation and sacrificial template methods.The regular and customizable microchannel patterns within the scaffolds(spacings:0.4 mm,0.5 mm,and 0.6 mm;diameters:0.8 mm,1 mm,and 1.2 mm)were made to investigate the effect of microchannel structure on angiogenesis and osteogenesis.The results of subcutaneous embedding experiment showed that 0.5/0.8-IPMs(spacing/diameter=0.5/0.8)and 0.5/1-IPMs(spacing/diameter=0.5/1)scaffolds exhibited more vascular network formation as compared with other counterparts.After loading with vascular endothelial growth factor(VEGF),VEGF@IPMs-0.5/0.8 scaffold prompted better human umbilical vein endothelial cells(HUVECs)migration and neo-blood vessel formation,as determined by Transwell migration,scratch wound healing,and chorioallantoic membrane(CAM)assays.Furthermore,the microangiography and rat cranial bone defects experiments demonstrated that VEGF@IPMs-0.5/0.8 scaffold exhibited better performance in vascular network formation and new bone formation compared to VEGF@IPMs-0.5/1 scaffold.In summary,our results suggested that the microchannel structure within the scaffolds could be tailored by an adjustable caramel-based template strategy,and the combination of interconnected perfusion microchannel networks and angiogenic factors could significantly enhance vascularization and bone regeneration.Jiani Gu Qianqian Zhang Mengru Geng Weizhong Wang Jin Yang Atta ur Rehman Khan Haibo Du Zhou Sha Xiaojun Zhou Chuanglong He 2021Bioactive Materials2021,6,10:1
5Heavy collecting induces smaller and deeper Fritillariae Cirrhosae Bulbus in the wild显示文摘Fritillariae Cirrhosae Bulbus is a well-known traditional Chine medicinal herb. However, the demand for this herb is leading to over-collection and its decline in the wild. This study aims to investigate the ecological conditions of Fritillaria cirrhosa under the influence of human disturbance, biotic species interactions and climatic conditions. We established a total of 78 plots at 14 sites of F. cirrhosa in its distribution center, the Hengduan Mountains area. At each site, we estimated the abundance of F. cirrhosa at different distances from roads. The diameter and height of F. cirrhosa fruit and bulbs were measured and compared to underground bulb depth. We then analyzed the effects of environmental conditions and human disturbance on the abundance of F. cirrhosa using variance partitioning. We found that(1)abundance of F. cirrhosa and their underground bulb depth showed a significant linear increase with the distance from the main road;(2) the diameter/height of fruits and the diameter/height of fruits/bulbs showed significantly different responses to the human disturbance;(3) the community associates,climate and spatial conditions can explain 58%, 22% and 27%, respectively, of the variance in the F. cirrhosa abundance. 7Multiplexed imaging of tumor immune microenvironmental markers in locally advanced or metastatic non-small-cell lung cancer characterizes the features of response to PD-1 blockade plus chemotherapy显示文摘Background:Although programmed cell death 1(PD-1)blockade plus chemotherapy can significantly prolong the progression-free survival(PFS)and overall survival(OS)in first-line settings in patients with driver-negative advanced non-small-cell lung cancer(NSCLC),the predictive biomarkers remain undetermined.Here,we investigated the predictive value of tumor immune microenvironmental marker expression to characterize the response features to PD-1 blockade plus chemotherapy.Methods:Tumor tissue samples at baseline were prospectively collected from 144 locally advanced or metastatic NSCLC patients without driver gene alterations who received camrelizumab plus chemotherapy or chemotherapy alone.Tumor immune microenvironmental markers,including PD-1 ligand(PDL1),CD8,CD68,CD4 and forkhead box P3,were assessed using multiplex immunofluorescence(mIF)assays.Kaplan-Meier curveswere used to determine treatment outcome differences according to their expression status.Mutational profiles were compared between tumors with distinct expression levels of these markers and their combinations.Results:Responders had significantly higher CD8/PD-L1(P=0.015)or CD68/PD-L1 co-expression levels(P=0.021)than non-responders in the camrelizumab plus chemotherapy group,while no difference was observed in the chemotherapy group.Patients with high CD8/PD-L1 or CD68/PD-L1 co-expression level was associated with significantly longer PFS(P=0.002,P=0.024;respectively)and OS(P=0.006,P=0.026;respectively)than those with low co-expression in camrelizumab plus chemotherapy group.When comparing survival in the camrelizumab plus chemotherapy with chemotherapy by CD8/PD-L1 co-expression stratification,significantly better PFS(P=0.003)and OS(P=0.032)were observed in high co-expression subgroups.The predictive value of CD8/PD-L1 and CD68/PD-L1 co-expression remained statistically significant for PFS and OS when adjusting clinicopathological features.Although the prevalence of TP53 or KRAS mutations was similar between patients with and without CD8/PD-L1 or CD68/PD-L1 co-expression,the positive groups had a significantly higher proportion of TP53/KRAS co-mutations than the negative groups(both 13.0%vs.0.0%,P=0.023).Notably,enriched PI3K(P=0.012)and cell cycle pathway(P=0.021)were found in the CD8/PD-L1 co-expression group.Conclusion:Tumor immune microenvironmental marker expression,especially CD8/PD-L1 or CD68/PD-L1 co-expression,was associated with the efficacy of PD-1 blockade plus chemotherapy as first-line treatment in patients with advanced NSCLC.Fengying Wu Tao Jiang Gongyan Chen Yunchao Huang Jianying Zhou Lizhu Lin Jifeng Feng Zhehai Wang Yongqian Shu Jianhua Shi Yi Hu Qiming Wang Ying Cheng Jianhua Chen Xiaoyan Lin Yongsheng Wang Jianan Huang Jiuwei Cui Lejie Cao Yunpeng Liu Yiping Zhang Yueyin Pan Jun Zhao LiPing Wang Jianhua Chang Qun Chen Xiubao Ren Wei Zhang Yun Fan Zhiyong He Jian Fang Kangsheng Gu Xiaorong Dong Tao Zhang Wei Shi Jianjun Zou Xuejuan Bai Shengxiang Ren Caicun Zhou 2022Cancer Communications2022,42,12:1

10Block copolymer functionalized quartz fibers/cyanate ester wave-transparent laminated composites显示文摘A block copolymer of PDMS-b-PGMA is synthesized by polymerizing glycidyl methacrylate(GMA)via reversible addition-fragmentation chain transfer(RAFT)polymerization applying a polydimethylsiloxane(PDMS)based macro-RAFT agent,which is then performed to functionalize the quartz fibers(QFs@PDMS-b-PGMA)via a simple coating process.Finally,the QFs@PDMS-b-PGMA/bisphenol A dicyanate ester(BADCy)wave-transparent laminated composites are fabricated by high-temperature molding.Nuclear magnetic resonance(NMR)spectroscopy,Fourier transform infrared(FT-IR)spectroscopy and size ex-clusion chromatography(SEC)demonstrate the successful preparation of PDMS-b-PGMA with expected structure.When the molar mass and coating amount of PDMS-b-PGMA are respectively 8100 g/mol and 2.0 wt.%,QFs@PDMS-b-PGMA/BADCy wave-transparent laminated composites present optimal mechan-ical properties and wave-transparent performance.The interlaminar shear strength(ILSS)and flexural strength are 53.6 and 552.0 MPa,respectively.Meanwhile,the dielectric constant and dielectric loss val-ues are 2.61 and 0.0028 at 1 MHz(wave transmittance of 93.8%),showing good stability at different frequencies(102-106 Hz and 8.4-12.4 GHz)and temperatures(25-250℃).Jiani Zhang Zheng Liu Muyi Han Junliang Zhang Yusheng Tang Junwei Gu 2023Journal of Materials Science & Technology2023,,8:0
11Assessment of complete lymph node dissection in patients with melanoma:A systemic review and meta-analysis显示文摘Background: Complete lymph node dissection(CLND) for patients with melanoma remains controversial. This meta-analysis aimed to compare the prognoses and complications between the CLND and control groups(patients who receive adjuvant treatment or observation only) in patients with sentinel lymph node(SLN)-positive melanoma.Methods: The Pub Med, Embase, Cochrane, and Web of Science databases were searched for cohort studies and randomized clinical trials(RCTs) conducted between 1964 and 2022, and the quality of the studies was assessed using the Cochrane risk-of-bias tool and Newcastle-Ottawa Scale. Hazard ratios(HR) or risk ratios(RR) with 95%confidence intervals(CIs) were calculated for each outcome. Heterogeneity and sensitivity tests were also conducted, and publication bias tests were performed when the pooled number of studies was >10.Results: Fifteen studies, including 11 cohort studies and 4 RCTs, were enrolled and assessed for quality. Analysis of overall survival showed no significant difference between the CLND and control groups(HR=1.02, 95% CI:0.69–1.51, P=0.922). Similarly, recurrence-free survival(HR=0.84, 95% CI: 0.6–1.16, P=0.287), disease-free survival(HR=1.06, 95% CI: 0.65–1.72, P=0.82), and disease-specific survival(HR=0.84, 95% CI: 0.59–1.21,P=0.355) showed no difference between the two groups. CLND did not reduce the risk of recurrence(RR=0.98,95% CI: 0.8–1.2, P=0.851).Conclusion: Remarkably, patients who underwent CLND were more likely to have complications such as flap necrosis and lymphedema than the controls. 14Re-Exploring Biomarkers and Therapeutic Targets in Primary Melanoma Patients: Insights from Network-Based Analysis of Microarray Data显示文摘Objective To identify novel biomarkers and therapeutic targets for primary melanoma using network-based microarray data analysis.Methods Eligible microarray datasets from the Gene Expression Omnibus(GEO)database were used to identify differentially expressed genes(DEGs).The protein-protein interaction(PPI)network,Gene Ontology(GO),and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses were performed to identify hub genes and pathways that might affect the survival of melanoma patients.Immunohistochemistry results obtained from the Human Protein Atlas(HPA)database confirmed the protein expression levels of hub genes.The Cancer Genome Atlas(TCGA)database was used to further verify the gene expression levels and conduct survival analysis.Results Three microarray datasets(GSE3189,GSE15605,and GSE46517)containing 122 melanoma and 30 normal skin tissue samples were included.A total of 262 common differentially expressed genes(cDEGs)were identified based on three statistical approaches(Fisher’s method,the random effects model(REM),and vote counting)with strict criteria.Of these,two upregulated genes,centromere protein F(CENPF)and pituitary tumortransforming gene 1(PTTG1),were selected as hub genes.HPA and TCGA database analyses confirmed that CENPF and PTTG1 were overexpressed in melanoma.Survival analysis showed that high expression levels of CENPF were significantly correlated with decreased overall survival(OS)(P=0.028).Conclusion The expression level of CENPF was significantly upregulated in melanoma and correlated with decreased OS.Thus,CENPF may represent a novel biomarker and therapeutic target for melanoma patients.Kangjie SHEN Chuanyuan WEI Yi XIE Lu WANG Shuyu WANG Ming REN Xinyi DENG Daohe WANG Zixu GAO Zihao FENG Jianying GU 2020Chinese Journal Of Plastic and Reconstructive Surgery2020,2,4:0

16No-Reference Quality Assessment of Enhanced Images显示文摘Image enhancement is a popular technique,which is widely used to improve the visual quality of images.While image enhancement has been extensively investigated,the relevant quality assessment of enhanced images remains an open problem,which may hinder further development of enhancement techniques.In this paper,a no-reference quality metric for digitally enhanced images is proposed.Three kinds of features are extracted for characterizing the quality of enhanced images,including non-structural information,sharpness and naturalness.Specifically,a total of 42 perceptual features are extracted and used to train a support vector regression(SVR) model.Finally,the trained SVR model is used for predicting the quality of enhanced images.The performance of the proposed method is evaluated on several enhancement-related databases,including a new enhanced image database built by the authors.The experimental results demonstrate the efficiency and advantage of the proposed metric.Leida Li Wei Shen Ke Gu Jinjian Wu Beijing Chen Jianying Zhang 2016China Communications2016,13,9:0
